Abstract

Shorea johorensis is one of the red meranti plants in Ketambe Research Station, Gunung Leuser National Park, Aceh Tenggara. Currently, Shorea johorensis also is well known as a major source of valuable commercial timber. This research aimed to analyze the phylogenetic of Shorea johorensis based on chloroplast and nuclear DNA in Ketambe Research Station so that it can be known the relationship of Shorea johorensis with other species Dipterocarpaceae in GenBank database. The research was conducted from July 2015 to August 2016 in Ketambe Research Station and Forestry and Forest Genetics Laboratory of Molecular, Bogor Agricultural University. The method used quadrat sampling technique with purposive sampling and experimental laboratory that consisted of DNA extraction, PCR, electrophoresis, and sequencing. The data analysis was done using BioEdit, MEGA6, and BLAST. The results showed that the phylogenetic tree of Shorea johorensis based on the rbcL and matK showed that Shorea johorensis was closely related with some species Hopea; but the phylogenetic tree based on psbA-trnH, 5.8S rRNA, ITS2, and 28S rRNA showed that Shorea johorensis was closely related with Shorea robusta.Keywords: Chloroplast dna, Ketambe Research Station,Nuclear DNA, Shorea johorensis

Abstract

Mushroom is one of living things with abundant diversity and has various beneficial potentials; one of them is as medicine. The use of mushrooms as medicine has long been known primarily as traditionally made herbs and the knowledge has been passed down for generations. The Pocut Meurah Intan Forest Park as a conservation forest, stores a variety of mushrooms in it. Data about the type of mushroom that has medicinal potential existed in the forest park has never been published so it is important to do so. This study aims to record the types of mushrooms that have benefits as medicines. The method used is an exploratory survey through a trail or tracking path then identified as a medicinal mushroom base on literature studies. The results of the study found 15 species of mushrooms that have potential as medicine, namely Cookeina tricholoma, Sarcoscypha coccinea, Tremella fuciformis, Auricularia auricula-judae, Flammulina velutipes, Schizophyllum commune, Scleroderma citrinum, Boletus edulis, Trametes versicolor, Lentinus tigrinus, Lentinus squarosulus, Ganoderma applanatum, Ganoderma resinaceum, Thelephora ganbajun and Lycoperdon echinatum. This shows that Pocut Meurah Intan Forest Park has great resources and potential that can be utilized for the benefit of the surrounding community, and it needs to be preserved so that diversity is maintained and can be used by future generations.

Abstract

DNA extraction is the first step in obtaining high-concentration and high-purity DNA that can use in the subsequent steps. Because the leaf structure is challenging and contains several secondary metabolites that can affect DNA extraction results, DNA extraction on dry leaves of Dipterocarpaceae is considered problematic. This research aims to find a suitable method for extracting DNA from the preserved Dipterocarpaceae leaves. Preserved leaves of Dipterocarpaceae in silica gel will dry up, making them tough to destroy. Perform DNA extraction is generally using CTAB buffers or extraction kits. According to the results of this study, extraction with modified CTAB buffer result in DNA with higher concentration and purity values than DNA extracted with the kit. DNA isolated from the modified CTAB can be used as a template in the PCR process to amplify the matK gene.

Abstract

The Tripa peat swamp forest is located on the west coast of Aceh province and falls administratively under the two districts of Nagan Raya and Aceh Barat Daya. Tripa peat swamp forest has a high diversity of plants. The previous research discovered that Myrtaceae is one of the dominant families in the forest. This study aimed to predict phylogenetic relationships among trees species within families Myrtaceae that live in the Tripa peat swamp forest using Internal Transcribed Spacer (ITS). The research was conducted at Laboratory of Forest Genetics and Molecular Forestry, Faculty of Forestry, IPB from June 2015 to December 2016. The methods were conducted with an experimental laboratory of molecular identification consisting of DNA extraction, amplification, and sequencing. Data was analyze using BioEdit, Clustal X, Mega 6, and BLAST. The morphology identification reported that there were four trees species within families Myrtaceae, i.e., Syzygium sp 1, Syzygium garciniifolium, Syzygium leptostemon and Tristaniopsis whiteana. Reconstruction of phylogenetic tree using the Neighbor-Joining (NJ) method 1000x showed that the ITS region was successfully to predict phylogenetic relationships within genus Syzygium and Tristaniopsis.Keywords: tripa peat swamp forest, tree species, phylogenetic, myrtaceae, internal transcribed spacer

Abstract

Artificial medium for in vitro culture is an alternative media to subtitute Murashige and Skoog (MS). Growmore added with organic substances as coconut water can be used as artificial medium. Coconut water can affect plant growth, so the research is to investigate the influence of coconut water in different concentration to chrysanth’s (Dendranthema grandiflora) growth should be done. The research was conducted in Laboratory of Molecular and Cell Biology, Department of Biology, Faculty of Mathematics and Natural Sciences, Syiah Kuala University Darussalam, Banda Aceh, from April until July 2013. The research was designed by using Randomized Block Design with sub sampling. Media act as group, while coconut water concentration as treatment. Observation was started from growing time up to a month after planting. The result showed that there was no significant different between using MS and artificial medium with different coconut water concentration to initiate shootlet.

Abstract

The Dipterocarpaceae family is one of the identified plant groups with high conservation value that exist in Weh Island Nature Tourism Park. However, the data availability on the species diversity and population structure of Dipterocarpaceae in Weh island in particular and Aceh in general is still very limited. Thus, it is necessary to conduct studies on species diversity and population structure of Dipterocarpaceae from Weh Island. This study aims to obtain data on species abundance and diversity, species composition of each stratum, population structure, and stand structure of Dipterocarpaceae in Weh Island Nature Tourism Park. This research utilized transect method with randomly selected plots according to topographic conditions. We found two Dipterocarpaceae species: Shorea sumatrana (Slooten) Desch and Dipterocarpus hasseltii Blume. Shorea sumatrana (Slooten) Desch has the greatest abundance of all types and strata. The diversity of Dipterocarpaceae species in Weh Island Nature Tourism Park is categorized as low because the diversity index value is in the range of H'1. This means that the presence of seedling and sapling strata from each species has a major effect on the survival of the Dipterocarpaceae family in the future. The stand structure of the Dipterocarpaceae family in normal conditions is indicated by an inverted “J” shaped diagram.

Abstract

The combination between Naphthalene acetic acid (NAA) (1-2 ppm) and Benzyl Amino Purine (BAP) (0, 5 and 1 ppm) for micro propagation of castrol oil plant (Jatropha curcas L.) had been studied to induce the callus formation had been studied to induce the callus formation by using the meristem shoot as the ex-plants. The result showed that the combination of NAA 1,5 ppm and BAP 0,5 ppm gave the best result.

Abstract

Abstrak. Medang Kuli (Actinodaphne nitida) merupakan  jenis kayu komersial dengan kayu yang kuat selain itu jenis tumbuhan ini mengandung alkaloid (obat-obatan). Medang kuli (Actinodaphne nitida) tidak ada dalam daftar kayu yang bisa diperdagangkan serta telah disahkan oleh Keputusan Mentri Kehutanan No.163/Kpts-II/2003. Tujuan dari dilakukannya penelitian agar bisa mengetahui ciri-ciri secara umum, makroskopis serta mikroskopis kayu Medang Kuli (Actinodaphne nitida) di kabupaten Gayo Lues. Dalam pengujian secara struktur anatomi mikroskopis digunakan standar internasional (Association of Wood Anatomist). Didapatkan hasil struktur anatomi darikayu Medang Kuli (Actinodaphne nitida) secara umum memiliki warna kuning jerami, tekstur, tekstur halus, arah seat berpadu, kusam, berat sedang, tidak terlalu keras, kesat, tidak bercorak, tidak berbau, tidak memiliki kulit tersisip dan tanda kerinyut. Berdasarkan ciri makroskopis dan mikroskopis kayu Medang Kuli (Actinodaphne nitida) memiliki Kayu Medang Kuli (Actinodaphne nitida) memiliki penyebaran pembuluh/pori tata baur, penyusunan pori diagonal dan atau radial, penggabungan pori bergabung, isi pori deposit, rata-rata luas pori 1.6101,24 µm, diameter pori 142,43 µm, dinding pori 8,24 µm, jarak antar pori 311,40 µm. Parenkim paratrakeal sepihak, parenkim paratrakeal alifrom sayap tipis, parenkim apotrakeal deretan tangensial pendek dan parenkim aksial lainnya bentuk tangga. Tipe jari-jari halus, ukuran jari-jari memiliki satu ukuran, lebar jari-jari 17,89 µm, panjang jari-jari 129,63 µm, tebal dinding jari-jari 4,54 µm, jarak antar jari-jari 61,64 µm, saluran intraseluler aksial deretan tangensial pendek. Diameter serat 32,82 µm, diameter lumen 12,77 µm, panjang serat 4.685,95 µm, tebal dinding serat 10.33 µm. Abstract. Medang Kuli (Actinodaphne nitida) is a commercial type of wood with strong wood besides that this plant species contains alkaloids (medicines). Medang coolie (Actinodaphne nitida) has not been registered in the timber trade which has been ratified by the Decree of the Minister of Forestry No. 163/Kpts-II/2003. This study aims to determine the general characteristics, macroscopic and microscopic characteristics of Medang Kuli wood (Actinodaphne nitida) in Gayo Lues district. In this study to test the microscopic anatomical structure refers to the standards of the International Association of Wood Anatomists. The results showed that the anatomical structure of Medang Kuli wood (Actinodaphne nitida) in general had a straw yellow color, texture, smooth texture, blended seat direction, dull, medium weight, not too hard, rough, not patterned, odorless, did not have inserted skin and wrinkle marks. Based on the macroscopic and microscopic characteristics of Medang Kuli wood (Actinodaphne nitida) Medang Kuli wood (Actinodaphne nitida) has diffuse distribution of vessels/pores, diagonal and/or radial pore arrangement, merged pores, deposited pore content, average pore area of 1,601, 24 m, pore diameter 142.43 m, pore wall 8.24 m, pore spacing 311.40 m. Unilateral paratracheal parenchyma, thin wing alifrom paratracheal parenchyma, short tangential row apotracheal parenchyma and other axial parenchyma in the form of stairs. Type of fine fingers, the size of the radius has one size, the width of the radius is 17.89 m, the length of the radius is 129.63 m, the wall thickness of the radius is 4.54 m, the distance between the fingers is 61.64 m, short tangential row axial intracellular channels. Fiber diameter is 32.82 m, lumen diameter is 12.77 m, fiber length is 4,685.95 m, fiber wall thickness is 10.33 m.

Abstract

Abstrak. Hutan Arul Relem merupakan hutan lindung yang terdapat di Kecamatan Pining yang masih menyimpan keanekaragaman hayati baik flora maupun fauna, salah satu keanekaragaman yang terdapat di hutan Arul Relem adalah suku Lauraceae. Suku Lauraceae merupakan kelompok tumbuhan yang memiliki kualitas kayu yang bagus dan nilai ekonomi yang cukup penting. Tujuan dari penelitian ini adalah untuk mengidentifikasi jenis suku Lauraceae dan memetakan keberadaan suku Lauraceae di hutan Arul Relem. Metode yang digunakan dalam penelitian ini adalah metode garis berpetak yang dibuat sebanyak 4 transek dan 20 plot. Di mana panjang setiap transek 180 m, dan setiap transek dibuat 5 petak ukur yang berukuran 20x20 m dengan jarak antar petak 20 m, dan pada setiap petak ukur dibuat sub-sub petak berdasarkan strata pertumbuhan. Berdasarkan hasil pengamatan di hutan Arul Relem terdapat 10 jenis suku Lauraceae yang berasal dari 5 genus yaitu genus Actinodapne, Litsea, Beilcsmiedia, Cinnamomum, dan Phoebe, dengan jumlah individu sebanyak 93 individu yang terdiri dari 30 individu pada strata pohon, 17 individu tiang, 12 individu pada strata pancang, dan 34 individu strata semai. Jenis yang paling banyak ditemukan adalah jenis Actinodaphne nitida yaitu sebanyak 19 individu dan strata pertumbuhan yang paling banyak ditemukan adalah strata semai. Suku Lauraceae tersebar pada beberapa ketinggian yaitu pada ketinggian 500 mdpl terdapat 31 individu suku Lauraceae, pada ketinggian 520 mdpl terdapat 38 individu,  pada ketinggian 550 mdpl terdapat 17 individu, dan pada ketinggian 575 terdapat 4 individu. Tingkatan yang paling dominan ditemukan pada setiap ketinggian adalah strata semai dan jenis yang paling dominan ditemukan adalah jenis Actinodaphne nitida, Actinodaphne glabra, dan Litsea sp.Abstract. Arul Relem Forest is a protected forest located in Pining District which still stores biodiversity both flora and fauna, one of the diversity found in Arul Relem forest is the Lauraceae tribe. The Lauraceae are a group of plants that have good wood quality and significant economic value. The purpose of this study was to identify the species of the Lauraceae tribe and to map the presence of the Lauraceae tribe in the Arul Relem forest. The method used in this research is the checkered line method which consists of 4 transects and 20 plots. Where the length of each transect is 180 m, and each transect is made 5 plots measuring 20x20 m with a distance between plots of 20 m, and in each measuring plot, sub-plots are made based on growth strata. Based on observations in the Arul Relem forest, there are 10 species of the Lauraceae family originating from 5 genera, namely the genus Actinodapne, Litsea, Beilcsmiedia, Cinnamomum, and Phoebe, with a total of 93 individuals consisting of 30 individuals in tree strata, 17 pole individuals, 12 individuals in the sapling strata, and 34 individuals in the seedling strata. The most common type found was Actinodaphne nitida, which was 19 individuals and the most common growth strata was the seedling strata. The Lauraceae tribe is spread over several altitudes, namely at an altitude of 500 meters above sea level there are 31 individuals of the Lauraceae tribe, at an altitude of 520 meters above sea level there are 38 individuals, at an altitude of 550 meters above sea level there are 17 individuals, and at an altitude of 575 there are 4 individuals. The most dominant level found at each altitude was the seedling strata and the most dominant species found were Actinodaphne nitida, Actinodaphne glabra, and Litsea sp.

Abstract

Kedah adalah salah satu nama Dusun di Desa Penosan Sepakat, Kecamatan Blangjerango, yang merupakan salah satu gerbang menuju Hutan Taman Nasional Gunung Leuser (TNGL). Hutan yang ada di Desa Penosan Sepakat dikenal dengan nama Rainforest Lodge Kedah yaitu salah satu hutan yang menjadi objek wisata dan penelitian bagi masyarakat lokal atau mancanegara, karena Rainforest Lodge Kedah menyimpan kekayaan sumberdaya alam hayati  baik itu flora maupun fauna. Penelitian ini bertujuan untuk mengidentifikasi jenis tumbuhan berbasis pohon pada tingkat pertumbuhannya, dan menghitung indeks keanekaragaman jenis tumbuhan. Pada penelitian ini metode yang di gunakan yaitu menganalisis tumbuhan pada setiap stara pertumbuhan yang berkayu. Metode analisis yang digunakan yaitu dengan mengkombinasikan jalur dan garis berpetak dengan ukuran 20 x 100 m, yaitu sebanyak 9 jalur. Peletakan 3 jalur pada masing-masing ketinggiaan yaitu pada ketinggian 1.300 m dpl, 1.400 m dpl dan 1.500 m dpl, jarak setiap jalur masing-masing  60 m, setiap jalur di bagi menjadi 4 petak contoh, setiap petak contoh dibagi lagi menjadi 4 ukuran yaitu dengan masing-masing memilki ukuran yang berbeda-beda sesuai stara pertmbuhan. Jenis tumbuhan yang terdapat di Rainforest Lodge Kedah terdapat 45 jenis tumbuhan kayu dari 21 suku, yang merupakan hasil seluruh jenis kayu yang terdapat disemua ketinggian. Jenis-jenis tumbuhan yang terdapat di Rainforest Lodge Kedah saat pengamatan dengan luas petak contoh 14.400 m² terdapat 45 jenis dengan jumlah 950 individu, di mana untuk strata pohon terdapat 248 individu, untuk strata tiang terdapat 144 individu, untuk strata pancang terdapat 335 individu dan untuk strata semai terdapat 223 individu. Indeks keanekaragaman tumbuhan di Rainforest Lodge Kedah untuk strata pohon diperoleh 3,184 untuk strata tiang diperoleh 3,096, Untuk strata pancang diperoleh 3,191, untuk strata semai di peroleh 2,919 Hal ini menunjukan bahwa komunitas jenis pohon, tiang, pancang dan semai bisa dikatakan termasuk ke dalam kondisi baik serta tinggi berdasarkan ilustrai penyebaran nilai indeks tersebut.
