Abstract

Eastern little tuna (Euthynnus affinis) is the main catch in WPP572. The caught E. affinis are still in their infancy. This research aims is to know the biological aspects, the fishing aspects and the distribution of the fishing gear per fishing gear, as a consideration for sustainable fisheries management. The research was carried out during the years 2019-2020. Taking samples for gonad observation was carried out by direct survey to the location in February, May, July and October 2019. The measurement of fish length was carried out in stratified random. Catching data were retrieved by requesting secondary data and interviews with related parties. The distribution of fishing grounds is mapped using the QGIS 3.4, the spatial distribution of catches is mapped. This study showed the size structure of E. affinis on purse seine, troll line and encircling gill netfishing gear from 16-55 cm FL, while on the boat/raft lift net it was 21-50 cmFL. These fish growth's is positive allometric on purse seine and encircling gillnet fishing gear, negative allometric on boat/raft lift net fishing gear, and isomeric on troll line. The sex ratio is in the ratio 1: 1.15 and balanced. Based on the maturity level, the gonads were mature in July and October and immature in February and May. Gonads ripened for the first time at a size of 42.032 cmFL. The distribution of the purse seine catchment areas at 3°-6°N and 93°-96°E, and 3°-6°S and 94°-95°E. Boat/raft lift net at 0°-3°S and 95° -101°E. Troll line 4°-6°N and 94°-95 E. Encircling gillnet 0°-2°S and 99°-101°E.

Abstract

Karbon tetraklorida (CCl4) merupakan senyawa kimia yang bersifat toksik, namun masih sering digunakan dalam industri bahan pendingin, alat pemadam kebakaran, dan pestisida. Penggunaan CCl4 secara terus menerus dapat memicu terbentuknya radikal bebas sehingga berpotensi merusak organ hati dan ginjal. Tanaman sirsak (Annona muricata L.) salah satu tanaman yang dapat digunakan sebagai obat alternatif karena mengandung senyawa antioksidan yang dapat mencegah pembentukan radikal bebas. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian ekstrak daun sirsak terhadap gambaran histologi hati dan ginjal mencit (Mus musculus L.) yang diinduksi karbon tetraklorida. Penelitian ini menggunakan Rancangan Acak Lengkap dengan 24 ekor mencit jantan yang dikelompokkan menjadi 4 perlakuan yaitu kontrol negatif (K-) diberi larutan Na-CMC 0,5%, kontrol positif (K+) diinduksi CCl4 0,007 mL/20g BB yang dilarutkan dalam minyak kelapa 0,1 mL, dan dua perlakuan yang diinduksi CCl4 0,007 mL/20g BB serta diberi ekstrak daun sirsak dosis 100 mg/kg BB (P1) dan 200 mg/kg BB (P2). Parameter yang diamati pada histologi hati yaitu degenerasi melemak, degenerasi hidropik, nekrosis, kongesti, serta infiltrasi sel radang. Parameter yang diamati pada histologi ginjal yaitu degenerasi melemak, nekrosis, infiltrasi sel radang, dan pembengkakan glomerulus. Hasil penelitian menunjukkan terjadi penurunan kerusakan yang berbeda nyata (p<0,05) pada P1 dan P2 dibandingkan dengan kontrol positif (K+) pada sayatan histologi hati dan ginjal, sehingga dapat disimpulkan bahwa pemberian ekstrak daun sirsak mampu memperbaiki kerusakan hati dan ginjal setelah diinduksi karbon tetraklorida.

Abstract

The immune system is important in dealing with foreign objects that enter the body. A good body defense system is very important in the existence of the corona virus disease 19 (COVID-19) pandemic caused by the SARS-CoV2. The immune system can be improved by consuming foods with balanced nutrition and various kinds of herbs, in the form of herbal medicine. Jamu is a product derived from medicinal plants that can function as an immune booster. The government has campaigned for the planting of family medicinal plants (TOGA) as an effort to maintain health. In Budaga Village, Klungkung Regency, several families have planted medicinal plants, but the types of are still limited and the processing carried out is still very simple, namely brewing the leaves or plant rhizomes. The purpose of this activity was to introduce cultivation techniques of various kinds of medicinal plants, as well as to train member of PKK of Budaga Village in processing medicinal plants into herbal jamu immune booster. The activity was held on September 1, 2021, attended by representatives of PKK in Budaga Village. The methods used are lectures, demonstrations and direct practice of seeding and planting medicinal plants and practice of preparing jamu. The results of this activity are that the community can cultivate medicinal plants and able to prepare jamu following recipe from Kementerian Kesehatan RI. This activity produced seeds of medicinal plants ginger, red ginger, Piper betle, lemongrass, turmeric, mint and three herbal products, namely jamu gula asam, kunyit asam and temulawak. Abstract

Advances in molecular biology has brought a new branch of multidiscipline study named biotechnology. To comprehend the complex nature of concepts in biology, practical and hands-on approaches are often required. This activity was a collaboration between Biology Teachers Working Group (MGMP Biologi) in Klungkung Regency with university as the facilitator, and aimed to train senior high school biology teachers in compiling learning modules as well as supplying them with the appropriate materials required in simple practical sessions, such as those in animal physiology and reproduction, as well as biotechnology. The event was held successfully, attended by total of 20 biology teachers from six senior high schools (SMA) in Klungkung District, Klungkung Regency, started by introductory lectures and followed by practical sessions on how to perform mice vaginal and sperm smear, human blood smear, and DNA extraction of plant tissues. Abstract

Oocyte developmental competence is one of the determining factors that in?uence the outcomes of an IVF cycle regarding the ability of a female gamete to reach maturation and be fertilized. A non-invasive approach using proteomic analysis of oocyte cumulus cells (CCs) can be used as a method to predict oocyte competence and viability that could potentially function as molecular predictors for IVF program prognosis. Our study was aimed at quantifying protein markers (HAS2, PTX3, BCL2 and BAX) that are expressed in human CCs between mature (MII) and immature (MI) oocytes. This research is an analytic observational study. Twelve samples of CCs from mature (MII) and immature (MI) oocytes were collected after ovum pick up from patients undergoing ICSI. CCs samples were isolated using an ultrasonicator and protein expressions were quantified using ELISA method. The Mann-Whitney test was used to compare the protein expressions from CCs between mature and immature oocytes. The results showed HAS2, PTX3 and BCL2 proteins expressions was increase in CCs from mature oocytes but had no significant effect compared to CCs from immature oocytes, however, BAX protein expression was significantly higher (P<0.05) in CCs from immature oocytes which CCs from mature oocytes. This present study shows that BAX proteins could be used as markers for oocyte quality and maturation. Key words: ELISA, maturation, proteins marker, cumulus cells (CCs)

Abstract

Wilayah Pengelolaan Perikanan Republik Indonesia di Samudera Hindia dengan kode WPP-RI 572 dan 573 kaya akan potensi sumber daya ikan yang didominasi oleh ikan Tongkol Komo (Euthynnus affinis (Cantor, 1849). Pemanfaatan ikan Tongkol Komo telah melampaui batas optimal dengan tekanan penangkapan yang tinggi. Biologi reproduksi Tongkol Komo perlu diketahui untuk memastikan keberadaannya di alam. Tujuan dari penelitian ini adalah Tahap Kematangan Gonad (TKG) dan pemijahan. Penelitian dilaksanakan pada bulan Februari, April, Juni hingga Desember 2020. Pengumpulan data dilakukan melalui pengamatan langsung di Pangkalan Pendaratan Ikan (PPI) Kedonganan. Teknik pengambilan sampel yang digunakan dalam penelitian ini adalah purposive stratified sampling. Analisis histologi di Laboratorium Penelitian Perikanan Tuna, Denpasar. Hasil yang diperoleh bahwa pola pertumbuhan tuna komo didominasi oleh pola alometrik positif dengan sebaran panjang 20-65cmFL. Ukuran pertama kali matang gonad (Lm) untuk betina 44,07 cmFL. Tingkat Kematangan Gonad (TKG) pada klasifikasi makroskopis dan mikroskopis perkembangan gonad didominasi oleh ikan yang belum matang gonad. Musim pemijahan pada bulan Agustus dan Desember.

Abstract

Phyllanthus niruri L. is a medicinal plant recognized for its wide range of therapeutic benefits, particularly its anti-inflammatory properties. This research focused on assessing the impact of P. niruri leaf extract on pro-inflammatory cytokines in vitro using RAW 264.7 macrophage cells, analyze its phytochemical composition, and validate its potential anti-inflammatory mechanisms through molecular docking studies. The extract was analyzed for its phytochemical composition using LC-MS, revealing the presence of sesquiterpene glycosides, flavonoid glycosides, and tannins among the identified compounds. Cell viability tests showed that the extract was not toxic to the cells at concentrations up to 50 µg/ml. Treatment with the extract significantly reduced the levels of pro-inflammatory cytokines TNF-α and IL-1β in LPS-induced RAW 264.7 cells, with the most effective concentration being 50 µg/ml. Molecular docking studies further supported the anti-inflammatory potential of the extract by demonstrating its ability to inhibit COX-2 and TNF-alpha. These findings suggest that P. niruri leaf extract has promising anti-inflammatory properties and needs further investigation as a potential treatment for inflammatory diseases. The in silico study provided insights into the molecular interactions between the extract's compounds and inflammatory targets, supporting its anti-inflammatory mechanism of action.

Abstract

Rokok elektrik dikembangkan untuk mengurangi kecanduan masyarakat terhadap rokok. Rokok elektrik mengandung bahan kimia yang bersifat karsinogen. Tanaman telang (Clitoria ternatea) memiliki aktivitas antioksidan. Penelitian ini bertujuan untuk mengetahui aktivitas antioksidan infusa bunga telang terhadap penyempitan diameter lumen trakea dan bronkus tikus diterpapar rokok elektrik. Penelitian ini menggunakan 30 ekor tikus jantan (Rattus novergicus) dengan umur sekitar 2-3 bulan dan berat badan sekitar 150-200 g. Penelitian ini bersifat eksperimental dengan Rancangan Acak Lengkap (RAL) yang dibedakan menjadi 5 kelompok, yaitu K+ (0,6 mL liquid vape), K- (normal), (0,6 ml liquid vapor + 9 mg/200 g BB infusa bunga telang), P2 (0,6 ml liquid vapor + 18 mg/200 g BB infusa bunga telang) dan P3 (0,6 ml liquid vapor + 36 mg/200 g BB infusa bunga telang). Perlakuan diberikan selama 35 hari. Hasil penelitian dianalisis dengan uji Oneway ANOVA dan dilanjutkan dengan uji Duncan. Hasil yang diperoleh menunjukkan bahwa infusa bunga telang terbukti memiliki aktivitas antioksidan dalam mencegah terjadinya penyempitan diameter lumen trakea dan bronkus, dibuktikan dengan hasil analisis statistik yang menunjukkan adanya perbedaan yang nyata (P<0,05). Infusa bunga telang dengan dosis 36/200 mg memberikan pengaruh yang optimal dalam mencegah penyempitan diameter lumen trakea dan bronkus tikus setelah diberi paparan asap rokok elektrik. E-cigarettes were developed to reduce people's addiction to cigarettes. E-cigarettes contain chemicals that are carcinogens. The telang plant (Clitoria ternatea) has antioxidant activity. This study aims to determine the antioxidant activity of telang flower infusa against the narrowing of the diameter of the lumen of the trachea and bronchi of rats exposed to e-cigarettes. This study used 30 male rats (Rattus novergicus) with an age of about 2-3 months and a body weight of about 150-200 g. This research was experimental with a Completely Randomized Design (CRD) which divided into five treatment groups, namely K+ (0.6 mL liquid vape), K- (normal), (0.6 ml liquid vapor + 9 mg/200 g BB infuse butterfly pea flower), P2 (0.6 ml liquid vapor + 18 mg/200 g BB of butterfly pea flower infusion) and P3 (0.6 ml liquid vapor + 36 mg/200 g BB of butterfly pea flower infusion). The treatment is given for 35 days. The result of the study was analyzed with Oneway ANOVA test and continued with Duncan test. The results showed that the lumen diameter in the K+ group was smaller than in the other groups. Butterfly flower infusion was proven to have antioxidant activity in preventing narrowing of the lumen diameter of the trachea and bronchi, as evidenced by the results of statistical analysis which showed a significant difference (P<0.05). Telang flower infusion given at a dose of 36/200 mg provided an optimal effect in preventing the narrowing of the lumen diameter of the trachea and bronchi of rats after being exposed to e-cigarette smoke.

Abstract

Highlights: This original research report on the differences in disease severity and blood test results between unvaccinated and vaccinated COVID-19 patients is quite novel, as the COVID-19 pandemic was still happening when this study was carried out. This original research report offers information on the advantages of the COVID-19 vaccines and provides data that the public may use as scientific evidence to counter hoaxes. Abstract The COVID-19 pandemic caused by SARS-CoV-2 is an unprecedented event in human history. Vaccines are a safe, long-term solution for addressing the COVID-19 pandemic. This study aimed to investigate the differences in disease severity and blood test results between unvaccinated and vaccinated COVID-19 patients. This study used an analytical observational method with purposive sampling. A total of 90 COVID-19 patients at Bhayangkara Hospital, Denpasar, Indonesia, were divided into three groups: unvaccinated group (V0), two-dose vaccinated group (Vp), and three-dose vaccinated group (Vb). Primary data were collected from July to December 2022, while secondary data were collected from January 2021 to June 2022. The data were analyzed using the Kruskal-Wallis test followed by the Mann-Whitney test, as well as one-way ANOVA test followed by Tukey's honestly significant difference (HSD) test with a confidence interval (CI) of 95% and α of 5%. The results revealed significant differences in disease severity (p<0.001). V0 had a higher percentage of severe (36.7%) and critical (6.7%) symptoms than Vp (severe=10.0%; critical, n=0) and Vb (severe and critical, n=0). The follow-up tests revealed significant differences in disease severity between V0 and Vp (p<0.001), V0 and Vb (p<0.001), as well as Vp and Vb (p=0.001). Blood test results revealed significant differences in lymphocytes (p=0.005), monocytes (p<0.001), monocyte-to-lymphocyte ratio (MLR) (p<0.001), and eosinophils (p=0.037). The follow-up tests revealed significant differences in these four indicators between V0 and Vb, in all parameters except for lymphocytes between V0 and Vp, and in lymphocytes only between Vp vs Vb. In conclusion, unvaccinated patients had a higher percentage of severe and critical symptoms than vaccinated patients. The blood test results revealed significant differences in lymphocytes, monocytes, MLR, and eosinophils. Unvaccinated patients had lower lymphocyte counts, higher MLR levels, and higher monocyte counts than vaccinated patients.

Abstract

Highlights: • This original study examined the antioxidant compounds derived from naturally sourced Hibiscus sabdariffa calyx. • The findings demonstrated that roselle tea offers a viable and cost-effective solution for repairing histological damage to the digestive organs induced by monosodium glutamate. Abstract Monosodium glutamate (MSG) is a commonly used synthetic additive for enhancing food flavor. Excessive use of MSG can cause cytotoxic effects, which disrupt the balance of oxidative stress and free radicals in the body, particularly in the human digestive system. Roselle (Hibiscus sabdariffa) is a plant with red petals renowned for its abundance of beneficial compounds, including polyphenols, flavonoids, anthocyanins, and other antioxidants that function as free radical antidotes. This study aimed to investigate the effect of administering roselle tea and determine the optimal dose for restoring the digestive organs of MSG-induced mice (Mus musculus). The research employed a completely randomized design with a random sampling method. A total of 25 mice were divided into five groups: a negative control group (K-) that received 0.3 mL of distilled water, a positive control group (K+) given 4 mg/g bw of MSG, and three treatment groups (P1, P2, and P3) administered with 4 mg/g bw of MSG along with varying doses of roselle tea (2.6 mg/g bw, 3.9 mg/g bw, and 5.2 mg/g bw, respectively). The treatment was orally administered via gavage for 30 days. The stomach, duodenum, and liver underwent histopathological examination using the paraffin method and hematoxylin-eosin staining. The observed parameters in the stomach and duodenum included necrosis, inflammatory cell infiltration, villous erosion, and epithelial desquamation. Meanwhile, the parameters examined in the hepatic organs were necrosis, inflammatory cell infiltration, and cell degeneration. The Kolmogorov-Smirnov normality test and one-way analysis of variance (ANOVA) were employed to assess the normal distribution and homogeneity of the data. If the data exhibited a normal distribution, Duncan's post-hoc test was conducted. The results revealed significant differences among the groups (p<0.05). In conclusion, the administration of roselle tea effectively recovered the histological damage in the stomach, duodenum, and liver of MSG-induced mice.
