Abstract

ABSTRACTThe Congaturi halfbeak (Hyporhampus limbatus), also known as the Valenciennes halfbeak, is an important economic fish in the fisheries sector of Indonesia, with a high level of diversity. The high diversity creates difficulties in the identification process for each species. The identification of species can be based on morphological and molecular methods. Therefore, this study aimed to compare the results of the morphological and molecular identification methods using a sample of H. limbatus. Morphological identification was carried out using morphometric techniques and for molecular identification following DNA Barcoding protocol. The results showed both morphological and molecular methods indicate that the sample was identified as Hyporhampus limbatus. The morphology of the Hyporhampus limbatus has an elongated body shape with a silvery color filled with scales all over his body except on the head. The lower jaw is taper in shape longer than the upper jaw and has an immarginate tail. While for the DNA Barcoding results show high similiraty (97%) and percentage of query coverage (98%) to identify Hyporhampus limbatus. Tingginya keragaman menyebabkan kesulitan dalam proses identifikasi dari setiap spesies. Metode yang dapat digunakan untuk identifikasi spesies adalah metode morfologi dan molekuler. Penelitian ini bertujuan untuk membandingkan hasil dari metode identifikasi morfologi dan molekuler dengan menggunakan sampel ikan julung-julung. Identifikasi morfologi dilakukan menggunakan teknik morfometrik sedangkan untuk molekuler diaplikasikan dengan DNA Barcoding. Hasil identifikasi dengan metode morfologi dan molekuler menunjukkan hasil yang sama yaitu Hyporhampus limbatus. Berdasarkan hasil identifikasi morfologi Hyporhampus limbatus memiliki bentuk tubuh yang memanjang dengan warna perak dengan sisik di seluruh tubuhnya kecuali pada bagian kepala. Rahang bawahnya berbentuk lancip dengan ukuran lebih panjang dibandingkan dengan rahang atas serta memiliki ekor imarginate. Hasil molekuler (DNA Barcoding) menunjukkan bahwa Hyporhampus limbatus  memiliki nilai yang tinggi untuk tingkat kemiripan (97%) dan Query Cover (98%). Abstract

ABSTRACTSardines are an important commodity in import activities. Excessive imports and lack of catch data are feared to reduce fish stocks. To complete the fish catch data required knowledge of body type and size. This study aims to identify and confirm commercial sardines species landed at Muara Angke Market, Jakarta using morphological, morphometric, and DNA barcoding methods targeting the Mitochondrial Cytochrome c oxidase-I (COI) gene. The results of morphological and morphometric analysis showed that the samples are the genus Dussumieria originating from the Dussumieriidae family, the Order Clupeidae, having silver-colored body characteristics with reflections resembling rainbows, slender bodies and rounded stomachs with a maximum length of up to 14.4 cm, then less abdominal fins advance from the midsection. Based on the relationship of body weight length, this fish is negative allometric. Molecular DNA barcoding analysis using COI, obtained by type of Dussumieria elopsoides fish with local name of rainbow sardines, similarity level with Query Cover percentage was 94% and Per percentage. Ident of 91.64%. DNA barcoding analysis has corroborated morphological and morphometric observations by obtaining fish samples identified as Dussumieria elopsoides.Keywords: Morphology, Morphometrics, DNA Barcoding, Dussumiera elopsoides.ABSTRAKIkan sardin merupakan komoditas penting dalam kegiatan impor. Impor berlebih serta kurangnya data penangkapan dikhawatirkan dapat menurunkan stok ikan. Untuk melengkapi data tangkapan ikan diperlukan pengetahuan mengenai jenis dan ukuran tubuhnya. Penelitian ini bertujuan untuk mengidentifikasi dan memastikan jenis ikan sardin komersial yang didaratkan di Pasar Muara Angke, Jakarta dengan menggunakan studi morfologi, morfometrik, dan metode DNA barcoding dengan target gen Mitokondrial Cytochrome c oxidase-I (COI). Hasil analisa morfologi dan morfometrik menunjukkan bahwa sampel merupakan genus Dussumieria berasal dari famili Dussumieriidae, Ordo Clupeidae, memiliki karakteristik tubuh yang berwarna silver dengan pantulan menyerupai pelangi, tubuh ramping dan perut membulat dengan panjang maksimal dapat mencapai 14,4 cm, kemudian sirip bagian perut kurang maju dari bagian tengah tubuhnya. Berdasarkan hubungan panjang berat badan, ikan ini bersifat alometrik negatif. Analisis DNA barcoding secara molekuler menggunakan COI, didapatkan ikan berjenis Dussumieria elopsoides dengan nama lokal ikan sardin pelangi, tingkat kemiripan dengan presentase Query Cover sebesar 94% dan presentase Per. Ident sebesar 91,64%. Analisis DNA Barcoding telah menguatkan pengamatan morfologi dan morfometrik dengan didapatkan sampel ikan yang teridentifikasi berjenis Dussumieria elopsoides.Kata Kunci: Morfologi, Morfometrik, DNA Barcoding, Dussumiera elopsoides.

Abstract

ABSTRACTCoral fish is one of the biota that live in coral reef ecosystems and its life is very dependent on thecondition of coral reefs. Pterocaesio chrysozona is a species of reef fish from the family Caesionidae that live in groups around coral reefs between 0-25 meters deep. Morphogenetic, morphological and DNA barcoding based measurements have advantages in identifying species that have been proven successful in identifying reef fish species. Morphometric and morphological analysis results showed that the overall specimens identified were Pterocasio chrysozona fish. It is directly proportional to the DNA Barcoding analysis which is read at the level of Pterocasio chrysozona fish species with a percentage of 93% Query Cover and Per. Pterocaesio chrysozona merupakan spesies ikan karang dari Famili Caesionidae yang hidup bergerombol di sekitar terumbu karang kedalaman antara 0-25 meter. Pengukuran yang berbasis morfogenetik, morfologi dan DNA Barcoding memiliki kelebihan dalam melakukan identifikasi spesies yang terbukti berhasil dalam melakukan identifikasi spesies ikan karang. Hasil analisis morfometrik dan morfologi menunjukkan secara keseluruhan spesimen yang teridentifikasi adalah ikan Pterocasio chrysozona. Berbanding lurus dengan analisis DNA Barcoding yang terbaca pada tingkat spesies ikan Pterocasio chrysozona dengan persentase Query Cover 93% dan Per. Abstract

Biorock® technique is the earliest methods to rehabilitate the damage of coral reef ecosystem. Its improvement is need to carry on since it is subject to some inhibitions in particular the dependence of expensively-imported Titanium (Ti) as the anode materials. The main purpose of this research was to find the best anode material as a possible subtitution which can be economically and easily to apply in Indonesia. Therefore we compared Titanium (Ti) with two other potential anodes material (Magnesium-Mg, and Aluminum (Al). The laboratory study was carried out for two days period (48 hours) in the stagnant sea water aquaria. Four aquarium tanks were treated by different electric current treatment (1 Ampere, 2 Ampere, 3 Ampere, and 5 Ampere, respectively). The reduction of electrode weigh (anode and cathode) was measured. During the experiment, water quality (i.e. pH, and salinity) and mineral waters (calcium) were collected every 6 hour in 48 hours. The solid form of calcium carbonate was analyzed using XDS (X-ray Dispersion Spectrophotometer). The accretion rates on cathode and anode decay were compared between anodes which were treated by different electrical current. Based on the study, three anodes (aluminum, magnesium and titanium) showed different respond. Titanium and Magnesium anode showed electric current affected accretion rate (P < 1), while for aluminum anode showed no differences on treatments.Keywords: mineral accretion, Biorock®, anode, coral reef rehabilitation, accretion rate, anode decay rate, mineral uptake

Abstract

Coral reef ecosystem management in Siantan Tengah District, Anambas Islands need to be improved to obtain the optimal results. Currently, the coral reef ecosystem management is conducted sectorally, therefore, it can cause a damage on coral reef ecosystem. The purposes of this study were to analyze sustainability status of coral reefs managements in the District Central Siantan and to  formulate the sustainability of coral reef ecosystem management. Multi Dimensional Scaling (MDS) with Rap-Insus COREMAG approach was used to analyze the sustainability status of coral reef management. The results showed that the sustainability status of coral reef management in Siantan Tengah District was sustainable with multidimensional index of 51.457. Dimensions that need to be improved to achieve the optimal management of coral reef ecosystems in a sustainable manner is social dimension to the value of sustainability by 42.324 and institutional dimensions of 49.85 which is classed as less sustainable. This results of this research are expected to be able to facilitate the stakeholders to arrange the sustainability of coral reef ecosystem management in the Siantan Tengah District. Abstract

Local ecological knowledge (LEK) refers to the knowledge of people in a local community that can be used in assessing environmental conditions as well as reflecting the interaction that occur in an ecosystem. The purpose of this study was to examine condition of coral reef ecosystem based on the LEK’s community in a marine protected area east coast weh island (KKPD PTPW) Sabang. Assessment of the coral reef condition based on the local’s ecological knowledge is justified from the suitability of the area by comparing map of the coral reefs condition based on LEK and observation map of the coral reefs condition during the study. Based on LEK, it is known that in the PTPW Sabang region coral reefs which falls under deteriorated class is found in the northern sea of Kelurahan Kota Atas, while those which falls under good condition class are found in Ie Meule and Ujung Kareng as well as in Anoi Itam except Benteng. Result from the observations done in 13 stations found that sea region near Sumur Tiga, Ujung Kareung and Benteng have a good coral reefs with a coverage range of 51-65 %. While the sea in Ujung Seuke, Anoi Itam and Reuteuk have a moderate coral reefs with a percentage of coverage ranging from 44- 49. Respondents’ perceptions of the coral reef condition during the last 10 years which include 55% respondents in Ie Meule and 74.29% of respondend in Anoi Itam stated that there is an increment of coral reefs coverage.

Abstract

Acceleration of the process of recovery of damaged reefs require rehabilitation, management and supervision of well-planned and sustainable. One of the rehabilitation process is to create artificial reef. This study aims to look at the rate of attachment of sessile organisms on cement media called “crypto”. A total of 270 cement media at each shelf. The size of cement media is 5x5x1 cm. The cement media were deployed at a depth of 6 meters and 10 meters. The rate of attachment of sessile organisms was compared between the upper and lower of the media side at each depth and between depths. Different organisms was observed colonized concrete media such as shells, sponges, worms, green algae, red algae, and brown algae. Percentage of coverage of sessile biota at a depth of 6 meters was significantly higher than 10 meters. Percentage of coverage of sponges at a depth of 6 meters and 10 meters, showed no significantly different (P = 0.0670). The other sessile biota, value of the test results showed significant differences between depths, such as the brown algae (P< 0.0001), red algae (P = 0.0003), and  green algae (P = 0.0022). Abstract

Makrozoobentos merupakan salah satu organisme yang keberadaannya banyak terdapat di ekosistem lamun. Tujuan penelitian untuk menganalisis perbedaan kelimpahan individu dan spesies makrozoobentos di bulan purnama dan perbani, melihat korelasi kepadatan makrozoobentos dengan penutupan lamun, dan mengetahui kontribusi spesies makrozoobentos pada habitat lamun. Penelitian ini dilakukan di pulau panggang dengan 3 stasiun pengamatan makrozoobentos di ekosistem lamun pada bulan Maret dan April 2016. Pengambilan data makrozoobentos di lamun secara sistematis menggunakan metode transek kuadran 1x1 m dan corer berdiameter 10 cm untuk infauna. Menganalisis nilai kerapatan, tutupan lamun dan kelimpahan makrozoobentos. Analisis data statistik kelimpahan makrozoobentos menggunakan uji ANOVA, korelasi kepadatan makrozoobentos dengan tutupan lamun menggunakan regresi linear sederhana dan kontribusi spesies pada bulan purnama dan perbani menggunakan analisis SIMPER. Hasil penelitian didapatkan tidak adanya perbedaan nyata kelimpahan individu makrozoobentos di bulan purnama dan perbani. Sedangkan spesies didapatkan perbedaan nyata yang signifikan. Korelasi positif antara kepadatan makrozoobentos dengan tutupan lamun dibulan purnama, sedangkan pada bulan perbani berkorelasi negative. Analisis SIMPER menunjukan kontribusi spesies makrozoobentos tertinggi di tiap-tiap stasiun di bulan purnama dan perbani adalah spesies Cerithium salebrosum. Kesimpulan kepadatan spesies makrozoobentos menunjukkan perbedaan nyata di bulan purnama dan perbani, namun tidak pada kelimpahan individu.

Abstract

Cendrawasih Bay is habitat for olive ridley (Lepidochelys olivacea) which is directly adjacent to the Pacific Ocean. The presence and the diversity of L. olivacea in Cendrawasih Bay has been threatened. Genetics can be the key to conservation because they play an important role in maintaining population and recovering from damage. This study aims to determine the genetic diversity of L. olivacea and its association to the current pattern in the waters of Cendrawasih Bay. Samples of L. olivacea (n=20) were collected in Kwatisore (n=8) and Yapen Island (n=12) in Cendrawasih Bay waters from August 2015 to December 2016. The molecular analysis was based on 791-bp fragment of D-Loop on the non-coding region gen. The current pattern analysis was performed through INDESO data and visualized by using Ferret software. Phylogenetic analysis showed that 2 groups of L. olivacea from a total of 2 haplotypes, whose population from Kwatisore was dispersed in two haplotypes, while the population from Yapen Island was only dispersed in one haplotype. Populations from Kwatisore showed higher variations than populations from Yapen Island. The current pattern analysis suggests that the two study sites, Kwatisore and Yapen islands are different. Both populations are only connected by the Northwest monsoon currents period that indicates a little geneflow between this populations. Abstract

Coral reef now are under threats due to sedimentation. Fatal effect of organic rich sediment, leading corals mortality. Therefore, the study was conducted to investigate effect of organic sediment content to the coral diversity in Karimunjawa Island, Central Java, Indonesia. Field data was conducted at 6 locations.  Three sediment traps were deployed at each sites to measure organic sediment content. Twenty replicated quadrant transects were used to estimate coral density and coral diversity. Analysis of variance (ANOVA) was used to examine the differences of organic sediment content in each station. The linear regression was used to assess the relationship between organic sediment content and coral diversity. Our field result demonstrated that sediment content significantly different between sites with value range from 0.42 mg/800 ml - 1.32 mg/800 ml. Based on the Simson’s Diversity Index, the highest coral diversity found at Alang-alang as site with low sedimentation while the lowest coral diversity was found at Legon Lele as site with high sedimentation. The study shown significant negative correlation between organic sediment content and coral diversity with the coefficient of regression 0.68. This study convinces that disturbance on coral reefs might affect coral diversity in Karimunjawa Island, in addition to other factors such as the influence of human activities, natural disruption and climate change. Keywords: sedimentation, organic, coral, diversity, Karimunjawa
