Abstract

Rhodamin B is dangerous for body due to its ability to improve oxidative stress on various reproduction tissues on female. However, there is no significant scientific proof that Rhodamin B exposure influences VEGF and the number of arteriole on endometrium. VEGF and arteriole is crucial to angiogenesis process of endometrium. Abstract

1(CA), 2, 3 1(CA), Indonesia;  (Corresponding Author) 2, Indonesia 4, Indonesia ABSTRACT Green tea extract (GTE), Camellia sinensis, known as a very effective antioxidant to repair the damage of organs exposed by stress oxidative. Cypermethrin (CYP) is a harmful substance that can accumulate in the body and cause oxidative stress. This study aimed to demonstrate the effect of GTE in increasing the number of granulosa cells and E2 serum levels. Female rats (Rattus norvegicus) were randomly selected and divided into 5 groups, control group (KN), a group given only CYP 20 mg/kg body weight/day (KP), groups given CYP 20 mg/kg body weight/day and then treated with GTE 7, 14, 28 mg/kg body weight/day (P1, P2, and P3 respectively). Treatment was given for 28 days. The number of granulosa cells was counted by HSel software, E2 serum levels were determined via ELISA ASSAY. Treatment results with GTE 7, 14, and 28 mg/kg body weight/day significantly (p < 0.05) increased the number of granulosa cells and E2 serum levels relative to KN. Administration of GTE 28 mg/kg body weight/day caused the greatest increase in the number of granulosa cells and E2 serum levels. Our result suggests that GTE has potential prevention to stress oxidative by CYP exposure in granulosa cells and E2 serum. Keywords: Cypermethrin, E2, Granulosa Cells, Green tea

Abstract

Latar Belakang: Sipermetrin adalah insektisida jenis piretroid sintetik yang dapat menganggu sistem reproduksi. Sipermetrin juga mampu menghasilkan  reactive oxygene species (ROS) sehingga bertanggung jawab menimbulkan terjadinya stres oksidatif. Teh hijau (Camellia sinensis) mengandung polifenol sehingga dapat menghambat radikal hidroksil dan enzim prooksidan. Tujuan: Penelitian ini bertujuan untuk membuktikan pemberian ekstrak etanol teh hijau dapat meningkatkan kadar FSH dan jumlah folikel antral pada tikus betina yang dipapar sipermetrin. Metode: Penelitian yaitu true experimental pada 25 ekor tikus betina galur wistar (Rattus norvegicus) yang dibagi menjadi 5 kelompok; KN: tanpa perlakuan, KP: sipermetrin 20 mg/kgBB/hari, P1: sipermetrin 20 mg/kgBB/hari + ekstrak etanol teh hijau 7 mg/kgBB/hari, P2: sipermetrin 20 mg/kgBB/hari + ekstrak etanol teh hijau 14 mg/kgBB/hari, P3: sipermetrin 20 mg/kgBB/hari + ekstrak etanol teh hijau 28 mg/kgBB/hari per oral selama 28 hari. Kadar FSH diukur dengan metode ELISA dan organ ovarium diambil untuk pemeriksaan jumlah folikel antral dengan metode pewarnaan hematoxylin eosin. Hasil: Kadar FSH dan jumlah folikel antral pada tikus betina menunjukkan hubungan yang positif dan kuat dengan nilai koefisien korelasi 0,742 yang berarti peningkatan kadar FSH diikuti penambahan jumlah folikel antral. Ada perbedaan bermakna pada kadar FSH (p=0,005) dan jumlah folikel antral (p=0,001) antara kelompok kontrol dengan kelompok perlakuan. Kesimpulan: Pemberian ekstrak etanol teh hijau dapat meningkatkan kadar FSH dan jumlah folikel antral pada tikus betina yang dipapar sipermetrin.

Abstract

BACKGROUND: The absence of potential biomarkers to detect the metastatic process at an early stage will consequently delay colorectal cancer (CRC) treatment. Some biomarkers including β-Catenin, E-Cadherin and N-Cadherin have been suggested as potential markers. However, there were opposite reports regarding expressions of these markers. Therefore, current study was conducted using CRC cell lines for early stage (SW-480 cells) and late stage (HCT-116 cells) of CRC.METHODS: SW-480 and HCT-116 cells were cultured and seeded on coverslip glasses for immunofluorescence staining to detect β-Catenin, E-cadherin, and N-cadherin. Expressions of β-Catenin, E-cadherin, and N-cadherin were observed and documented under a fluorescent microscope and analyzed with Image J software. Measured results were then statistically analyzed. RESULTS: All β-catenin, E-Cadherin and N-Cadherin expressions were observed in SW-480 and HCT-116 cells. β-catenin MFI averages of SW-480 (47.157±3.479) and HCT-116 (47.240±4.107) cells were similar. E-Cadherin MFI average of SW-480 cells (45.104±4.107) was higher than the one of HCT-116 cells (40.191±3.702). N-Cadherin MFI average of HCT-116 cells (43.702±8.219) was significantly higher (p=0.009) than the one of SW-480 cells (72.506±5.297).CONCLUSION: Taken together, N-Cadherin could be suggested as an important metastasis marker in CRC since the N-Cadherin expression was significantly higher in HCT-116 cells as the late-stage CRC model than SW-480 as the early-stage of CRC model. Further research is still needed by comparing several biomarkers from various clinical samples at all clinical stages of CRC.KEYWORDS: CRC, β-Catenin, E-Cadherin, N-Cadherin, Metastasis, Biomarker

Abstract

Bawang Dayak (Eleutherine sp) merupakan salah satu tanaman yang tumbuh liar di hutan Kalimantan yang berpotensi dikembangkan menjadi Obat Herbal Terstandard. Berdasarkan studi etnofarmakologi, tanaman ini dimanfaatkan masyarakat suku Dayak sebagai obat diabetes, stroke, kanker payudara, antihipertensi, mempercepat penutupan luka, dan pengobatan jantung. Meski sudah digunakan turun temurun dan riset yang berkelanjutan, bawang Dayak belum dimanfaatkan dokter formal untuk pengobatan klinis. Hal ini disebabkan belum adanya uji y akurat dan terstandar yang dapat digunakan sebagai bukti ilmiah. Pada sisi lain, masyarakat berpendapat bahwa penggunaan bawang Dayak sebagai jamu dapat berbahaya karena komplikasi akibat bercampur dengan Bahan Kimia Obat (BKO). Dengan demikian, bawang Dayak perlu diupayakan dalam Program Saintifikasi Jamu untuk mewujudkan jamu, obat herbal terstandar, dan fitofarmaka yang berkualitas, bermutu serta aman. Oleh karena itu, pada artikel ini akan dibahas beberapa riset tentang bioaktifitas bawang Dayak mulai fitokimia hingga tahapan uji praklinik

Abstract

This study aimed to evaluate the effects of ciplukan (Physalis angulata L.) leaf ethanol extract on L-NG-nitroarginine methyl ester (L-NAME)-induced hypertensive rats. We randomly divided twenty-five Wistar rats into five groups. The sham group was given a PBS injection. The hypertensive group was injected with L-NAME on days 1 to 28. Three groups of hypertensive rats were given the extract on days 4 to 28. Blood pressure was measured using the tail-cuff method on days 0, 4, 10, and 27. The endothelial progenitor cells (EPCs) in the blood were measured by flow cytometry as a percentage of circulating angiogenic cells (CACs, CD34+/CD309+/CD45+) and endothelial colony-forming cells (ECFCs, CD34+/CD309+/CD45-). Serum NO and MDA levels, as well as serum SOD activity, were measured colorimetrically. Serum TNF-α levels were measured by the ELISA method. The ciplukan leaf extract reduced systolic and diastolic blood pressure, reduced the percentage of EPCs in the blood, increased serum NO levels, reduced MDA levels, increased serum SOD activity, and reduced serum TNF-α levels in L-NAME-induced hypertensive rats. It is concluded that ciplukan ethanol leaf extract exerts protective effects on L-NAME-induced hypertensive rats. These study results can strengthen the scientific basis of using ciplukan leaf ethanol extract to treat hypertension.

Abstract

Objective: Tamarindus indica is one of the famous natural plant that has various effects on health. Recent research has begun to examine the effect of Tamarindus indica on cell malignancy. One of the things that happens in cell malignancy is an increasing ability of cell migration, which is characterized by increasing expression of Snail2 and MMP-9. This study was conducted to determine the effect of extract of Tamarindus indica Fruit Extract (TIFE) towards Snail2 and MMP-9 expression in JEG-3 cells.Methods :.TIFE were extracted by using methanol. JEG-3 cells were grown in culture medium treated with TIFE (at 0, 25, 50 and 100 µg/ml). Immunofluorescence staining was carried out, and observed under Confocal Laser Scanning Microscope.Result: Snail2 expression after treatment with TIFE 50 and 100 µg/ml higher than control- (P=0.015 ; P=0.001) whereas at MMP-9 expression there was a biphasic pattern, in which the MMP-9 expression peaked at concentration of 50 µg/ml (P=0.002), then MMP-9 expression decreased at 100 µg/ml concentration. Abstract

Background: Meningioma is the most common intracranial tumor in adults. Progesterone plays a role in the cell development and proliferation of hormone-associated tumors, such as meningiomas. Significant morbidity and mortality can occur, when the lesion is localized in an area that is difficult to undergo surgical resection or is highly infiltrative in the surrounding tissue. Many studies have been published on the molecular properties associated with meningioma aggressive behavior, including matrix metalloproteinase (MMP), but research on the relationship between PR expression and MMP-9 has not been widely carried out. Methods: This study used a cross-sectional analytic observational study design. The analysis used in this study includes descriptive analysis and bivariate analysis. The data obtained was processed using SPSS software. Results: Spearman's correlation test for PR expression with WHO grading in meningioma, obtained p = 0.492 and correlation coefficient -0.117. This shows a correlation that is not significant. Spearman's correlation test for MMP expression with WHO grading in meningioma, obtained p = 0.16 and a correlation coefficient of 0.236. This shows a correlation that is not significant. Spearman's correlation test between MMP9 expression and WHO grading in meningioma obtained p = 0.008 which indicated that the correlation between PR and MMP9 values ​​was significant with a correlation value of -0.427. Conclusion: There is no significant correlation between PR expression and WHO grading. There was no significant correlation between MMP9 expression and WHO grading. There is a significant correlation between PR expression and MMP9.

Abstract

Background: The high number of Out of Hospital Cardiac Arrest (OHCA) worldwide, and Indonesia is not followed by high life expectancy and bystander. The role of bystander influences the high life expectancy on OHCA. The flipped learning method can increase knowledge and skills and the success of flipped learning.Objective: This study aimed to develop and test Aplikasi Hadapi Henti Jantung (Sipitung) app on the flipped classroom method in learning Basic Life Support more effectively to increase knowledge, skills, and self-efficacy.Method: The design of this research was Research and Development (R&D). This design consisted of 3 stages; stage I (literature study), stage II (application development), and stage III (application trial).Results: Based on the results of the literature review conducted following the American Heart Association guidelines on cardiac arrest, basic life support on adults required learning media that combines text, video, and audio. 4 features were included in developing and creating applications. In the experimental process, the researchers compared learning methods using flipped classroom with simulations in increasing knowledge, skills, and self-efficacy.Conclusion: "SIPITUNG" application in the flipped classroom method was an effective basic life support learning medium to improve knowledge, skills, and self-efficacy.

Abstract

Objective: In this study we aimed to know further mechanism and relation between the long-term usage of tamsulosin with the apoptotic mechanism on prostatic cells of BPH patients qualitatively. Material & Methods: A Case-control study from prostate resection specimens. Subjects were 20 male who underwent the TUR-P procedure, were divided into two groups equally, a group who got long-term administration of tamsulosin 0.4 mg, and one without tamsulosin. TGF-β1 and Smad-4 expression was measured by Enzyme-Linked Immunosorbent Assay (ELISA), meanwhile expression of NF- kB measured by immunohistochemistry assay. Results: There was a significant difference on the Independent T-Test that showed an increase in expression of TGF-β1 and Smad-4 expression and a decrease in expression of NF- kB p= 0.003 (p<0,05) from a group of patients who got long-term oral administration of tamsulosin 0.4 mg, compared with a group of patients who didn’t get any treatment. Conclusion: Long-term tamsulosin would induce an apoptotic process in the prostate cells shown by the activity of TGF-β1, Smad-4, and NF- kB.
