Abstract

Ikan Ekor Kuning (Caesio cuning) merupakan salah satu target ikan tangkapan nelayan yang didaratkan di Kaliadem yang selanjutnya di jual di Pasar Ikan Muara Angke, Jakarta. Caesio cuning mempunyai nilai ekonomis tinggi dan telah banyak dikonsumsi oleh berbagai kalangan masyarakat. Penelitian ini bertujuan untuk mengetahui hubungan berat terhadap variasi morfometrik Caesio cuning yang menghasilkan pola pertumbuhan ikan. Pengambilan sampel ikan sebanyak 30 ekor dilaksanakan pada bulan April 2018 dengan metode survei di Pasar Ikan Muara Angke. Pengamatan morfologi dilakukan secara tradisional menggunakan penggaris dengan ketelitian 0.01 mm dan ditimbang menggunakan timbangan analitik dengan ketelitian 0.01 gr. Analisa yang digunakan untuk data pengukuran morfometrik dan berat ikan adalah regresi linear berganda. Hasil yang diperoleh bahwa karakter morfometrik secara keseluruhan berpengaruh terhadap berat badan ikan sebesar 98.4%, dimana karakter morfometrik pada panjang mulut (PM) yang berpengaruh positif terhadap berat badan ikan dengan nilai p-value sebesar 0.048 (>0.05), t Stat sebesar 2.113 (>1.96) dan coefficients sebesar 21.843 (+). Angka tersebut dapat diasumsikan bahwa setiap adanya penambahan pada panjang mulut sebesar 1cm maka akan bertambah 21.8gr berat badan ikan dengan kategori pola pertumbuhan alometrik positif.

Abstract

Over the past five decades, there has been a growing trend in the capture of kawakawa (Euthynnus affinis), which, alongside its potential to support small-scale commercial fisheries, highlights its significance.However, there is inadequate data on this species for suitable management and conservation status. This study aimed to investigate the genetic diversity, population structure, and connectivity of the kawakawa in the three major fishing port in Sumatra, namely Belawan , Padang, and Lampung by using DNA mitochondria control region (d-loop) Sequence. In total, 78 sequences of kawakawa with an average size of 303 bp, we found 36 polymorphic sites and 56 haplotypes from all population were identified with DNA compatibility values of 97-100%. We found the genetic diversity values in Sumatera Island were high (h = 0.950; π = 0.034), with the highest genetic diversities value in Padang (h = 0.990; π = 0.082) and the lowest in Belawan (h = 0.929; π = 0.082). AMOVA and Fst analyses revealed no differentiation in each population (Fst = 0.005). The haplotype distribution and connectivity analyses showed genetic mixing among the three populations. This study showed a single stock at the study sites and suggests management measures at a regional level to maintain the population.

Abstract

Green mussels are marine organisms that are threatened due to heavy metal pollution such as lead and copper in marine waters. In addition, to threatening shell organisms heavy metals are also a threat to symbiont organisms. Bacteria exposed to heavy metals continuously will later be able to adapt (resistance) to heavy metal contamination. This study aims to determine the concentration of heavy metals lead (Pb) and copper (Cu) in green mussels from Lampung Bay and to test the resistance of symbiotic bacteria to Pb and Cu as well as to conduct molecular identification to determine the type of selected symbiotic bacteria. This research was conducted by analyzing the content of heavy metals in water samples and green mussels then isolated the symbiotic bacteria and selected using Luria Bertani agar by adding heavy metal concentrations, then tested the level of resistance to Pb and Cu which were added continuously from concentrations of 100 ppm to 1000 ppm to bacteria can no longer grow optimally. After that, molecular identification was carried out to determine the type of bacteria and reconstructed to see the molecular proximity. The results showed that the water and green mussels had exceeded the quality standard and were classified as polluted. Bacterial isolates were resistant to Pb in the range of 100-1000 ppm and Cu in the range of 100-700 ppm. Molecular identification of the selected samples, namely STL09 and STL11, showed that the symbiont bacteria were a type of bacterial species Bacillus sp.

Abstract

Tridacna (kima) merupakan bivalvia yang berukuran besar dan memiliki peran penting dalam ekologi. Ukuran dan warna menarik yang menjadi daya tarik hingga mengalami overexplotation. Langkah perlindungan perlu dilakukan, salah satunya pengukuran kepadatan, variasi ukuran yang banyak ditemukan dan identifikasi spesies berdasarkan morfologi dan DNA barcoding. Tujuan penelitian ini adalah untuk mengidentifikasi kima dengan metode DNA barcoding serta mengevaluasi ukuran dan kepadatan populasinya di Perairan Maluku Utara. Metode penelitian menggunakan DNA barcoding dan identifikasi morfologi berupa warna mantel dan variasi ukuran cangkangnya serta pengukuran kepadatan kima yang ditemukan di Perairan Maluku Utara. Hasil identifikasi berdasarkan DNA barcoding dan identifikasi morfologi menunjukan hasil yang sama, yaitu terdeteksi tiga jenis kima, Tridacna crocea, Tridacna squamosa, dan Tridacna maxima. Perbedaan ukuran dan bentuk cangkang serta warna mantel yang timbul, memperkuat ketiga jenis tersebut sebagai spesies yang berbeda. Selain itu, bentuk clade pohon filogenetik antara sampel dengan data genbank membentuk clade yang sama. Kondisi kepadatan kima pada perairan ini menunjukan hasil yang sama dengan lokasi lain, yaitu nilainya kurang dari 1 /m2. Nilai kepadatan < 1 m2 dapat mengarahkan bahwa spesies tersebut mengalami penurunan atau mengarah pada overexploitatation. Sehingga, data penelitian ini dapat dijadikan sebagai referensi penilaian untuk kegiatan konservasi kima.

Abstract

Biodiversity assessment surveys are necessary for establishing conservation areas. However, such surveys are typically expensive, primarily if they cover a large area and take a long time. The survey difficulty increases when applied to cryptic, sparse, and fast-moving organisms. In addition, it requires expertise in taxonomic-biota classification. The breakthrough environmental DNA (eDNA) metabarcoding technique promises to overcome all the hurdles of assessing the potential for marine biodiversity in a non-invasive, rapid, extensive, and more effective way. We evaluated the ability of the eDNA survey to reveal the potential diversity and character of marine eukaryotes in the Lombok Island Marine Protected Area. A sampling of seawater and sediment eDNA in pore size fractions of 0.4-12 ?m and >12 ?m was carried out in the Core Zone, Non-Core Zone, and Non-Conservation Area, in east, north, and west Lombok, respectively. The detection and classification of eukaryotes using bioinformatics analysis were accomplished following extraction, amplification, and DNA sequencing. We identified 20,478 unique sequences of potential species classified in five kingdoms to 654 marine eukaryotes families. The comparison results show differences in community structure between locations, as well as differences in diversity between media and factions. The eDNA survey can assess marine biodiversity at a macro level and has implications for management in conservation areas.Keywords: Biodiversity, Marine eukaryotes, environmental DNA, the Primary V9-SSU 18S rRNA gene

Abstract

Sponges are symbiotic organisms that play a significant role in coastal ecosystems, such as filtering water, decomposing organic material into nutrients, providing shelter for microorganisms, and contributing to sediment stability and binding. This study aims to identify the composition and species diversity of sponges found in seagrass habitats on Panggang Island, Thousand Islands, DKI Jakarta. The research was conducted at three locations using a 50-meter line transect and 11 square transects measuring 0.5 x 0.5 meters along each line. Sponge samples were collected by cutting small pieces from each sponge for spicule analysis. The results indicate that a total of 10 sponge species were recorded across all research locations, dominated by Chondrilla caribensis forma, with sponge density values ranging from 0 to 13 individuals/m². The sponge diversity index on Panggang Island falls within the moderate category, indicating that the distribution of genera within the sponge community is relatively even. Phosphate concentration and current velocity showed a strong positive correlation with sponge density, while temperature exhibited a positive correlation with seagrass density. Conversely, the correlation between sponge density and seagrass density was relatively weak, supporting the findings that these two components do not have a close relationship within the observed ecosystem.

Abstract

AbstractTerdapat beberapa faktor fisik dan kimia yang dapat mempengaruhi distribusi pertumbuhan karang diantaranya suhu perairan, kecepatan arus, salinitas, kecerahan perairan, pH, dissolved oxygen (DO), nitrat, fosfat, sulfida, dan TSS . Faktor lingkungan tidak hanya menentukan pola sebaran dan kelimpahan karang, tetapi juga mempengaruhi bentuk morfologi pertumbuhannya. Penelitian ini memiliki dua tujuan utama yaitu mengetahui sebaran dan kelimpahan karang pada tiap kedalaman, dan menganalisis pengaruh kedalaman terhadap life form karang. Metode yang digunakan adalah metode kuantitatif dengan pendekatan meta-analisis. Analisis data menggunakan uji statistik non- parametrik Mann-Whitney. Data yang digunakan berasal dari data sekunder yang diperoleh dari beberapa jurnal atau karya tulis ilmiah lainnya. Hasil penelitian menunjukkan sebaran dan jenis karang yang ditemukan di seluruh lokasi pengamatan yaitu Acropora Branching, A. Encrusting, A. Submassive, A. Digitae, A. Tabulate, Coral Encrusting, C. Foliose, C. Massive, C. submassive, C. Heliopora, C. Millepora, dan C. Mushroom. Kelompok terumbu karang jenis C. Massive merupakan jenis yang paling mendominasi dengan nilai persentasi sebesar 7,90%. Berdasarkan hasil uji statistik, Z hitung lebih besar dari Z tabel maka H0 ditolak dan dapar disimpulkan bahwa terdapat perbedaan bentuk pertumbuhan karang terhadap perubahan kedalaman perairan.

Abstract

Whale shark occurrence in Probolinggo differs from other Indonesian locales, suggesting a link to zooplankton availability. Zooplankton composition and whale shark emergence are the focus of this study. From December 2017 to November 2018, six observation points were made each month. A plankton net filters and lugol preserves water. Olympus CX23 microscope observations were repeated twice. The spatial analysis revealed varying whale shark numbers at each station (Chi-square test, X2 = 1418.6, P <0.05), with six sharks observed at station PR_5. Zooplankton numbers were similar at each location. Temporal analysis revealed significant differences in whale shark appearance each month (Chi-square test, X2 = 81.04, P <0.05), with March and November having the highest appearance among the three individuals. The amount of zooplankton varied (Chi-square test, X2 = 148.61, P <0.05), with the highest abundance in April and March. Whale shark appearance and zooplankton composition were not correlated (r = 0.01, P< 0.05) both geographically and temporally. Whale sharks were linked to zooplankton kinds. Results indicate whale sharks are particularly interested in Acartia sp. (r = 0.3, P < 0.05). This suggests that whale sharks' appearance is determined by their demand for food, not zooplankton availability.

Abstract

Raja Ampat conservation area is divided into three zones: the core zone, the tourism zone, and the open zone. This region is crucial for sustainable fisheries and environmental management, yet it is under significant anthropogenic pressure. Accurate species detection is essential for inventory and diversity surveys, and environmental DNA (eDNA) methods have been shown to be more effective than conventional techniques. This study aimed to evaluate whether V9 primers could detect multispecies invertebrate compositions, assess community structure and contributions within each zone, and identify significant differences in invertebrate diversity among the zones. A total of 66 eDNA samples were collected from water and sediment columns across the three zones. Samples were extracted using the NucleoSpin„¢ kit (Macherey-Nagel) protocol, amplified with universal eukaryote primers targeting the 18S gene, and sequenced using Illumina MiSeq. Taxonomic analysis was performed using QIIME2 software and the SILVA database. The study identified 19 invertebrate species. The Shannon-Wiener (H') and Simpson (D) indices revealed greater species diversity in the core zone compared to the tourism and open zones. However, the Kruskal-Wallis test indicated no significant differences in species diversity across the zones. SIMPER analysis demonstrated a high percent contribution of species in each zone. This study highlights the effectiveness of eDNA methods for biodiversity assessment and offers valuable insights into invertebrate community structures within the Raja Ampat conservation area.
