Abstract

The aim of this study was to determine the prevalence and risk factors Dirofilariaimmitis (D. immitis) infection in dogs slaughtered in Yogyakarta. A total of 151 dogs that were slaughtered from May – November 2013 were examined their heart in order to determine the presence of D. immitis infection. Blood samples were tested using Modified Knott’s Technique for microfilariae examination. The results showed that based on the heart and blood examination the prevalence of D. immitis infection was 14.6 % and 7.9 %, respectively. The risk factors for D. immitis infection were the age and origin of the dog.

Abstract

Heart worm (Dirofilaria immitis) is the causative agent of a serious parasitic disease in dogs.Dirofilariasis is generally diagnosed by microfilariae examination and specific antigen testing. Microfilariaeexamination has low sensitivity due to occult infections. The available antigen test at this time is able todetect circulating antigens secreted by adult female worms only. The aim of the present study was toidentify male (MES) and female (FES) heart worms excretory-secretory antigens which have the potentialas a diagnostic targets. Identification of antigen was done by sodium dodecyl sulphate polyacrylamide gelelectrophoresis (SDS-PAGE) and Western Blotting analysis. The results of this study indicated that therewere differences between the MES and the FES profiles. The results showed 12 bands in MES (14–118kDa) and 18 bands in FES (10–205 kDa). Protein with a molecular weight of 59 kDa has the potential asdiagnostic markers of dirofilariasis.

Abstract

Feline Infectious Peritonitis (FIP) merupakan penyakit fatal yang terjadi pada kucing di seluruh dunia yang disebabkan oleh Feline Coronavirus. Diagnosis antemortem FIP sulit akibat tanda klinis dan laboratoris yang kurang spesifik. Diagnosis yang cepat dan andal sangat penting untuk tujuan prognostik. Tujuan dari penelitian ini adalah mengevaluasi profil biokimia serum dan cairan asites kucing penderita FIP efusif. Penelitian ini menggunakan lima ekor kucing yang diduga kuat menderita FIP efusif. Diagnosis ditegakkan berdasarkan pemeriksaan klinis, uji Rivalta dan rapid test antibodi FIP terhadap sampel cairan asites. Kucing diambil serum darah dan cairan asites untuk pemeriksaan total protein, albumin, globulin, dan rasio albumin/globulin (A/G). Hasil pemeriksaan klinis menunjukkan 5/5 (100%) kucing mengalami asites. Uji Rivalta terhadap sampel cairan asites didapatkan hasil positif eksudat dan rapid test antibodi positif FIP. Profil biokimia serum dan cairan asites menunjukkan rata-rata total protein serum dan cairan asites masing-masing sebesar 7,14 ± 1,36 g/dL dan 6,12 ± 0,33 g/dL (P<0,05); terjadi hipoalbuminemia pada sampel serum dengan rata-rata sebesar 1,94 ± 0,33 g/dL dan albumin cairan asites sebesar 1,69 ± 0,32 g/dL (P<0,05), globulin serum memiliki rata-rata sebesar 5,06±1,32 g/dL dan globulin asites sebesar 4,80 ± 0,88 g/dL (P>0,05). Semua kucing memiliki rata-rata rasio A/G yang rendah, yaitu 0,37 ± 0,36 pada serum dan 0,34 ± 0,01 pada cairan asites (P>0,05). Disimpulkan bahwa profil biokimia serum dan cairan asites pada FIP efusif kucing antara lain total protein serum dan asites normal, hipoalbuminemia, globulin serum normal dan penurunan rasio albumin-globulin. Namun demikian, perbedaan profil biokimia serum dan asites hanya terjadi pada total protein dan albumin, tetapi tidak pada globulin dan rasio A/G.
