Abstract

A phylogenetic relationship of Indonesian banana cultivars was conducted using trnL-F intergenic spacer of chloroplast DNA sequences. The ingroup was 39 banana accessions from various genomic groups in Indonesia consisting of 9 AA/AAA; 11 AAA; 2 BB; 13 AAB; and 4 ABB cultivars and 12 sequences of different taxa of Musa downloaded from GenBank consisting of 4 AA wild; 1 AA; 3 AAB; and 4 ABB cultivars. Two accessions from section Rhodoclamys and Australimusa were used as the outgroup. The 39 accessions of the ingroup and the 2 accessions of the outgroup were analyzed using primers of the trnL-F intergenic spacer. The results showed that sequence of the trnL-F region can clearly distinguish 40 banana accessions having the A-type chloroplast (AA wild; AA; AA/AAA; AAA; AAB; and ABB) from those 11 accessions having the B-type chloroplast (BB, AAB, and ABB). The A-type chloroplast accessions are most probably derived from female Musa acuminata, while the B-type chloroplast accessions may be derived from female M. balbisiana. However, the specific M. acuminata ancestor of each cultivar remains unidentified.

Characterization of Local Durian Accession Collected Hortimart Agro Centre, Central Java Using Molecular Markers Inter Simple Sequence Repeats (ISSR). Floribunda 5(7): 267–276. — The lack of morphological markers encourage the use of other markers that are considered more accurate, i.e molecular markers. The purpose of this study is to analyze the genetic diversity as well as determine the identity of the local durian accession collection of Hortimart Agro Centre, Central Java. Leaves of 41 durian accessions were isolated using CTAB method. DNA amplification was conducted with four primers of ISSR PKBT and the results were separated by agarose gel. Data were analyzed using NTSYSpc program version 2: 02. The results showed that DNA amplification using four primers of ISSR BKPT yielded 66 polymorphic bands. The results of the analysis showed that the genetic diversity of local durians of Hortimart collection is high because 100% of polymorphic DNA bands with variations of size 250–1500 bp. No local durian accession was found to have the same DNA100% profile, although four pairs of accessions were known to have a 96% high similarity. Five accessions, namely Petruk 2, Ontoseno, Semar, Arjuno, and Gondomono have specific DNA bands. The only accession of durian apart from the other group is Gondomono because it has only one band on ISSR PKBT 8 with size 750 bp which is not owned by other accession. Floribunda 5(7): 267–276. — Keterbatasan ciri morfologi mendorong penggunaan penanda lain yang dipandang lebih akurat, yaitu penanda molekuler. Tujuan penelitian ini adalah menganalisis keanekaragaman genetika sekaligus menentukan identitas aksesi durian lokal koleksi Hortimart Agro Centre, Bawen, Jawa Tengah. Daun 41 aksesi durian diisolasi menggunakan metode CTAB. Amplifikasi DNA dilakukan dengan empat primer ISSR PKBT dan hasilnya dipisahkan melalui gel agarose. Data dianalisis menggunakan program NTSYSpc versi 2:02. Hasil penelitianmenunjukkan bahwa amplifikasi DNA menggunakan empat primer ISSR BKPT menghasilkan 66 pita polimorfik. Hasil analisis menunjukkan bahwa keanekaragaman genetika durian lokal koleksi Hortimart tergolong tinggi karena 100% pita DNA polimorfik dengan variasi ukuran 250–1500 bp. Tidak ditemukan aksesi durian lokal yang memiliki profil DNA100% sama, meskipun empat pasang aksesi diketahui memiliki kemiripan tinggi 96%. Lima aksesi, yakni Petruk 2, Ontoseno, Semar, Arjuno, dan Gondomono memiliki pita DNA spesifik. Satu-satunya aksesi durian yang terpisah dari kelompok lainnya adalah Gondomono karena hanya memiliki satu pita pada ISSR PKBT 8 dengan ukuran 750 bp yang tidak dimiliki aksesi yang lain. Meskipun demikian, dendrogram memperlihatkan tidak ada identitas yang 100% sama sehingga setiap profil pita DNA dapat menjadi identitassetiap aksesi durian.  Kata kunci:  Durio zibethinus, penanda molekuler, keanekaragaman genetika.

Abstract

Efforts to overcome global warming and climate change are integrated with science learning with the socio-scientific issues method. Materials on global warming and climate change are included in an interactive e-book that integrates the peat ecosystem's socio-scientific issues (SSI). This study aims to determine the effectiveness of the Integrated Interactive E-book SSI Ecosystem Peat on learning outcomes and students' environmental care attitudes. The research subjects were 30 grade VII students of SMPN 5 Palangkaraya, Central Kalimantan. The result is that SSI's integrated interactive e-book is effective in improving learning outcomes. The results of the SPSS test with a significance value of 0.05 showed a value of 0.008. There is a difference in the mean of the pretest and posttest results, with an N-gain value of 1.66 with low criteria. SSI's Integrated Interactive E-book is effective to improve students' environmental care attitude with an average score of 89.8% with the criteria of being very caring

Abstract

The aim of community service carried out by KKN students in Kebondowo Village and Demakan Village, Banyubiru District, located around Rawapening, is 1) to provide easy use of water hyacinth; 2) provide practical skills in the use of social media for marketing; 3) reduce the skill level of the skill briefing. The methods used in completing the objectives are 1) training in material assistance, 2) practical assistance to finishing, 3) business analysis and marketing. The results of these mentoring activities are: 1) increased workforce capable of making handicrafts from water hyacinth in a number of forms; 2) product marketing at the Rawa Pening lake attraction, Bukit Cinta and provincial level bazaar and expo events; also 3) helps businesses make social media also join in and fill content with product images. Overall, the outputs from this service were obtained and strived to be owned, strived to be independent, production, and consultation with UNNES.

Abstract

Plantae material consists of three primary materials, namely moss, ferns, and seed plants. Moss plant material is difficult for students to understand. Hence, It is necessary to develop an e-book on moss plants that can provide learning solutions related to moss plant material. This study aims to measure the level of validity, feasibility assessment by High-School Biology Subject Teacher Forum (STF) in Semarang City, and student readability. The research and development method used is the 4-D development model. Several stages in the development research are define, design, develop and disseminate. The development stage includes expert validation, feasibility, and student readability tests. The research subjects are students of class X SMA N 12 Semarang. The data collection instruments are expert validation sheets, assessment sheets, and readability sheets. The analysis technique used is descriptive data analysis. The result of the e-book validity test for the material content, presentation, and language assessment aspects are 96% (very good), 96% (very good), and 94% (very good), respectively. The results of the assessment by learning media experts is 93% (very good). The e-book feasibility test by STF in Semarang is 95% (very good), and the e-book readability test is 90% (strongly agree).

Abstract

Pembelajaran di MAN Indramayu belum mengaitkan pada lingkungan sekitar, padahal kondisi sekolah dapat dijadikan pokok bahasan materi pencemaran lingkungan, karena terdapat lingkungan yang rentan terhadap pencemaran lingkungan. Oleh karena itu dibutuhkan pembelajaran dan tugas yang nyata pada lingkungan sekitar, sehingga siswa dapat aktif dan memenuhi ketuntasan hasil belajar. Tujuan penelitian ini menguji keefektivan pembelajaran bervisi SETS melalui praktikum identifikasi bioindikator Sungai Cimanuk terhadap ketuntasan hasil belajar aspek keterampilan siswa. Metode penelitian yang digunakan adalah eksperimen dan subjek penelitian adalah kelas X matematika ilmu alam (MIA) sebanyak 88 siswa (tiga kelas). Hasil uji keefektivan diperoleh nilai ketuntasan klasikal hasil belajar aspek keterampilan mencapai 93%. Data hasil belajar aspek keterampilan diperoleh nilai rerata 3,48 atau predikat (B+). Hasil ini menunjukkan pembelajaran bervisi SETS melalui praktikum identifikasi bioindikator Sungai Cimanuk efektif terhadap ketuntasan hasil belajar aspek keterampilan siswa.Learning in MAN Indramayu has not related the material in the surrounding environment yet. In fact, the condition of the school can be set as a subject matter of environmental pollution because there is an environment that is vulnerable toward the environmental pollution. Therefore it needs learning and the real assignment in the surrounding environment, so that the students can be active and achieve the completeness of learning outcomes. The purpose of research is to test the efectiveness by learning SETS-vision through practical bio-indicator identification of Cimanuk River will be effective toward the completeness of learning outcomes of students' skills aspects . The method used was experimental and research subjects are grade X math science (MIA) as many as 88 students (three classes). The effectiveness result of learning in getting the classical completeness score of learning outcome of skill aspect reach 93%. Learning outcome of skills aspect gets a mean score of 3.48 or predicate (B+). These results show that learning SETS-vision through practical bio-indicator identification of Cimanuk River effective toward the completeness of learning outcome of students’ skill aspect.

Abstract

Pembelajaran IPA di SMP Negeri 13 Magelang selama ini belum menerapkan pembelajaran bervisi konservasi dengan pendekatan scientific skill. Pembelajaran masih menggunakan metode ceramah sehingga kurang aplikatif dan kurang memotivasi siswa dalam belajar Hal ini menyebabkan karakter konservasi siswa belum berkembang dengan baik. Oleh sebab itu perlu dikembangkan perangkat pembelajaran biologi bervisi konservasi dengan pendekatan scientific skill melalui pengolahan sampah organik di sekolah dan menguji efektifitas serta kepraktisan perangkat pembelajaran untuk menanamkan nilai-nilai konservasi siswa. Metode yang digunakan adalah Research and Development dengan menganalisis kebutuhan kemudian mengembangkan produk dan diuji validitas, efektivitas, dan kepraktisanya hingga diperoleh produk final. Produk yang dikembangkan adalah perangkat pembelajaran bervisi konservasi  dengan pendekatan scientific skill yang  meliputi silabus, rencana pelaksanaan pembelajaran (RPP), bahan ajar, lembar diskusi siswa (LDS), dan lembar penilaian scientific skill. Hasil penelitian berupa perangkat pembelajaran IPA materi lingkungan hidup bervisi konservasi dengan pendekatan scientific skill pada pengolahan sampah organik di sekolah cukup valid, praktis dan efektif untuk diterapkan dalam proses pembelajaran. Hasil uji coba menunjukkan rata-rata respon siswa 97,57% atau sangat baik,  rata-rata nilai  scientific skills siswa  3,53 atau baik, hasil  t-test menunjukkan sig = 0,001 = 0,1% 5 % dan nilai rata-rata  konservasi siswa sebesar 89,43 berada dalam kategori sangat baik.   Science teaching learning at Sekolah Menengah Pertama Negeri  13 Magelang at present has not applied yet the conservation  vision, using scientific skill approaches. Teaching learning process still uses lecturing methods, consequently it is not applicative and motivating students. It also makes student conservation character is not developed yet. Because of the needs, this study was aimed at developing science teaching tools grounded in conservation using scientific skill approaches through student activity ‘making compost’ at schools, and  testing its effectiveness and practicability, in terms of  student conservation values.  The method used is research and development by analyzing needs and then develop products and test their validity, effectiveness, and convenience to the final product obtained. products developed are learning devices visionary approach to conservation with scientific skills include syllabi, lesson plans, teaching materials, sheet student discussions, and scientific skills assessment sheet. The results of the research are: the science learning device of living space in conservational vision using scientific skill approach is valid, practice and effective to apply in teaching learning process. The result of the experiment shows the average of students responses 97. 57% (very good), the average of students scientific skill values 3.53 (good), The result of T-test shows sig = 0.001 = 0.1% 5% and The average of students conservation values 89.43 in very good category.

Abstract

Abstrak: Desa Gondang, Kecamatan Karangreja, merupakan salah satu Desa penghasil kopi arabika dan robusta bermutu tinggi di Kabupaten Purbalingga. Produk kopi selama ini belum optimal dikembangkan untuk mencapai produk premium, karena keterbatasan pengetahuan dan teknologi proses. Kopi premium mempunyai nilai jual tinggi pada pasar nasional maupun internasional, baik dalam bentuk green bean, roast bean dan serbuk. Kopi yang dihasilkan di Desa ini bercirikan butir yang besar, bersih dan merata, berpotensi dikembangkan menjadi produk kopi premium. Penerapan teknologi tepat guna (TTG) proses pengolahan kopi premium, dengan melakukan penyuluhan, pelatihan penggunaan TTG, studi banding dan mengundang narasumber pengolah kopi dari UKM yang sudah maju. Mesin pengolah kopi premium yang dibantu kepada kelompok terdiri dari: (1) TTG mesin roaster kapasitas 20 kg/proses, dengan monitoring Internet of Thing (IoT) berbasis android; (2) TTG mesin sachet otomatis dengan variasi berat (30-250 gr); (3) TTG Sealing Continues untuk paking kopi ukuran diatas 500 gr; dan (4) Dry House dilengkapi panel solar sel, untuk stabilisasi suhu dan kelembaban. Selain itu dalam rangka penerapan mesin produksi otomatis, untuk layanan dan monitoring perkembangan kelompok telah dikembangkan Website modern untuk meningkatkan branding produk yang terintegrasi layanan penjualan online. Hasil pemberdayaan kelompok tani ini menunjukkan bahwa dari 11 orang anggota kelompok yang dilatih khusus pengolahan kopi premium, setelah dilakukan post test, diketahui 8 orang (73%) hasil sangat baik, 2 orang (18%) baik, dan 1 orang (9%) cukup. Hasil penerapan TTG menunjukkan 11 orang anggota kelompok Tani Bawono Lestari mampu meningkatkan variasi produk kopi arabika dan robusta masing-masing 3 jenis rasa khas yang diproses untuk dipasarkan pada premium market. Dampak penerapan proses pengolahan kopi premium meningkatkan nilai ekonomis produk sebesar 57%, harga produk kopi roastbean arabika meningkat dari rata-rata per kilogram Rp. 175.000/kg meningkat menjadi Rp. 235.500-250.000/kg, kopi robusta dari Rp. 75.000/kg menjadi Rp 117.000-120.000/kg, peningatan kapasitas produksi kopi olahan sebesar 73%, dari 175 kg/hari menjadi 306,25/hari.Abstract: Gondang Village, Karangreja District, is one of the villages producing high-quality Arabica and Robusta coffee in Purbalingga Regency. Premium coffee has a high selling value in national and international markets, both in the form of green beans, roasted beans, and powder. The coffee produced in this village is characterized by large, clean, and even grain, which has the potential to be developed into premium coffee products. application of appropriate technology for premium coffee processing by conducting counseling, training on the use of appropriate technology, comparative studies, and inviting resource persons from advanced SMEs. The premium coffee processing machines assisted by the group consist of: (1) an appropriate technology roaster machine with a capacity of 20 kg/process, with Android-based Internet of Things (IoT) monitoring; (2) appropriate technology automatic sachet machines with weight variations (30–250 gr); (3) appropriate technology sealing machines for packing coffee sizes above 500 gr; and (4) a dry house equipped with solar cell panels to stabilize temperature and humidity. In addition, in the context of implementing automatic production machines for service and monitoring group developments, a modern website has been developed to improve product branding that is integrated with online sales services. The results of the empowerment of this farmer group showed that out of 11 group members who were specifically trained in premium coffee processing, after the post-test, it was found that 8 people (73%) had very good results, 2 people (18%) had good results, and 1 person (9%) had enough. The results of the application of appropriate technology showed that 11 members of the Bawono Lestari Farmer Group were able to increase the variety of Arabica and Robusta coffee products, each of which had three distinctive flavors and was processed to be marketed at the premium market. The impact of using the premium coffee processing process raises the economic value of the product by 57%, raising the price of Arabica roast bean coffee products from an average of Rp. 175,000 per kilogram to Rp. 235,500 to 250,000 per kilogram and raising the price of robusta coffee from Rp. 75,000 per kilogram to IDR 117,000-120,000 per kilogram, resulting in a 73% increase in processed coffee production capacity from 175 kg per day to 306.25 kg per day.

Abstract

Permasalahan industri kecil kertas handmade bersumber dari belum optimalnya kuantitas dan kualitas produk yang pada akhirnya berdampak pada kontinuitas. Produksi kertas ini dipengaruhi cuaca, keterbatasan bahan dan teknik pembuatan kertas handmade yang pada akhirnya mempengaruhi kontinuitas produk. Konsumen kertas handmade yang umumnya pengrajin juga menghendaki kertas yang bervariasi dalam pembuatan produk berbahan kertas. Tujuan kegiatan adalah meningkatkan kemampuan industri kecil untuk meningkatkan kuantitas dan kualitas kertas handmade serta produk berbahan kertas handmade. Implementasi ipteks dalam pembuatan kertas handmade menghasilkan 6 varian kertas yang dibuat dari bahan baku tunggal maupun campuran dari mendong, rumput, limbah kertas dan debog pisang dengan pewarnaan sintetis maupun alami dan corak menggunakan berbagai bentuk daun. Pemanfaatan alat potong yang bervariasi dan tajam juga telah menghasilkan 6 jenis paper box berbahan kertas handmade, termasuk bentuk yang cukup sulit dibuat seperti bulat, oval atau bentuk jantung. Pembuatan produk untuk jumlah tertentu telah dikuasai sehingga industri kecil mampu menerima pesanan dalam waktu relatif cepat sesuai kebutuhan konsumen.

Abstract

Teknik PCR mempunyai kemampuan yang sensitif untuk deteksi keberadaan daging babi di dalam daging segar maupun produk olahan yang dicampur dengan bahan lain. Tujuan penelitian ini untuk mengetahui apakah produk bakso yang dijajakan di pusat kota Salatiga mengandung daging babi. Teknik stratified random sampling digunakan untuk mengambil sampel bakso yang dijajakan 13 warung bakso dari 25 warung bakso yang tersebar di pusat Kota Salatiga. Isolasi dan purifikasi DNA sampel bakso, daging sapi, dan daging babi menggunakan metode isolasi DNA jaringan hewan. DNA hasil isolasi dilanjutkan proses PCR menggunakan primer p14 untuk mengamplifikasi lokus PRE-1 pada genom babi. Proses amplifikasi DNA dengan program denaturasi awal pada suhu 93 °C selama 2 menit, diikuti 45 siklus terdiri atas denaturasi 93 °C selama 1 menit, annealing 62 °C selama 30 detik, ekstensi 72 °C selama 1 menit, kemudian diakhiri ekstensi 72 °C selama 2 menit. Produk PCR yang diharapkan muncul berukuran 481bp. Hasil elektroforesis gel agarose 1,2% pada produk PCR menunjukkan adanya pita DNA spesifik berukuran 481 bp pada daging babi dan sampel bakso nomor tiga belas, sehingga disimpulkan warung bakso nomor tiga belas produk baksonya mengandung daging babi.PCR technique has the ability to be sensitive to the detection of the presence of pork in fresh meat and processed products are mixed with other materials. The aim of this research to determine whether the product meatballs are sold in downtown Salatiga containing pork. Stratified random sampling technique is used to take samples of meatballs stall which sold 13 of the 25 meatballs stalls in the Salatiga City centre. Isolation and purification of DNA samples of meatballs, beef, and pork using DNA isolation method of animal tissue. DNA isolation results continue the process of PCR using primers to amplify p14 locus PRE-1 in the pig genome. DNA amplification process with initial denaturation program at a temperature of 93 °C for 2 min, followed by 45 cycles consisting of denaturation 93 °C for 1 min, annealing 62 °C for 30 seconds, extension 72 °C for 1 minute, then topped extension 72 °C for 2 minutes. PCR products were expected to appear sized 481bp. Results of a 1.2% agarose gel electrophoresis of PCR products indicate a specific DNA band sized 481 bp on pork and meatball sample number thirteen, so it concluded meatball stall number thirteen baksonya products containing pork.
