Abstract

Bali cattle plays an important role in supplying beef in Indonesia because it has good quality, high fertilization and low fat percentage. The aim of this research was to characterize the Bali cattle genetic using molecular marker of microsatellite BM1329 and its relation with calving interval. The data was taken from 19 cattles that belong to UD Sari Laba, Bangli, Bali. The association between BM1329 marker and calving interval was analyzed using General Linear Models concept. The result showed that the duration of calving interval for the Bali cattle was 314 – 451 days. There were 4 alleles found in the locus of microsatellite BM1329. The alleles size varied from 144 to 150 bp, in which, the allele 146 had the highest frequency (71.05%). The observed heterozigosity (Ho) and the expected heterozigosity (He) and the PIC were 0.474, 0.478, and 0.435 respectively. The statistical analysis showed that microsatellite BM1329 has no assocition with the calving interval (P>0.05).

Abstract

The aim of this study was to determine the profile of red blood cells in obese long-tailed macaques. A totalof 16 long-tailed macaques was arrested, and then blood samples were collected aseptically and stored inEDTA vacutainer. Twelve out of 16 were classified obese which is determined by body mass index (BMI).The results showed that the average number of erythrocytes 6.08±0.87 (x1061,2*/mm), the levels ofhemoglobin 11.98±1.59 (g/dl), hematocrit value 37.5±3.55 (%), MCV 62.12±5.35 (fl), MCH 19.91±2.06(pg), and MCHC 32.15±4.22 (g/dl) in average. It can be concluded that the red blood cell profile of theobese long-tailed macaques is still in normal range.

Abstract

Bali cattle has an important role to fulfill the meat demands in Indonesia. The improvement of molecular technology will facilitated bali cattle genomic selection. The aim of this research was to verify association between BMS1282 microsatellite locus and repeat breeding in bali cattle. Totalling 52 bali cattle blood samples was taken from UD Sari Laba, Bangli, Bali as DNA resources. DNA was extracted using blood mini Kit qiagen and microsatellite locus was amplified by Polymerase Chain Reaction (PCR). Alleles were separated with 6% PAGE and visualized using silver stain. There were 5 alleles in BMS1282 locus which the alleles size varied from 148 to 156 bp. Allele 152 was the higest frequency (54.81%) and allele 156 was the lowest frequency (1.92%). The value of heterozygosity (Ho), He, and PIC was 0.346, 0.626, and 0.569. consecutively. Statistical analysis revealed that BMS1282 locus has no assosiation with the repeat breeding (P>0.05). This result indicates that BMS 1282 locus has no important role in regulating the repeat breeding.

Abstract

The observational study was directed to find out the number of populations, large of habitat, home range, and density of long tailed macaques (Macaca fascicularis) in Badung Peninsula. Population site was recorded using GPS program. Data were analyzed using ArcView 3.2 and GmapCatcher programs. There were four population sites of long tailed macaques, namely Pura Dalem Karang Boma, Pura Gunung Payung, Pura Batu Pageh, and Pura Luhur Uluwatu. The average of population density was 114 individuals per hectare and the average of home range was 852,5 hectare. The study indicates that the population densities of long tailed macaques in Badung Peninsula are very high and their home ranges are very large. It is suggested that the population growth of the long tailed macaque in Badung Peninsula must be controlled for decreasing the negative impact.

Abstract

Long tailed macaque (Macaca fascicularis) has distributed throughout the regions in Indonesia, however,they, now, live separately to many local populations within an island. This research aimed to explore thegenetic diversity within and between populations of long tailed macaque in the Region of East Java. Thegenetic diversity was examined by molecular marker of d-loop region mtDNA using a pair of specificprimers. A total of 29 blood samples (15 samples from Alas Purwo population and 14 samples fromBaluran population) was extracted to find out a total DNA. The d-loop region mtDNA was amplifiedthrough PCR tecnique. The PCR products were sequenced using dideoxynucleotide chain terminationmethod at Macogen Inc. Korea. The research found 6 haplotypes, in which 3 haplotypes were found in eachpopulation. Haplotype and nucleotide diversities varied from 0.257 to 0.275 and 0.0012 to 0.0025repectively. Genetic differentiation between populatioon was F=44.1%. Phylogenetic reconstructionshowed that all haplotypes constructed  two different groups (haplogroups)  and there was no sharedhaplotype betwen populations. It can be concluded that both populations (Alas Purwo and Baluran) areuique and have low genetic diversities.

Abstract

Genetic polymorphisms are variations in the genetic structure of populations that represents the base level of biodiversities.  This research aimed to assess the polymorphism of microsatellite locus of D10S1432 in the population of long-tailed macaques in Sangeh, Bali.  A total of 18 blood samples collected as a source of DNA. DNA was extracted using QIAmp® DNA Blood Mini Kit. Microsatellite locus was amplified by polymerase chain reaction technique (PCR) with a total of 30 cycles, and a 57oC of annealing temperature. Furthermore, the alleles were separated by 7% polyacrylamide gel electrophoresis, and emerged with silver staining. The results showed that there were three types of alleles found at D10S1432 locus in the long-tailed macaques population in Sangeh. The alleles size varied from 170 bp to 186 bp. The allele frequencies were  0.28, 0.67, and 0.05 for allele 170, allele 174, and allele 186 consecutively. The heterozygosity of the locus was 0.48. It can be concluded that microsatellites locus D10S1432 is polymorphic in the long-tailed macaques population in Sangeh.

Abstract

Performance test is a preliminary selection to choose Bali cattle stud based on qualitative and quantitative characteristics, including measurement, weighing and observation. Performance test is  applied  on rapid growth of bull  (about 1 year of age) by comparing individual performance with the performance of the group average. The number of bulls that were subjected  the  Performance test  in 2011  was 61  bulls.  Based on Performance test, the best three of the performances  namely the score of 113.30, 109.46,and 104.2  were s selected to be potential stud candidates for next Zuriat test (progeny test).

Abstract

Population growth and aggressiveness are important factors on macaque populations in which their habitats are used as tourist destinations. Controlling those two factors may decrease the negative impact of population both to its surrounding environment and to the internal circumstance of population. This Community Service was packed as a vasectomy and canine cutting services on the adult male of long tailed macaques occupying a tourist destination of Uluwatu, Badung Regency. Vasectomy is a surgical division of all or part of vas deferens to induce sterility. Surgical resection of part of vas deferens used a bilateral slicing method. Additional medical treatments were also performed according to cases encountered in field. During this performance, one selected adult male was vasectomy-surged and four adult males subjected to canine cutting. An operation of removing rubber encircling the neck of a juvenile macaque was done successfully. Moreover, uprooting snare tied the left leg (near tarsal region) of sub adult male was also performed successfully. A week later, Uluwatu manajement reported that all individuals formerly subjected to medical treatments were in good health and their wounds went to be healing completely.

In this program, one selectively adult male was successfully vasectomized and the four canines were also cut successfully. Later inspection showed that there was no any problem with the individual that formerly subjected to medical treatments and even the wounds went to be healing completely.

Abstract

ABSTRACT Growth population control on the animal population dwelling a restricted habitat is very important to decrease, especially the negative impact on its surrounding environment. This Community Service was conducted as an application of vasectomy on an adult male of long tailed macaque occupying the tourist destination of Sangeh. This was the choosen method to overcome the overcrowded population. Vasectomy is surgical division of all or part of the vas deferens to induce sterility. One selected adult male monkey was anesthetized successfully using blowpipe. Surgical resection of part of vas deferens used a bilateral slicing method. Vasectomy was fully done nearly in one hour, and the monkey was released to habitat in the following three days of captivity.
