Abstract

A myokine is one of the proteins that are produced and released by myocytes in response to muscular contractions when doing physical exercise. One protein that is thought to function as myokine is FGF23.The purpose of this study was to determine the effect of acute physical exercise with moderate intensity on the expression of FGF23 gene in Wistar rat heart. This was an animal experimental study using 24 male Wistar rats that were divided into 4 groups:treatment groups that performed 30 minute acutephysical exercise with moderate intensity (20 m/min) for 3 days, 6 days, and 15 days and a control group without physical exercise. The study was conducted in the Animal Laboratory and Central Laboratory of Universitas Padjadjaran during the period of February to July 2019. Data observed were the FGF23 gene expressions in Wistar rats heart. Data were analyzed using Kruskal-Wallis and Mann-Whitney tests. The results from the Kruskal-Wallis test showed that acute physical exercise with moderate intensity did not increase the FGF23 gene expression in Wistar rat heart (p>0.05), and the average of relative ratios of FGF23/GAPDH gene expression were as follows: control (0.970±0,03), 3 days (0.992±0.03), 6 days (1.014±0.05), and 15 days (1.056±0.02). GAPDH was used in this study as a housekeeping gene since its expression is very constant. This study proves that FGF23 is more likely to take a role in the cardiac remodeling process, especially those associated with cardiac hypertrophy after chronic exercise with no effect observed after acute physical exercise with moderate intensity in Wistar rat heart. Pengaruh Latihan Fisik Akut Intensitas Sedang terhadap Ekspresi Gen FGF23 pada Jantung Tikus Galur WistarMiokin diproduksi dan dilepaskan oleh miosit sebagai respons terhadap latihan fisik. Salah satu protein yang diduga berfungsi sebagai miokin adalah FGF23. Penelitian ini bertujuan mengetahui pengaruh latihan fisik akut intensitas sedang terhadap ekspresi gen FGF23 pada jantung tikus galur Wistar. Penelitian ini menggunakan desain eksperimental dengan tikus galur Wistar jantan yang berjumlah 24 tikus. Tikus dibagi menjadi 4 kelompok untuk diberi perlakuan latihan fisik intensitas sedang (20 meter/menit) dengan durasi 30 menit, selama 3 hari, 6 hari, dan 15 hari serta kelompok tanpa latihan fisik. Penelitian ini dilakukan di Lab Hewan dan Lab Sentral Universitas Padjadjaran pada bulan Februari hingga Juli 2019. Ekspresi gen FGF23 pada jantung tikus galur Wistar dilihat dengan PCR. Analisis data menggunakan uji Kruskal-Wallis dan Mann-Whitney. Uji statistik tidak mendapatkan peningkatan ekspresi gen FGF23 setelah dilakukan latihan fisik akut intensitas sedang selama 3 hari, 6 hari dan 15 hari (p>0,05) dan didapatkan rasio relatif ekspresi gen FGF23/GAPDH otot jantung pada : kontrol (0,970±0,03), hari 3 (0,992±0,03), hari 6 (1,014±0,05), hari 15 (1,056±0,02). GAPDH digunakan dalam studi ini sebagai gen ‘housekeping’ karena ekspresinya yang sangat konstan. Studi ini membuktikan bahwa FGF23 mungkin lebih berperan dalam proses remodeling jantung, terutama yang berhubungan dengan hipertrofi jantung setelah latihan fisik kronik. Simpulan, tidak terdapat pengaruh latihan fisik akut intensitas sedang terhadap ekspresi gen FGF23 pada jantung tikus galur Wistar.

Abstract

BACKGROUND: Recently, coffee is widely used for preventing photoaging because of its antioxidant capacity. Among two kinds of coffee, robusta coffee has higher content of antioxidant such as chlorogenic acid (CGA) and caffeine. Researchs about robusta coffee bean effect on photoaging due to UVB radiation is still limited. Therefore, the aim of this study was to examine the effect of robusta extract cream (RE cream) on preventing wrinkle in mice induced by ultraviolet-B (UVB) radiation.METHODS: RE cream was made by mixing RE coffee with moisturizing cream in different concentration (10%, 20%, and 40%). Twenty-five male of Mus musculus Balb/c strain mice aged 4 weeks were divided into five groups; control group, UVB group, UVB + 10% RE group, UVB + 20% RE group, and UVB + 40% RE group. The UVB groups were given UVB radiation three times a week with an exposure duration of 100 seconds per time for ten weeks. At the end of the treatment, skin samples were excised and statined histologically, also were analyzed for their protein expression. Evaluation of wrinkles was carried out using the Bissete method before and after treatment. To evaluate the thickness of the epidermis, HE staining was performed, while masson Trichome staining was performed to determine the collagen content.RESULTS: RE cream-treated groups showed lower wrinkle score compared to the control group. Furthermore, in UVB + 10% RE group, the RE cream application reduce wrinkle formation. In UVB + 10% RE group and UVB + 20% RE group, the RE cream application increased epidermal thickness and collagen content (p=0.00). While collagenase, matrix metalloproteinase-1 (MMP-1) expression was lower in UVB + 20% RE group compared to the UVB group (p<0.05), however the MMP1 expression in UVB + 40% RE group was higher than other treatment group.CONCLUSION: RE cream prevents wrinkle by maintaining epidermal thickness and collagen contain. RE cream also decreases MMP-1 expression in mice.KEYWORDS: coffee, collagen, MMP-1, robusta, wrinkle

Abstract

Highlight:AKT or protein kinase B increase protein synthesis and control the degradation of proteins. Satellite cells was provided remarkable ability to regenerate skeletal muscle.Stimulation of protein synthesis is effective therapy to maintain muscle mass, prevent muscle wasting to reduce risk sarcopenia and improve quality of life in the elderly.Abstract:Muscles have an important role as a regulator of glucose and triglyceride metabolism. Some researches show the correlation between skeletal muscle mass and metabolic diseases, such as diabetes. Skeletal muscle mass decrease occurs due to chronic illness or physiological process of aging, thus increasing the risk of metabolic diseases as well as motion difficulty in the elderly. Skeletal muscle mass depends on balanced protein synthesis and degradation, controlled through a variety of signal transduction pathways including the AKT. AKT or protein kinase B increases protein synthesis through the mTOR and GSK3β and controls the degradation of proteins through FoxO transcription factors. Another factor that has an alleged role in the regulation of skeletal muscle is the satellite cells which provide remarkable ability to regenerate skeletal muscle. A comprehensive understanding of the biomolecular mechanism of muscle mass regulation is important to develop effective treatment or prevention of muscle atrophy in many cases, either caused by pathological conditions, such as chronic diseases, or the process of aging.

Abstract

One of the potential public health issues in Indonesia is adolescent reproductive health (ARH) issue, particularly premarital sex and promiscuity behaviors which create the risk for unwanted pregnancies and sexual transmitted diseases such as HIV and AIDS. This study aimed to increase in student’s motivation and learning satisfaction through the application of integrated reproductive health learning module in junior high school. This was a pre- and post-test quasiexperimental study with control group design on 358 seventh grade junior high school students in Bandung city. Sampling was performed using multistage random sampling method and subjects were divided equally into treatment group (n=179) and control group (n=179). Data collected were analyzed using chi-square test, Wilcoxon test, and Mann-Whitney test. Results showed that the application of integrated reproductive health learning module influenced motivation and satisfaction, which was evident from the increase in motivation and learning satisfaction scores in the treatment group (21.9% and 6.23%) when compared to the control group (2.2% and 6.1%). In conclusions, the use of integrated reproductive health learning module significantly influences student’s motivation and learning satisfaction among seventh grade junior high school students. PENGARUH PENERAPAN MODUL PEMBELAJARAN KESEHATAN REPRODUKSI TERINTEGRASI TERHADAP MOTIVASI DAN KEPUASAN BELAJAR SISWA DI SEKOLAH MENENGAH PERTAMASalah satu permasalahan kesehatan masyarakat yang potensial di Indonesia adalah masalah kesehatan reproduksi remaja (KRR) khususnya perilaku seks pranikah dan pergaulan bebas yang berisiko kehamilan yang tidak diinginkan dan penyakit infeksi menular seksual (IMS) seperti HIV dan AIDS. Penelitian ini bertujuan meningkatkan motivasi dan kepuasan belajar siswa dengan menerapkan modul pembelajaran kesehatan reproduksi terintegrasi di sekolah menengah pertama (SMP). Penelitian ini menggunakan pre- and post-test quasi-experimental study dengan desain kelompok kontrol pada 358 siswa kelas tujuh SMP di Kota Bandung pada Maret–April 2017. Pengambilan sampel dilakukan menggunakan metode multistage random sampling dan subjek dibagi secara merata menjadi kelompok perlakuan (n=179) dan kelompok kontrol (n=179). Data yang dikumpulkan dianalisis menggunakan uji chi-square, uji Wilcoxon, dan uji Mann-Whitney. Hasil penelitian menunjukkan bahwa penerapan modul pembelajaran kesehatan reproduksi terintegrasi memengaruhi motivasi dan kepuasan belajar yang terbukti dari peningkatan motivasi dan skor kepuasan belajar pada kelompok perlakuan (21,9% dan 6,23%) bila dibanding dengan kelompok kontrol (2,2% dan 6,1%). Simpulan, penerapan modul pembelajaran kesehatan reproduksi terintegrasi secara signifikan memengaruhi motivasi dan kepuasan belajar siswa kelas tujuh SMP.

Abstract

Tujuan: Melihat hubungan antara pola kardiotokografi dan hasil luaran (cara persalinan dan status asfiksia neonatus).Metode: Penelitian ini observasional analitik retrospektif dengan desain case-control. Penelitian melibatkan pasien-pasien yang melahirkan di RS Hasan Sadikin Bandung antara 2019-2020. Kriteria inklusi adalah pasien dengan kehamilan normal dan/atau dengan kehamilan patologis, pasien dengan usia gestasi 37-40 minggu, kehamilan dengan janin tunggal dan pasien dengan induksi persalinan. Kriteria eksklusi mencakup data pemeriksaan tidak lengkap  anamnesa, pemeriksaan fisik terkait kehamilan dan persalinan, rekaman kardiotokografi, dan skor APGAR menit ke-5; hambatan pertumbuhan janin intrauterine (IUGR); kelainan kongenital (cacat bawaan) dan infeksi/demam pada ibu. Penelitian ini menggunakan data sekunder dari rekam medis. Metode perekrutan sampel menggunakan consecutive admission sampling. Analisis data secara deskriptif dan analitik dilakukan. Hasil: Rerata usia pasien adalah 26,83±7 tahun. Pasien paling banyak memiliki latar belakang pendidikan tamat SMA (51,7%). 48,3% pasien belum pernah bersalin. 53,3% pasien sedang hamil 39-40 minggu saat bergabung dengan penelitian ini. Rata-rata berat bayi lahir sebesar 2923,33±385,229 gr. Untuk APGAR Score 5’ memiliki rata-rata sebesar 7,68±2,347. Ditemukan bahwa kategori kardiotokografi patologis secara signifikan meningkatkan terjadinya asfiksia (p=0,024). Kemudian, temuan kardiotokografi patologis juga secara signifikan meningkatkan pemilihan seksio sesarea sebagai metode persalinan (p<0,001).  Kesimpulan: Pola kardiotokografi patologis secara signifikan meningkatkan risiko munculnya asfiksia pada neonatus. Association between Cardiotocographic traces, and mode of delivery and asphyxiaAbstractObjective: the aim of this study is to evaluate the association between cardiotocographic traces and maternal and neonatal outcomes (mode of delivery and neonatal asphyxia status).   Method: This is a retrospective observational analytical case-control study involving patients delivering at Hasan Sadikin General Hospital between 2019-2020. The inclusion criteria were patients with normal pregnancies and/or pathologic pregnancies at 37-40 weeks of gestation, singleton pregnancies and patients with induced labor. The exclusion criteria were the following: incomplete case notes (patient history, physical examination, CTG record and 5-minute APGAR score), intrauterine growth restriction (IUGR); fetal with congenital anomalies dan maternal infection and/or fever. This study utilized secondary data from patient case notes. This study employed consecutive admission sampling. Descriptive and analytical statistics were performed. Results: Mean patient age was 26,83 ±7 years. 51,7% patients were high-school graduates. 48,3% were nulliparous. 53,3% were at 39-40 weeks of gestation. Mean neonatal birth weight was 2923,33±385 gr. Mean 5-minute APGAR score was 7,68±2. Pathologic CTG traces significantly increased perinatal asphyxia (p=0,024). Pathologic CTG traces also significantly increased the use of caesarean section as method of delivery (p<0,001). Conclusion: Pathologic CTG traces significantly increased the likelihood of neonatal asphyxia. Key words: Cardiotocography, Asphyxia, Delivery, APGAR

Abstract

A high-fat diet (HFD) is associated with bone inflammatory processes that can affect bone remodeling balance. Osteocrin produced by periosteal osteoblasts correlates with osteoblast activity, is expressed on osteoblast-derived cells, and is localized in osteoblasts and young osteocytes. Propolis is an antioxidant and anti-inflammatory in bone remodeling by inhibiting proinflammatory factors NF-κB and COX-2, reducing inflammatory suppression of cytokines responsible for osteoclast differentiation and osteoblast apoptosis. The flavonoid content increases the production of nitric oxide and osteoprotegerin, which enhances osteoblastogenesis. This study determined the effect of propolis administration on bone formation and resorption in bone previously damaged by an HFD. This research was conducted in the Animal Laboratory of Postgraduate Building Dipati Ukur and Genetics and Molecular Laboratory Eycman Building, Faculty of Medicine, Universitas Padjadjaran. The research time was from January 2023–May 2024. Male Wistar rats were divided into four groups: normal chow diet (NCD), NCD with propolis administration, HFD, and HFD with propolis administration. The 12-week-old rats were given an HFD for 12 weeks and then treated with propolis at a 300 mg/kgBW dose for nine weeks. The administration of propolis increased the ratio of osteoblasts-osteoclast cells in the femur of the HFD rats but did not affect periosteal osteocrin expression.

Abstract

Selenium is a trace element that has antioxidant properties and is a component of selenoproteins. In certain circumstances, such as injury, selenium supplementation is required to regulate the inflammatory response. Contusion injury increases the inflammatory response to a muscle or tissue that begins with vasodilation, increases blood flow, increases vascular permeability, and leads to edema formation at the site of injury. Objectives: To determine the effect of selenium supplementation on changes in blood markers in rats treated with contusion injury. Methods: Experimental design, using experimental male rats (Rattus norwegius), eight weeks old, weighing 200-220 grams, divided into three groups, namely: The Control group, the contusion group, the contusion + Selenium group. Selenium supplementation was given orally on days 1-3 after injury at a dose of 0.0153 mg into 2% PGA. Examination of markers such as White Blood cells (WBC), lymphocytes, monocytes, and neutrophils was carried out on day three after treatment. Results: This study found that the administration of post-muscular selenium supplementation based on examination of blood markers such as WBC, Lymphocytes, Monocytes was higher in the contusion group and lower in the selenium group. Neutrophils were higher in the contusion and selenium groups. This was not significantly different from the control group. The result of blood marker examination tended to change, but it was not statistically significant. Conclusion: Selenium supplementation in post-contusive injury mice did not provide significant changes in blood markers such as WBC, lymphocytes, monocytes, and neutrophils.Keywords:  antioxidant, blood marker, contusion, selenium

Abstract

Abstract: Pregnancy is a critical period involving significant physiological and psychological changes, particularly in primigravida women who are more susceptible to anxiety and stress. Elevated stress levels during pregnancy may increase the risk of complications like high blood pressure, preeclampsia, and premature delivery. Non-pharmacological interventions like Hatha Yoga and gymnastic prenatalhave been widely recommended to improve maternal well-being. The purpose of this research was to evaluate and compare the impacts of two different interventions on maternal psychological and physiological health through a Systematic Literature Review (SLR) conducted in accordance with PRISMA guidelines. Relevant literature was retrieved from databases including Pubmed, Scopus, ScinceDirect, Google Scholar, focusing on research published between 2014 and 2024. The findings indicate that Hatha Yoga is more effective in reducing anxiety, cortisol levels, and systolic blood pressure, while prenatal gymnastic better supported diastolic blood pressure regulation and physical endurance for labor. Both interventions contribute to better cardiovascular function, increased oxygen saturation, and a smoother labor process. Additionally, yoga is associated with improved neonatal outcomes, while studies on prenatal exercise’s effects on newborn health remain limited. Future research should explore the long-term effects and key differences between these interventions to determine the most effective approach for maternal and neonatal health. These findings support integrating yoga and gymnastic prenatalinto antenatal care programs to enhance pregnancy outcomes.

Abstract

Background Protein is essential for skin integrity, while exercise modulates cellular and molecular processes that regulate autophagy, mitochondrial biogenesis, and collagen synthesis. This study evaluates the combined effects of dietary protein restriction and varying exercise intensities on epidermal thickness, autophagy markers (LC3, p62), mitochondrial biogenesis (PGC1α), and collagen synthesis (Col1a1, Col3a1) in male Wistar rats.    Methods An experimental laboratory study was conducted involving 32 male Wistar rats. They were randomized into 8 groups: control, low protein, low intensity, group IV, moderate intensity, moderate intensity low protein, high intensity, and high intensity low protein. Low protein diet (5%) compared to the control (24%) was given for 60 days, while the exercise regimen was eight weeks of treadmill exercise (low/10 m per minute, moderate/20 m per minute, and high intensity/30 m per minute) for 30 minutes a day, five days a week. Epidermal thickness, LC3, p62, PGC1α, Col1a1, and Col3a1 mRNA expression were measured at the end of the study. One-way ANOVA/Kruskal-Wallis followed by Tukey HSD/Mann–Whitney test was used to analyze the data.   Results A low-protein diet significantly reduced epidermal thickness (p=0.048), increased LC3 (p=0.043) and p62 (p=0.011), indicating impaired skin regeneration and autophagy. However, exercise, especially at high intensities with low protein, mitigated this effect by increasing both LC3 (p=0.011) and PGC1α (p=0.011).   Conclusions These findings highlight the compensatory role of exercise in maintaining tissue integrity under protein-deprived conditions in male rats. High-intensity exercise potentially enhances autophagy and mitochondrial biogenesis.

Abstract

ABSTRAK Latar belakang : Anemia postpartum masih menjadi masalah kesehatan global yang berdampak pada pemulihan fisik dan psikologis ibu setelah melahirkan. Penurunan kadar hemoglobin dapat menurunkan kualitas hidup ibu nifas dan menghambat proses adaptasi terhadap peran keibuan. Kajian ini diperlukan untuk memahami sejauh mana kadar hemoglobin berpengaruh terhadap kesejahteraan ibu pascapersalinan. Metode : Penelitian ini merupakan scoping review berdasarkan pedoman PRISMA-ScR. Pencarian literatur dilakukan pada PubMed, Scopus, ScienceDirect, dan Google Scholar untuk periode 2015–2025. Sebanyak 10 artikel memenuhi kriteria inklusi dan dianalisis secara tematik. Hasil : Analisis menghasilkan tiga tema utama, yaitu kesehatan fisik, kesejahteraan psikologis, dan adaptasi peran ibu. Kadar hemoglobin rendah berhubungan dengan kelelahan, penyembuhan luka yang lambat, depresi postpartum, serta penurunan kepercayaan diri dan kelekatan ibu–bayi. Kesimpulan : Kadar hemoglobin postpartum memengaruhi kualitas hidup ibu secara multidimensi.
