Abstract

Indonesia is an important habitat for olive ridley (Lepidochelys olivacea) that it is necessary for sustainable management with respect to biological and ecological aspects as well as its environment. Genetic diversity and connectivity of L. olivacea can be calculated by using mitochondrial DNA analysis approach. The water circulation pattern are greatly affects to the distribution of L. olivacea that can be a transport medium for its migrations. This study aims to determined the genetic diversity of L. olivacea and its relation to water circulation across western and center of Indonesian waters. Samples of L. olivacea were collected in 6 locations across western and center of Indonesian waters from August 2015 to December 2016. The molecular analysis was conducted on control region gen based on 791-bp fragment. The water circulation analysis was performed through INDESO data and visualized by using Ferret software. Phylogenetic analysis showed that there are population connectivity of L. olivacea between western and central Indonesian waters. While population of L. olivacea in Kapoposang Island tend to be different compared to other populations. Genetic diversity analysis showed that L. olivacea from western Indonesia are more higher than center Indonesia. The water circulation analysis suggests that there is an established connection between western and center indonesian waters that connected by Northwest and Southeast seasons current, which the current vector indicates the movement of alternating water throughout Aceh, Pariaman, Panggul, Serangan, Kapoposang Island and Tuafanu populations.

Abstract

Gaining extensive knowledge of prey sources is an essential approach for understanding trophic structure and relationships, especially in highly diverse coral reef ecosystems. Groupers are a major Asian reef fish commodity, making it important to study grouper prey and the trophic relations involved. The wide distribution of groupers across different environments could lead to distinctive predatory behaviour. Therefore, this study was conducted to investigate and compare the taxonomic classification and composition of prey in the diets of two common groupers (Epinephelus areolatus and E. malabaricus) based on DNA metabarcoding of stomach contents and potential prey detection using environmental DNA tools at sites in the coral reefs of Raja Ampat, Indonesia. DNA recovered from the water column comprised taxa from the Arthropoda, Chordata, Cnidaria and Mollusca, several of which were also found in grouper guts, with Cnidaria the most abundant class. Diversity was high for potential prey species in the environment and prey consumed by each grouper species. The high overlap in prey identified from gut contents indicates these two epinephelids have a similar feeding strategy. However, nMDS ordination showed segregation between the prey consumed by each species and potential prey available in the environment. The results indicate a low likelihood of competition between the two grouper species, related to the abundance and wide choice of potential prey in the highly biodiverse Raja Ampat coral reef ecosystem.

Abstract

Abstract. Research on the sponge barnacle of Indonesia is very rare, and this study is aimed to describe the morphological characteristics of sponge barnacles and their specific relationship with their sponge host species. This research was a survey and sponge samples were collected by tearing apart any available sponge found in three sampling locations, typically coral reef areas of Weh Island, Seribu Islands, and Karimunjawa Islands. Sponge barnacles contained in sponge samples were observed using stereo microscope and scanning electron microscope, and species identification was determined based on the morphological description of Darwin (1854), Pilsbry (1916), Martin dan Davis (2001), and Kolbasov (1993). Four species of sponge barnacles were found consisting of Acasta cyathus, Acasta fenestrata, Euacasta dofleini, and Membranobalanus longirostrum. Specific relationships of barnacle and its sponge host were found between Euacasta dofleini and Haliclona sp. and between Membranobalanus longirostrum and Suberites sp. respectively. Hasil penelitian didapatkan empat spesies teritip spons yaitu Acasta cyathus, A. fenestrata, Euacasta dofleini, dan Membranobalanus longirostrum. Abstract

Abstract. Yellowfin tuna (Thunnus albacores) is a large, pelagic, and migratory species of tuna that inhabits Moluccas Sea in Indonesia, and most sea environment worldwide. However, high fishing activities tend to happen in the Indonesia region and catch product appear to be decreasing. A better understanding of yellowfin tuna genetic diversity is required to plan better conservation strategy of tuna. The study was conducted to infer the genetic diversity of yellowfin tuna (T. albacores) in the Moluccas Sea. A total of 41 tissue samples of yellowfin tuna were collected from two regions in the Moluccas Sea (North Moluccas and Ambon) during an expedition in February 2013. The results showed that genetic diversity and nucleotide diversity of yellowfin tuna from North Moluccas population was 0.984 and 0.021, respectively; while in Ambon population, the genetic and nucleotide diversities were 1.00 and 0.018, respectively. The high genetic diversity (0.990) and nucleotide diversity (0.020) between two populations were observed. Based on phylogenetic analysis, no genetic differentiation between the two populations in Moluccas Sea was revealed .Keywords :  Population genetics; Haplotype diversity; Coral Triangle; Phylogenetics; Polymerase Chain Reactions (PCRs) Abstrak. Tuna sirip kuning (Thunnus albacores) adalah ikan komersial penting dan ditemukan di Laut Maluku, Indonesia. Tetapi, aktivitas penangkapan ikan tuna sirip kuning dapat menurunkan kualitas dan kuantitas stok ikan, sehingga perlu adanya pengkajian keragaman genetik ikan tuna sirip kuning. Pemahaman yang baik tentang keragaman genetika dibutuhkan untuk merencanakan strategi konservasi tuna yang lebih baik. Penelitian ini bertujuan untuk mengetahui keragaman genetik  ikan tuna sirip kuning dari dua populasi di Laut Maluku. Sebanyak 41 sampel jaringan dari tuna sirip kuning dikumpulkan dari dua populasi di Laut Maluku (Maluku Utara dan Ambon) selama ekspedisi pada bulan Februari 2013. Hasil penelitian menunjukkan bahwa keragaman genetik populasi tuna sirip kuning pada perairan Maluku Utara dan Ambon masing-masing sebesar 0,984 dan 1,00 sedangkan nilai keragaman nukleotida masing-masing bernilai 0,021 dan 0,018. Nilai keragaman genetik dan keragaman nukloetida yang tinggi didaptkan antar kedua populasi masing-masing sebesar 0,990 dan 0,020. Abstract

Abstract. Yellowfin tuna (Thunnus albacares) is a large pelagic fish that have high economic value and inhabits the Moluccas Sea, Indonesia. Tuna catches in the Moluccas sea was very high and might decrease the yellowfin tuna population in this region. The research on population genetic of yellowfin tuna is fundamental to answer the problem. This information can be used as baseline data for future management, utilization, and basis of genetic conservation. The objective of this research was to infer the genetic population structure of two populations (North Maluku and Ambon) in the Moluccas Sea, Indonesia. In total, 41 tissue samples from pectoral fins of yellowfin tuna were collected in this study (North Maluku 33 samples and Ambon 8 samples). The results showed that genetic distances were low between the two populations. Additionally, the comparison of genetic distance between the Moluccas population and Indian Ocean waters also showed no significant differences. The Fst analysis showed the high gene flow between these two populations. Furthermore, haplotype network analysis showed that these two populations were the panmixia population. The overall result showed that no refraction genetic in the yellowfin tuna population from two populations in the Moluccas Sea.Keywords: Haplotype, genetic distance, Moluccas Sea, index fixation analysis, yellowfin tuna, population genetic structure. Abstrak. Tuna sirip kuning (Thunnus albacares) merupakan jenis pelagis besar bernilai ekonomis tinggi yang hidup di perairan Laut Maluku, Indonesia. Tangkapan ikan tuna di Laut Maluku berstatus tinggi, sehingga dapat menurunkan jumlah populasi. Penelitian tentang genetika populasi ikan tuna sirip kuning penting dilakukan untuk menjawab permasalahan ini. Informasi ini dapat menjadi sumber data untuk pengelolaan, pemanfaatan dan pelestarian untuk konservasi genetik. Tujuan penelitian untuk mengetahui struktur populasi genetik pada dua populasi di Laut Maluku (Maluku Utara dan Ambon). Secara total, 41 sampel jaringan dari sirip pectoral Tuna sirip kuning dikumpulkan dalam penelitian ini (Maluku Utara 33 sampel dan Ambon 8 sampel). Hasil penelitian menemukan jarak genetik yang dekat antar kedua populasi. Perbandingan jarak genetik pada populasi Perairan Maluku dan Samudera Hindia tidak menunjukan perbedaan signifikan. Analisis fiksasi indeks (Fst) memperlihatkan aliran genetik kuat antar populasi. Analisis jaringan haplotipe menunjukan kedua populasi merupakan populasi panmiksia. Penelitian ini secara umum menunjukkan belum terjadi perubahan struktur genetik populasi ikan tuna sirip kuning pada dua populasi di Laut Maluku.Kata kunci: Haplotipe, jarak genetik, Laut Maluku, analisis fiksasi indeks, tuna sirip kuning, struktur populasi genetik.

Abstract

Soft corals of the genus Clavularia (Blainville 1830) have complex and difficult to distinguish morphological characteristics, making them rather difficult to identify. A paucity of information exists for Indonesian coral reefs, a complex biogeographic region at the confluence of the Pacific and Indian Ocean that covers an extensive geographic area and several marine ecoregions, is subject to several ocean current systems, and includes a high diversity of habitats. Specimens of Clavularia inflata were collected from eleven study sites across the Indonesian archipelago, morphologically described based on their sclerite characteristics, and compared among sites. Sclerite observations were divided into four parts for each colony, namely pinnules, crowns, calyces, and stolons. The sclerites found in C. inflata comprised various forms such as rods, fingers, sticks, and brackets. Morphometric measurements of C. inflata showed specimens from Kepulauan Seribu had the largest pinnule, calyx, and stolon sclerites compared to other locations. The maximum mean size of crown sclerites was observed for specimens from Maluku Tenggara Barat. Similarities in the shape of sclerites resulted in specimens from Kangean, Morotai, Tanjung Lesung, Bontang, Lombok and Natuna forming a cluster, while those from Maluku Tenggara Barat and Arborek were dissimilar to other sites. Smaller sclerite sizes in specimens from Arborek are likely related to strong currents at that site. In conclusion, there were variations in the size of sclerites across the eleven study sites, with several sites forming a cluster of similar sclerite dimensions. Further studies using molecular markers are suggested to further explore the possibility of cryptic species.

Abstract

The current research was conducted to investigate the effect of light on growth of soft coral Lobophytum strictum. This species was transplanted and reared in two different ponds, uncovered pond (with light penetration) and covered pond (no light penetration. A total of 16 coral fragments was placed on each pond. Both, the survival rate and the growth rate were significantly different on the effect of light (P<0.05). The soft coral on the uncovered pond was survive up to 12 weeks (100%), followed with increased length (from 5,95 to 10,04 cm) and width (from 5,27 to 6,84 cm) of the transplanted coral fragments. Conversely, the soft coral in the covered ponds showed survival rate of 62,5% (up to 8th week), with decreased length (from 8,25 to 5,25 cm) and width (from 9,14 to 4,86 cm) of each fragments during the period of study.

Abstract

Ikan julung-julung (Hyporhamphus dussumieri) termasuk salah satu ikan yang kerap ditemui dalam kegiatan penangkapan ikan di perairan sekitar Muaro Angke, hal ini menunjukan bahwa potensi ikan julung-julung masih sangat tinggi. Untuk mengetahui hubungan panjang berat ikan tersebut maka dilakukan penelitian pada bulan April 2018. Sebanyak 30 ikan yang tertangkap dengan jumlah ikan betina 9 ekor dan ikan jantan 29 ekor. Metode yang digunakan pada penelitian ini adalah metode survey studi kasus. Hasil analisis hubungan panjang berat ikan julung-julung yang didapat dari perairan Muara Angke menunjukan koefisien korelasi (r) yaitu 0.9305. Ini mengartikan bahwa pertumbuhan ikan julung-julung menunjukan sifat isometrik. Adanya hasil korelasi yang erat memiliki arti bahwa semakin bertambah panjang baku (PB) pada ikan julung-julung maka morfometrik pembandingnya juga bertambah. Hasil korelasi tersebut diduga dipengaruhi oleh ketersediaan makanan. Faktor ketersediaan makanan saat berperan dalam proses pertumbuhan. Dari hasil korelasi ini menunjukan bahwa ketersediaan makanan di perairan Muara Angke masih mendukung kehidupan ikan julung-julung.

The high genetic diversity (0.990) and nucleotide diversity (0.020) between two populations were observed. Based on phylogenetic analysis, no genetic differentiation between the two populations in Moluccas Sea was revealed .Keywords :  Population genetics; Haplotype diversity; Coral Triangle; Phylogenetics; Polymerase Chain Reactions (PCRs) Abstrak. Tuna sirip kuning (Thunnus albacores) adalah ikan komersial penting dan ditemukan di Laut Maluku, Indonesia. Tetapi, aktivitas penangkapan ikan tuna sirip kuning dapat menurunkan kualitas dan kuantitas stok ikan, sehingga perlu adanya pengkajian keragaman genetik ikan tuna sirip kuning. Pemahaman yang baik tentang keragaman genetika dibutuhkan untuk merencanakan strategi konservasi tuna yang lebih baik. Penelitian ini bertujuan untuk mengetahui keragaman genetik  ikan tuna sirip kuning dari dua populasi di Laut Maluku. Sebanyak 41 sampel jaringan dari tuna sirip kuning dikumpulkan dari dua populasi di Laut Maluku (Maluku Utara dan Ambon) selama ekspedisi pada bulan Februari 2013. Hasil penelitian menunjukkan bahwa keragaman genetik populasi tuna sirip kuning pada perairan Maluku Utara dan Ambon masing-masing sebesar 0,984 dan 1,00 sedangkan nilai keragaman nukleotida masing-masing bernilai 0,021 dan 0,018. Nilai keragaman genetik dan keragaman nukloetida yang tinggi didaptkan antar kedua populasi masing-masing sebesar 0,990 dan 0,020. Berdasarkan analisis filogenetik, dua populasi di Laut Maluku ini memiliki kedekatan secara genetik.Kata kunci : Genetika populasi; Keragaman haplotipe; Segitiga Terumbu Karang; Filogenetika; Polymerase Chain Reaction (PCR)
