Abstract

Diabetes mellitus (DM) is a metabolic disease contributes to the health problem in Indonesia. The inhibition of α-glucosidase is one of the mechanisms of antidiabetic treatment. α-glucosidase inhibitor can be found in the duwet plants (Syzygium cumini L. Skeels)  that have been used traditionally in Indonesia as antidiabetic drug. However, due to the insufficient quantity and the long harvesting time, the forthcoming appliclation of duwet is consideraly not promising. The natural tendencies of endophytic microorganisms are identicl to the host plant. In this case the endophytic bacteria of the duwet plant is studied to determine its potential as an alternative to producing α-glucosidase inhibitors. A total of 14 isolates of endophytic duwet bacteria isolated tested the ability of α-glucosidase inhibitors using p-nitrophenyl-D-α-glucopyranoside. The result of  the absorbance based on breakdown of substrate that produce colored product and analyzed by spectrophotometric technique. All isolates had α-glucosidase inhibitor activity, the highest activity produced by isolates A21 and A22 with value of 69,18% and 69,22%. Keywords : inhibitor α-glucosidase, endophytic bacteria, Syzygium cumini

Abstract

Complete genome of chili has not been reported.The first step to study the genome is DNA isolation, so it is necessary to optimize the protocol to get an optimum DNA. This research aimed to optimize chili DNA isolation by variate the quantity and quality of chili leaves as row material and variate the grinding technique. DNA isolation was done using commercial kit without liquid nitrogen, and analyzed by agarose gel electrophoresis. The results showed that frozen chili leaf yields more DNA than fresh leaf, 0,1 g of leaf got optimum DNA, and grinding in mortar was better than in microtube.   Key words: DNA,  isolation, Capsicum annuum

Abstract

Diarrheal diseases is an endemic desease in Indonesia and potential desease of Kejadian Luar Biasa (KLB) which often leads to death. Enteropatogenic Escherichia coli (EPEC) is one of pathogenic bacteria which can cause diarrhea. Brotowali plant (Tinospora crispa, L. Miers) is one of traditional medicinal plants which is widely used to treat various diseases and and relatively have no side effects than chemical drugs. he purpose of this research is to know the antibacterial activity of Brotowali (Tinospora crispa, L. Miers) stems ethanol and ethyl acetate extract in inhibiting the growth of EPEC bacteria causing diarrheal disease. Brotowali extract was obtained by maceration method using ethanol and ethyl acetate solvent. The condensed extract obtained was dissolved in DMSO 100% until the concentration of the extract to 20%; 40% and 60%. Test of antibacterial activity used diffusion method using paperdisk. The ethanol and ethyl acetate extract of brotowali stems were each tested on EPEC bacteria. The results in this study are the inhibition zone on ethanol and ethyl acetate extract stem of brotowali plant to EPEC bacteria. The antibacterial activity of brotowali stem ethanol extract at 60% concentration showed the best effect compared to the concentration of 20% and 40%. The antibacterial activity of ethyl acetate extract of brotowali stem was as good at concentration of 20%; 40% and 60%. The antibacterial activity of brotowali stem ethyl acetate extract showed better results than brotowali stem ethanol extract

Abstract

Some traditional medicinal plants are known to have efficacy as a medicine for diabetes. Active compoundsproduced by a plant can be derived from endophytic microbes that live in these plants. One way diabetes drugs workis to prevent digestion of complex carbohydrates into glucose so that glucose intake is reduced. Alpha-glucosidaseinhibitor is a compound that can prevent the digestion of carbohydrates, especially starch into glucose. This studyaimed to test the inhibitory activity of alpha gluosidase PR-3 isolate, an endophytic bacteria from Momordicacharantia. The results showed that the crude extract (supernatant) from PR-3 has the capability of the enzyme alphaglucosidase inhibition that is equal to 61.2% compared with positive control compound acarbose 1 mg / ml. Theresults also showed that the use of maltose as carbon source produce the highest an alpha glucosidase inhibitor(54,97%), followed by the starch (47.77%), glucose (31.97%), fructose (44.14%) and sucrose (27.7%).

Abstract

Onions (Allium cepa L.) is one of the leading horticultural commodities in Indonesia and is often used as seasoning and traditional medicine. Onion has a high economic value and fluctuating prices so that domestic onion production needs to be improved, one of them with a presence of endophytic bacteria that act as plant growth promoting agent or Plant Growth Promoting Bacteria (PGPB). Endophytic bacteria isolated from the root, leaves, and bulbs. In this research has been tested endophytic bacteria of onion plants from Garut regency which has PGPB factors such as able to dissolve phosphate, and produce HCN. The results obtained 251 isolates of endophytic bacteria. Based on the characterization results, the superior isolates capable of dissolving phosphate with an average diameter of 0.45 cm is isolate II.B.1D.3, and 11 isolates capable of producing high HCN. These isolates can be used as PGPB agents so that they can be useful in increasing plant growth and onion production and biocontrol in suppressing pathogens. Keywords: PGPB, endophyte, onion, phosphate, HCN

Abstract

Tea Mistletoe (S. Atropurpurea BL. Dans) is a medicinal plant species used as anticancer because it contains some flavonoids compounds are chalcones, c-glycoflavonols, chatechin and quercetin. The ability of endophytic bacteria to produce similar bioactive compounds with its host plant is potential source to get flavonoids compounds. This research aims to know ability of endophytic bacteria in produce flavonoids compounds in vitro. Each isolate was fermented in submerged culture with 0.1% soluble starch, 0.5% pepton, and 0.15% yeast extract medium for 5 days, then extracted with ethyl acetate. Flavonoid content of the extract then was tested qualitatively and confirmation test used thin layer chromatography. Qualitative test results showed that crude extract from isolates B4, B5, B10, B17, and B19 positive containing flavonoids. The most potent extract were B10 and B19 tested by thin layer chromatography. Two of these extracts had the same Rf value with quercetin, thus endophytic bacteria from tea mistletoe can produce flavonoids in vitro.

Abstract

Microalgae Chlorella pyrenoidosa and Dunaliella salina have been used as natural aquaculture food supplement because the previous contains 60,5% proteins and 180,8 mg/100g β-carotene and the other  were accumulated β-carotene by 95% from their total carotenoid.  Carotenoid  production can be improved by protoplast fusion technique. The aim of the research was conducted protoplast fusion of from C. pyrenoidosa and D. salina in order to gaining boarder salinity spectrum for natural aquaculture food supplement . The research metodology consist of protoplast  isolation followed by protoplast fusion process  induced by PEG6000 and regeneration of  fusant.  Protoplast fusion was done in three different PEG incubation time that are 15, 30 and 45 minutes.  The fusants were grown in 2 different medium, sea water media and fresh water media.  Research result shows that optimal fusion incubation time with PEG6000 is at 30 minutes.  Fusant can grown in both  medium and revealed higher β-carotene contents 2,008 µg/ml comparing with their parents.   Keywords: Protoplast fusion, Chlorella pyrenoidosa, Dunaliella salina, β-carotene.

Abstract

Pengendalian populasi nyamuk Aedes aegypti sangat penting dalam rangka pencegahan terjadinya wabah penyakit demam berdarah. Beberapa bakteri memiliki aktivitas kitinolitik sehingga berpotensi digunakan sebagai agen biokontrol terhadap nyamuk Aedes aegypti, karena komponen eksoskeleton larva nyamuk tersebut tersusun dari bahan kitin.Tujuan dari penelitian ini adalah untuk mendapatkan isolat bakteri kitinolitik akuatik lokal yang dapat digunakan sebagai biokontrol larva nyamuk Aedes aegypti.  Bakteri kitinolitik diisolasi menggunakan media selektif agar kitin. Sumber isolat diperoleh  dari berbagai sampel air yang diperoleh dari daerah Klaten, Boyolali dan Jepara. Isolat yang diperoleh diseleksi untuk mendapatkan isolat dengan aktivitas tinggi. Uji aktivitas bakteri terhadap larva nyamuk menggunakan media air mineral. Hasil penelitian diperoleh satu isolat dengan kode B6 yang mampu menyebabkan kematian larva nyamuk sebesar 97% dalam waktu 108 jam. Isolat ini berpotensi digunakan sebagai agen  bioinsektisida untuk pengendalian larva nyamuk A. aegypti. Keywords: Bakteri kitinolitik, demam berdarah, Aedes aegypti, biokontrol

Abstract

The intraspecies protoplast fusion of P. manshurica DUCC-015 was conducted in searching the fusant with greater inulinase production. Inulinase on Dahlia variabilis Willd tuber from Baturraden-Purwokerto showed inulinase activity 0,683 IU/mL. Inulinase is an enzyme that catalyzes the hydrolysis reaction of inulin polysaccharides into fructose and or fructooligosacarides. The aims of this research was revealing fusant from intraspecies protoplast fusion process of P. manshurica DUCC-015 followed by analyzing of their inulinase productivity and activity. The research metodology protoplast fusion process consist of lysis of cell wall, protoplast fusion and regeneration of intraspecies fusant P. manshurica DUCC-015. Analysis of fusant were done by Schiff Basic Fuchsin staining of fusant nuclei, also measuring the inulinase activity and inulinase production comparing with their parent. The inulinase activity will be analyzed by T-Test Independent Two Sample on 95% Confidence interval of the difference use Statistical Product and Service Solution programme (SPSS). The result of experiment gaining fusant with regeneration capability, ploidi diversity  of fusant, inulinase activity about 0,965 IU/mL while their parent 0,622 IU/mL and inulinase production 0,736  IU/mL comparing with 0,731 IU/mL during 42 hours incubations. The fusant indicated the increase of inulinase activity and production  compared with their parent.   Keywords: Pichia manshurica DUCC-015, fusant, inulinase    Fusi protoplas intraspesies Pichia manshurica DUCC-015 telah dilakukan untuk mencari fusan dengan produksi inulinase yang lebih tinggi. Inulinase pada umbi tanaman Dahlia variabilis Willd dari Baturraden-Purwokerto memperlihatkan aktivitas sebesar 0,683 IU/mL. Inulinase merupakan enzim yang mengkatalisis reaksi hidrolisis polisakarida inulin menjadi fruktosa dan atau fruktooligosakarida. Penelitian ini bertujuan untuk mendapatkan fusan hasil fusi protoplas intraspesies P. manshurica DUCC-015 yang memiliki aktivitas inulinase lebih tinggi. Rancangan percobaan fusi protoplas terdiri dari isolasi protoplas, fusi protoplas dan regenerasi fusan. Analisis fusan menggunakan pewarnaan  Fuchsin pada inti sel, mengukur aktivitas dan produksi inulinase fusan. Aktivitas inulinase dianalisis dengan Uji T Test Dua Sampel Independen pada taraf kepercayaan 95% menggunakan program Statistical Product and Service Solution (SPSS). Hasil penelitian menunjukkan fusi protoplas intraspesies P. manshurica     DUCC-015 menghasilkan aktivitas inulinase mencapai 0,965 IU/mL dibandingkan induk sebesar 0,622 IU/mL dan produksi inulinase 0,736  IU/mL pada inkubasi selama 42 jam. Fusan  mengindikasikan kenaikan aktivitas dan roduksi inulinasi dibandingkan induk. Kata kunci : Pichia manshurica DUCC-015, fusan, inulinase.

Abstract

Starch is an abundant carbon source in nature, and ?-amylase (1, 4-?-D-glucanohydrolase; EC 3.2.1.1), which hydrolyzes ?-1, 4-glucosidic linkage in starch-related molecules. Microbe ?-amylase production is a hydrolytic enzyme and one ofinterest in its microbial production has increased dramatically due to its wide spread use in food, textile, baking anddetergent industries in recent years. Here we report ?-amylase from marine bacterium which was purified andcharacterized, as well as analyzed its hydrolysis product on starch. The enzyme of Arthrobacter arilaitensis partiallypurified by acetone precipitation with 90% and ion exchange chromatography produced specific activity 0.25 U/mg and0.38 U/mg, and it?s purity rate increased until 1.14 fold compared with former crude extract. Purifed extracelluler amilasehad an optimum activity at temperature 50°C and pH 9.0. An apparent molecular mass was between 50-75 kDa, asestimated by zimogram electrophoresis.
