Abstract

This research aims to isolate and characterize the fcin enzyme from Ficus septica stem latex. Ficin from Ficus septica stem latex was isolated using column chromatography. Then enzyme activity was tested at different temperature (40oC, 50oC, 60oC, 70oC) and pH (6.0, 7.0, 8.0) levels. Ficin enzyme activity of joint treatment with variations in temperature and pH was analyzed using two-way ANOVA with a factorial pattern followed by Least Signifcant Difference (LSD) test. The results showed that temperature treatment signifcantly affects enzyme activity. However, the treatment of pH and the interaction between temperature and pH did not signifcantly affect the fcin enzyme activity. There was no signifcant difference in fcin activity at the incubation temperatures of 40oC and 50oC, as well as 60oC and 70oC. However, comparing the incubation temperatures of 40oC and 50oC with treatment 60°C and 70°C showed a signifcant difference in fcin enzyme activity. In the treatment of incubation at pH 6, 7 and 8 for fcin enzyme activity showed no signifcant difference. We concluded that the Ficus septica plant latex contained fcin enzyme with an optimum temperature of 60°C and optimum pH of 6, 7, and 8.

Abstract

Diabetes mellitus (DM) merupakan penyakit metabolik dengan karakteristik hiperglikemia. Pada DM  mudah  terjadi pembentukan  radikal  bebas. Antioksidan dari flavonoid diketahui dapat mengatasi penyakit disebabkan radikal bebas, seperti DM. Beberapa penelitian melaporkan ekstrak daun sirih merah memiliki aktivitas antioksidan dan antihiperglikemik. Belum ada data yang menunjukkan potensi tersebut secara in vivo.  Penelitian ini bertujuan menganalisis pengaruh ekstrak daun sirih merah terhadap kadar glukosa darah dan kadar antioksidan glutation peroksidase (GPx) tikus jantan hiperglikemik. Pada penelitian ini terdapat lima kelompok perlakuan yang terdiri dari kontrol positif (K+), kontrol negatif (K-), perlakuan ekstrak daun sirih merah dosis 100, 200, dan 400 mg/kgBB. Hewan coba yang digunakan adalah tikus jantan strain Wistar berumur 8-12 minggu dengan berat badan minimal 180 gram. Prosedur penelitian meliputi pengkondisisan hiperglikemik pada hewan uji dengan induksi aloksan 120 mg/kgBB secara intraperitoneal,  ekstraksi daun sirih merah menggunakan metode maserasi dengan pelarut etanol 70%, perlakuan dengan pemberian ekstrak daun sirih merah secara oral selama 28 hari, serta pengukuran kadar glukosa darah dan kadar GPx. Hasil penelitian menunjukkan pemberian ekstrak daun sirih merah berpengaruh signifikan pada penurunan kadar glukosa darah dan kenaikan kadar GPx. Pengaruh yang diberikan daun sirih merah terhadap penurunan kadar glukosa darah sebesar 37,5%, sedangkan pengaruh yang diberikan daun sirih merah terhadap kenaikan kadar GPx sebesar 92,9%. Simpulan dari penelitian ini adalah pemberian ekstrak daun sirih merah pada tikus jantan hiperglikemik berpengaruh terhadap penurunan kadar glukosa darah dan peningkatan kadar GPx. Dosis 100 mg/kgBB merupakan dosis efektif dalam menurunkan kadar glukosa darah dan menaikkan kadar GPx.

Abstract

Tempe known as a traditional fermented food originated from Indonesia. It has a unique flavour and texture. It also contains high protein and usually serves to substitute meat, fish, or egg as a complement to rice. The manufacture process of Tempe is quite complex and mostly, the traditional process has not employed the hygienic standard. In the process of Tempe making, there are two critical stages of the whole process; i.e. soaking of soybeans and solid state fermentation by Rhizopus sp. During the process, foodborne pathogen bacteria such as Escherichia coli could contaminate the product of Tempe. The bacterial contamination could be revealed through culture dependent methods which is costly, laborious, and time consuming. Therefore, the culture-independent method such as polymerase chain reaction using a specific primer could be applied to detect target microorganism to save time and labour. In this study, thirty-one Tempe samples collected from different manufacturers in Semarang, Central Java, Indonesia were analysed by PCR. In order to obtain the bacterial genomic DNA, a modified Chelex 100-Microwave method was employed. The results of DNA extraction showed that the method was an applicable method. It gave high quantity and quality of DNA; therefore, it could be applied in the PCR reaction. The DNA samples were employed in PCR for detection of Escherichia coli using Ecoli706F/R. It was found that 27 out of 31 samples were detected having Escherichia coli contamination showed by the presence of the amplified product size 706 bp. The application of this method could significantly reduce costs and time of analysis in the laboratory. Further response after E. coli were detected could be employed, including investigation of the critical factors in Tempe manufacturing process which allowed E. coli contamination.

Abstract

During the pandemic, healthy life behavior is needed in an attempt to reduce the transmission risk.  One of them could be done by washing hands. Various human activities may not allow them to wash their hands frequently, for example, using hand sanitizer to wash hands. Hand sanitizer or Isopropyl based alcohol are recommended materials by WHO to clean bacteria on hands. Storing hand sanitizer for a longer time leads to inaccurate quality of the hand sanitizer to the written quality on the package. A simple alcohol meter is designed to test the liquid percentage of hand sanitizer or Isopropyl Alcohol (IPA) based alcohol. The required materials are easy to get from the surrounding environment. Thus, this device could be established at home without ignoring the health protocol. Although it has strength IPA-based alcohol or hand sanitizer is also risky if it is incorrectly used

Abstract

The importance of water management for milkfish cultivation determines the yields. This research aims to find the performance specification and pH, TDS, and Salinometer measurement (PTS) design validity and relevance. These tools are combinations from various sensors connected by Arduino nano 328 system, pH meter analogue, turbidity, and salinity measurement sensors complemented by Liquid Crystal Display and battery. They are connected by data cable. The findings showed that 1) the system’s performance specification was applying copper electrodes as the salinity, turbidity, and pH meter sensors. The Arduino system did the readings of the data. 2) the device-design specification was valid and relevant to measure the pond water quality for cultivating milkfish (Chanos chanos Forsk).

Abstract

The liquid organic fertilizer (LOF) formula from peanut and banana peels. This research aims to: (1). Analyze the effect of LOF provision from peanut and banana peels on the vegetative growth of spinach; (2). Analyze the best LOF dose for vegetative growth of spinach, and (3). Analyze the effect differences between NPK fertilizer and LOF from peanut and banana peels toward the vegetative growth of spinach. The researchers promoted this study from November to December 2020 in a greenhouse in Banjarbaru. The researchers applied RAL design by providing fertilizer since the spinaches were 10, 17, and 24 days. The vegetative growth parameters were: Plant height, number of leaves, leaf width, and the leave color. The result proved that: (1). LOF provisions from peanut and banana peels significantly influenced the height and leave numbers of spinach; (2). The best dose for LOF provision is 50ml of peanut peel LOF + 50ml of banana peel LOF to get excellent height and leave numbers of the spinach; (3). The growth of the spinach, in terms of height, numbers of leaves, and leaf width during NPK fertilizer provision, was in line with the provisions of banana and peanut peel LOF.

Abstract

Background: A laboratory is a learning tool for conducting experiments or observations. Knowledge of workplace safety procedures and laboratory tools and materials can simplify and streamline the learning process. This study aims to analyze the knowledge of high school students on laboratory work safety material. Methods: This study is a quantitative study, namely research that describes students' knowledge of laboratory work safety. The research subjects were students of class X MIA 1 and MIA 2 SMAN 7 Jambi City, class X students of SMAS Insan Madani Jambi. Primary data on students' abilities about work safety in the laboratory were measured by a multiple-choice test consisting of 4 alternative choices, namely A, B, C, and D. Secondary data were taken by using an interview sheet for class X biology. Data were analyzed according to Miles and Huberman, namely data reduction, display data, and conclusions. Results: High school students' knowledge of work safety materials in the laboratory is still low, namely 61.86%. Students' knowledge of how to use the tools and their functions showed the lowest percentage, namely 55.36% (poor category), while the safety aspect of laboratory work showed the highest percentage, namely 73.71% (sufficient category). Conclusion: High school students have low knowledge in understanding work safety material in the laboratory. It is necessary to deepen the material so that students' knowledge of laboratory work safety increases.

Abstract

This research was conducted by making a rectangular slit using a razor blade as a narrow gap in the Fraunhofer diffraction experiment. The intensity distribution measurement on the resulting diffraction pattern uses a voltage divider circuit. This circuit takes advantage of the nature of the LDR, which changes resistance when exposed to light. Experiments show that a diffraction pattern screen when a 560 nm coherent light passes through a narrow rectangular slit made of razor cut. We measured the narrow gap using a tracker application with the resulting gap size (0.3564 x 0.4677) mm. The ratio of the intensity distribution of the x-axis bright pattern on the Fraunhofer diffraction by a rectangular slit with slit size height x width = (0.4677 x 0.3564) mm from b = -5p to 5p (maximum 4, maximum 3, maximum 2, maximum 1, central maximum, maximum 1, maximum 2, maximum 3, maximum 4) is 0.000873; 0.000763; 0.005395; 0.020583; 1; 0.039658; 0.008088; 0.002554; 0.001218. The ratio of the intensity distribution of the y-axis bright pattern on the Fraunhofer diffraction by a rectangular slit with slit size height x width = (0.4677 x 0.3564) mm from g = -5p  to 5p (maximum 4, maximum 3, maximum 2, maximum 1, central maximum, maximum 1, maximum 2, maximum 3, maximum 4) is 0.001890; 0.001469; 0.002447; 0.040516; 1; 0.037141; 0.006482; 0.001690; 0.000440. This study indicates that diffraction experiments and the measurement of the diffraction pattern’s intensity can be carried out with simple materials and equipment and can be used in the correct experiment.

Abstract

Research has been carried out with a device to observe the phenomenon of Rayleigh Benard convection, made of a glass box bounded by a lower plate and an upper plate. The bottom plate is hotter than the top plate which functions to heat the horizontal fluid layer from below. This study aims to observe the phenomenon of water molecule movement. The observation medium was water mixed with teak sawdust as a representation of water particles with an average density of mean=0.99 g/cm3. The variation of water thickness (d) used 5 cm and 6 cm with variations in temperature difference (DT) with an average increase of 50°C. Data was collected with a cellphone camera and run with Windows Movie Maker software which has a time accuracy of up to 0.001 seconds. It was observed that an increase in the temperature difference (DT) between the two plates resulted in an increase in the velocity of motion of water particles in convection which is depicted by a v-DT diagram. Due to the difference in temperature (DT) also results in the movement patterns of water molecules, namely laminar and turbulent. The turbulent phase with the Rayleigh number value R~107, indicated that the flow of water particles is getting faster, the direction of the flow of the particles starts to become irregular and sometimes signs of a water vortex appear. At different fluid immersion results in the start of a turbulent phase. The turbulent phase for d=5cm is observed at DT~30°C, and d=6cm at DT~20°C. Furthermore, this RBC phenomenon is used as a representation of learning natural phenomena in air fluids, namely whirlwinds as an event due to differences in temperature and pressure of a fluid

Abstract

Experimental activities are part of physics learning process with the aim of gaining knowledge and having good understanding of physics concepts. Sound and organ pipes are the main subjects discussed in physics and engineering lessons. These materials contain facts accompanied by actualization in everyday life in social life. However, the fact in high school (SMA) physics learning is that experimental activities in supporting students’ learning and understanding have not been widely carried out. This is due to the lack of practical support tools. The purposes of this research are to design a sound experimentation tool design of an open and closed organ pipes and to determine the value of the speed of sound which propagates through air medium using a data acquisition technique assisted by sound analysis oscilloscope software. This type of research is a Research and Development research with five stages referring to the ADDIE model. The first stage was analysis, that was done by conducting a study of the problems of physics learning at senior high school level, especially in sound chapter. The second stage was design, namely designing experimentation tools. The third stage was development, that was done by providing materials that were used in the experiment. The fourth stage was implementation, that was testing the tool which had been made combined with sound analysis oscilloscope software, and the fifth was conducting an evaluation and feasibility test based on the validity of the experimental data. The data analysis technique used average and linear regression by utilizing the results of data from frequency analysis using sound analysis oscilloscope software. The results showed that the experimentation tool in this study was able to be used as learning media for the sound and open and closed organ pipes materials. The experimental results obtained that the speed of the sound waves propagation in air for an open organ pipe was v = 332.19 m/s with an accuracy rate of 97.70%. While the value of the speed of the sound waves in air for a closed organ pipe was v = 334.69 m/s with an accuracy rate of 98.44%.
