Abstract

Aspergillus parasiticus, the causative agent of Aspergillus cob rot, and Mucor circinelloides, which causes Mucor stem rot, are types of pathogenic fungi that have the potential to reduce the quality of corn (Zea mays L) harvests. An effective and environmentally friendly approach to control these pathogens is to use biological agents, such as Trichoderma harzianum. This study aims to evaluate the inhibitory potential of T. harzianum against A. Parasiticus and M. circinelloides using antagonism tests with dual culture methods in vitro. The parameters used in this study were measurements of the percentage of the inhibition rate of the biological agent T. harzianum against pathogenic fungi. The results of the antagonism test showed that the biological agent T. harzianum had moderate inhibition against A. parasiticus and M. circinelloides with the percentage of each inhibition during the five days incubation period are 32.5% and 42.38%. This indicates that T. harzianum has the potential as a biological agent in controlling A. parasiticus and M. circinelloides in corn plants.

Abstract

Red chili (Capsicum annuum L.) is a plant that is widely used for food needs, medicine, and also has the potential as an antimicrobial agent. Fusarium oxysporum is known as a soil-borne phytopathogen that causes fusarium wilt disease that infects many plants, causing major losses in infected plants. This research aims to determine the antifungal potential of ethanol, acetone and chloroform extracts from red chili fruit (Capsicum annuum L.) against Fusarium oxysporum mold which infects potato plants. Red chilies (Capsicum annuum L.) were extracted using the maceration method. The yields of ethanol, acetone and chloroform extracts were respectively 15.3%, 13.43% and 14.56%. Based on antifungal tests on Potato Dextrose Agar (PDA) media, the three extracts in the same concentration, namely 100%, were unable to inhibit Fusarium oxysporum. This is possibly because Capsicum annuum L. is known to have antimicrobial abilities in a narrow spectrum, while Fusarium oxysporum is generally a stress-adaptive pathogen. Controlling Fusarium oxysporum by using red chili extract cannot be an alternative for controlling fusarium wilt disease.

Abstract

Fusarium and Colletotrichum are pathogenic fungi that attack potato and chili plants. Farmers often use synthetic fungicides to control the disease, while the continuous use of synthetic fungicides can cause negative impact to environment. This study aimed to test the effectiveness of plant disease control by using coconut shell liquid smoke. Inluded rejuvenation of isolates, characterization of tested isolates, and liquid smoke antifungal tests using the well diffusion method, and data analysis. Five formula concentrations were applied namely 0% (negative control), 10%, 20%, and 30% concentration, and antracol as positive control. The measurement of inhibition (mm) was carried out after 7 days of incubation. The result of in vitro effectiveness test showed that the liquid smoke had the ability to inhibit the growth of pathogenic fungi Fusarium foetens, F. moniliforme, and Colletotrichum capsici with the best concentration of 30%.

Abstract

Plant Growth Promoting Rhizobacteri (PGPR) memiliki kemampuan untuk meningkatkan pertumbuhan serta meningkatkan ketahanan terhadap hama dan penyakit pada tanaman. Bacillus altitudinis P-10 dari rhizosfer tanaman padi persawahan organik di Kecamatan Banyubiru, Kabupaten Semarang, Jawa Tengah memiliki potensi sebagai PGPR yang memiliki kemampuan menghambat fitopatogen Xanthomonas oryzae dan Xanthomonas campestris, serta diprediksi memiliki gen untuk produksi antibiotik peptida non ribosomal. Karakterisasi B. altitudinis P-10 dilakukan untuk mengetahui potensinya sebagai PGPR melalui uji kualitatif yang meliputi aktivitas kitinolitik dan selulolitik, produksi IAA (Indole Acetic Acid), produksi HCN (Hidrogen Sianida), kemampuan melarutkan fosfat dan produksi senyawa antibiotik. Hasil uji kualitatif menunjukkan B. altitudinis P-10 memiliki aktivitas selulolitik, kemampuan menghasilkan IAA dan kemampuan melarutkan fosfat serta produksi senyawa antibiotik dari golongan peptida non ribosomal yaitu bacitracin, gramicidin, surfactin, fengycin dan bacillomycin D. Bacillus altitudinis P-10 memiliki karakteristik sebagai PGPR melalui kemampuannya dalam memproduksi IAA, melarutkan fosfat, aktivitas selulolitik dan produksi antibiotik. Hal tersebut menunjukkan potensi B. altitudinis P-10 sebagai PGPR.

Abstract

Trichoderma harzianum merupakan jamur antagonis yang dimanfaatkan sebagai agen hayati pengendali penyakit pada tanaman. Peranan T. harzianum yang begitu besar membutuhkan media yang efektif dan murah. Tujuan dari penelitian ini adalah mengetahui hasil media organik terbaik yang dapat dijadikan media alternatif pengganti PDB pada perbanyakan miselium jamur T. harzianum. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) dengan empat perlakuan, yaitu media Potato Dextrose Broth (PDB), sekam padi, tongkol jagung serta campuran sekam padi dan tongkol jagung dengan tiga kali ulangan. Suspensi konidia Trichoderma harzianum sebanyak 2 x 108 ml/propagul diinokulasikan pada 150 ml media perlakuan kemudian miselium jamur dipanen pada hari keenam. Hasil analisis menunjukkan, penggunaan media PDB menghasilkan berat kering miselium T. harzianum yang paling besar yaitu 0.62 gram, sedangkan pada media sekam padi, tongkol jagung, campuran keduanya berturut turut yaitu 0,59 gram; 0,18 gram; 0,42 gram. Berdasarkan hasil tersebut maka dapat disimpulkan bahwa sekam padi dapat dijadikan media alternatif untuk perbanyakan miselium T. harzianum. Abstract

Dusun Indrokilo merupakan salah satu dusun yang terletak di Desa Lerep, Kecamatan Ungaran Barat, Kabupaten Semarang. Dusun Indrokilo memiliki potensi sumber daya alam yang besar khususnya di bidang pertanian. Mayoritas masyarakat sebagai petani kopi, gula aren, jahe, dan cengkeh. Dengan berlimpahnya hasil perkebunan kopi, maka dibutuhkan produk olahan kopi yang dapat meningkatkan perekonomian masyarakat Dusun Indrokilo. Tujuan diadakan pengabdian masyarakat ini yaitu untuk memberikan pengenalan dan pembuatan dalam pemanfaatan kopi menjadi masker wajah, meningkatkan pemahaman mengenai pemanfaatan komoditas kopi serta memberikan pelatihan dalam pembuatan masker wajah dari kopi. Pengabdian masyarakat ini menggunakan metode kaji tindak dengan tahap perencanaan, tahap pelaksanaan dan rancangan evaluasi. Hasil yang di dapat peserta dapat mengetahui manfaat dan cara pembuatan masker kopi serta antusias masyarakat dalam kegiatan pembuatan masker kopi. Hasil evaluasi menunjukkan bahwa 100% peserta puas setelah mengikuti kegiatan ini. Dari kegiatan ini dapat disimpulkan bahwa pengetahuan tentang pemanfaatan kopi sebagai masker wajah sangat dibutuhkan bagi masyarakat Dusun Indrokilo. Selain itu, masyarakat perlu mengetahui kandungan senyawa pada kopi yang dapat dimanfaatkan dalam bidang kecantikan.

Abstract

This study aimed to isolate and characterize fungi from the rhizosphere of carrots grown in non-organic systems that potentially produce indole-3-acetic acid (IAA), and to evaluate their biostimulatory potential in supporting environmentally friendly agriculture. The methods employed included rhizospheric sample collection from non-organic carrot fields, fungal isolation and purification using PDA media, morphological identification, as well as qualitative screening and spectrophotometric quantification of IAA production using Salkowski reagent. The results revealed six fungal isolates capable of producing IAA, namely Rhizopus stolonifer, Rhizopus oryzae, Trichoderma longibrachiatum, Trichothecium roseum, Pythium aphanidermatum, and Pythium inflatum, with varying levels of IAA production. Among these, R. oryzae produced the highest IAA concentration at 46.8 ppm, followed by P. aphanidermatum (41.8 ppm), T. longibrachiatum (27.3 ppm), P. inflatum (24.5 ppm), R. stolonifer (18.4 ppm), and T. roseum (17.9 ppm). These findings suggest that rhizospheric fungi from the non-organic carrot rhizosphere hold promising potential as IAA-producing bioagents to support sustainable and environmentally friendly agricultural practices.

Abstract

This research focused on the evaluation of raw broiler chicken meat texture and pathogenic antibacterial effect under the treatment of grade 2 coconut-shell liquid smoke (CSLS) as a food preservative. Compound identification of grade 2 CSLS was conducted using the GC-MS method. The main chemical compounds of CSLS were polyunsaturated fatty acid derivates, fatty acid and phenol. The evaluation of texture was conducted on raw broiler chicken meat under the treatment of grade 2 CSLS during storage at 25 ℃ for 7 days. The effect of antibacterial activity of grade 2 CSLS with different concentration (i.e., 5%, 25%, 50%, and 75%) were tested using Gram-positive bacteria on several pathogenic bacteria such as L. monocytogenes, S. aureus, P. Aeruginosa, and E. coli. The results indicated that grade 2 CSLS influenced the inhibited zone of L. monocytogenes, S. aureus, P. Aeruginosa, and E. coli respectively with linear correlation. The optimal concentration of grade 2 CSLS resulted in a concentration of 50%, which was the most optimal against L. monocytogenes, S. aureus, P. Aeruginosa, and E. coli as pathogenic bacteria strains.

Abstract

Datura metel L. is a type of shrub that has a wooden stem, hard, and thick. Datura leaves contain chemical compounds alkaloids, saponins, flavonoids, and phenols. Based on the chemical content, datura leaves potentially as an alternative insecticide that can replace the use of synthetic insecticides. The use of synthetic insecticides in controlling the Aedes aegypti L. population has had a negative impact, such as environmental pollution, public health problems, and vector resistance. The purpose of this study was to study the effect datura leaf extract as a biochemical insecticide on inhibiting egg hatching and life cycles of Ae. aegypti. The research was conducted at Entomology Laboratory, Public Health Faculty, Universitas Diponegoro. This type of research was a true experiment with four replications and with concentration treatment i.e. 125, 250, 500, 750, 1000, and 1250 ppm. The subjects of this study were egg fertile of Ae. aegypti which were 25 eggs in each container and the total number of samples was 800 eggs. The used method for extraction was maceration. The result of probit analysis showed the activity of amethyst extract insecticide with LC50 = 199.34 ppm and LC90 = 749.08 ppm. The result of ANOVA showed a difference in the average number of non-hatching eggs due to exposure of datura leaf extract (P = 0,001). The percentage of Ae. aegypti egg hatching failure was lowest at concentration 125 ppm (41%) and the highest was at concentration 1250 ppm (98%). The survival of larvae, pupa, and mosquitoes was highest at concentration 125 ppm (49.18%, 55.17%, and 43.75% respectively). In conclusion, the datura leaf extracts potentially as a biochemical insecticide that inhibits eggs hatching and life cycles of Ae. aegypti.
