Abstract

Iced tea is a popular and refreshing beverage commonly sold by street vendors. However, because the production process utilizes water and relies on the level of hygiene during processing, there remains a risk of bacterial contamination. This study aims to determine the level of bacterial contamination, specifically Coliform and Escherichia coli, in iced tea samples sold in X City, in accordance with food safety standards. This study was conducted using the Most Probable Number (MPN) method with three dilution series. The presence of Coliform was identified using a presumptive test with Lactose Broth (LB) media and a confirmatory test with Brilliant Green Lactose Bile Broth (BGLB) media. The presence of Escherichia coli was identified using Eosin Methylene Blue Agar (EMBA) media and biochemical tests using Triple Sugar Iron Agar (TSIA) media. Samples were taken from 10 different locations spread across 5 subdistricts in X City, with 2 samples taken from each subdistrict. The results showed that there was Coliform bacterial contamination in 10 samples and Escherichia coli bacterial contamination in 6 of the 10 iced tea samples in X City with MPN values ranging from 3-460 MPN/100 mL. Based on the Mann-Whitney test, a p-value of 0.004 (p < 0.05) was obtained, indicating a significant difference in MPN values between Escherichia coli-positive and negative samples. These results indicate that all samples did not meet the microbiological quality requirements based on Peraturan Menteri Kesehatan Republik Indonesia No. 2 of 2023, which stipulates that the maximum allowable levels of total Coliform and Escherichia coli in drinking water is 0 MPN/100 mL.

Abstract

B-cell lymphoma extra-large (Bcl-xL) is an anti-apoptotic protein that is crucial for cancer cell survival and resistance to chemotherapy. Flavonoids have shown potential as anticancer agents through various pathways, including apoptosis. However, molecular interactions of flavonoids with Bcl-xL remain unknown. This study aims to evaluate the binding affinity and stability of myricetin and fisetin as Bcl-xL inhibitors using in silico approaches, including molecular docking and molecular dynamics (MD) simulations. Molecular docking was performed by AutoDock Vina software to evaluate the binding affinity of myricetin and fisetin to the Bcl-xL protein. MD simulations were conducted using the AMBER 2022.1 package to analyze the stability and dynamic behavior of the flavonoids-Bcl-xL complexes over a 100 ns trajectory. Docking analysis revealed strong binding affinities for both flavonoids, with fisetin exhibiting a slightly higher affinity (-7.6 kcal/mol) compared to myricetin (-7.2 kcal/mol). MD simulations confirmed the stability of both complexes, with myricetin forming a more extensive hydrogen bonding due to its additional hydroxyl groups, contributing to lower RMSD fluctuations and higher structural stability. Binding free energy calculations further supported the favorable interaction of myricetin with Bcl-xL (-61.542 kJ/mol), suggesting its potential as a potent inhibitor. In silico analysis indicates that both myricetin and fisetin have promising inhibitory potential against Bcl-xL, with myricetin demonstrating better stability and binding efficiency. These findings provide a basis for further experimental validation and the potential development of flavonoid-based Bcl-xL inhibitors for targeted cancer therapy.

Abstract

The background that underlies this activity is that the cooperation between Indonesia and South Korea in the field of education has opened up opportunities for Indonesian students to continue their studies at several universities in Korea, such as Hankuk University for Foreign Studies and Busan University for Foreign Studies. However, Indonesian students in Korea face challenges in maintaining their health, especially dental and oral health. Language barriers and high treatment costs are the main obstacles in accessing dental health services. Answering existing problems, activities are carried out with methods in the form of community service in the form of health classes and health communication rooms. The targeted goal is to increase the knowledge and awareness of Indonesian students' health in Korea. The course of the activity includes need assessment, preparation of educational materials, implementation of health classes and introduction of teledentistry, as well as evaluation of activities. The results showed that the activity succeeded in increasing students' understanding of the importance of dental and oral care and the use of remote health communication technology. This activity also strengthens the academic network between the University of Muhammadiyah Surakarta and universities in Korea. It is hoped that this activity will become a model of sustainable international service that can be imitated in various other countries.

Abstract

A prevalent issue among the elderly is cognitive deterioration. Modifications in behavior and the adoption of a healthy lifestyle have been demonstrated to mitigate cognitive decline in the elderly. The objective of community service initiatives is to disseminate knowledge regarding cognitive health in the elderly and to offer brain workouts to the community, particularly targeting women who are PKK activists in Mliwis Village. This community service initiative employs an instructional methodology that incorporates PowerPoint presentations and leaflets, utilizing a one-group pre-test and post-test design. The event took place at the Posyandu Sidorejo in Mliwis Village, Boyolali. The target audience for this activity was PKK mothers in Mliwis Village, Boyolali Regency, Central Java. To measure the level of knowledge about cognitive health, a questionnaire developed by the community service team was used. This activity consisted of six stages, namely opening, pretest, education, group brain exercises, discussion, and post-test. 51 participants, comprising PKK mothers, attended this activity. The most extensive age range was 31-40 years old, with 16 (31.4%) participants, and the largest occupation was housewife, with 36 (70.6%) participants. The education and discussion activities went well. The enthusiasm of the participants also increased with the joint brain exercises. The average pretest and posttest scores were 90.26 ± 10.78 and 93.68 ± 7.50, respectively. There was a significant difference in knowledge between the pretest and post-test measurements (p = 0.034; < 0.05). It is hoped that this educational activity will help form independent and happy elderly people.
