Abstract

Increasing the number of heavy metals in the aquatic environment results in the accumulation of contaminants in fish body tissue, making fish an ideal bio-indicator of environmental pollution. The spleen included in the hematopoietic tissue contains macrophages. Macrophages form aggregates that contain pigments referred to melanomacrophage centres (MMCs). This research aims to analyse the metric profiles of MMCs on the common carp (Cyprinus carpio) spleen as a non-specific cellular immune response against mercury chloride exposure. This study used a Completely Randomized Design method with four treatments at different concentrations of mercury chloride (HgCl2) of 0 mg.l-1 , 0.01 mg.l-1 , 0.05 mg.l-1 , and 0.1 mg.l-1 . Each treatment was replicated five times. Data on the number and size of MMCs were analyzed by Analysis of Variance (ANOVA) and continued by using Duncan's Multiple Range Test (DMRT) to determine the differences between treatments. The result showed that the concentration of mercury chloride in water affects the metrics profile of spleen MMCs of carp. The number and size of MMCs of carp spleen increased at the concentration of 0.01 mg.l-1 and 0.05 mg.l-1 but decreased at the level of 0.1 mg.l-1 . The results suggest that the profile of MMCs as a non-specific cellular immune response can function as bio-indicators of environmental pollution.

Abstract

Artemia salina is a natural food source known for its high nutritional quality. It is commonly used in larval rearing due to its ability to produce long-lasting cysts that can hatch at any time. These characteristics make A. salina very suitable for sustainable cultivation. In this study, the feed consisted of rice bran suspension enriched with Isolated Soy Protein (ISP) meal. The aim was to investigate the effect of different protein concentrations in the rice bran suspension feed on cyst production in A. salina broodstock. This study used a Completely Randomized Design (CRD) with five treatments and four replications. The treatments included P0 (without ISP meal, 25% protein concentration), P1 (ISP meal, 40% protein concentration), P2 (ISP meal, 45% protein concentration), P3 (ISP meal, 50% protein concentration), and P4 (ISP meal, 55% protein concentration). Data analysis was performed using ANOVA followed by Duncan's multiple range test. The results showed that variations in protein concentration with the addition of ISP flour had a significant effect (P < 0.05) on total cyst production and cyst production per broodstock of A. salina. The highest total cyst production (710.67 cysts/L) was achieved in P4 with a protein concentration of 55%, while the highest cyst production per broodstock (28.67 cysts/broodstock) occurred in P2 with a protein concentration of 45%.

Abstract

White leg shrimp (Litopenaeus vannamei) is a widely cultivated commodity in Indonesia. The high demand of white leg shrimp requires increased production. High stocking density in intensive cultivation and excessive amounts of feed are obstacles in white leg shrimp productiom. It will cause the shrimp to become stressed and susceptible to pathogen attack , one of them is ectoparasite. This research was conducted from August to September 2023 with the aim of determining the intensity and infestation degree of ectoparasite on white leg shrimp that cultivated in intensive ponds. The samples of white leg shrimp in this study were 50 individuals by purposive random sampling method. Observations were carried out by natif method in the Cultivation and Anatomy Laboratory, Faculty of Fisheries and Marine, Airlangga University. The results showed that white leg shrimp cultivated in intensive ponds were infested by Zoothamnium sp. with intensity 184 individuals/head, Vorticella sp. with intensity 268 individuals/head and Epistylis sp. with intensity 235 individuals/head. Abstract

Macrognathus aculeatus is a local fish from the Bengawan Solo and Brantas Rivers threatened with extinction. Due to the lack of literacy on the biology of this fish. Biological parameters can be used to determine the condition of this fish in nature. Field observations at five stations from the two rivers were conducted to obtain samples. Biological and reproductive parameters such as weight and length, gonad maturity level (GML), Gonadosomatic Index (GSI), Hepatosomatic Index (HSI), and Condition Factor (CF) were the parameters observed in this study. All data obtained were presented as mean and standard deviation. Furthermore, analysis of variance was conducted using ANOVA (p < 0.05) to determine differences in samples from each observation station. The results showed that some biological factors had significant results (p < 0.05) on Gonadosomatic Index and Hepatosomatic Index). However, there was no significant difference (p > 0.05) in the condition factor. On the other hand, there were significant differences (p < 0.05) in body weight, total length, ovary weight, and fecundity parameters across all sampling sites.

Abstract

Whiteleg shrimp (Litopenaeus vannamei) is one of the main fishery commodities since it has a high economic value. Vaname shrimp cultivated is done intensively where the stocking density and the amount of feed required are high. High stocking densities can trigger stress, increase possibility of physical contact among shrimps, and the spread of disease in shrimp becomes faster. The most common shrimp disease is Vibriosis where whiteleg shrimp are infected by Vibrio sp.. Whiteleg shrimp has an innate immunity system thus hemolymph has an important role in shrimp body defense. In this study, the research method used was purposive random sampling and was conducted from August to September 2023 by 50 shrimps sample. Assay of phagocytosis index (IF) which is a cellular response of the vaname shrimp body defense system was conducted at the Center for Brackish Water Aquaculture (BBPBAP) Jepara by applying a bacterial suspension of Staphylococcus sp. which has been attenuated and bacterial staining. Calculation of Total Vibrio Count (TVC) was conducted at the Laboratory of Bacteriology and Mycology, Faculty of Veterinary Medicine, Universitas Airlangga, by using TCBS (Thiosulfate Citrate Bile Salt Sucrose Agar) agar media from Vibrio sp. bacteria cultured from the hepatopancreas of shrimp samples. The results showed that vaname shrimp cultured in intensive ponds were infected with Vibrio sp. with a TVC value of 3.3x105 cfu/ml and an IF value of 0.87.

Abstract

One of the failures of freshwater fish farming is often caused by the attack of A. hydrophila bacteria. This bacterial attack can be prevented by administering immunostimulants derived from G. verrucosa extract. This study aims to determine the non-specific immune response of striped catfish to Aeromonas hydrophila infection following supplementation of G. verrucosa extract in feed. Five doses were administered in this study: 1 ml, 1.5 ml, and 2 ml per kg of feed, as well as a positive control and a negative control. Blood samples were collected every week after infection, and the parameters observed were total erythrocyte and leucocyte counts, as well as leucocyte differential counts. The results showed that G. verrucosa extract significantly increased blood cell counts. The highest dose was found to be the most effective in increasing the non-specific immune response of striped catfish against A. hydrophila infection
