Abstract

Decreased of libido is illustrated with disinterest in sexual activity caused by erectile dysfunction, impotence, and infertility. It can be treated by aphrodisiac agents. Katuk or Sauropus androgynus (L). Merr has long been used as a medicinal plant. The aim of this research was to evaluate which fraction of katuk leaf ethanol extract that had the aphrodisiac activity with parameters of climbing, introduction and the weight of testicular and seminal vesicle of male rat. Sprague-Dawley male rats as animals model divided into five groups: the normal control group, the positive control group (X-gra®), the n-hexane, ethyl acetate and water fraction groups in which each fraction group given a dose of 11.85 mg/kg body weight. The number of climbing and introductions were calculated on 0, 1st, 3th, and 5th day. The data were tested statistically with one-way ANOVA test followed by Tukey test. The weight of testicular and seminal vesicle of male rat were observed on the 15th day. Previously, rats were anesthetized using ketamine and then performed surgery. The results showed that the n-hexane fraction (11.85 mg/kg body weight) increased libido with the average number of climbing was 16.5 times and the average number of introductions was 27.75 times. It was also able to increase the weight of testicular and seminal vesicle of male rat compare to positive control (X-gra® 51.37 mg/kg body weight). The compounds contained in n-hexane fraction are terpenoids and steroids.

Abstract

Sauropus androgynus (L.) Merr. (Phyllanthaceae) is a green vegetable. It is rich in natural antioxidant compounds such as phenolics and flavonoids. The pre-extraction preparation can affect the results of extracting compounds from natural materials. This study aims to determine the effect of pre-extraction procedures, including sample form (fresh/dried), drying process (oven-drying/indirect sunlight-drying/air-drying), and particle size (ground/powdered) on total phenolic and flavonoid levels of ethanolic extracts of S. androgynus leaves and their antioxidant activity against DPPH radicals. S. androgynus leaves were extracted using the cold maceration method. The total phenolic and flavonoid contents of the extract were determined by the colorimetric method using a UV-Vis spectrophotometer. Antioxidant activity was measured based on the ability of the extracts to reduce DPPH free radicals. The results showed that the pre-extraction preparation influenced the antioxidant components obtained in S. androgynus leaves. According to sample form, fresh leaves contained a higher phenolics and flavonoids (33.66 mg GAE/g and 11.61 mg QE/g, respectively) than dried leaves. Whereas according to particle size, powdered dried leaves have higher phenolics and flavonoids content (45.12 mg GAE/g and 12.54 mg QE/g, respectively) than ground dried leaves. The leaves drying method that produced the highest phenolic content was oven-drying (47.08 mg GAE/g), while the one with the highest flavonoid content was air-drying (8.87 mg QE/g). All extracts had IC50 values against DPPH radicals around 85.71-93.91 ppm. Pre-extraction preparation in optimum condition makes the extraction process more efficient and effective in obtaining the target compounds with antioxidant activities.

Abstract

Pluchea indica L. (beluntas) from the Asteraceae is a plant known to be rich in phenolics. Studies on the phenolic content of P. indica stems have not been explored much. The purpose of this study is to qualitatively identify the presence of phenolic compounds and to screen for the radical scavenging ability of DPPH in P. indica stem extracts using the TLC method. Extraction was carried out with the ultrasonic directly and sequentially using 50% ethanol as solvent. Separation was carried out on a silica gel plate GF254 with ethyl acetate: water: formic acid: toluene (20:2:2:1) as the mobile phase. Gallic acid and chlorogenic acid were used as comparisons. Identification of the presence of phenolic using FeCl3 5% as a spray reagent, while screening for radical scavenging activity using DPPH 0.1 mM as a spray reagent. The results showed that the 50% ethanol extracts of P. indica stems, both obtained from direct and stratified extraction, indicated the presence of phenolic compounds as antioxidants. The phenolic compounds detected were identical to gallic acid and chlorogenic acid, respectively, at hRf values of 89.4 and 55.3. The development of phenolic utilization from P. indica stems as a source of antioxidants still needs to be studied.

Abstract

Epiphyllum oxypetalum (DC.) or Wijaya Kusuma (Cactaceae) is an ornamental plant which is knownto contain polyphenol compounds. The purpose to determine the levels of total phenolic and flavonoidcompounds in the fractions of Wijaya Kusuma leaves using microplate-based assay. Each sample wasdetermined phenolic content with Folin-ciocalteu reagent and flavonoid content using AlCl3 on themicroplate. Absorption was measured using a microplate reader. Screening of DPPH radical scavengingactivity was identified using the TLC method. The results showed that the extract and fractions ofthe leaves of Wijaya Kusuma contained phenolic and flavonoid compounds. The ether fraction hadthe highest phenolic content (35,912±0,54776 mgGAE/g) and flavonoids (0,773±0,008 mgQE/g)compared to other fractions (ether>water>ethyl acetate fraction). There is one yellow spot with an Rfvalue of 0,85 in the ether fraction which is thought to be a compound capable of scavenging the DPPHradicals. The ether fraction of Wijaya Kusuma leaves has potential as a source of polyphenols withantioxidant activity. However, further research is needed to evaluate the antioxidant capacity in theether fraction of Wijaya Kusuma leaves. Keywords: Antioxidant, Epiphyllum oxypetalum (DC.) Abstract

Pluchea indica (L.) Less is traditionally utilized to treat postpartum women in Indonesia. The plant has many pharmacological properties, so that it can be further developed as herbal medicine. In that development process, plant authentication is needed to ensure the quality of raw materials. A simple microscopical and thin-layer chromatography (TLC) analysis might be a way to authenticate the plant, but it has never been reported. So, this study evaluates the microscopical and TLC analysis for P. indica authentication in standardized herbal medicines production. Plant microscopic observation, fluorescence analysis, and polyphenol screening were conducted. n-Hexane, ethyl acetate, and methanol extracts of plant organs were then analyzed by TLC. Here, we reported that in microscopical analysis the simplicia of P. indica contains trichomes and tannin-containing cells. In addition, chlorogenic acid as a marker was present in TLC analysis by ethyl acetate-water-formic acid-acetic acid (8.5:1.5:1:1, v/v). The results of this evaluation might provide additional information in the identification, authentication, and quality control of P. indica as a raw material for herbal medicine.
