Abstract

Plant Nutmeg (Myristica fragrans) is an endemic plant in Indonesia originating from Banda Maluku Province. Nutmegs have long been known to people as spices. The nutmeg plants have been cultivated in North Sulawesi for a long time. This research has been conducted to obtain the position of species and the reconstruction of the phylogenetic tree of nutmeg plant (Myristica fragrans) from North Minahasa based on chloroplast DNA of matK gene. This research uses a descriptive method with a laboratory experiment. Total DNA was obtained by leaf tissue extraction and purification. The matK gene was amplified using the PCR method and visualized with automatic electrophoresis, sequencing was performed at FIRST BASE Singapore. The result of the analysis of the gene sequence of North Min Minerals using Barcode Of Life Database (BOLD) Systems shows that Pala Wori and Pala Patokaan have 99% similarity with Myristica maingayi and Myristica glubosa. While the reconstruction of the phylogeny tree shows the North Minahasa Pala has the closest resemblance to Myristica fatua.

Abstract

Honey, royal jelly, bee pollen, bee wax, propolis, bee glue, and bee venom/ apitoxin are among the items produced by honey bees. Honey, pollen, bees, eggs, larvae, and pupae are all stored in the A. dorsata Binghami beehive. Secondary metabolites and bioactive phenolic compounds (phenols, phenolic acids, esters, flavors, dihydroflavones, flavanols, chalcones, and phenolic glycerides), essential oils (mono and sesquiterpenes), polysaccharides, amino acids, amines, lactones, quinones, steroids, and vitamins) can all be found in honeycomb. The goal of this study is to see how A. dorsata Binghami honeycomb tea affects blood lipid levels in humans after 14 days of consumption in a small group of male and female volunteers who were given a 2 gram tea bag extract of A. dorsata Binghami honeycomb simplicia to drink once a day to see how it affects lipid levels (total cholesterol, LDL, HDL, and TAG) in human blood. Total cholesterol (140.56 mg/dL), triglycerides (140.93 mg/dL), LDL (85.92 mg/dL), and HDL (21.49 mg/dL) were all found to be high in humans. As a result, "The Use of A. dorsata Binghami's Honeycomb Extract on the Effect of Reducing Human Blood Lipids" can lower lipid levels.

Abstract

Bee Honey (Apis dorsata Binghami) Is a subspecies of A. dorsata which is only found in sulawesidan island and surrounding islands and until now has not been successfully cultivated. Various studies have been conducted species of trees used for nesting places, nesting behavior, biotic conditions, physical surrounding nesting trees and nesting characteristics. This study aims to obtain secondary metabolite components and scientific data of antioxidant activity of honeybee extract (Apis dorsata Binghami) derived from Bogani Nani Wartabone Bolaang Mongondow National Park, North Sulawesi Province. The method of extraction in this research was conducted using maceration with methanol solvent, phytochemical test and antioxidant activity analysis using method (1,1-definil-2-picrilhidrazil) by measuring absorbance absorption at maximum wavelength using UV-Vis spectrophotometer. The results of the phytochemical test show that the honeycomb (Apis dorsata Binghami) contains alkaloids, flavonoids, saponins and tannins. From the analysis results showed that with the increasing concentration of extracts, the greater the antioxidant activity is marked by decreasing the value of IC50. The honeycomb (Apis dorsata Binghami) has IC50 of 212.42 ppm in comparison with vitamin C IC50 of 59.34 ppm

Abstract

Nyamuk Aedes sp. adalah vektor utama penyakit demam berdarah dengue yang mempunyai kiprah besar pada terhadap penyebaran penyakit DBD di Indonesia. Pengendalian penyebaran vektor nyamuk dibutuhkan agar dapat menurunkan populasi vektor nyamuk Aedes sp. sehingga dapat menurukan masalah penyebaran DBD. Salah satu pengendalian vektor penyakit DBD yaitu mengunakan larvasida yang berasal dari tumbuhan. Ekstrak alkohol daun eceng gondok (Eichhornia crassipes (Mart.) Solms) diduga memiliki pengaruh larvasida Aedes sp., sebab mempunyai kandunan flavonoid, saponin dan tanin yang mengganggu perkembanan larva nyamuk Aedes sp. Tujuan dari penelitian ini untuk mengetahui efetivitas ekstrak (crude extract) alkohol 70% daun eceng gondok (Eichhornia crassipes (Mart.) Solms) terhadap mortalitas larva nyamuk Aedes sp. Penelitian eksperimental dilakukan dengan subyek dari penelitian yaitu 150 larva nyamuk Aedes sp. instar 3. Subyek penelitian dibagi menjadi lima grup pelakuan, konsentrasi kandungan yang digunakan di penelitian adalah 1000 ppm, 1500ppm, 2000 ppm, 2500ppm dan untuk kontrol positif adalah abate 1%. Pengamatan dilakukan 1x24 jam di ke 24 jam setelah perlakuan, perlakuan dilakukan replikasi sebanyak 3 kali. Data dianlisis serta dilakukan penghitungan nilai Lethal consentration mengunakan analisis probit. Berdasarkan analisis probit tidak ditemukan nilai dari LC50 di seluruh konsentrasi. Ekstrak (crude extract) alkohol 70% daun eceng gondok (Eichhornia crassipes (Mart.) Solms) tidak mempunyai efek larvasida nyamuk Aedes sp. Kesimpulan ekstrak (crude extract) alkohol 70% Daun eceng gondok (Eichhornia crassipes (Mart.) Solms) tidak mampu membunuh nyamuk vektor penyakit DBD nyamuk Aedes sp. di seluruh tingkat konsentrasi sesudah dilakukan pengamatan selama 24 jam.

Abstract

North Sulawesi local pigs have good potential because they have a high adaptability to the environment and low-quality feed obtained from processed food remnants of the population. The samples used in this study were local sows from North Sulawesi which were spread on traditional farms. The purpose of this study was to obtain data on body weight of livestock including the weight of organs in the digestive system (stomach, intestine and liver). Body weight data will be correlated with organ weight and intestinal length data. The results showed that the average body weight of the local pigs used in this study ranged from 16.65 ± 0.66 kg with the age of the cattle ranging from 1 year. The results of statistical analysis of the correlation test between body weight and organ weight of local pigs showed a very significant correlation value (P < 0.01). The correlation value shown in the study shows a very strong value in the correlation between body weight and the weight of an empty stomach and a strong correlation value is shown in the correlation between body weight and liver weight and intestine length, meaning that the weight of organs in the body in the digestive system will increase with increase in body weight. Body weight is one indicator of livestock productivity that can be estimated based on the size of the animal's body.

Abstract

One of the producers of honey bees in North Sulawesi comes from the Manembo Forest, South Minahasa. Today many ordinary people do not know the quality of good honey. This study aims to study the characteristics of bee honey from the South Minahasa Manembo Forest based on SNI. The research was conducted in April-July 2022. The research method used for testing the quality of honey was using qualitative research methods. The data obtained were then analyzed using descriptive data analysis. Determination of total flavonoid levels using quercetin as a comparison. The absorbance value was measured by UV-Vis spectro-photometer. The absorbance value is then entered into the linear regression equation. The results showed that the characteristics of Manembo forest honey, South Minahasa Regency based on SNI 2018 parameters met the standards and quality with the percentage of reducing sugar 68.53%, sucrose content 3.07%, insoluble solids 0.14%, diastase enzyme DN=4 and 0.03% ash content while those that do not meet the standard are water content with a percentage of 25%. The results of the analysis of the total flavonoid content showed a value of 1.6 mgQE/g. For further research, it is recommended to make an antidiabetic or anti-bacterial analysis in forest honey.

Abstract

Honeycomb can be used as a source of antibacterial, this is due to the content of secondary metabolites in the form of flavonoids in the honeycomb which serves as a protective determinant of honey quality. This study aims to isolate, identify and test antibacterial isolates of Indigenous bacteria from Apis mellifera nests against gram-positive bacteria S. aureus and gram-negative bacteria E. coli. This research used descriptive method and the research data were obtained through laboratory experiments. The results of the isolation stage of indigenous bacteria obtained seven (7) isolates AM1, AM2, AM3, AM4, AM5, AM6, AM7 Indigenous bacteria from Apis mellifera bee hives. AM1 and AM4 isolates were similar to the Enterobacter genus, AM2 isolates to Corynebacterium genus, AM3 and AM6 isolates to Paracoccus genus, and AM5 and AM7 isolates to Azotobacter Sp. The seven isolates of Indigenous bacteria have potential as antibacterial and the diameter of the inhibition zone against E. coli bacteria is AM1 (8.88 mm), AM2 (8.65 mm), AM3 (8.03 mm), AM4 (6.41 mm) , AM5 (9.07 mm), AM6 (9.53 mm) and AM7 (9.44 mm) while against S. aureus AM1 (9.08 mm), AM2 (9.23 mm), AM3 (9.15 mm), AM4 (8.70 mm), AM5 (10.44 mm), AM6 (11.56 mm) and AM7 (9.0 mm).

Abstract

Nutmeg leaf, lemongrass, and bee nests contain bioactive compounds that have potential as mosquito repellents. The purpose of this study was to learn about the bioactivity of a combination extract of nutmeg leaf, lemongrass, and bee nests, as well as to determine the efficient mosquito repellent lotion recipe from a combination of nutmeg leaf, lemongrass, and bee nests extract. The maceration process was used for extraction. Total flavonoid content was estimated by comparing it to the flavonoid quercetin. The absorbance value was measured with a UV-Vis spectrophotometer. The concentration of the extract combination in the lotion used was 0%, 2.5%, 5% and 10%. The total flavonoid concentration was found to be 16.67 mgQE/g. The 2.5% formula lotion has a protective power of 22%, the 5% formula lotion has a power of 62.96%, and the 10% formula lotion has a power of 82.41%. From these results, it can be seen that a formula with a concentration of nutmeg leaf, lemongrass, and bee nests as much as 10% can be the most effective mosquito repellent. The outcomes of this study are expected to help the community select environmentally friendly and safe insect repellents.

Abstract

Apis Dorsata Binghami is a honey bee that is often found in the forest, which is conserved by the community, but there are also honey bees that are just left alone; the community preserves these honey bees because their honey can be traded and can also be used as medicine. This study aims to determine the plants that are a feed source for Apis Dorsata Binghami in the Pinabetengan Forest, Minahasa Regency. This research was conducted in June-August 2022. The data collected came from field survey data using descriptive methods and identified plant species with values of Reactive Density (KR), Relative Frequency (FR), Relative Dominance (DR), and Index Significant Value (INP). Plant diversity as a food source for Apis Dorsata Binghami has different types of plants based on their habitus or the characteristics of the plants themselves.

Abstract

Research on insect phylogenetics is intricated by their similar morphology and significant genetic diversity. The cytochrome oxidase subunit 1 (CO1) gene is the most widely utilized mitochondrial DNA gene in the identification and study of animal molecular biodiversity. This study aims to identify and reconstruct the phylogeny of fruit flies from North Sulawesi using the cytochrome oxidase subunit 1 (CO1) gene. Fruit flies were obtained from 5 (five) areas in North Sulawesi, namely Siau (L1), North Minahasa (L2), Minahasa (L3), Southeast Minahasa (L4), and Bolaang Mongondow (L5). Fruit fly imago limbs were used as a tissue source for genomic DNA extraction. Genomic DNA extraction was carried out using the Quick-DNAâ„¢ Miniprep Kit manufacture protocol. The CO1 gene amplification was carried out by the PCR method, and the visualization of the amplicons was carried out by the 1.5% gel electrophoresis method. Nucleotide sequencing used a sequencing service at First BASE Singapore with a bidirectional sequencing method. CO1 gene amplification of each sample was visualized at 690 bp to 702 bp length. After analyzing the CO1 gene concession area using the MEGA XI program, it is found that Drosophila at L1 has 702 bp, L2 has 703 bp, L3 has 698 bp, L4 has 700 bp, and L5 has 697 bp. Based on alignment analysis using the BLAST method, it is found that the L1 fruit fly has a similarity rate of 99.29% (E=0.0) to Drosophila parapallidosa [MK659836.1]. The L2 fruit fly also has a similarity rate of 96.86% with Drosophila parapallidosa [MK659836.1]. The L3 fruit fly has a similarity level of 94.94% with Drosophila parapallidosa [MK659836.1]. The L4 fruit fly has a similarity rate of 94.43% with Drosophila parapallidosa [MK659836.1]. However, the L5 fruit fly shows a similarity rate of 96.86% with Drosophila rubida [EU493593.1]. The reconstruction results with the MEGA XI program using the Minimum Evolution model obtain two monophyletic groups where the fruit fly in Bolaang Mongondow is in a monophyletic group different from other fruit flies. The results of this study prove the variation in fruit fly species in North Sulawesi based on the identification of the CO1 gene.
