Abstract

This research aimed to find relationship between numbers of Vibrio sp in sediment and in blood cockle. This research was conducted from March until May 2017. Survey method was used in this research. The data was analyzed descriptively and statistically using simple linear regression. The highest average log number of Vibrio sp in sediment was find at sampling point 2 (7.9033 CFU/g) and the lowest was at sampling point 4 (7.3187 CFU/g). Meanwhile in blood cockle the highest Vibrio number was at sampling point 3 (7.6889 CFU/g) and the lowest was at sampling point 1 (6.3411 CFU/g). Simple linear regression indicated the r value (correlation coefficient) was 0.31, showed was low which meant that the relationship between number of Vibrio sp in sediment and in blood cockle.Key words: Relationship, Vibrio sp, Sediment, Blood Cockle, Coastal waters

Abstract

Lactic acid bacteria (LAB) has been applied as probiotic mainly to prevent pathogens. This group of bacteria can be found in marine fish products including fermented, shrimp product, cincaluk. The objective of this research was to isolate LAB from cincaluk, and examined the activity against pathogen V. alginolyticus and A. hydrophila. The research was conducted on March-Juny, total of 12 isolates were obtained from cincaluk survey method. The result of research showed that all isolates round, creamy white, smooth-edged, raised surface, and the size is 1.0 to 3.0 mm. The physiological and biochemical test showed that get all isolates had a Gram-positive, catalase positive and are indole negative, 10 isolates were motile, as many as five isolates have citric positive, 10 isolates is Methyl Red (+), all isolates ferment sucrose and lactose, while only 9 isolates ferment glucose, and as much as 6 isolates that produce sulfide. All isolates were found to have inhibitory effects against bacteria V. alginolyticus and A. hydrophila. The highest antimicrobial activity is indicated by B isolates with an average diameter of 12.3 mm and inhibition zone antivibrio activity of 6.3 units. Isolates B with an average diameter of inhibition zone of 9.7 mm and 3.7 units antiaeromonas activity.Keywords: Isolation, Lactic Acid Bacteria, Cincaluk, V. alginolyticus, A. hydrophila

Abstract

                                                ABSTRACT Heterotrophic bacteria is a group of organism that play important role in biogeochemical processes in aquatic ecosystem.The bacteria are able to decompose and remineralize organic materials into a simple inorganic components which is turn to nutrients for phytoplankton, perifiton, and other aquatic microflora. This Study was conducted from September to October 2018. The purpose of this study wos to obtain bacterial isolates that can be used as an antibacterial against pathogenic bacteria and to identify bacteria found in River estuary Masjid Dumai of Riau Province. Based DNA character the result found10 bacterial isolates. The bacterial (1A, 1B, 1C, 1D, 1E, 1F,1J, 2C, 2D, and 2G). These bacteria were able to inhibit the pathogenic growth. DNA identification using PCR and 16S rRN sequence technique resulting found6 types of bacteria, namely Bacillus cereus strain ATCC 14579, B. anthracis strain ATCC 14578 16S,B. methylotrophicus strain CBMB205 16S, B. thuringensis strain YBT-1518, B. pseudomycoides strainNBRC, and B. anthracis strain C10.Keywords: Heterotrophic Bacteria, Antibacteria, 16S rRNA Sequence, Pathogens.

Abstract

ABSTRACT  granosa.This research was conducted from January to April 2019 by taking samples of blood cockles from the waters of the Asahan river, Tanjungbalai city, North Sumatra Province. The purpuse of this research were to know the metal concentrations of Pb, Cu and Zn in different station and different sizes of blood cockle’s shell (Anadara granosa), as well as to find out the level of metal pollution based on MPI (Metal Pollution Index). Analysis of metal concentrations of heavy metal was done by following procedurs outline by Yap et al., (2003). The results showed that the highest Pb metal concentration in meat of small blood clams was found at station 3 namely, 0.7100 μg / g and the low concentration was 0.4716 μg / g at Station 1. While the highest concentration at medium size was found at Station 2 namely , 1.3683 μg / g and low concentration was 0.8000 μg / g at Station 1. The highest Pb concentration at large size was found at Station 3 ie, 2,2200 μg / g and the lowest concentration was 1,2266 μg / g at Station 1. The calculated MPI value for the Asahan estuary research from this study was 3,7260  which was considered quite low compared to other regions. Keywords: Heavy Metal, Pollution, North Sumatra, Industry, Anadara

Abstract

ABSTRACT The aims of this study are to determine the condition of the coral reefs including percentage live coral cover, the diversity index, the uniformity index, and the dominance index of  lifeform coral at waters of Tidung Island Kepulauan Seribu Jakarta. This research has been conducting from 2018 May 7th - 17th at the waters of Tidung Island.The research station was divided into 3 stations based on environmental characteristics with 2 different depths ; 3 and 7 meters. The research station is a coral conservation area, snorkeling area, and natural area. Data of coral reef was obtained by using the Line Intercept Transect (LIT) method. The results showed that the conditions of coral reefs at stations 1 and 2 respectively in the bad category with the percentage of cover area are 3.52% and 10.84%. Whereas at station 3 the condition of coral reefs is in the medium category with the percentage of cover area is 30.01%. The low percentage of coral cover is caused by rising sea water temperatures, port activities and human activities. Water quality such as temperature, salinity, pH, current velocity and brightness at the study site is quite good for coral recovery and growth. Keyword : LIT, Coral Recovery, Diversity, Uniformity, and Dominance Index

Abstract

ABSTRAK Pseudomonas merupakan salah satu bakteri patogen opurtunistik yang menyerang makhluk hidup khususnya manusia dengan sistem kekebalan yang lemah.Tujuan penelitian ini adalah unutuk mengetahui ketahanan bakteri Pseudomonas terhadap antibiotik dan mengetahui kekerabatan antar spesies Pseudomonas sp yang diisolasi dari Perairan Laut Dumai menggunakan analisis 16S rRNA.Penelitian ini dimula dari bulan Januari 2019 – Februari 2019. Dari hasil penelitian Ciprofloxacin paling sensitif dalam menghambat pertumbuhan Pseudomonas dengan zona hambat terbesar 24,00 mm sedangkan yang resisten Sterptomycin dengan zona hambat 8,5 mm dan Erythromycin dengan zona hambat 4,7 mm. Setelah dilakukan analasis sekuensing DNA terhadap ketujuh isolat tersebut, diketahui isolat memiliki kekerabatan dengan bakteri Pseudomonas dengan strain yang berbeda. Tingkat homolog isolat terssebut berkisar 86,01% – 93,01 %.  Isolat P7 FPK Dumai, P21 FPK Dumai, dan isolat P22 FPK Dumai identik dengan Pseudomonas geniculate (89,69%, 93,01% dan 93,00%) sedangkan isolat P9 FPK Dumai identik dengan P. aeruginosa (92,40%). Isolat P1 FPK Dumai identik dengan P. stutzeri (86,16%). Isolat P13 FPK Dumai identik dengan P. monteilii (86,01%) sedangkan P20 FPK Dumai identik dengan P. fluorescens (89,70%) hal ini berarti isolat P7 FPK Dumai, P9 FPK Dumai, P13 FPK Dumai, P20 FPK Dumai dan isolat P1 FPK Dumai merupakan strain baru yang urutan basa dan nitrogennya belum masuk kedalam database sekuen genbank. Sedangkan isolat P21 FPK Dumai dan isolat P22 FPK Dumai mewakili identitas pada tingkat genus.    Kata Kunci : bakteri Pseudomonas, antibiotik, 16S rRNA

Abstract

Probiotic is an alternative subtance to prevent fish diseases which is environmentally friendly. The research purpose was to isolate potential probiotic to control bacteria pathogen in barramundi (Lates calcarifer) to control the bacteria pathogen. The probiotic bacteria candidates were isolated from intestine of barramundi by using the spread and streak methods on MRS and TSA media. The antibacterial activity was examinined by using disc diffusion agar method on Zobell 2216E medium. The results showed thirty eight isolates were found which consisted of coccus, white, orange and yellow colonies. Twelve isolates had inhibition against Vibrio alginolyticus,two isolates showed highest inhibition zone and antivibrio activities. The highest inhibition zone (10.04 ± 1,48 mm) and (9,50 ± 2,12 mm) and the antivibrio activities (4.04 mm) and (3.50 mm) were indicated by KPBU2-5 and KPMU2-6 isolates respectively.Keywords: Probiotics, Isolation, Barramundi, Antagonistic.

Abstract

                                          ABSTRACT   Petroleum pollution can originiate from oil spills during drilling, production, refining and transportation activities. One of the hardly to decomposed petroleum contaminants is hydrocarbons, once thehydrocarbons enter the marine environment, the chemical componend can be reduced by biological processes (bioremediation) using bacteria. The purpose of this study was to examine the ability of bacterial isolates from Selat Baru coastal sediments to degrade crude oil and produce biosurfactants. This research was conducted in August to October 2018. The ability of bacterial isolates to degrade oil and biosurfactant production was carried out in the Marine Microbiology Laboratory and Marine Chemistry Laboratory, Department of Marine Sciences, Faculty of Fisheries and Marine, University of Riau. Experimental method was used in this research.. Nine isolates of crude oil degrading bacteria were provided from previous researchers, namely isolates 11p31 (1A), 33p31 (3A), 22p41 (10A), 22p42 (4B), 33p31 (5B), 12p32 (9B), 22p31 (1C), 11p41 (9C), 31p41 (12C). Each isolate was examined the ability to degrade crude oil at  concentration of 1% (0.5 ml), 2% (1 ml) and 3% (1.5 ml). The highest crude oil degradation ability was at 1% concentration by isolates 22p41 (10A) and 11p41 (9C), and for 2% concentrations by isolates 33p31 (5B) and 3% concentrations by bacterial isolates 33p31 (3A). All bacterial isolates were found to produce biosurfactan through the Oil Speading Assay. Emulsifikation test using kerosene indicated kerosene. where the highest emulsification in test 1 is isolate 33p31 (3A). Then in test 2 the highest emulsification value was shown by isolate 33p31 (3A). Abstract

ABSTRACT               Azolla michrophylla is beneficial for Anabaena azollae because it can act as a host through the supply of oxygen and nitrogen that can be used. Nitrogen fixation carried out by A. Azollae is atmospheric nitrogen gas (N2) converted into ammonia (NH3). This research was conducted at the Laboratory of Marine Microbiology and Marine Chemistry, Department of Mariene Science, Faculty Of Fisheries And Marine Science, University of Riau in April to June 2018. The purpose of this research was to determine the abundance of A. azollae in the growth of A. microphylla plants and determine the levels of nitrate contained in the growth of A. microphylla in brackish water culture. Factorial randomized group design (RAK) was applied in this experimental. Temperature, salinity, and pH were water quality parameters measured. The results showed that the best growth of A. microphylla was obtained at a salinity of 0 ppt with a starter of         A. microphylla 3 gram with a value of 50.42 gram. The highest abundance of A.  azollae is found in salinity 3 ppt with a 3 gram level with the abundance of 5643 cells / ml. A. microphylla and A. azollae had sufficient nitrate levels for their growth in brackish water cultured at 0, 1, 2, and 3 ppt salinitis of in combination with  A. microphylla 1, 1.5 and 3 gram as a starters. The highest nitrate level was obtained on the 14th and 21st day with a value of 1.2625 mg / L and 1.4500 mg / L.   Keywords: Azolla microphylla, Anabaena azollae, Growth, Abundance, Nitrate.

Abstract

ABSTRACT This research was conducted from March until April 2019. The purpose of this study was to examine the resistance of E. coli bacteria toward antibiotics (ciprofloxasin, streptomycin and erythromycin) and to identify isolated from Dumai seawaters the bacteria genotypes. Resistance test showed that isolated  E1, E4, E6, E8, E11, E13, and E15 were resistant to antibiotic streptomycin and erythromycin with inhibitory zones ranging from 2.9 to 5.9 mm. While sensitivity to ciprofloxasin indicated the inhibition zones of 22.1 mm.  Analysis of E. coli  by using PCR method technique 16S sequences found three strains of bacteria had the highest homologi levels, those were E. coli strain NBRC 102203, E. coli strain JCM 1649 and E. coli strain U 5/41. E1-2 isolate has similarity to E. coli strain NBRC 102203, E4-2 and E6-2 isolates are similar to E. coli strain JCM 1649, E8-2 isolate to strain NBRC 102203, E11-2 isolates to E. coli strain U 5/41, E13-2 and E15-2 isolates to strain NBRC 102203.  Keyword : Escherichia coli, Resistance, Antibiotics, Genotype,  Sekuens 16S rRNA
