Abstract

Mahkota dewa plant (Phaleria macrocarpa (Scheff.) Boerl.) which is included into family of Thymelaeaceae is one of Indonesia's traditional medicines. Chemical constituent has been isolated from ethyl acetate extract of leaves of mahkota dewa. Sample was extracted with methanol, concentrated then extracted by n-hexane, chloroform and ethyl acetate. The ethyl acetate extract was separated and fractionated by column chromatography. The first fraction was purified by TLC preparative and recrystalization. Compound was isolated as red-brown spherical crystal in 8 mg (m.p. 129-131 °C). Its spot gave dark fluoroscence at TLC plate (UV366) with Rf of 0.3 at TLC chromatogram with eluent of n-hexane : ethyl acetate (7:3); 0.6 with n-hexane : ethyl acetate (1:1); 0.9 with -hexane : ethyl acetate (4:6). This compound was dissolved in methanol. Compound was identified by UV, IR, 1H NMR, 13C NMR and NMR 2 dimension (HMQC, COSY, HMBC and DEPT-135) spectroscopic as 2,6,4'-trihydroxy-4-methoxybenzophenon. This compound as well as the ethyl acetate extract showed antioxidant activity on DPPH with IC50 was 10.57 and 101.06 μg/mL, respectively. This compound showed strong antioxidant activity on DPPH, almost to the standard antioxidant activity of quercetin (IC50 of 2.93 μg/mL)

Abstract

Fe (III), Cr(III), Cu(II), Ni(II), Co(II), and Pb(II)  metal ions had been separated using poly(eugenyl oxyacetic acid) as an ion carrier by bulk liquid membrane transport method. The effect of pH, polyeugenyl oxyacetic acid ion carrier concentration, nitric acid concentration in the stripping solution, transport time, and metal concentration were optimized. The result showed that the optimum condition for transport of metal ions was at pH 4 for ion Fe(III) and at pH 5 for Cr(III), Cu(II), Ni(II), Co(II), and Pb(II) ions. The carrier volumes were optimum with concentration of 1 x 10-3 M at 7.5 mL for Cr(III), Cu (II),  Ni(II), Co(II) ions and at 8.5 mL for Fe(III) and Pb(II) ions. The concentration of HNO3 in stripping phase was optimum at 2 M for Fe(III) and Cu(II) ions, 1 M for Cr(III), Ni(II) and Co(II) ions, and 0.5 M for Pb(II) ion. The optimum transport times were 36 h for Fe(III) and Co(II) ions, and 48 h for Cr(III), Cu (II), Ni(II), and Pb(II) ions. The concentration of metal ions accurately transported were 2.5 x 10-4 M for Fe(III) and Cr(III) ions, and 1 M for Cu (II), Ni(II), Co(II), and Pb(II) ions. Compared to other metal ions the transport of Fe(III) was the highest with selectivity order of Fe(III) > Cr(III) > Pb(II) > Cu(II) > Ni(II) > Co(II). At optimum condition, Fe(III) ion was transported through the membrane at 46.46%.

Abstract

Polyeugenyl oxyacetic acid has been synthesized from polyeugenol and chloroacetic acid and applied to separate metal Fe(III), Cr(III), Cu(II), Ni(II), Co(II), and Pb(II) using solvent extraction method. pH effect, polyeugenyl oxyacetic acid ion carrier concentration, extraction time, and metal concentration optimized to gain optimum condition. The result of experiment indicated that polyeugenyl oxyacetic acid resulted the yellowed brown synthesis with melting point 125 °C and yield 68.9%. The Characterization of spectroscopy IR attributed by absorption to the area 1735,8 cm -1 as carbonyl extend vibration (C=O) acid and 1H-NMR that was attributed by chemical shift at 4.6 ppm as carboxy proton (CH2-C=0). The metal separation optimum condition gained pH 3-6 for ion Fe(III), pH 5 for ion Cr(III), Ni(II), and Co(II) and pH 6 for ion Cu(II), and Pb(II). Optimum carrier concentraction 5 mL, 10 mL, and 15 mL for ion Fe(III), and ion Cr(III), Ni(II), Co(II), and ion Cu(II), Pb(II) respectively. Extraction time 2.5 h for ion Fe(III), 20 h for ion Cr(III), and 36 h for ion Cu(II), Pb(II), Ni(II), and Co(II). Metal concentration that would extracted correctly (0.75-5 x 10-4 M) for ion Fe(III), (0.75-2.5 x 10-4 M) for ion Cr(III), Ni(II), and Co(II) and (0.75-1 x 10-4 M) for ion Cu(II) and Pb(II). The best polyeugenil oxyacetic acid response to separate ion Fe(III) than other ions with selectivity order Fe(III)> Cr(III)> Cu(II)> Pb(II)> Ni(II)> Co(II).

Abstract

Chitinase is enzyme that hydrolyzes chitin, a polimer of b-1,4-N-Acetilglucosamine which is the most abundant natural resource after cellulose. Chitinolytic microorganism can be found in  environment like soil and water that contain chitin, or in extreme environment which is known as thermofilic microorganism. Chitinolytic microorganism is identified by recognizing the morphological and physiological properties based on Bergey's Manual of Systematic Bacteriology. The sequence data of the 16S rRNA genes is determinated in the GeneBank nucleotide sequence database. Chitinase activity can be qualitatively determined from the clearance zone around the colony formed in agar medium containing colloidal chitin. Chitinase can be utilized as biocontrol agent and derivate chitin as the result of chitinase degradation which can be used in the fieltd of health, food, industry and waste management

Abstract

Synthesis of analog L-α-metil-Dopa from eugenol has been achieved through conversion of allyl group to ketone, followed by reaction with NH3 and KCN and by hydrolisis. The addition reaction of methyleugenol with formic acid yield methyleugenyl formate (60,69%). The hydrolis of methyileugenylformate with KOH in aqaueous-ethanolic solution produced methyleugenyl alcohol (73,68%). The oxidation of methyleugenyl alcohol with PCC yield methyleugenyl ketone (67,71%). The reaction of methyleugenyl ketone with NH3 and KCN yield D,L-α-amino-α-(3,4-dimetoxybenzyl) propionitril (84,14%). The hydrolisis of D,L-α-amino-α-(3,4-dimetoxybenzyl) propionitril with concentrated hydrochloric acid gave Analog L-α-metil-Dopa as a main target (91,98%). Abstract

Synthesis of flavanoid compounds of chalcone and flavanone groups have been conducted. Flavanoid Is one of the group natural products which is mostly found in plants and have been proved to have physiological activity as drug. In this research, chalcone proup compounds that being synthesized are: chalcone, 3,4-dimethoxychalcone, 2'-hidroxy-3,4-dimethoxychalcone where as compound of flavanone group that being synthesized is 3',4'-dimethoxyflavanone. The synthesis of chalcone group are carried out based on Claisen-Schmidt reaction by using raw material of aromatic aldehydes and aromatic ketones. The synthesis in carried out by stirring at the room temperature using alkali solution as catalyst and ethanol as solvent. The synthesis of 3',4'-dimethoxyflanone is made based on the nucleophilic 1,4 addition of the unsaturated α,β ketone. The synthesis is made by refluxing 2'-hydroxy-3,4-dimethoxychalcone in alkali condition for 12 hours. The identification of flavanoid compound is carried out by using spectroscopic IR, GC-MS and 1H-NMR methods. The result of each synthesis chalcone group are follows: chalcone as yellowish solid with m.p= 50 °C and the yield is 83.39%; 3,4-dimethoxychalcone as yellow solid with m.p= 57°C and the yield is 76.00% ; 2'-hydroxy-3,4-dimethoxychalcone as orange solid with m.p= 90 °C and the yield is 74.29%, for 3',4'-dimethoxyflavanone as pale yellow solid with m.p= 80 °C and the yield is 72.00%.

Abstract

Research on the isolation, toxicity test, antimalarial test, and identification of the active compound from the ethyl acetate fraction of Stylissa massa sponge from Oenggae waters, Rote Island, has been conducted. This study aimed to investigate the antimalarial activity of the ethyl acetate fraction of the Stylissa massa sponge. Isolation was carried out by the extraction method using a mixed solvent of methanol: dichloromethane of 3: 2 (v/v), then the extract was partitioned in a solvent mixture of ethyl acetate: water of 1: 2 (v/v). The ethyl acetate extract obtained was separated by column chromatography using the gradient polarity system method. The toxicity test of each fraction was carried out by the Brine Shrimp Lethality Test (BSLT) method, and the antimalarial test was carried out by the haematin polymerization inhibition method. Identification of compounds from the active fraction in the antimalarial test was carried out using Liquid Chromatography-Mass Spectrometry (LC-MS). The extraction yield was 1.14 g (0.23%) of the ethyl acetate extract in the form of a dark brownish-yellow oily solid. Separation by column chromatography resulted in 15 fractions. Toxicity test results showed the four most active fractions with LC50 values, which are very promising for new drug discovery. The IC50 value in the antimalarial activity test of the four fractions indicated that the Stylissa massa sponge ethyl acetate extract was more active than the standard chloroquine compound (115 μg/mL). The LC-MS analysis indicates that fraction 11 contains two compounds that have been reported, and 1 compound is unknown. In contrast, fraction 14 indicates that it contains three compounds that have been reported and one unknown compound.

Abstract

Cassava peel was determined to be underutilized by the community that it turned out to be wasted. Cassava peel contains starch which has a potential as an edible coating. During post-harvest, Tomatoes are easily to loose Phenol and Vitamin C. Therefore, there should be solutions to keep Phenol and Vitamin C level on Tomatoes through edible coating application of Cassava Peel Starch. This study aims to find out the effect of edible coating application of cassava peel starch with concentrations of 1%, 2% and 4% on the Total Phenolic Content and Vitamin C in tomatoes on day 3, 6 and 12; as well as to find out the present research’s potential as learning resources for grade VIII of Junior High School in learning Additive Substance. Total Phenolic Content was tested using spectrophotometric, while Vitamin C content was tested using Iodine titration. The results show that the highest Total Phenolic Content and Vitamin C occured on day 12 in the 4% of edible coating treatment, respectively: 0.0525% and 33.2210mg /100g. According to the results, a conclusion could be made that edible coating of cassava peel starch is able to maintain Total Phenolic Content and Vitamin C on Tomatoes, compared to the control. The results of the study could be used as learning resources for grade VIII of Junior High School in learning Additive Substance. Abstrak. Kulit singkong kurang dimanfaatkan oleh masyarakat sehingga menjadi limbah. Kulit singkong mengandung pati yang berpotensi sebagai edible coating. Tomat mudah mengalami kerusakan pasca panen sehingga kadar total fenol dan vitamin C menurun, maka perlu adanya alternatif untuk mempertahankan kadar total fenol dan vitamin C tomat dengan cara aplikasi edible coating pati kulit singkong. Tujuan dari penelitian ini adalah untuk mengetahui pengaruh adanya aplikasi edible coating pati kulit singkong konsentrasi 1%, 2% dan 4% terhadap kadar total fenol dan kadar vitamin C tomat hari ke 3, 6 dan 12 serta mengetahui potensi hasil penelitian sebagai sumber belajar IPA SMP kelas VIII materi zat aditif. Kadar total fenol diuji menggunakan metode spektrofotometri dan kadar vitamin C menggunakan metode titrasi iodium. Hasil penelitian menunjukan kadar total fenol dan kadar vitamin C tertinggi terjadi pada hari ke 12 perlakuan edible coating 4% berturut-turut sebesar 0.0525% dan 33.2210mg/100g, maka edible coating pati kulit singkong dapat mempertahankan kadar total fenol dan vitamin C tomat dibandingkan kontrol. Hasil penelitian dapat digunakan sebagai sumber belajar IPA SMP kelas VIII materi zat aditif.

Abstract

This study aimed to investigate the potential of hydroxy chalcone and flavone derivatives as inhibitors of the epidermal growth factor receptor (EGFR) with anticancer properties. Molecular docking simulations were conducted using Autodock Tools 1.5.6 and Discovery Studio visualizer. The EGFR protein structure with the PDB code 1M17 was utilized as the receptor, explicitly targeting the binding pocket. Redocking of the reference ligand erlotinib yielded a binding energy of -7.51 kcal mol-1 with an RMSD of 0.54 Å, confirming the accuracy of the docking protocol. The hydroxy chalcone and flavone derivatives exhibited binding energies ranging from -6.50 to -7.67 kcal mol-1 when interacting with the EGFR protein. Among the studied compounds, compound 2',5'-dihydroxy-3,4-dimethoxychalcone (1g) displayed the lowest binding energy. Interactions involving amino acids such as Met769, Ala719, Thr766, Lys721, and Glu738 were identified as crucial hydrogen bonding interactions between the ligands and the EGFR protein. These findings suggest that 2',5'-dihydroxy-3,4-dimethoxychalcone holds strong potential as a tyrosine kinase inhibitor, positioning it as a promising candidate for further development as an anticancer agent. The outcomes of the computational analysis conducted through the pkCSM online platform indicated that the chemical 2',5'-dihydroxy-3,4-dimethoxychalcone had favorable pharmacokinetic characteristics and showed low toxicity levels. Abstract

The synthesis derivate acetophenone from fenol and eugenol had been carried out. The acylation of fenol was performed by anhydride acetate and the fries rearrangement reaction was carried out using AlCl3 catalyst at temperature of 50oC produce ortho-hydroxyacetophenone (35,29 %), purity was tested by GC, structure elucidation of these product ware analized by FTIR and 1H-NMR. Acylation reaction of eugenol was performed by anhydride acetate in base condition at temperature of 120oC for 3 h produce 5-allyl-2-hydroxy-3-metoksi acetophenone (21,42%), purity was tested by GC, structure elucidation of these product ware analized by FTIR, 1H-NMR and MS.
