Abstract

Background: Breast cancer (BC) is the leading cause of death cancer in women. Cancer therapies using TNFα and IFNγ have been recently developed by direct effects and activation of immune responses. This study was performed to evaluate the effects of TNFα and IFNγ directly, and TNFα and IFNγ secreted by Conditioned Medium-human Wharton’s Jelly Mesenchymal Stem Cells (CM-hWJMSCs) toward apoptosis of BC cells (MCF7).Materials and Methods: BC cells were induced by TNFα and IFNγ in 175 and 350ng/mL, respectively. CM-hWJMSCs were produced by co-culture hWJMSCs and NK cells that secreted TNFα, IFNγ, perforin (Prf1), granzyme B (GzmB) for treating BC cells. The BC cells were treated with CM-hWJMSCs in 50%. The expression of apoptotic genes Bax, p53, and the antiapoptotic gene Bcl-2 were determined using RT-PCR.Results: TNFα and IFNγ at concentration of 350 ng/mL induced higher Bax expression compared to 175 ng/mL. TNFα and IFNγ 350 ng/mL, 175 ng/mL induced p53 expression, whilst TNFα and IFNγ at 350 ng/mL decreased Bcl-2 expression. Perf1, GzmB, TNFα and IFNγ-containing CM-hWJMSCs induced significantly apoptosis percentage, induced Bax expression, but did not effect p53, Bcl-2 expression.Conclusion: TNFα and IFNγ directly induce Bax, p53, decrease Bcl-2 gene expression. The Prf1, GzmB, TNFα, IFNγ-containing CM-hWJMSCs induce apoptosis and Bax expression.Keywords: breast cancer, Wharton’s Jelly mesenchymal stem cells, TNFα, IFNγ

Abstract

This study aims to reveal the anticancer mechanism of the bioactive compound of Pisangulin angulata and to assess the feasibility of that results as teaching material. This research includes descriptive explorations. In the first stage, an in silico analysis was performed by molecular docking method between physalin compounds and GLI1 protein. The second steps of this study aim to develop teaching materials based research using the Analysis, Reorganizing, Piloting Class, and Evaluating (ARPE models). Feasibility test was carried out by experts and practitioners. Think Pair Share was used in the pilot project. Student motivation and misconception were recorded using SMI and CRI instrument. This study reveals that physalin B has higher activities than controls. The type of chemical bond that is formed between GLI1 amino acids residues with physalin is hydrogen bonds and hydrophobic bonds. The visualization of the types of bonds in that molecular docking between GLI1 amino acid residues and physalin has a high degree of feasibility (89) and can be used to enrich Chemistry for Biology lectures. The visualization of these chemical bonds can increase learning motivation and can improve the understanding of the concept of chemical bonds.

Abstract

Zona pellucida is one of the protective layer of the egg cell and has a function as an intermediary species-specific fertilization. Glycoproteins of human and bovine zona pellucida is composed of three types, namely ZP1, ZP2 and ZP3. ZP3 gene has amino acid sequence homology with other mammals. Oligosaccharides components of the zona pellucida glycans are composed from units of asparagine residues (N -linked) and serine/threonine (O -linked). The aims of this study was to analyze the DNA sequences of human and bovine and further predicts glycans site on amino acid sequence of human and bovine ZP3. In this study, ZP3 gene fragments have been isolated from bovine and humans were analyzed in Silico. This work were conducted by comparing the data of DNA sequence from human and bovine PCR product using NCBI BLAST. The results showed that there were similarities at amino acid positions number 23-38. bZP3 sequence had three glycans site (Asn-X-Thr/Ser) and one site on hZP3 glycans. One of the sites was conserved between the two species.

Abstract

Objective: Tamarindus indica is one of the famous natural plant that has various effects on health. Recent research has begun to examine the effect of Tamarindus indica on cell malignancy. One of the things that happens in cell malignancy is an increasing ability of cell migration, which is characterized by increasing expression of Snail2 and MMP-9. This study was conducted to determine the effect of extract of Tamarindus indica Fruit Extract (TIFE) towards Snail2 and MMP-9 expression in JEG-3 cells.Methods :.TIFE were extracted by using methanol. JEG-3 cells were grown in culture medium treated with TIFE (at 0, 25, 50 and 100 µg/ml). Immunofluorescence staining was carried out, and observed under Confocal Laser Scanning Microscope.Result: Snail2 expression after treatment with TIFE 50 and 100 µg/ml higher than control- (P=0.015 ; P=0.001) whereas at MMP-9 expression there was a biphasic pattern, in which the MMP-9 expression peaked at concentration of 50 µg/ml (P=0.002), then MMP-9 expression decreased at 100 µg/ml concentration. Conclusion : The expression of Snail2 influence MMP-2 expression after treat with TIFE, so it might increase the potential for JEG-3 cell migration Keywords : Methanolic extract, Tamarindus indica, Snail2, MMP-9

Abstract

The purpose of the study was to determine the influence of the concentration of ethylene glycol (EG) and length of exposure to levels of bovine oocytes in vitro fertility. This research was conducted using a completely randomized design factorial 5x3 with 7 replicates. The first factor was the concentration of cryoprotectants EG 10, 20, 30, 40, and 50%. The second factor was the length of exposure 1, 3, and 5 minutes. The results showed that the concentration of EG and the length of exposure were effect on fertilized oocytes (P0.05). The level of oocytes in vitro fertility after vitrification in 30% EG and long exposure to 3 minutes did not different (P0.05) compared to fresh oocytes, while the EG treatment 10, 20, 40, and 50% significantly showed a lower fertility rate than the 30% EG (P0.05). It was concluded that the concentration of EG and length of exposure were effect on fertilized oocytes. The highest oocytes in vitro fertility level were found in 30% EG concentration with 3 minutes exposure time.

Abstract

Tujuan penelitian ini untuk mengetahui pengaruh induksi EGF terhadap lokalisasi Cx43 dan gap junction selama ekspansi sel kumulus. Hasil penelitian ini menunjukkan bahwa ekspansi sel kumulus mempunyai signifikansi dengan meningkatnya konsentrasi EGF dan waktu kultur. Ekspansi sel kumulus diindikasikan dengan perubahan bentuk sel menjadi memanjang dan menyebar. Ekspresi Cx43 meningkat pada kultur jam ke-5 dan semakin meningkat pada kultur jam ke-10 dengan semakin padatnya protein pada badan sel. Pada kultur jam ke-15 terjadi penurunan ekspresi Cx43 pada badan sel. EGF berpengaruh terhadap peningkatan ekspansi sel kumulus tetapi tidak pada ekspresi Cx43 dengan bertambahnya konsentrasi dan waktu kultur. Hal ini menunjukkan bahwa komunikasi antar sel kumulus menurun, karena jarak antar sel yang semakin jauh.

Abstract

21st century learning focuses on problem-solving skills. A higher level of education requires an understanding of a more systematic concept to elaborate problems. Problem-solving skill is one of the fundamental cognitive processes. It involves higher order thinking to discover solutions or ideas. This research aims to analyze the relationship between concept mastery and problem-solving skill of pre-service biology teacher in human physiology courses. This is descriptive correlational research conducted in Sulthan Thaha Saifuddin State Islamic University Jambi. The sample was selected using the purposive sampling technique. Respondents in this study were 33 students in semester VI. The instrument used in the study was an essay test of concept mastery integrated with problem-solving. The results showed that the average scores of the students' concept mastery and problem-solving were 56.8 and 58.0, respectively. Both included in the poor category. It also showed that there was a relationship between students' concept mastery and problem-solving skill. The correlation value was 0.76, with a significance value of p0.05; therefore, it included in the strong category.

Abstract

This research is a type of descriptive research with the aim of knowing the critical thinking ability profile of prospective biology teachers at the University of Papua. The population in this study were all prospective biology teacher students at the University of Papua. The sampling technique is purposive sampling, namely sampling based on a specific purpose. The number of respondents was 42 prospective biology teachers. The instrument is in the form of essay questions. Based on research data analysis, the average proportion of critical thinking skills for biology teacher candidates is 55.03. Furthermore, the proportions based on critical thinking criteria are 10% uncritical, 53.33% less critical, 36.67% critical and 2.38% very critical. This in general it can be interpreted that the critical thinking skills of prospective biology teachers at the University of Papua are in the less critical category, this is very important to be empowered as a provision to face competition in the era of the industrial revolution 4.0.Keywords: Analysis, Critical Thinking Skills, Prospective Biology TeachersAbstrak Penelitian ini merupakan jenis penelitian deskriptif dengan tujuan untuk mengetahui profil kemampuan berpikir kritis calon guru biologi di Universitas Papua. Populasi dalam penelitian ini adalah seluruh mahasiswa calon guru biologi di Universitas Papua. Teknik pengambilan sampel dengan cara Purposive Sampling yakni pengambilan sampel berdasarkan tujuan tertentu. Jumlah responden sebanyak 42 calon guru biologi. Instrumen berupa soal essay. Berdasarkan  analisis data hasil penelitian  rata-rata persentase  keterampilan berpikir kritis calon guru biologi yaitu 55,03. Selanjutnya persentase berdasarkan kriteria berpikir kritis yaitu  kategori tidak kritis 10%, kurang kritis 53,33%, kritis 36,67 % dan sangat kritis 2,38%. Dengan demikian secara umum dapat disimpulkan bahwa keterampilan berpikir kritis calon guru biologi Universitas Papua berada pada kategori kurang kritis, hal ini kiranya sangat penting  untuk diberdayakan sebagai bekal untuk menghadapi persaingan di era revolusi industry 4.0.Kata Kunci:  Analisis, Keterampilan Berpikir Kritis, Calon Guru Biologi

Abstract

Colorectal carcinogenesis induced by cigarette smoke requires at least 30-40 years. This long time duration causes an animal research conducted becomes relevant. This research was carried out to observe colorectal carcinogenesis due to cigarette smoke exposure in Wistar Rat. The observations focused on changes in epithelial morphology and expression of APC, MSH2 and MLH1. Twenty male Wistar rats inbreed strain were randomly allocated into control group and experimental group exposure to cigarettes smoke for 14 weeks and 28 weeks sequentially.  Colorectal epithelial morphology was assessed on the histopathology examination, whereas the expression of APC, MSH2 and MLH1 was assessed on aspect of immunohistochemistry. The comparative analysis between the two groups was performed using non-parametric Mann-Whitney U test. Histology of colorectal epithelium showed pattern of colitis associated cancer that was significant both in 14 weeks and 28 weeks of treatment.  This research indicated negative expression of APC, MSH2 and MLH1 in the colorectal cancer that were significant at 28 weeks of exposure. This research implies that chronic exposure to cigarette smoke can induce colitis associated colorectal cancer through decreased expression of APC, MSH2 and MLH1.

Abstract

This research aims to determine the effect of PBL integrated with Igya Ser Hanjob's local wisdom on students' problem solving skills in the Conservation Biology course. The sample for this research was Biology Education Semester VII students taking Conservation Biology courses, totaling 48 students, namely class A (experimental class) with 24 students and class B (control class) with 24 students. This type of research is quasi-experimental research using a nonequivalent control group design. The instruments used in this research were description tests and questionnaires. The data obtained were analyzed using descriptive and inferential statistics which had previously met the requirements for normality and homogeneity, then the Anacova test was carried out using the SPSS IMB 20 program. The results of the research showed 1) there was an increase in the average score of students' problem solving skills after implementing PBL containing local wisdom 2 ) there is a difference in student problem solving skills between the group of students who took part in PBL containing Igya ser hanjob local wisdom and the group of students who took part in conventional learning as evidenced by a significance value of 0.000 < 0.05. Therefore, it can be concluded that PBL learning integrated with Igya Ser Hanjob's local wisdom can improve students' problem solving skills.Keywords: PBL, Problem solving skills, Igya ser hanjop, Local wisdom, Biology Conservation
