Abstract

Upaya peningkatan kualitas gizi bahan pakan, terutama yang berkaitan dengan serat kasar yang tinggi dan terdapatnya zatantinutrisi seperti asam sianida (HCN) dalam kulit singkong dapat dilakukan dengan melakukan proses degradasi mengguna-kan agen biologis (biodegradasi). Agen biologis yang mampu bersifat selulolitik dan sekaligus mampu meningkatkan kualitas nilai gizi bahan pakan, salah satunya adalah mikroorganisme dalam cairan rumen sapi. Penelitian telah dilaksanakan di Laboratorium Mikrobiologi Fakultas Matematika dan Ilmu Pengetahuan Alam (MIPA). Penelitian dilakukan dalam dua tahap, yaitu: 1) Tahap isolasi dan pemilihan bakteri yang memiliki indeks selulolitik terbaik dan 2)Tahap pengujian kemampuan degradasi pada kulitumbi singkong. Penelitian tahap 1 dilakukan menggunakan metode pengenceran berseri dan cawan tuang, selanjutnya data dianalisis secara deskriptif, sedangkan tahap 2 dilakukan menggunakan Rancangan Acak Lengkap (RAL) pola Faktorial dengan 3 (tiga) perlakuan yang masing-masing diulang sebanyak tiga kali. Analisis data dilakukan dengan ANAVA dan dilanjutkan dengan uji Duncan bila terjadi pengaruh perlakuan. Parameter yang diamati pada tahap 1 adalah indeks selulolitik terbesar pada isolat bakteri, sementara pada tahap 2 adalah kandungan serat kasar, pada produk biodegradasi menggunakan bakteri. Hasil penelitian tahap 1 diperoleh 13 isolat bakteri selulolitik. Bakteri cairan rumen sapi aerob yang memiliki kemampuan selulolitik tertinggi adalah Bacillus megaterium, Bacillus mycoides. Berdasarkan uji iodium didapatkan dua kandidat yang memiliki kemampuan selulolitik terbesar, yaitu CM2 dan CM5, dengan indeks selulolitik masing-masing 3 dan 3,5. Hasil penelitian tahap 2 menunjukkan terjadi penurunan serat kasar setelah proses fermentasi. Perlakuan B. megaterium dosis 1% menghasilkanpenurunan kandungan serat kasar kulit umbi singkong tertinggi yaitu sebesar 30,14%, dimana nilai kandungan serat kasar pada produknya adalah sebesar 9,11% .

Abstract

Untuk mengetahui apakah membran alginat dapat digunakan sebagai produk alternatif tekstil medispembalut luka primer, maka telah dilakukan uji pre klinis untuk mengobati luka pada kulit kelinci albino jantan,sesuai dengan standar yang berlaku (OCDC Guidelines for the testing of Chemicals, 404). Percobaan dilakukanterhadap tiga ekor kelinci albino jantan, yaitu dengan melukai bagian kiri dan kanan punggung kelinci. Bagiankanan punggung kelinci ditempelkan pembalut luka yang kontak langsung dengan luka, sedangkan bagian kirinyatidak (sebagai kontrol). Adapun waktu pengamatannya adalah 1 jam, 24 jam, 48 jam dan 72 jam. Dari hasilpercobaan diketahui bahwa membran dapat mempercepat penyembuhan luka dan tidak menyebabkan iritasi kulit,bahkan setelah tiga hari hampir tidak terlihat adanya goresan bekas luka.

Abstract

A series of studies on the hydrolysis of Sago starch for ethanol fermentation had been conducted. Hydrolysis of sago starch was carried out using sulfuric acid 2.5% and amylase(s) enzymes. The concentrations of sago starch used in this experiment were  5, 10, 15, 20, and 30% (w/v). The highest hydrolyzate  containing reducing sugar was used as substrate for ethanol fermentation by Saccharomyces cerevisiae FNCC 3012. The results indicated  that hydrolysis using 2.5% sulfuric acid for 120 min at 121 °C produced 6.6%  (w/v)  reducing sugar and hydrolysis using α-amylase and Dextrozyme DX produced more reducing sugar, 7% (w/v) and 17.1% (w/v), respectively. The fermentation of hydrolyzed sago starch by S. cerevisiae FNCC 3012 produced ethanol 7.98% (v/v).

Abstract

BACKGROUND: Iron-overload cardiomyopathy (IOC) is a major comorbidity in patients with chronic repetitive blood transfusion due to myocardial iron uptake that facilitated by calcium channels. As cardiac compensatory mechanism to IOC, we hypothesized the cardiac calcium channels expression would be increased and involved in cardiomyopathy progressivity. This study was aimed to investigate the gene expression of calcium channels in the heart of the iron overload mice model.METHODS: Mice were divided into three groups according to iron administration doses 0, 0.1, and 0.3 mg/day. Systolic blood pressure (SBP), diastolic blood pressure (DBP), and mean arterial pressure (MAP) were measured for the representation of cardiovascular outcomes. The heart tissues were harvested. Further mRNA levels of L-type calcium channels (LTCCs) and T-type calcium channels (TTCCs) were examined using semi-quantitative PCR. The expressions of cardiac calcium channels and blood pressure among the three groups were compared.RESULTS: The expressions of TTCCs in the two iron-injected groups were higher than the control group (p=0.018). The expressions of LTCCs were not different (p=0.413) among groups. SBP, DBP, and MAP of the iron-injected group were lower than the control group (p=0.025, p=0.011, and p=0.008, respectively).CONCLUSION: Iron administration affects the expression of TTCCs but not the LTCCs, accompanied by decreasing of systolic and diastolic blood pressure.KEYWORDS: cardiomyopathy, iron overload, L-type calcium channel, T-type calcium channel.

Abstract

Bacillus and Lactobacillus are bacteria that can be used in dry-stock probiotics in vannamei shrimp feed. In this study, it was performed to encapsulate Bacillus and Lactobacillus probiotic bacteria in several carrier materials, including rice flour, talc, and maltodextrin. The purpose of this study was to determine the type of carrier material that can provide the best probiotic character in the encapsulation process which will then be used in vannamei shrimp feed. The method used in this study is experimental method. The study was conducted with the preparation stage and the initial probiotic characterization test. Furthermore, Bacillus and Lactobacillus probiotic encapsulation process used spray drying method to produce powder inoculum, as well as advanced characterization test of bacteria that have been encapsulated. The parameters observed were the viability, resistance to temperature, pH and bile salts. Data were analyzed using ANAVA and Duncan Multiple Range Test. The results showed that all types of carrier materials used maintained the probiotic character of Bacillus and Lactobacillus after the encapsulation process. This can be seen from the character of probiotics that are in good category and deserve to be used as a probiotic agent after encapsulation process in the material of rice flour, maltodextrin, and talc.

Abstract

The purpose of this research is to isolate and characterize chemical compound from bark of mangosteen. Bark of mangosteen was macerated with methanol. The extract from that process was fractionated with n-hexane, methylen chloride, and ethyl acetate. Ethyl acetate fraction was separated and purified by vacuum column chromatography, gravitation column chromatography, and preparative thin layer chromatography. The relative pure compound was afforded from ethyl acetate fraction of mangosteen bark (8.5 mg) resulting from 1.5 kg of mangosteen bark. The yellow amorphous powder of compound melts at 114 ~ 116ºC (uncorrected). The purity of compounds was tested by 1 and 2 dimension thin layer chromatography which gave one spot on TLC plate. The ultraviolet-visible (in CH3OH solvent) spectrum showed absorption maximum at 318 nm (sinamoyl group/band I), 258 (shoulder), and 243 nm (benzoyl group/band II). Addition of NaOH caused batochromic shift of band I and band II predicted as two hydroxyl at C-4’ and C-7, respectively. The infrared spectrum displayed absorption bands of OH stretching at  3436 cm-1, C-H stretching at 2920 cm-1, C=O stretching at 1631 cm-1, C-O-C stretching at 1094 cm-1, and C-H aromatics bending at  970-800 cm-1. A molecular ion in the FABMS at m/z 271.36 [M+H] + was consistent for the molecular formula C15H10O5. The 1H NMR spectrum showed characteristic resonances of a flavone. Based on the result of phytochemical test and analysis of the spectrum, it is predicted that the compound belongs to flavone, a kind of flavonoids which has hydroxyl at C-5, C-7, and C-4.

Abstract

Sappan wood (Caesalpinia sappan L.) is a plant used by the community to mix drinking. The antioxidant index of Sappan wood extract has a higher value than commercial antioxidants to counteract oxidative free radicals and improve the body's defense system. The use of Sappan wood drinking in society is often used in daily so it is necessary to do a chronic toxicity test to observe the response to prolonged use of the system in the body. Kupffer cells are an important part of the innate immune system, acting as “scavengers” and phagocytes. A study that aims to see the chronic toxicity studies of the ethanolic extract of Sappan wood on the number of Kuffer cells in the liver has been carried out on 60 Wistar rats divided into 12 groups. The group was the negative control group, doses 100 mg/kgBW, 200 mg/kgBW, 300 mg/kgBW, 400 mg/kgBW and 500 mg/kgBW of extract Sappan wood which were divided into male and female groups. Each group was given ethanol extract of Sappan wood for one year orally. The results showed a significant increase in Kupffer cells in the female group at a dose of 100 mg/kgBW and the male group at 200 mg/kgBW with significant values, respectively, p<0.001 p = 0.004.
