Abstract

Xylose and arabinose are pentosesugars that present in hemicellulose, part of lignocellulose biomass. These pentose sugars can be fermented by yeast into ethanol. The aim of this research was to utilize yeast isolated from durian fruit (DuriozibethinusL.) in fermentation of xylose and arabinose to produce bioethanol.Phenotypic test of isolates was conducted by growingthe isolates in various agar media, i.e.YPD (Yeast Peptone Dextrose), YPA (Yeast Peptone Arabinose), and YPX (Yeast Peptone Xylose) containing dextrose, arabinose, xylose, respectively, assole carbon source to see cell growth.  The yeast isolates were further identified using API AOC 20C kit method. Yeast isolates were applied for fermentation of glucose, arabinose, and xylosein incubated cultures.  Ethanol production in the fermentation was analyzed bygaschromatography. Yeast isolates were identified as Kodamaea ohmeri, Candida famata, Candida guilliermondii, and Crytococcuc laurentii. Based on gas chromatography data, it was found that ethanol produced in the fermentation for three days, the highest ethanol content on xylose substrate was fermented by Candida famata-Awhich is0.021% (v/v) ethanol resulted from initial concentration of 5% xylose (w/v).  While on arabinose substrate, the highest ethanol content was fermented by Crytococcus laurentii-Bwhich is 0.0034% (v/v) ethanol resulted from initial concentration of 5% arabinose (w/v).

Abstract

It had been done a biodegradation polyblend from blending polypropilene-palm oil-amylum with three composition of polyblend i.e polyblend A 80% polypropilene- 19.5% palm oil- 0.5% amylum, poliblend B 80% polypropilene- 19% palm oil- 1% amylum and polyblend C 80% polypropilene-18% palm oil- 2% amylum by B. subtilus and C. botulinum, time incubation was twenty five days. The characterization of polyblend before and after biodegradation has done with FTIR, DTA, Viscometre and tensile strength of polyblend. The result showed that Bacillus subtilus and Clostridium botulinum can biodegradate polyblend and make holes as well as chink on polyblend especially polyblend C, because it has more carbohidrat than polyblend A and B. Analysis from FTIR showed compatible of poliblend because it did not have a new function group and did not change of wavelength. Data of tensile strength showed lower value after biodegradation at polyblend C and from DTA and Viscometre showed lower melting point and lower average molecule weight, respectively.

Abstract

The purpose of this research was to isolated DNA from the yeast C. tropicalis with freeze thawing method -200 C conducted on 3 colonies of C. tropicalis.  Each colony   threated variations of cooling, 3x15 minutes, 3x25 minutes and 3x35 minutes, to break the cell walls.  Subsequently all the samples amplified with 3 variations of PCR cycles, 15 cycles, 25 cycles and 35 cycles, after all of the samples isolated by freeze thawing method -200 C. Its was known that sample A15 has the smallest concentration of DNA yeast C. tropicalis, ie 50 µg/mL, while sample C35 had the largest concentration of DNA yeast C. tropicalis, ie 225 µg/mL. The result of the research indicated that the best condition can be reached in 3x35 minutes. On 35th cycle has clearer C. tropicalis DNA bands than the 25th and 15th PCR cycle. C. tropicalis DNA bands at 35th cycles there were 7 DNA bands were detected and bright bands on a long 35 minutes cooling. In the 25th and the 15th cycle, there was no DNA bands were detected in all samples. Based on the results obtained, the amplification process must be carried out at least 35 times cycles so that the C. tropicalis DNA bands can be detected.

Abstract

This paper described the progress of phytochemical, especially alkaloids, work on Fagraea fragrans species (tembesu), Loganiaceae. A dimeric-pyridine alkaloid with molecular formula C20H20O4N2 has been successfully isolated from diethyl ether extracts from this plant’s barks. The alkaloid was explored from diethyl ether extracts using sulphuric acid (2%), then basified with ammonium hydroxide (25%) until the solution had a pH of 10.5. The later solution was extracted again with chloroform. The alkaloid residue was subjected to column chromatography and eluted with 10% acetone in chloroform. The chemical structure of the isolated dimeric-pyridine alkaloid was determined by LCMS/MS, 13C-NMR, and 1H-NMR spectroscopies. As a result, the isolated dimeric-pyridine alkaloid was closely similar to the oliveramine alkaloid, and this is also the first report about oliveramine from Fagraea fragrans (F. Fragrans).

Abstract

Background: Diarrhea can be treated with probiotic bacteria such as Bifidobacterium bifidum, which decreases the intestinal environment's pH to become acidic so that pathogenic bacteria cannot thrive. Objective: To make fermented coffee that can increase the number of probiotic bacteria Bifidobacterium bifidum and has antidiarrheal activity against pathogenic bacteria Escherichia coli. Methods: Robusta coffee (20.25% and 19.75%) was fermented using Saccharomyces cerevisiae, and then the probiotic bacteria Bifidobacterium bifidum was added. Unfermented coffee was compared with the same concentration (20.25% and 19.75%) to obtain four formulas. Organoleptic panelists tested all formulas to determine the best formula for fermented and non-fermented coffee. The number of Bifidobacterium bifidum and antibacterial activity was calculated on the optimum formulation using the Total Plate Count and Disc Diffusion Method. Result: The optimum formula obtained at fermented and unfermented coffee concentration was 20,25%. The number of probiotic bacteria Bifidobacterium bifidum growing in fermented and non-fermented coffee was 7.3 x 108 ± 32.4 and 3.1 x 108 ± 30.7 (p < 0.05). The diameter of the inhibition zone of the best fermented and non-fermented coffee was 11.5 ± 0.5 mm and 8.5 ± 0.5 mm, respectively (p < 0.05). Conclusion: Fermented coffee can increase the growth of the probiotic bacteria Bifidobacterium bifidum and has strong antibacterial activity against Escherichia coli bacteria.

Abstract

An unbalanced diet can have a negative impact on people's nutritional status. Therefore, nutrition training is considered an effective intervention strategy to improve diet and nutritional status. This research aims to evaluate the effect of nutrition training on people's eating patterns and nutritional status through an intervention study. This research method uses an intervention approach with a pre-test and post-test design involving people randomly selected from a certain area. Nutrition training is held with a focus on understanding balanced nutrition and implementing healthy eating patterns. Dietary pattern data was collected through questionnaires, and nutritional status analysis was carried out by measuring nutritional and anthropometric parameters before and after the intervention. The research results showed a significant increase in people's understanding of balanced nutrition and positive changes in their eating patterns after attending nutrition training. Apart from that, there have been substantial improvements in people's nutritional status, such as increasing body mass index (BMI) and optimal nutritional levels. This research concludes that nutrition training is effective in improving healthy eating patterns and the nutritional status of the community. This shows the importance of a focused intervention approach to increase nutritional awareness and behaviour in an effort to improve overall community health. This research makes an important contribution to the development of nutritional intervention strategies that can be widely applied to enhance community welfare. Therefore, it is recommended to pursue further research with a focus on the long-term effects of nutrition training and increasing accessibility to nutrition resources.
