Abstract

Ulva lactuca is rich in bioactive compounds, which are beneficial to humans and other organisms. This study aimed to determine the tyrosinase inhibitory activity of an ethanol extract of U. lactuca seaweed in vitro. This research was carried out in stages, namely, the maceration process of U. lactuca with an ethanol solvent. The extract was liquid-liquid fractionated using n-hexane, chloroform, and water as solvents. The ethanol extracts and fractions were tested for total phenol, total flavonoid, and tyrosinase inhibitory activities. The highest total phenol and flavonoid values ​​were obtained from the n-hexane fraction with values ​​of 9.43±0.27 mgGAE/g and 9.20±0.49 mgQE/g. The highest tyrosinase inhibitory activity was obtained from the n-hexane fraction, with an IC50 value of 127.74±6.47 µg/mL. Active compounds that play an important role in tyrosinase inhibitory activity in the ethanol extract and n-hexane fraction include chalcone, ferulic acid, 4-nitrocinnamic acid, 4-aminobenzoic acid, 2TMS derivatives, 4-hydroxybenzoic acid, 4-tert-butylfenol, 1-tetradecanol, n-hexadecanoic acid methyl ester, palmitic acid, 2-(2-Aminophenyl)-1H-benzimidazole, and 5-methylsalicylic acid. Thus, the n-hexane fraction of U. lactuca could be used as a natural tyrosinase inhibitor.

Abstract

Seaweed contains various bioactive compounds rich in pharmacological potentials, such as antioxidants and anti-arthritic. This study aims to determine the antioxidant and anti-arthritic activities of the methanolic extract of Halimeda tuna. Halimeda tuna samples were extracted using the maceration method and methanol. The methanol extract was tested for antioxidant (2,2-diphenyl-1-picrylhydrazyl (DPPH), ferric reducing antioxidant power (FRAP)) and anti-arthritic activity using denatured bovine serum albumin (BSA). The methanol extract was also subjected to phytochemical and Gas Chromatography-Mass Spectrometry (GC-MS) analysis. Research results revealed that the extract yield was 0.50 ± 0.06%. Phytochemical analysis showed that H. tuna methanolic extract contained flavonoids, steroids, and alkaloids. GC-MS analysis showed that H. tuna methanolic extract contained steroids, fatty alcohols, and fatty acids. The antioxidant activity tests revealed an IC50 value of 299.98 ± 121.15 ppm and a FRAP value of 200.02 ± 8.50 M/g, categorized as a weak antioxidant. Anti-arthritic activity of H. tuna methanolic extract showed an IC50 value of 877.29 ± 209.84 ppm. The anti-arthritic activity of H. tuna methanolic extract was lower than sodium diclofenac as a standard drug. The extract of H. tuna has antioxidant and anti-arthritic activities; hence, further exploration is needed as an alternative potential in the pharmacological field.

Abstract

Gracilaria sp. merupakan jenis rumput laut merah yang kaya akan kandungan iodium. Pemanfaatan Gracilaria sp. bisa diolah menjadi bubuk yang dapat difortifikasikan pada beberapa makanan. Inovasi penambahan bubuk Gracilaria sp. pada opak singkong dapat dilakukan sebagai salah satu alternatif makanan untuk mencukupi kadar iodium bagi tubuh. Tujuan penelitian ini adalah untuk mengetahui pengaruh fortifikasi rumput laut Gracilaria sp. terhadap karakteristik mutu opak singkong dan tingkat penerimaan konsumen terhadap opak singkong dengan fortifikasi Gracilaria sp. Penelitian dilakukan dengan membuat opak singkong dengan pencampuran bubuk Gracilaria sp. (0%; 2,5%; 5%; 7,5%; dan 10%) (b/b). Opak singkong dibuat untuk diuji kadar iodium, uji kerenyahan, uji daya kembang, uji kadar air, kadar abu, kadar lemak, kadar protein, kadar karbohidrat dan uji hedonik. Fortifikasi Gracilaria sp. pada opak dapat meningkatkan kadar iodium, menurunkan tingkat kerenyahan dan daya kembang opak, serta meningkatkan kadar air, kadar abu, dan kadar protein serta menurunkan kadar lemak dan karbohidrat dari opak singkong. Gracilaria sp. is a type of red seaweed that is rich in iodine content. Iodine is one type of essential mineral needed for the body which if in a state of deficiency will result in various kinds of health problems. Utilization of Gracilaria sp. can be processed into a powder that can be fortified in some foods. The innovation of adding Gracilaria sp powder. in cassava opaque can be done as an alternative food to meet the levels of iodine for the body. The purpose of this study was to determine the effect of fortification of Gracilaria sp. on the quality characteristics of cassava opaque and the level of consumer acceptance of cassava opaque with fortification of Gracilaria sp. The research was conducted by making cassava opaque by mixing powdered Gracilaria sp. (0%; 2.5%; 5%; 7.5%; and 10%) (w/w). Cassava opaque was made to be tested for iodine content, crunch test, swellability test, water content test, ash content, fat content, protein content, carbohydrate content and hedonic test. Fortification of Gracilaria sp. in opaque can increase iodine content, reduce the level of crispness and opacity of opaque, and increase water content, ash content, and protein content and reduce fat and carbohydrate content of opaque.

Abstract

Archaeological research in various parts of Indonesia has helped the construction of local and national identities as a unifying narrative of the nation. The results of archaeological research on the Srivijaya Kingdoms have been used as a historical basis for building Indonesia's identity as a maritime and agrarian nation spread across the archipelago. Archaeological research has grown rapidly since the 1800s and focuses on material culture such as artifacts and features without seeing other non-material objects around them. This paradigm produces a limited narrative that only views humans as the solo actors in the process of cultural formation in civilization, while non-material objects such as the natural environment have been marginalized from the process of identity reconstruction. This phenomenon raises the question of what approach can accommodate material and non-material culture used in one whole perspective in reconstructing identity. This article is theoretical with a desk study of Witmore's thoughts. From the review, it is known that the symmetrical archaeology approach places material and non-material culture as equal data in interpreting the past which produces a more in-depth narrative with various perspectives. Symmetrical archaeology is also able to see the role of non-material culture such as the environment, and thought as active actors in the process of cultural formation. Symmetrical archaeology bridges the connectivity between identities in comprehensive narrative.

Abstract

Acne is a prevalent skin disorder, particularly among adolescents, causing significant physical and psychological impacts. Ongoing efforts are dedicated to acne treatment by developing therapeutic agents with enhanced effectiveness while minimizing side effects. In this study, we aimed to assess the potential of Sargassum cristaefolium sourced from Teluk Awur Jepara, extracted using ethanol, in combating acne through its in vitro antibacterial and antioxidant attributes. Utilizing the disk diffusion method, our findings demonstrated the extract's efficacy against Cutibacterium acne, Staphylococcus epidermidis, and Staphylococcus aureus. The minimum inhibitory concentration and minimum bactericidal concentration were determined to be 50 and >100 mg mL-1 against C. acnes, 25 and 100 mg mL-1 against S. epidermidis, and 25 and 100 mg mL-1 against S. aureus, respectively. Additionally, bacteriolytic assays confirmed the extract's ability to partially lyse bacterial cells, particularly S. aureus and S. epidermidis at 2× MIC, while C. acne exhibited partial lysis until the 6th hour, followed by an increase in absorbance, possibly due to cellular debris aggregation or bacterial persistence due to incomplete lysis. Furthermore, the extract exhibited notable free radical scavenging properties against 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS), with IC50 values of 645.35 ± 9.52 and 574 ± 33.52 µg mL-1, respectively. The S. cristaefolium ethanol extract exhibited a total phenol content of 14.17 ± 0.39 mg GAE g-1 and a sulfate content of 10.99 ± 0.45%. Liquid chromatography-mass spectrometry (LC-MS) analysis identified bioactive compounds, including carotenoids, terpenoids, steroids, flavonoids, chromenols, and fatty acids, all of which demonstrate substantial potential as antioxidants and antibacterial agents against acne-causing bacteria. Therefore, ethanolic extract of S. cristaefolium demonstrates potential as an anti-acne treatment due to its antibacterial and antioxidant properties.

Abstract

Ulva lactuca, a widely available green seaweed, has demonstrated promising bioactivity as a tyrosinase inhibitor. This study aimed to evaluate the tyrosinase inhibitory activity of U. lactuca extracts and identify the active compounds responsible. The seaweed was extracted using methanol via maceration, followed by partitioning with hexane, chloroform, and water. Total phenolic and flavonoid contents were determined using UV-VIS spectrophotometry, and tyrosinase inhibition was assessed in vitro. Among the tested fractions, the hexane fraction exhibited the highest tyrosinase inhibitory activity with an IC50 value of 584.34 ± 35.07 µg/mL, classifying it as an active inhibitor. Although this activity was weaker than kojic acid (IC50 = 11.07 ± 0.86 µg/mL), the hexane fraction showed relatively high phenolic (11.66 ± 0.53 mg GAE/g) and flavonoid (9.75 ± 0.30 mg QE/g) contents. Gas Chromatography-Mass Spectrometry (GC-MS) analysis identified several bioactive compounds, notably 2,4-dimethoxycinnamic acid, chalcone, and derivatives such as 2,6-dihydroxybenzoic acid and phloretin. These findings highlight the potential of U. lactuca, particularly its hexane fraction, as a source of natural tyrosinase inhibitors. This opens avenues for its application in the pharmaceutical and cosmetic industries, especially in formulations targeting hyperpigmentation and skin brightening.

Abstract

Cholesterol is a significant contributor to mortality rates worldwide. The quest for effective cholesterol treatments continues, aiming to develop preventive agents with minimal adverse effects. This study seeks to elucidate the anti-cholesterol properties of extracts and components fractionated from water chestnut (Eleocharis dulcis). The research encompassed several phases, including sampling, extraction and fractionation, qualitative and quantitative flavonoid analysis, anti-cholesterol activity assays, and gas chromatography-mass spectrometry (GC-MS) analysis. The quantitative flavonoid assessment revealed that the highest flavonoid concentration was present in the ethyl acetate:methanol (5:5) fraction, totaling 79.224 mg/mL. The water chestnut extract demonstrated notable anti-cholesterol activity with an IC50 value of 13016.27 ± 5648.31 ppm, whereas the water chestnut fraction exhibited enhanced anti-cholesterol activity with an IC50 value of 42.22 ± 9.79 ppm, comparable to simvastatin. The bioactive compounds in water chestnut attributed to its anti-cholesterol effects include 7,9-Di-tert-butyl-1-oxaspiro(4,5)deca-6,9-diene-2,8-dione, n-Hexadecanoic acid, Octadecanoic acid ethyl ester, 9-Octadecenamide (Z)-, 9,12,15-Octadecatrienoic acid, 2,3-dihydroxypropyl ester, 3,4-Dimethoxycinnamic acid, and 2H-1-Benzopyran-2-one, 7-methoxy-. These findings suggest that the fraction derived from E. dulcis exhibits significant anti-cholesterol activity and holds promise for further development in the treatment of hypercholesterolemia.

Abstract

Arthrospira platensis uses phycocyanin, its main pigment, for its antioxidant properties in the food and cosmetic fields. This study aims to determine the effect of biomass conditions on the characteristics (yield, phycocyanin concentration, purity index) and antioxidant activity of A. platensis phycocyanin extracted using dimethyl sulfoxide (DMSO). Wet and dry biomass of A. platensis were extracted using 50% DMSO with a ratio of 1:10 to obtain phycocyanin extract and then purified using ammonium sulfate. Phycocyanin extract and purified phycocyanin were analyzed for yield, concentration, purity index, functional groups, and antioxidant activity (1,1-diphenyl-2-picrylhydrazyl (DPPH) and ferric reducing antioxidant power (FRAP) methods). The results indicated that the conditions of A. platensis biomass (wet and dry) extracted using DMSO affected the characteristics of phycocyanin and antioxidant activity. Phycocyanin from wet biomass has characteristics (yield 145.14±5.59 mg/g, concentration 0.40±0.02 mg/mL, purity index 0.19±0.06) and antioxidant activity (IC50 DPPH 30,628.89±886.81 ppm, FRAP 1,695.57±5.18 μM/g) higher than that from dry biomass (yield 39.50±20.3 mg/g, phycocyanin concentration 0.08±0.04 mg/mL, purity index 0.03±0.02) and antioxidant activity (IC50 DPPH 162,820.59±1,446.30 ppm, FRAP 1,691.01±50.29 μM/g). Fourier transform infrared (FTIR) results show the typical absorption of functional groups found in phycocyanin. The antioxidant activity of phycocyanin from wet biomass using the DPPH method (primary antioxidant) is classified as a weak antioxidant, while based on the FRAP method (secondary antioxidant), the antioxidant is classified as a strong antioxidant.

Abstract

This community service activity was motivated by the declining interest of students in local history due to the dominance of national history in the curriculum and the lack of innovation in history learning. In the digital era, the preservation of local history can be achieved through creative and technology-based approaches that actively involve students. The purpose of this program is to enhance historical awareness, creativity, and digital literacy among students of SMKN 6 Kota Jambi through training in history digitalization by creating creative content. The implementation consisted of preparation, training, digital content production, and monitoring and evaluation stages. The results show that students’ skills improved in integrating local history into digital media such as short videos, podcasts, digital comics, and infographics. The digital products created have the potential to serve as history learning resources as well as promotional media for Jambi’s cultural tourism. Therefore, this activity contributes to strengthening local identity, developing 21st-century competencies, and preserving historical values through digital innovation within the educational environment

Abstract

This community service activity aims to enhance the educational function of the Siginjei Museum in Jambi and to raise young people’s awareness of the historical and symbolic value of the rupiah as a marker of national economic identity. The program, entitled “Cinto Duit Kito” (Our Beloved Money), focuses on promoting love for the rupiah through public education activities based on the museum’s numismatic collection. The program was initiated due to students’ low interest in ancient currency collections and the lack of engaging educational narratives in museums. The implementation consisted of preparation, execution, and evaluation stages. The execution phase included cataloging numismatic collections, developing educational materials integrating history and economics, and conducting interactive learning sessions for student visitors. The results indicate improved understanding and enthusiasm among students regarding the history of Indonesia’s currency, accompanied by the creation of educational worksheets handed over to the museum as sustainable learning media. This program also strengthened collaboration between the University of Jambi and the Siginjei Museum in public education and contributed significantly to the university’s Key Performance Indicators. Thus, “Cinto Duit Kito”serves as a model for synergy between higher education and cultural institutions in developing history and economic education based on local wisdom.
