Abstract

Ipomoea aquatica F. are plant that has a hypoglycemic effect. Ipomoea aquatica F. was  formulated into tablet dosage forms to produce benefits in terms of practicality and accuracy of therapeutic doses.Disintegrant has effect to release the active substance rate of the tablet so that drug effects can be felt by patients with diabetic immediatly. The purpose of this study was to determine disintegration time of Ipomoea aquatica F. extract tablets with various levels of amilum manihot and determine the best concentration of amilum manihot based on the evaluation of physical properties of tablets.Ipomoea aquatica F. extracts was obtained from the maceration using 96% ethanol, than concentrated by rotary evaporator. The extracts were tested qualitatively by TLC with petroleum eter: aceton: dietil amin mobile phase (10:4:1), the phase silica gel F254 with Rf value about 0,625.  Tablets were formulated with amilum manihot concentration variation of 10%, 15% and 20% by wet granulation method. Physical properties of granules were tested such as powder flow, angle of repose, porosity and compressibility index.Tablets were tested to know physical properties such as weight uniformity, sizeuniformity, hardness, friability and disintegration time.The results showed that variaton levels of amilum manihot decrease disintegration time. Variation of amilum manihot was affected in disintegration time, and the optimumt formula with was achieved by 15% amilum manihot.

Abstract

Gummy candy parasetamol adalah sediaan permen kenyal mengandung parasetamol yang ditujukan untuk hancur dan larut di dalam mulut. Tujuan penelitian untuk mengetahui pengaruh variasi campuran pemanis sorbitol dan glukosa cair terhadap sifat fisik sediaan gummy candy parasetamol. Pembuatan sediaan menggunakan metode cetak tuang dengan perbandingan konsentrasi sorbitol dan glukosa cair: 50:50, 40:60, 30:70, 20:80, dan 10:90%. Data yang diperoleh dianalisis dengan deskriptif non analitik. Hasil uji sifat fisik dan uji stabilitas fisik sediaan dengan parameter bentuk, rasa, aroma, tekstur, pengamatan kekakuan, pengamatan kristal, dan pH sediaan. Formula 1 (50:50%) memiliki rasa yang sedikit manis dan stabil dalam penyimpanan selama sebulan di climatic chamber suhu 25oC dibandingkan formula yang lain. Hasil persentase uji hedonik (bentuk, rasa dan aroma) pada 28 responden dari formula 1 sampai formula 5 berturut-turut: 25; 60,7; 3,5; 0, dan 10,7%. Semakin tinggi kadar sorbitol maka sediaan yang dihasilkan semakin manis dan meningkatkan kekerasan. Sedangkan semakin tinggi kadar glukosa cair maka sediaan semakin kenyal dan mencegah kerusakan tekstur gummy candy parasetamol

Abstract

Oral drug absorption was highly influenced by dissolution rate, especially for poorly and insoluble drugs. Piroxicam is a nonsteroidal anti-inflammatory drug that is practically insoluble in water. The oral absorption rate of piroxicam is dependent on its dissolution rate in the GI tract. Polyvinyl Pyrrolidone (PVP) as surfactant can increase drug solubility by means of a micelle forming mechanism. The aim of the study was to know the influence of addition and variation of Polyvinyl Pyrrolidone (PVP) on Piroxicam absorption with everted intestinal sac method.  The  concentration of piroxicam solution was prepared by PVP in  1.0,2.0 and 3.0 % respectively. Crane and Wilson tube containing 75 ml of themucosal fluid was taken at 37 0 C in waterbath. Than, serosal solution of 1.5 mlwas added to intestinal sac by turned upside down and tied to a cannula, then put into the tube containing the mucosal fluid and constantly flowing oxygen gas.Serosal solution of 1 ml were taken every 15 minutes and then diluted with 2 ml of Ba (OH) 2 and 2 ml of ZnSO4 then centrifuge until 25 minute. The absorbant of supernatant was measured by UV spectrophotometer and data analyse wascalculated by one-way ANAVA.  PVP at 1%,2% and 3% increased piroxicamabsorption from the phosphate buffer pH 7.5 compared with negative control.According to P app , the values were 2.52 ± 0.43 cm/minute (negative control), 3.41 ± 2.17 cm/minute (1% PVP), 2.75± 1.14 cm/minute (2% PVP) and 4.77 ± 4.93 cm/minute (3% PVP) respectively. In conclusion. Lower doses of the surfactant (1%, 2%, and 3% PVP) significantly increased absorption of the drug by altering the membrane permeability.

Abstract

Ekstrak etanolik bunga kembang sepatu warna merah telah terbukti dapat menurunkan viskositas mukus secara in vitro. Tujuan dari penelitian ini adalah untuk mengetahui besarnya pengaruh varian konsentrasi ekstrak dalam menurunkan viskositas mukus dalam sirup secara in vitro dan untuk mengetahui besarnya konsentrasi ekstrak yang sebanding dengan sirup asetilsistein 2,00%. Ekstrak etanolik diperoleh dengan menggunakan metode maserasi kemudian dibuat dalam sediaan sirup  dengan konsentrasi ekstrak yang bervariasi (1,00; 1,25; 1,50; 1,75 dan 2,00%). Hasil penelitian menunjukkan bahwa sirup dengan konsentrasi ekstrak 1,00; 1,25; 1,50; 1,75; dan 2,00% tidak  menunjukkan adanya aktivitas mukolitik secara in vitro dengan menurunkan viskositas mukus usus sapi dan tidak ada yang sebanding dengan aktivitas mukolitik asetilsistein 2,00%.

Abstract

Kembang sepatu flower (Hibiscus rosa-sinensis L.) was fractionally used as expectorant. Based on Bioassay Guided fractionation, an active fraction was separated, and the fraction was identified is Alkaloid was the major compound based on TLC analysis. Viscosity value measured by viscometer was used as a Bioassay model of expectorant activity in vitro  and asetyl cysteine was used as positive control. Alkaloid content determination of the ethanolic extract was measured by TLC-Densitometric compared with standard curve of isolated alkaloid as the selected marker (Y=12.1360X+2901.4474). The alkaloid content in the ethanolic extract was determined as 2.35 ± 0.67 %.

Abstract

Essential oils of basil leaves that have spesific aroma can be used as a mosquito repellent. Lotions are more practical, easy to apply, easy to pour, and more efficient so repellent made in dosage form of lotion. The purpose of this study was to determine the repellent activity of volatile oil of basil leaf (Ocimum basilicum (L.) f. Citratum Back) against Aedes aegypti in lotion. Basil leaves are air-dried, then distilled by water and steam distillation for ± 6 hours. Essential oils with some concentrations(15% v / v, 25% v / v, and 35% v / v in 95% ethanol)are formulated in lotion. Physical properties (homogeneity, viscosity, dispersive power, and adhesion power) and repellent activity of lotions are tested. The data obtained are analyzed using the Kolmogorov-Smirnov test analysis, 2-way ANOVA test, and Turkey HSD test. The repellent activity is analyzed using One-Way ANOVA and Turkey test in level of 95%. The results showed that lotions with increasedessential oil concentration (15% v / v, 25% v / v, 35% v / v in 95% ethanol) are homogeneous and easily poured. Lotion with that increased concentration of essential oils of basil leaves (15% v / v, 25% v / v, 35% v / v in 95% ethanol) showed repellent activity with time protection of 3522, 4611, and 5435 seconds.

Abstract

This research aims to isolate and determine relative concentration of alkaloid in Hibiscus rosa-sinensis L flowers. using spectrodensitometry. Ethanolic extract of H. rosa-sinensis flowers was made by maceration methode using etanol 70% as solvent. Fractionation of alkaloid compound was using VLC (Vaccum Liquid Chromatography) and using organic solfent mixture of methanol-ethyl acetate in variation concentration. Isolate was used as standard curve, then the concentration of H. rosa-sinensis flowers extract was determined using spectrodensitometry with silica gel as stationer phase and ethyl acetate: methanol ( 1:5 v / v) as mobile phase. The relative concentration of alkaloid was calculated by using standard curve Y = 29023.75 X + 5809.35, r = 0.996. Determination of isolate showed the relative concentration of alkaloid was 0.210 ± 0.035 % and this can be used as marker compound.

Abstract

Hibiscus flower (Hibiscus rosa-sinensis L.) is efficacious in cough remedies. This flower has many variations , one of which is a red flower with a crown subject.  Ethanolic extract of the hibiscus flower has been studied and shown to have mucolytic activity. Fractionation will be closer in the process of finding active compounds. This study aimed to determine the effect of fractions containing alkaloids of red hibiscus flower varieties subject as mucolytics in vitro and determine the range of concentrations of the fractions equivalent to acetylcysteine effect of 0,1 %. This study included maceration, fractionation by VLC, the identification of fractions containing alkaloids, and mucolytic activity test fractions containing alkaloids. Mucolytic activity assay performed in vitro to decrease the viscosity of mucus cow. Test solutions were made with concentrations of fractions 0,4;0,6; and 0,8 %. Acetylcysteine 0,1 % was used as a positive control. Viscosity values were analyzed statistically using one -way ANOVA test , followed by the LSD test level of 95 % to determine differences between treatment groups. The results showed that the fraction containing alkaloids of hibiscus flower with a concentration of 0,4 % has not shown mucolytic activity, whereas at a concentration of 0,6 % and 0,8 % has been demonstrated in vitro mucolytic activity. Fractions containing alkaloids of hibiscus flower with a concentration of 0,6 % and 0,8 % have mucolytic activity equivalent to 0,1 % acetylcysteine in vitro.

Abstract

Ethylacetate insoluble fraction containing alkaloids from the hibiscus flower (Hibiscus rosa-sinensis L.) with a concentration of 0.6% w/v has mucolityc activity which is equivalent to 0.1% acetylcysteine based on the capasity to decrease viscosity of the mucus. The use of hibiscus as a new cough medicine done traditionally. Syrup of ethylacetate insoluble fraction containing alkaloids was the appropiate liquid dosage form that has some advantages for examples easy to use and fast absorption of the active substance. The study aims to determine the optimum formulation syrup ethanolic fractions containing alkaloids from hibiscus flowers using the method of Simplex Lattice Design. Ethylacetate insoluble fraction containing alkaloids from the hibiscus flower is obtained by using the Vacuum Liquid Chromatography (VLC). Simplex Lattice Design Methods used to optimize the formulation syrup ethylacetate insoluble fraction containing alkaloids from hibiscus flower with seven formulas with variations in the amount of glycerin, sorbitol solution 70%, and mucilago 0.5% CMC-Na. Physical stability was analized to find higher total response value (R total) as parameter of the optimum formula by Simplex Lattice Design Methods with Design Expert ® software version 8.0.2. Optimum formula syrup ethylacetate insoluble fraction containing alkaloids from the hibiscus flower obtained with the proportion of 25.376% glycerine, 51.985% of 70% sorbitol solution, and 22.639% of 0.5% CMC Na mucilago. Data obtained physical characteristics test compared with prediction value using Design Expert ® software version 8.0.2. Physical characteristics of optimum syrup ad the zero week compared with physical characteristics on the forth week using paired t-test. The result showed that physical properties of syrup formula optimum didn’t different significantly for easy for pouring and tasted response except for viscosity and acidity. Ethylacetate fraction syrup containing alkaloids from hibiscus flower optimization results are less stable during the four weeks of storage.

Abstract

Previous  study  indicated  that  ethylacetate  insoluble  fraction  of  ethanol extract  of  Hibiscus  flowers  (Hibiscus  rosa-sinensis L.)  were  able  to  dilute intestine cattle mucous in vitrothat was often associated with mucolytic activity.In  this  study,  syrup  preparation  of  various  concentrations  of  these  fractions (1.20,  1.60  and  2.00%)  was  tested  for  its  ability  to  dilute  of  mucous.  Each  of syrup  concentrations  was  mixed  with  20%  buffer  Phosphate  (pH,  7)  at  equalproportion (1:1 v/v), the resulting mixtures were tested for their ability to dilute mucous  in  vitro. Mucous  dilution  was  observed  by  flow  time  recorded in  the viscosimeter.  The  study  showed  that  all  syrup  concentrations  (1.20,  1.60, 2.00%) exhibited mucous dilution effect compared tonegative control and syrup concentration  at  2.00%  was  comparable  to  positive  control  acetylsistein  0.1% (p<0.05).
