Abstract

Background: Thermophilic bacteria are potential sources of thermostable enzymes with important applications in biotechnology and industry. Among these enzymes, metalloproteases are of particular interest because their catalytic activity depends on metal ions. This study aimed to screen thermophilic bacterial isolates from hot springs in Solok Regency, West Sumatra, Indonesia, for metalloprotease production and to evaluate the effects of trace elements on metalloprotease activity. Methodology: Four thermophilic bacterial isolates (TUA-104,                   TUA-106, TUA-109, and TUA-113) were cultivated in protease production medium (pH 8.0) at 50oC for 24 hours, and crude enzymes were obtained by centrifugation. Metalloprotease screening was conducted using EDTA inhibition assays through well diffusion and specific protease activity analyses. The isolate showing the strongest metalloprotease characteristics was further evaluated using 5 ppm Ca²⁺, Mn²⁺, and Zn²⁺ supplementation. Findings: The results showed that isolates TUA-106, TUA-109, and TUA-113 exhibited characteristics of metalloprotease-producing bacteria. Among them, TUA-106 showed the most consistent inhibition pattern, with specific activity decreasing from 0.780 U/mg in the control to 0.522 U/mg after treatment with 10 mM EDTA. Zn²⁺ produced the highest specific activity (0.284 U/mg), followed by Mn²⁺ (0.263 U/mg), whereas Ca²⁺ resulted in the lowest activity (0.036 U/mg). Contribution: These findings indicate that TUA-106 is a potential Zn-dependent metalloprotease-producing thermophilic bacterium and provide new insight into the metalloprotease-producing potential of thermophilic bacteria from Solok geothermal environments for future biotechnological applications

Abstract

Seed-borne diseases caused by Colletotrichum and Fusarium can reduce the viability and vigor of chili seeds. An environmentally friendly alternative for disease management is the use of Trichoderma atroviride as a biopriming agent. This study aimed to evaluate the effectiveness of T. atroviride biopriming on the germination of Kopay chili seeds infected with Colletotrichum scovillei and Fusarium oxysporum. The experiment was arranged in a Completely Randomized Design (CRD) consisting of eight treatments with four replications. Observed parameters included germination percentage, root length, shoot length, fresh weight, dry weight, vigor index, and root emergence time. Data were analyzed using analysis of variance (ANOVA) followed by Duncan’s New Multiple Range Test (DNMRT) at the 5% significance level. The results showed that T. atroviride treatment produced the highest germination percentage (100%), which was not significantly different from the positive control (95%) and negative control (90%). The T. atroviride treatment also resulted in the greatest root length (2.54 cm), shoot length (1.74 cm), highest vigor index (100%), and the fastest root emergence (2 days). In contrast, the C. scovillei treatment exhibited the lowest germination percentage (30%) and vigor index (45%). These findings suggest that biopriming with T. atroviride has the potential to enhance seed vigor and support early seedling growth of chili under seed-borne pathogen stress.

Abstract

This study aimed to isolate, screen, and partially characterize protease-producing thermophilic bacteria from the Padang Dama hot spring, Solok, Indonesia. The main issue addressed in this study is the limited information regarding thermophilic bacteria capable of producing protease enzymes from this geothermal environment, despite their potential use in industrial processes requiring stable enzymes. Isolation was carried out using Nutrient Agar followed by incubation at 50 °C for 24 hours. Screening of protease activity was performed using Skim Milk Agar based on clear zone formation, and the proteolytic index (PI) was calculated to evaluate enzyme activity. Selected isolates were further characterized through macroscopic, microscopic, and biochemical analyses. The results showed that three isolates exhibited proteolytic activity, with isolate TUA-109 showing the highest proteolytic index, indicating superior enzymatic potential compared to other isolates. Partial characterization revealed that isolate TUA-109 is a Gram-negative, rod-shaped bacterium with positive catalase activity and motility, which are important traits supporting its adaptability and potential for enzyme production under extreme conditions. These findings highlight the potential of thermophilic bacteria from the Padang Dama hot spring as sources of protease enzymes, with isolate TUA-109 identified as a promising candidate for further investigation, particularly for industrial enzyme production. Future studies are recommended to optimize enzyme production and evaluate its application in industrial processes.

Abstract

Lecturer and community based science and technology activities (IbDM) were carried out with the aim of disseminating technology for Gurame culture intensively in an effort to increase fisheries productivity in Limau Manis, Pauh Subdistrict, Padang City and increase the knowledge and skills of fish farmers (community) in Koto Tangah Village in the field of Gurame fish maintenance business in West Sumatra, so that they are expected to be able to carry out maintenance in a good / environmentally friendly manner. Besides this IbDM activities are also expected to be able to reduce operational costs in maintaining Nirwana Tilapia and in the end can increase the income of the farmers (community) of Limau Manis Village through the development of sustainable fish farming. To achieve optimal technology dissemination results, the main target of IbDM in Pauh Subdistrict is fish farmer groups and community leaders / leaders of Limau Manis Village who are expected to be able to motivate farmers to socialize the method of making alternative formula feeds for intensive development of Nirwana Tilapia aquaculture and environmentally friendly. The methods used in this activity are (1) counseling, with the main material: (a) providing knowledge of various kinds of raw materials that can be used for fish food, (b) providing knowledge about the preparation of artificial food formulas / compositions, ( c) provide knowledge about the process of making artificial food (d) increase public knowledge about how to store pellets that are good to be more durable and (e) provision of material on testing the quality of feed, both physically, chemically and biologically; and (2) Intensive demonstration of making alternative pellet feeds for Nirwana Tilapia aquaculture. From the results of the IbDM activity, it was found that the knowledge and skills of farmer groups in Limau Manis Sub-district, Kec. Pauh, Padang before the IbDM activities were carried out was still very low. However, the motivation or desire of the community of IbDM partner farmer groups to carry out the culture or maintenance of fish is very large. This can be seen from the desire of the IbDM partner farmer group to master the technology of making fish feed from the results of leaf waste and taro stems which are processed into flour relatively high. Furthermore, the results of IbDM activities in the community can increase the knowledge and skills of the community in finding alternative feed for fish feed so that culture activities can run well in IbDM partner areas. In order for aquaculture to run well in partner areas of IbDM, it is necessary to carry out IbDM activities continuously on the techniques of making alternative fish pellets and efficient and effective fish-raising techniques. Besides that, it needs financial support or credit by the authorities to the community or fish farmers for capital to develop their businesses.

Abstract

Fish cultivators generally experience difficulties providing quality artificial feed (pellets) due to a lack of knowledge and skills in processing technology and making alternative feeds. Besides that, the costs incurred for feed procurement are relatively large, reaching 70 - 80% of the total production costs. However, the provision of feed is often an obstacle because, in addition to the increasingly high price, the quality of available feed does not always match the nutritional needs needed by Fish. The provision of quality feed for intensively cultivated tilapia dramatically affects the resulting growth and productivity. The higher the quality of the feed, the production will also increase. One of the technologies that can be applied to overcome this problem is the manufacture of alternative formulated diets using local raw materials. The method used in this service activity is the transfer of knowledge presented in the socialization activities in the form of 1) introduction to the types of alternative fish feed ingredients sourced from the area, (2) preparation of artificial tilapia feed formulations, (3) manufacturing technology artificial feed (pellets) and 4) monitoring and assistance. The socialization was carried out in the form of a lecture and was followed by a question and answer session. The results of farmer activities recognize the types of local feed that can be used as fish feed, know the preparation of feed formulations, and alternative artificial feed manufacturing technologies. The knowledge and skills possessed by fish cultivators are expected to overcome the difficulties of fish farmers in providing feed so that fish production and cultivation business increase.

Abstract

Jamur endofitik yang berasosiasi dengan mangrove Sonneratia sp. diketahui menghasilkan metabolit sekunder yang berpotensi sebagai antioksidan. Kawasan pesisir Mandeh merupakan habitat alami yang kaya akan Sonneratia sp., sehingga menjadi lokasi strategis untuk eksplorasi jamur endofitik.Tujuan dari penelitian ini adalah untuk mengeksplorasi jamur endofitik yang berasosiasi dengan mangrove Sonneratia sp., melakukan skrining aktivitas antioksidan secara in vitro, serta mengidentifikasi isolat potensial berdasarkan karakteristik makroskopis dan mikroskopis.Penelitian ini menggunakan metode eksperimen dengan tahapan sebagai berikut: isolasi jamur dilakukan melalui teknik direct planting, skrining aktivitas antioksidan dilakukan secara in vitro menggunakan metode DPPH, dan identifikasi isolat dilakukan berdasarkan karakteristik morfologi makroskopis dan mikroskopis. Hasil penelitian menunjukkan bahwa sebanyak 20 isolat jamur endofitik berhasil diperoleh. Berdasarkan hasil skrining aktivitas antioksidan, ditemukan empat isolat yang menunjukkan aktivitas antioksidan dengan kategori kuat, sedang, dan lemah. Isolat dengan kode T4L1C4 dan T3L2C3 menunjukkan aktivitas antioksidan kuat, sedangkan T3L2C2 menunjukkan aktivitas sedang. Berdasarkan karakteristik mikroskopis, ketiga isolat tersebut teridentifikasi sebagai anggota genus Fusarium sp. Kata kunci: Antioksidan; Endofitik; Jamur; Mandeh; Skrining

Biodiversitas 25: 3441-3448. Antibiotics previously used to treat skin infections have recently been ineffective due to pathogenic bacteria resistance, and the discovery of antibacterial agents derived from medicinal plants is a breakthrough. Eurya acuminata DC. is a member of the Pentaphylacaceae family and has efficacy values as a medicinal plant used in traditional medicines. This study aims to analyze the antibacterial activity of fractions (hexane, ethyl acetate, and water) of E. acuminata against bacteria causing skin infections and identify the chemical components of active fractions. A diffusion test was conducted for the antibacterial test. TLC (Thin Layer Chromatography) was used to determine the chemical components of the fractions. The fraction with the highest level of antibacterial activity was subjected to additional analysis using diffusion and LC-MS/MS (Liquid Chromatography-Mass Spectrometry/Mass Spectrometry). The antibacterial test showed that hexane, ethyl acetate, and water fractions were active against bacteria, with the ethyl acetate fraction having the highest activity. The TLC profile and the diffusion assay of the ethyl acetate fraction showed that spot number 4 with an Rf 1,00 had antibacterial activity. Identification of the compounds of the ethyl acetate fraction by LC-MS/MS resulted in identified compounds such as acacetin, kaempferol, peonidin, chrysoeriol, nobiletin, luteolin, and antroquinonol. This study demonstrates a new hope for further research into natural antibacterial agents, and it is a breakthrough with these phytochemicals.

Protease enzymes, the hydrolase enzymes that break down proteins into simpler molecules, are a vital component of the food, pharmaceutical, and other chemical industries. By selecting microorganisms as enzyme producers, we can meet the high demand for enzymes and promote sustainable production. This study aimed to determine the optimum conditions for protease production by protease-producing PUA-14 bacterial isolates from mangrove waters and to identify the isolates through biomolecular analysis. We conducted laboratory experiments to optimize protease production by varying carbon sources, nitrogen sources, trace elements, and inoculum concentrations. The optimization process employed the One Factor at A Time (OFAT) approach and the Central Composite Design (CCD) within the framework of Response Surface Methodology (RSM) using Design Expert 13 software. Furthermore, biomolecular analyses, including DNA isolation, amplification, and sequencing, were performed to identify the bacterial isolates. The study identified the optimum conditions for protease production by the PUA-14 bacterial isolate are 0.5% lactose, 2.5% NaNO3, Zn as the trace element, and 2.5% inoculum concentration. Molecular analysis confirms the similarity of the PUA-14 bacterial isolate to Bacillus pseudomycoides strain LB-AsDX1-3. These research findings not only provide a foundation for future research but also have immediate practical implications, highlight the potential for sustainable enzyme production under optimized conditions, and support a wide range of industrial applications.

Abstract

Tumbuhan mangrove Ceriops tagal diketahui sebagai salah satu sumber jamur endofitik yang mampu menghasilkan senyawa metabolit bioaktif dengan berbagai aktivitas biologis, termasuk potensi sebagai antibiotik untuk mengatasi masalah resistensi antimikroba yang terus meningkat secara global. Penelitian ini bertujuan untuk mengisolasi jamur endofitik Aspergillus spp. dari daun, akar, dan ranting C. tagal, serta mengevaluasi potensinya sebagai penghasil senyawa antibiotik terhadap Staphylococcus aureus, Escherichia coli, dan Candida albicans. Isolat dikultivasi pada suhu ruang selama 7 hari dalam kondisi statis, kemudian metabolit sekundernya diuji menggunakan metode difusi cakram (Kirby-Bauer). Sebanyak delapan isolat Aspergillus spp. berhasil diperoleh, yaitu AS7, AS8, AS12, AS26, AS47, AS51, AS53, dan AS85, yang menunjukkan variasi morfologi koloni serta potensi antibakteri. Hasil uji menunjukkan sebagian besar isolat mampu menghambat pertumbuhan E. coli dan S. aureus, tetapi tidak menunjukkan aktivitas terhadap C. albicans. Aktivitas terhadap E. coli umumnya lemah, kecuali AS8 (10,45 mm) yang tergolong kuat. Aktivitas terhadap S. aureus lebih tinggi, dengan tiga isolat AS7 (11,89 mm), AS12 (11,45 mm), dan AS51 (10,92 mm) masuk dalam kategori kuat. Dari delapan isolat yang diperoleh, AS7, AS8, AS12, dan AS51 menunjukkan aktivitas kuat terhadap Staphylococcus aureus dan Escherichia coli, namun tidak menunjukkan aktivitas terhadap Candida albicans. Temuan ini menegaskan potensi jamur endofitik mangrove sebagai sumber kandidat antibiotik baru.

Abstract

 Thermostable protease enzymes can be produced by thermophilic bacteria that act as biocatalysts in protein hydrolysis and have extensive industrial applications. The supply of stable and high-performance protease enzymes is still insufficient, encouraging exploration of thermophilic bacteria from extreme environments as potential producers. The aim of this study was to identify and characterize the macroscopic, microscopic, physiological, and biochemical features of thermophilic bacterial isolates that indicated protease production. This study was conducted using descriptive methods through rejuvenation, screening, and characterization of thermophilic bacteria isolated from the Sapan Hot Spring, South Solok Regency, as potential thermostable protease producers. Based on the results, six protease-producing thermophilic bacterial isolates, namely TUA-01, TUA-02, TUA-11, TUA-21, TUA-26, and TUA-31, with the highest Proteolytic Index (PI) value being isolate TUA-26. Characterization of the six protease-producing thermophilic bacterial isolates revealed differences in colony morphology characteristics, Gram-positive, rod-shaped cells, subterminal spores, motility, and catalase-positive. The thermophilic bacterial isolate TUA-26 has the potential to produce thermostable protease enzymes at high temperatures, making it highly promising for use in various biotechnology and industrial applications.
