Abstract

Soft corals of the genus Clavularia (Blainville 1830) have complex and difficult to distinguish morphological characteristics, making them rather difficult to identify. A paucity of information exists for Indonesian coral reefs, a complex biogeographic region at the confluence of the Pacific and Indian Ocean that covers an extensive geographic area and several marine ecoregions, is subject to several ocean current systems, and includes a high diversity of habitats. Specimens of Clavularia inflata were collected from eleven study sites across the Indonesian archipelago, morphologically described based on their sclerite characteristics, and compared among sites. Sclerite observations were divided into four parts for each colony, namely pinnules, crowns, calyces, and stolons. The sclerites found in C. inflata comprised various forms such as rods, fingers, sticks, and brackets. Morphometric measurements of C. inflata showed specimens from Kepulauan Seribu had the largest pinnule, calyx, and stolon sclerites compared to other locations. The maximum mean size of crown sclerites was observed for specimens from Maluku Tenggara Barat. Similarities in the shape of sclerites resulted in specimens from Kangean, Morotai, Tanjung Lesung, Bontang, Lombok and Natuna forming a cluster, while those from Maluku Tenggara Barat and Arborek were dissimilar to other sites. Smaller sclerite sizes in specimens from Arborek are likely related to strong currents at that site. In conclusion, there were variations in the size of sclerites across the eleven study sites, with several sites forming a cluster of similar sclerite dimensions. Further studies using molecular markers are suggested to further explore the possibility of cryptic species.

Abstract

Abstract. Yellowfin tuna (Thunnus albacores) is a large, pelagic, and migratory species of tuna that inhabits Moluccas Sea in Indonesia, and most sea environment worldwide. However, high fishing activities tend to happen in the Indonesia region and catch product appear to be decreasing. A better understanding of yellowfin tuna genetic diversity is required to plan better conservation strategy of tuna. The study was conducted to infer the genetic diversity of yellowfin tuna (T. albacores) in the Moluccas Sea. A total of 41 tissue samples of yellowfin tuna were collected from two regions in the Moluccas Sea (North Moluccas and Ambon) during an expedition in February 2013. The results showed that genetic diversity and nucleotide diversity of yellowfin tuna from North Moluccas population was 0.984 and 0.021, respectively; while in Ambon population, the genetic and nucleotide diversities were 1.00 and 0.018, respectively. The high genetic diversity (0.990) and nucleotide diversity (0.020) between two populations were observed. Based on phylogenetic analysis, no genetic differentiation between the two populations in Moluccas Sea was revealed .Keywords :  Population genetics; Haplotype diversity; Coral Triangle; Phylogenetics; Polymerase Chain Reactions (PCRs) Abstrak. Tuna sirip kuning (Thunnus albacores) adalah ikan komersial penting dan ditemukan di Laut Maluku, Indonesia. Tetapi, aktivitas penangkapan ikan tuna sirip kuning dapat menurunkan kualitas dan kuantitas stok ikan, sehingga perlu adanya pengkajian keragaman genetik ikan tuna sirip kuning. Pemahaman yang baik tentang keragaman genetika dibutuhkan untuk merencanakan strategi konservasi tuna yang lebih baik. Penelitian ini bertujuan untuk mengetahui keragaman genetik  ikan tuna sirip kuning dari dua populasi di Laut Maluku. Sebanyak 41 sampel jaringan dari tuna sirip kuning dikumpulkan dari dua populasi di Laut Maluku (Maluku Utara dan Ambon) selama ekspedisi pada bulan Februari 2013. Hasil penelitian menunjukkan bahwa keragaman genetik populasi tuna sirip kuning pada perairan Maluku Utara dan Ambon masing-masing sebesar 0,984 dan 1,00 sedangkan nilai keragaman nukleotida masing-masing bernilai 0,021 dan 0,018. Nilai keragaman genetik dan keragaman nukloetida yang tinggi didaptkan antar kedua populasi masing-masing sebesar 0,990 dan 0,020. Berdasarkan analisis filogenetik, dua populasi di Laut Maluku ini memiliki kedekatan secara genetik.Kata kunci : Genetika populasi; Keragaman haplotipe; Segitiga Terumbu Karang; Filogenetika; Polymerase Chain Reaction (PCR)

In- formation regarding the presence of Selaroides leptolepis fish in the waters allows for effective conservation, and management of marine resources. A morphological identification of fish species is still considered inaccurate, so the molecular anal- ysis is necessary. This study aims to identify commercially important fish species deriving from Muara Baru Modern Fish Market, North Jakarta, employing mor- phological, and molecular analysis. A total of 30 specimens were collected from the field. The morphological analysis utilized a visual identification method, and morphometric measurement, while molecular analysis with DNA barcoding em- ployed the mitochondrial cytochrome oxidase subunit I (COI) gene. The results of the morphological analysis indicated that the fish species were yellowstripe scad (S. leptolepis). It also followed the results of molecular analysis of DNA barcoding that the fish was a S. leptolepis species. Therefore, the combination of morphological and genetic analysis has succeeded in identifying the fish species of S. leptolepis.

Abstract

Highlight Research The very first attempt to investigated and revealing biodiversity of crustacean crab in coral reef ecosystem at Tidung Island, Indonesia Conservation area (Tidung Kecil) has more abundant crab species than tourism area (Tidung Besar). A microhabitat that suffers anthropogenic pressure may alter the composition of crustacean biodiversity, especially in crab group. Different characteristics and complexity of coral habitat affect the dynamics of cryptic crustacean communities   Abstract   Recent studies have been widely carried out on the important role and ecological function of coral reefs. However, little attention has been given to small and cryptic crustaceans organisms living within different microhabitat in dead corals. This study aimed to reveal the diversity of crustaceans that inhabit dead corals in Tidung Islands, Indonesia. A total of 134 individuals from 41 species in 13 families were recorded within eight dead corals taken from two different sites. Furthermore, the species identified from Tidung Kecil were significantly different and more diverse than Tidung Besar. It was suggested that differences in anthropogenic pressure between Tidung Besar and Kecil might have an important role in the spread of crab crustaceans' organisms. Xanthidae from infraorder Brachyura were observed to be the most dominant family, while Galatheidae and Porcellanidae from infraorder Anomura spread almost equally in both sites. Additionally, Psaumis cavipes and Chlorodiella nigra were species with the largest contribution of crab crustaceans' communities in the death of coral's ecosystem in the Tidung Islands. This result indicated that dead corals might play a key role in supporting and providing a habitat for crustacean cryptic.

Abstract

Groupers are one of the most important and commercial fishes; many of the species are classified as Life Reef Fish Trade (LRFT). The number of grouper species in these locations may be more diverse than at other sites. Identifying the species number of a grouper is essential for mapping reef fitness. However, morphological identification of groupers is relatively difficult. Molecular methods such as DNA barcoding could be an option and even serve as reliable validation. Therefore, this research was conducted to identify groupers belonging to the Serranidae family. The grouper samples were obtained from the fish market in Waisai, Raja Ampat, Indonesia. The DNA barcoding employed the COI genes to accurately identify groupers. The Qiagen DNeasy 96 tissue extraction kit was used to extract DNA, and Primer Fish F1/R1 was applied. The sequences were analysed using ClustalW in MEGA 7.0 and phylogenetic tree was generated using the NJ (Neighbour Joining) method. Eleven species of groupers belonging to five taxa were discovered in Waisai, Raja Ampat. Anyperodon, Cephalopholis, Epinephelus, Plectropomus, and Variola are the grouper genera. Epinephelus was the predominant genus within the serranidae family, with 51.4% of the total. The International Union for Conservation of Nature's (IUCN) Red List category puts the majority of grouper species in the 'Least Concerned' category. This study confirmed the effectiveness of DNA barcoding for species identification and the value of the COI gene for Grouper's phylogenetic research. The outcome offers information about fisheries resources, which should be helpful in managing fisheries in reef ecosystems.

Abstract

Ikan Ekor Kuning (Caesio cuning) merupakan salah satu target ikan tangkapan nelayan yang didaratkan di Kaliadem yang selanjutnya di jual di Pasar Ikan Muara Angke, Jakarta. Caesio cuning mempunyai nilai ekonomis tinggi dan telah banyak dikonsumsi oleh berbagai kalangan masyarakat. Penelitian ini bertujuan untuk mengetahui hubungan berat terhadap variasi morfometrik Caesio cuning yang menghasilkan pola pertumbuhan ikan. Pengambilan sampel ikan sebanyak 30 ekor dilaksanakan pada bulan April 2018 dengan metode survei di Pasar Ikan Muara Angke. Pengamatan morfologi dilakukan secara tradisional menggunakan penggaris dengan ketelitian 0.01 mm dan ditimbang menggunakan timbangan analitik dengan ketelitian 0.01 gr. Analisa yang digunakan untuk data pengukuran morfometrik dan berat ikan adalah regresi linear berganda. Hasil yang diperoleh bahwa karakter morfometrik secara keseluruhan berpengaruh terhadap berat badan ikan sebesar 98.4%, dimana karakter morfometrik pada panjang mulut (PM) yang berpengaruh positif terhadap berat badan ikan dengan nilai p-value sebesar 0.048 (>0.05), t Stat sebesar 2.113 (>1.96) dan coefficients sebesar 21.843 (+). Angka tersebut dapat diasumsikan bahwa setiap adanya penambahan pada panjang mulut sebesar 1cm maka akan bertambah 21.8gr berat badan ikan dengan kategori pola pertumbuhan alometrik positif.

Abstract

Over the past five decades, there has been a growing trend in the capture of kawakawa (Euthynnus affinis), which, alongside its potential to support small-scale commercial fisheries, highlights its significance.However, there is inadequate data on this species for suitable management and conservation status. This study aimed to investigate the genetic diversity, population structure, and connectivity of the kawakawa in the three major fishing port in Sumatra, namely Belawan , Padang, and Lampung by using DNA mitochondria control region (d-loop) Sequence. In total, 78 sequences of kawakawa with an average size of 303 bp, we found 36 polymorphic sites and 56 haplotypes from all population were identified with DNA compatibility values of 97-100%. We found the genetic diversity values in Sumatera Island were high (h = 0.950; π = 0.034), with the highest genetic diversities value in Padang (h = 0.990; π = 0.082) and the lowest in Belawan (h = 0.929; π = 0.082). AMOVA and Fst analyses revealed no differentiation in each population (Fst = 0.005). The haplotype distribution and connectivity analyses showed genetic mixing among the three populations. This study showed a single stock at the study sites and suggests management measures at a regional level to maintain the population.

Abstract

Biodiversity assessment surveys are necessary for establishing conservation areas. However, such surveys are typically expensive, primarily if they cover a large area and take a long time. The survey difficulty increases when applied to cryptic, sparse, and fast-moving organisms. In addition, it requires expertise in taxonomic-biota classification. The breakthrough environmental DNA (eDNA) metabarcoding technique promises to overcome all the hurdles of assessing the potential for marine biodiversity in a non-invasive, rapid, extensive, and more effective way. We evaluated the ability of the eDNA survey to reveal the potential diversity and character of marine eukaryotes in the Lombok Island Marine Protected Area. A sampling of seawater and sediment eDNA in pore size fractions of 0.4-12 ?m and >12 ?m was carried out in the Core Zone, Non-Core Zone, and Non-Conservation Area, in east, north, and west Lombok, respectively. The detection and classification of eukaryotes using bioinformatics analysis were accomplished following extraction, amplification, and DNA sequencing. We identified 20,478 unique sequences of potential species classified in five kingdoms to 654 marine eukaryotes families. The comparison results show differences in community structure between locations, as well as differences in diversity between media and factions. Abstract

Spons merupakan organisme simbion yang memiliki peran penting dalam ekosistem pesisir, seperti menyaring air, menguraikan bahan organik menjadi nutrien, menyediakan tempat berlindung bagi mikroorganisme, serta berperan dalam stabilitas dan pengikatan sedimen. Penelitian ini bertujuan untuk mengidentifikasi komposisi, keanekaragaman spesies spons yang terdapat di habitat lamun di Pulau Panggang, Kepulauan Seribu, DKI Jakarta. Penelitian dilakukan di tiga lokasi menggunakan transek garis sepanjang 50 dan 11 transek kuadrat berukuran 0,5 x 0,5 meter di setiap garis transek. Sampel spons diambil dengan memotong sebagian kecil dari setiap spons untuk dianalisis spikulanya. Hasil penelitian menunjukkan bahwa spons yang ditemukan di seluruh lokasi penelitian terdata sebanyak 10 spesies dan didominasi oleh Chondrilla caribensis forma dengan nilai kerapatan spons sebesar 0-13 ind/m². Indeks keanekaragaman spons di Pulau Panggang berada pada kategori sedang yang menunjukkan bahwa distribusi genus dalam kumpulan spons di Pulau Panggang cukup merata. Kandungan fosfat dan kecepatan arus menunjukkan korelasi yang kuat dan positif terhadap kepadatan spons, sementara suhu memiliki korelasi positif terhadap kerapatan lamun. Di sisi lain, korelasi antara kepadatan spons dan kerapatan lamun relatif lemah, yang mendukung temuan bahwa kedua komponen ini tidak memiliki keterkaitan erat dalam ekosistem yang diamati.

Abstract

AbstractTerdapat beberapa faktor fisik dan kimia yang dapat mempengaruhi distribusi pertumbuhan karang diantaranya suhu perairan, kecepatan arus, salinitas, kecerahan perairan, pH, dissolved oxygen (DO), nitrat, fosfat, sulfida, dan TSS . Faktor lingkungan tidak hanya menentukan pola sebaran dan kelimpahan karang, tetapi juga mempengaruhi bentuk morfologi pertumbuhannya. Penelitian ini memiliki dua tujuan utama yaitu mengetahui sebaran dan kelimpahan karang pada tiap kedalaman, dan menganalisis pengaruh kedalaman terhadap life form karang. Metode yang digunakan adalah metode kuantitatif dengan pendekatan meta-analisis. Analisis data menggunakan uji statistik non- parametrik Mann-Whitney. Data yang digunakan berasal dari data sekunder yang diperoleh dari beberapa jurnal atau karya tulis ilmiah lainnya. Hasil penelitian menunjukkan sebaran dan jenis karang yang ditemukan di seluruh lokasi pengamatan yaitu Acropora Branching, A. Encrusting, A. Submassive, A. Digitae, A. Tabulate, Coral Encrusting, C. Foliose, C. Massive, C. submassive, C. Heliopora, C. Millepora, dan C. Mushroom. Kelompok terumbu karang jenis C. Massive merupakan jenis yang paling mendominasi dengan nilai persentasi sebesar 7,90%. Berdasarkan hasil uji statistik, Z hitung lebih besar dari Z tabel maka H0 ditolak dan dapar disimpulkan bahwa terdapat perbedaan bentuk pertumbuhan karang terhadap perubahan kedalaman perairan.
