Abstract

Food spoilage can caused by pathogenic bacteria and can cause disease (food-borne pathogens). Foods that are vulnerable to pathogenic bacteria are foods that contain lots of protein, such as meat, milk and eggs. Listeria monocytogenes and Escherichia coli are commonly found in fresh meat. The solution for the problem is microbiological preservation by using Lactic Acid Bacteria (LAB). The purpose of this study is to isolate LAB from Apis mellifera honey, find out its characteristics, and test for antibacterial growth of L. monocytogenes and E. coli. LAB have been isolated with pour plate. Morphological characterization have been done by observing the colonies macroscopically and microscopically. LAB have been enumerated by Total Plate Count (TPC). Antibacterial Test have been done by well diffussion method. Experimental design was using Completely Randomized Factorial Design (CRFD). Statistical analysis have been done by univariate test with SPSS 16.0. 1st factor is LAB, 2nd factor is pH of supernatant and 3rd factor is pathogenic bacteria. Total 9 isolates of LAB were obtained from A. mellifera honey. LAB population is 9.6 x 105 CFU/mL. The result of screening LAB antibacterial assay is 2 potential isolates, they are MA 3 and MA 4. The best supernatant of LAB that could inhibit the growth of L. monocytogenes and E. coli is MA 3 with average diameters 12,05 mm and 12,07 mm. Pathogenic bacteria that is more sensitive is E. coli, because of the antibacterial compound from LAB is composed by organic acid that can break down the outer membrane and lower the pH, so pathogenic bacterial cannot be adapted to acidic environment.

Abstract

Food contaminated by pathogenic bacteria is very dangerous to consume especially if the bacteria can produce toxins. Staphylococcus aureus andSalmonella typhimurium are two examples of pathogenic bacteria that cause demage to food because they produce enterotoxins that can cause food poisoning to consumers. The use biological agents as antibacterial agents can be used as a solution to overcome this. Lactic acid bacteria (LAB) can produce compounds thet can inhibit the growth of pathogenic bacteria in food. This study aims to obtain local LAB isolates from the stomach of Apis mellifera in Indonesia and determine their characteristics as potential biopreservatif foods, to test the inhibition of LAB bacteriocin against Staphylococcus aureus and Salmonella typhimurium in vitro. The isolation method was carried out using multilevel dilution and streak plates on MRSA media. Morphological identification was carried out macroscopically on BAL and microscopic colonies with gram staining. Antibacterial test using the well method. LAB isolation result obtained eight isolatates. Antibacterial activitytest results for cells, all isolates showed inhibition of   Staphylococcus aureus and Salmonella typhimurium, for further antibacterial testing using 2 potential isolates namely PAM 3 and PAM 4. The two isolates were further tested for antibacterial with cell free supernatant using two treatments namely pH acidic and neutral, both isolates showed antibacterial activity at acidic ph better than neutral ph, wheares for the bacteriosin analysis test all isolates had no inhibition.

Abstract

Empowerment of Micro, Small and Medium Enterprises (MSME), requires model empowerment, which includes increasing existing MSME to be more advanced in order to be able to compete tightly in free market competition. Since ancient times, empon empon has been proven to have medicinal properties as herbal medicine, which is not only used as a medicine but also as a beauty treatment. One of the MSME that processes empon-empon into instant herbal medicine is Tiga Dara MSME which is located at Jalan Urip Sumoharjo No. 45 East Ungaran Subdistrict, Semarang Regency. Instant herbal powders produced by UD Tiga Dara include instant powders of Ginger, Turmeric, Temulawak, Mahkota Dewa, Kencur etc. The importance of maintaining body immunity during the COVID-19 pandemic is closely related to the availability of instant herbal powder products with empon-empon raw materials, especially ginger. During this pandemic, the demand for instant herbal medicine powder has increased, but Tiga Dara MSME still have limitations in the sales system. The method used in this community service is in the form of training and mentoring MSME in making web and social media as a means of marketing instant herbal powder products. This service aims to assist MSME in marketing their products in order to improve the economy of Tiga Dara MSME. With the existence of the Web and social media as one of the media for product marketing, it is hoped that it can increase sales from MSME and can improve the community's economy so that it can achieve community welfare.

Abstract

Kegiatan Pengabdian Kepada Masyarakat Program PPPUD ini bertujuan untuk merintis dan membina UMKM Karya Bumi dalam mengatasi pemasaran susu hasil peternak di Mojosongo Boyolali. Mengingat banyaknya susu yang tidak terjual, maka sebagai alternative dengan dibuat susu pasturisasi dengan berbagai rasa yang mempunyai daya simpan lebih lama. Dengan diproduksinya sebagai susu pasturisasi ini akan meningkatkan nilai jual, daya simpan dan memperluas pemasarannya.Program juga bertujuan untuk mempercepat difusi teknologi dan manajemen UKM, serta mengembangkan proses link and match antara Perguruan Tinggi, UKM, Pemerintah dan masyarakat.Program ini diawali dengan perancangan dan pabrikasi alat produksi, utamanya adalah pasturizer dengan kapasitas 50 liter yang dilengkapi sinar UV agar produk yang dihasilkan steril. Adanya alat tersebut perlu diberikan bimbingan teknis kepada pegawai yang terlibat pada proses produksi. Disamping itu juga diberikan penyuluhan tentang produksi bersih dan halal. Guna menjamin kualitas produksinya maka diupayakan untuk mendapatkan sertifikat MD dari Balai Pengawasan Obat dan Makanan Provinsi Jawa Tengah. Untuk memenuhi persyaratan agar diperoleh sertifikat MD tersebut diperlukan pembinaan dan penataan ruang produksi dan perlengkapannya serta laboratorium yang diperlukan. Ruang produksi harus selalu steril dengan perlengkapan yang memadahi untuk keperluan produksi, pengepakan hingga siap untuk dipasarkan.Adanya program ini diharapkan dapat memberikan solusi dalam mengatasi masalah penjualan susu hasil ternak petani dan akan meningkatkan nilai jualnya yang berarti akan meningkatkan pendapatan peternak sapi perah yang ada.

Abstract

Susu adalah salah satu bahan makanan sumber hewani yang sangat penting bagi manusia dengan kandungan gizi yang tinggi dan lengkap. Sentra peternakan sapi perah terbesar di Provinsi Jawa Tengah terletak di Kabupaten Boyolali sehingga sebagai tempat yang tepat untuk produksi susu sapi segar dan produk diversifikasinya. Karena kandungan gizi yang tinggi dan lengkap produksi susu menjadi perhatian yang sangat penting bagi pemerintah untuk menjamin bahwa susu tersebut aman dikonsumsi seluruh konsumen. Salah satu bentuk jaminan yang dikeluarkan pemerintah adalah NIE (Nomor Izin Edar). Susu dan produk olahannya seperti susu pasteurisasi, es krim, yougurt sebelum diedarkan harus mendapat izin Edar dari BPOM (Badan Pengawasan Obat dan Makanan). Permasalahan yang dialami oleh UKM Karya Bumi saat ini adalah belum memiliki NIE untuk produk susu pasteurisasi dan es krimnya sehingga masih kesulitan dalam memasarkan produknya. Program dari pengabdian masyarakat ini bertujuan untuk membantu usaha UKM Karya Bumi yang berada di Kabupaten Boyolali untuk mendapatkan NIE dari BPOM. Metode yang digunakan untuk mencapai tujuan program yaitu memberikan sosialisasi tentang prosedur mendapatkan NIE dari BPOM dan melakukan pendampingan dalam mengurus pengajuan NIE untuk produk susu pasteurisasi dan es krim. Hasil dari kegiatan ini adalah usaha mitra mendapatkan penyuluhan tentang prosedur pembuatan produk susu pasteurisasi dan es krim yang aman dikonsumsi, higienis, sehat dan bersih sehingga UKM Karya Bumi mendapatkan surat izin produk layak edar yaitu berupa NIE.

Abstract

Datura metel L. is a type of shrub that has a wooden stem, hard, and thick. Datura leaves contain chemical compounds alkaloids, saponins, flavonoids, and phenols. Based on the chemical content, datura leaves potentially as an alternative insecticide that can replace the use of synthetic insecticides. The use of synthetic insecticides in controlling the Aedes aegypti L. population has had a negative impact, such as environmental pollution, public health problems, and vector resistance. The purpose of this study was to study the effect datura leaf extract as a biochemical insecticide on inhibiting egg hatching and life cycles of Ae. aegypti. The research was conducted at Entomology Laboratory, Public Health Faculty, Universitas Diponegoro. This type of research was a true experiment with four replications and with concentration treatment i.e. 125, 250, 500, 750, 1000, and 1250 ppm. The subjects of this study were egg fertile of Ae. aegypti which were 25 eggs in each container and the total number of samples was 800 eggs. The used method for extraction was maceration. The result of probit analysis showed the activity of amethyst extract insecticide with LC50 = 199.34 ppm and LC90 = 749.08 ppm. The result of ANOVA showed a difference in the average number of non-hatching eggs due to exposure of datura leaf extract (P = 0,001). The percentage of Ae. aegypti egg hatching failure was lowest at concentration 125 ppm (41%) and the highest was at concentration 1250 ppm (98%). The survival of larvae, pupa, and mosquitoes was highest at concentration 125 ppm (49.18%, 55.17%, and 43.75% respectively). In conclusion, the datura leaf extracts potentially as a biochemical insecticide that inhibits eggs hatching and life cycles of Ae. aegypti.

Abstract

ABSTRACTInstant herbal medicine is a powder or granule form of herbal medicine that is usually made from sugar and spices which are combined with or without the addition of other food ingredients and food additives. Powdered herbal medicine has the advantage that it is practical and fast in serving, and has a relatively long shelf life. However, poor processing of herbal powder can lead to microbial contamination such as mold and yeast. This study aims to determine the presence of mold and yeast contamination contained in the red ginger and ginger powder herbs. The steps taken in this study were sampling, preparation of tools and materials, homogenization and sample dilution, and testing for mold and yeast levels. This research was conducted for 2 months from August to September 2020 at the Biotechnology Laboratory, Department of Biology, Faculty of Science and Mathematics, Diponegoro University. The results showed that the mold and yeast levels found in the red ginger powder herbal sample were 1 x 101 colonies / g, while the mold and yeast levels found in the ginger powder herbal medicine were 21 x 101 colonies / g. of the two samples have met the standards of BPOM RI Regulation Number 32 of 2019 so that it can be concluded that the red ginger and ginger powder herbs are still safe for consumption.Keyword :  Herbal powder, Contaminants, Molds and Yeasts

Abstract

Rhizospheric fungi can have serve as biological agents against plant pathogens Alternaria alternata, which infects carrot plants. Rhizospheric fungi are more abundant in organic lands. This study aims to isolate fungi from organic carrot’s rhizosphere that are able to suppress A. alternata’s growth and determine isolate rhizospheric fungi ability to inhibit A. alternata to identify potential biological control agents against A. alternata. This research was conducted at the Laboratory of Biotechnology and Basic Biology, Diponegoro University, between January–June 2023. Research methods include survey location, soil sampling, rhizospheric fungi isolation, pathogenic fungi isolation, creating growth curve, pathogenicity tests, antagonism tests, and fungi identification. This study used a complete randomized design (CRD) and was analyzed using ANOVA at the 5% level of significance and Duncan's hoc-posttest. Fungi isolation from organic carrot plant’s rhizosphere resulted in 13 isolates consisting of six genera: Penicillium, Aspergillus, Paecilomyces, Myrothecium, Trichoderma, and Simplicillium. The antagonism test showed that eight antagonistic isolates, including the genus Penicillium (WO1, WO6, WO10), Paecilomyces (WO5), Myrothecium (WO7), Trichoderma (WO9), and Aspergillus (WO11, WO13), where WO1, WO6, WO10, and WO11 have a medium percentage of inhibition (44.12-57.84%), while WO5, WO7, WO9, and WO13 have a high percentage of inhibition (64.29-76.06%). The eight isolates showed antagonism mechanisms, including antibiosis (WO1, WO5, WO6, WO7, WO10, and WO11) and parasitism (WO9 and WO13).

Abstract

Kegiatan ini bertujuan agar Kelompok UMKM Ngudi Makmur 1 dapat memproduksi asap cair sendiri dan dapat mengaplikasikannya dengan benar. Kegiatan dilakukan dengan metode survei sekaligus observasi, sosialisasi, praktik, dan pendampingan produksi. Dengan terlaksananya sosialisasi terkait pengaplikasian asap cair ke pertanian dan peternakan di UMKM Ngudi Makmur 1 dan dilaksanakannya sesi tanya jawab hal tersebut mampu menjawab semua kebingungan dan keresahan anggota UMKM dalam pengaplikasian dan manfaat dari digunakannya produk asap cair, anggota juga jadi bisa mengetahui bagaimana efektifitas yang didapat dari asap cair itu sendiri, bagaimana cara produksi dan bahan apa saja yang diperlukan dalam proses produksi. Setelah dapat memproduksi dan mengaplikasikan asap cair, kemudian dilakukan pendampingan pemasaran produk. Pendampingan yang dilakukan di antaranya yaitu perhitungan harga pokok produksi (HPP), pembuatan dan pengoperasian marketplace dan digital marketing dan pembuatan label kemasan produk. Kegiatan sosialisasi asap cair berlangsung dengan baik dengan tingkat antusiasme anggota UMKM yang tinggi dan sesi diskusi yang berjalan dengan interaktif.
