Abstract

Sintesa 3,4-dimetoksi flavanon dengan menggunakan bahan dasar veratraldehida dan orto hidroksiasetofenon melibatkan dua tahap reaksi, yaitu reaksi Claisen-Schmidt dan siklisasi (adisi nukleofilik keton tak jenuh α, β). Pada reaksi Claisen-Schmidt, veratraldehida dan orto-hidroksiasetofenon bereaksi dalam suasana alkali menghasilkan 2’-hidroksi-3,4-dimetoksikalkon. Pada reaksi siklisasi, senyawa 2’-hidroksi-3,4-dimetoksikalkon mengalami siklisasi dalam kondisi alkali menghasilkan 3,4- dimetoksiflavanon. Analisis struktur senyawa flavanoid dilakukan dengan menggunakan spektrometer IR, UV-Vis dan 1H-NMR. Senyawa 2’-hidroksi-3,4-dimetoksikalkon yang dihasilkan berwama kuning dengan titik lebur 57°C dan rendemen 74,3%, sedangkan senyawa 3,4-dimetoksiflavanon yang dihasilkan berwama kuning pucat dengan titik lebur 80°C dan rendemen 72,0%.

Abstract

As one of countries which has the large tropical forests in the world and the largest in ASEAN, Indonesia has been facing serious deforestation problems. Eventhough some scholars have found causes of  the deforestation and have tried to give solutions, however, identifications of deforestation causes are still worth studying as the deforestation still becomes an important issue in Indonesia. This study tries to extend and explore the causes of deforestation in Central Kalimantan, one of Provinces in Indonesia which has been facing the highest deforestation problem caused by many factors. This study applied a qualitative approach. Data were collected by using interview and focus group discussions. It involved about 27 informants representing 14 organizations including goverment officers and non-governmental organization leaders such as WALHI,AMAN, Save Our Borneo, Yayasan Bentang Borneo, Ampuh, TUK Jakarta, Greenpeace, SIEJ, SPKS, Sawit Watch, Forest Watch Indonesia, Mongabay, Institute Dayakologi, LINK-AR Borneo, WWF and Academia as informants. The study found that as a socio political construct, deforestation in Indonesia emerges at the intersection between the biophysical changes of the politically defined forest and the political economic struggles around access and control over forestland resources by domestic and international actors. Hardly is a given issue, the term deforestation itself is still subject to debate as state definition of deforestation is continuously being challenged by the definition developed by civil society members. Deforestation, in many of its aspects, is also associated with governance failure and complex political economic settings that shape its emergence.

Abstract

Pengembangan obat antibiotik dalam menanggulangi penyakit akibat infeksi bakteri semakin meluas. Hal ini diiringi dengan semakin meningkatnya prevalensi retensi terhadap antibiotik. Banyaknya kasus retensi terhadap antibiotik menyebabkan perlu dilakukan pengembangan agen antibakteri yang salah satunya berasal dari senyawa turunan kalkon. Sintesis senyawa turunan kalkon yang memiliki aktivitas dalam menghambat bakteri Bacillus subtilis dan Escerichia coli telah dilakukan. Sintesis senyawa (E)-2’-hidroksi-2-bromo-4,5-dimetoksikalkon melalui reaksi kondensasi Claisen-Schmidt dari senyawa 2-hidroksiasetofenon dengan 2-bromo-4,5-dimetoksibenzaldehida.. Sintesis dilakukan dengan metode refluks dalam kondisi basa menggunakan katalis NaOH dan metanol sebagai pelarut selama 24 jam. Struktur hasil sintesis senyawa turunan kalkon dianalisis dengan FTIR, KLT-scanner, GC-MS, MS-direct, 1H-NMR dan 13C-NMR. Uji antibakteri dilakukan dengan metode difusi cakram (sumuran) terhadap bakteri Bacillus subtilis dan Escherichia coli. Seri konsentrasi senyawa meliputi 1%, 5%, 10%, dan 20% serta digunakan kontrol positif 4-isopropil-3-metilfenol 1%. Hasil penelitian menunjukkan bahwa sintesis senyawa turunan kalkon menghasilkan padatan berwarna kuning dengan rendemen sebesar 78,45%. Daya hambat senyawa kalkon dan 4-isopropil-3-metilfenol sebagai kontrol positif hampir sama untuk kedua bakteri yaitu 15 mm untuk Bacillus substilis dan 13 mm untuk Escherichia coli. Selain itu, uji daya hambat senyawa turunan kalkon terhadap bakteri Bacillus substillis lebih baik dibandingkan pada bakteri Escherichia coli pada konsentrasi 20% dengan zona hambat berturut-turut sebesar 19,7 dan 17,7 nm.

Abstract

Titration acid-base needs indicator  to show the change of color on interval of hydrogen exponent/degree of acid (pH). Indicator of synthetic which always be used have disadvantages like chemical pollution,  stock and expensive of pro- duction  cost. The research has been carried out to substitute the synthetic indicator with herbal indicator extracted from flower crown of Hibiscus rosa sinensis L extract. The herbal indicator was extracted from the flower crown Hibiscus rosa sinensis L using a mixture methanol-acetic acid. Then it was evaluated with phenolphthalein and methyl orange(E merck) comparer to titration the acid-base, they are strong acid-strong base, weak base-strong acid and weak acid-strong base. The result of research show that herbal indicator  of flower crown Hibiscus rosa sinensis L to show theequivalent point in all titrations give peer result with the comparison. With the research result hoped that indicatortitration acid-base flower crown Hibiscus rosa sinensis L is able to as replace synthetic indicator  (metyl orange andphenolphtalein) which always be used before.ABSTRAKTitrasi asam-basa memerlukan indikator untuk menunjukkan perubahan warna pada setiap interval derajad keasaman (pH). Indikator sintetis yang digunakan selama ini mempunyai beberapa kelemahan seperti polusi kimia, ketersediaan dan biaya produksi mahal. Upaya penelitian sudah dilakukan untuk menggantikan indikator sintetis dengan indikator dari ekstrak mahkota bunga sepatu. Indikator herbal tersebut dibuat dengan cara mengekstrak mahkota bunga Hibiscus rosa sinensis L dengan mengunakan pelarut metanol-asam asetat. Kemudian dievaluasi dengan indikator pembanding fenolftalein dan metil oranye (produksi E merck) untuk titrasi asam-basa yaitu asam kuat-basa kuat, basa lemah-asam kuat dan asam lemah-basa kuat. Dari hasil penelitian diketahui bahwa indikator dari mahkota bunga sepatu untuk menunjukkan titik ekivalen dalam titrasi tersebut memberikan hasil yang setara dengan indikator pembandingnya. Hasil penelitian menunjukkan, indikator dari mahkota bunga sepatu dapat sebagai pengganti indikator sintetis (metil oranye dan fenolftalein) yang selama ini digunakan.

Abstract

Fries Rearrangement of eugenyl acetate in order to study allilic substituentgroup influence has been done. Fries rearrangement of eugenyl acetate was done byheating at 120°C for 3 hours under AlCl3, dichloromethane as the catalyst and solvent,respectively. The structure of the compound was identified using Infra Redspectrometry (IR) and GC-MS. The result of te research showed that rearrangementfries of eugenyl acetate are not formed, but yielded lightly brown oil of dimmer.

Abstract

The Synthesis of acetophenone derivative from 1-(4-acetoxyphenyl-3-methoxy)-2-propanyl formate through Fries Rearrangement in order to produceortho hidroxy acetophenone derivative as starting material of Flavanoid compound hasbeen done. The reaction of 1-(4-acetoxyphenyl-3-methoxy)-2-propanyl formate wasdone by heating at 120 °C for 3 hours under AlCl3, dichloromethane as the catalyst andsolvent, respectively. The structure of the compound was identified using Infra Redspectrometry (IR) dan GC-MS. Fries rearrangement of 1-(4-acetoxyphenil-3-methoxy)-2-propanyl formate produce 1-(2-hyidroxy-3-methoxy-5-propenyl)-acetophenone and 1-(2-hyidroxy-3-methoxy-5-propanyl)-acetophenone as sideproduct with product rendemen were 43.26% and 9.48%, respectively.

Abstract

Chalcone is one of the phenolic group secondary metabolic with numerous biological activity. Many studies have shown that chalcone derivatives compound has anticancer, anti-inflammatory, antimalarial, and antibacterial activities. The purpose of this research was to study the prediction potency unsaturated carbonyl system of chalcone derivative against the HeLa cell by MTT assay. Those activities assumed can inhibit the mechanism action of NF-kB that caused cervical cancer. The 2,4-dihydroxide-4’-methoxychalcone has done synthesis as a target compound by a sonochemical for 7 hours. The results showed that chalcone derivative most active against the HeLa cell.

Abstract

A chitinolytic microorganism had been isolated from mud fields in Ketintang Surabaya area by using a minimal media containing 0.4% colloidal chitin. Activity assay is based on released N-acetyl-glucosamine which reacted with 3,5 dinitrosalicyclic acid. Mud fields produce 63 bacterial isolates with chitinase activity (TNH1 – TNH63 isolates). The highest activity was shown by TNH11 isolate with specific activity 1.27 U/mg. TNH11 isolates was Gram negative, rod-cocoid cell, has a colony of yellow, round shape, convex elevation, flat margin and the colony diameter 3–5 mm.

Abstract

Enzim lipase mempunyai peranan penting dalam katalis berbagai reaksi industri satu diantaranya pembuatan flavor melalui reaksi esterifikasi. Lipase adalah biokatalis yang berperan besar dalam aplikasi bioteknologi, seperti dalam sintesis biopolimer, biodiesel, produksi obat, dan produksi flavor. Peningkatan penggunaan lipase untuk industri mendorong dilakukan penelitian untuk mendapatkan sumber-sumber lipase baru. Sumber lipase yang potensial salah satunya adalah bakteri Azospirillum sp.PRD1 dari isolat lokal Laboratorium Mikrobiologi, Fakultas Biologi Universitas Jenderal Soedirman. Tujuan penelitian adalah untuk mendapatkan ekstrak kasar lipase dan menentukan karakteristik sifat-sifat biokimiawinya. Metode yang digunakan antara lain peremajaan bakteriAzospirillum sp.PRD1, dan produksi inokulum, penentuan waktu produksi optimum dan fase pertumbuhan bakteri, ekstraksi dan produksi ekstrak kasar lipase dan penentuan karakteristik sifat-sifat biokimiawinya. Hasil penelitian diperoleh ekstrak kasar lipase dari inokulum berumur 7 jam dan medium produksi dengan induser minyak zaitun yang diinkubasi selama 3 jam memiliki aktivitas spesifik 7,0547 Unit/mg. Lipase ekstrak kasar optimum pada pH 7, suhu 40 oC dan waktu inkubasi selama 25 menit. Lipase merupakan metaloenzim dengan kofaktor Zn2+ , Mn2+, Hg2+, Ca2+, Co2+ and Mg2+.

Abstract

Breast cancer is one of cancer causes of death in woman. Chemotherapy is one cancer treatment give toxic effects on normal cells. Alternative of cancer treatment by using flavonoid derivative have potent anticancer to reduce side effects of cancer. Chalcone is family of flavonoid that have biological activity. Chalcone derivatives have potential compound as anticancer agent. Chalcone with the presence halogen, metoxy group in ring B is know to inhibit cancer cells. The aims of this research were to synthesize chalcone derivate with bromo, methoxy, and hyroxy group in ring chalcone and to determine the anticancer activity of chalcone derivative. The chalcone derivative was synthesized from 2-hydroxyacetophenone with 2-bromo-4,5-dimethoxybenzaldehyde by Claisen-Schmidt reaction. In vitro cytotoxicity against breast cancer cell was tested by MTT assay method. The compound of 2’-hydroxy-2-bromo-4,5-dimethoxychalcone was yield in 78% as yellow solid. The IC50 of 2’-hydroxy-2-bromo-4,5-dimethoxychalcone was 42,19 µg/mL as a moderate activity to inhibiting breast cancer cell line. Cytotoxity of docorubicin againts breast cancer cell line more active than 2’-hydroxy-2-bromo-4,5-dimethoxychalcone with IC50 10,61 µg/mL. Doxorubicin as drug standar had better anticancer activity than 2’-hydroxy-2-bromo-4,5-dimethoxychalcone. Based on the IC50 value, the compound 2’-hydroxy-2-bromo-4,5-dimethoxychalcone has a moderate activity towards breast cancer cell lines. This study can recommend as candidate for anticancer againts breast cancer cell lines.
