Abstract

Tomatoes are climacteric fruits that experience a surge in respiration rate and ethylene production. This condition affects the fruits’ physiological deterioration, shortens their shelf-life, and degrades the quality of tomatoes. The study aimed to enhance the shelf-life of commercial tomatoes with different genetic backgrounds by utilizing the Sletr1-2 mutant, which has a lower sensitivity to ethylene. A randomized block design (RBD) was applied to achieve this goal, where 12 different plant genotypes were considered as treatments, and each treatment was replicated four times.  The results showed that fruit shelf-life was extended by crossing commercial tomatoes with Sletr1-2 mutant tomatoes. Genetic variability for all shelf-life characteristics were narrow. In contrast, there were broad phenotypic variability for fruit hardness and weight loss characteristics.

Abstract

ZmES4 is a plant-derived antimicrobial peptide (AMP) from maize that shows promise as a biocontrol agent against plant pathogenic organisms. In the context of growing challenges in sustainable agriculture, AMPs like ZmES4 represent innovative alternatives to chemical pesticides. This study focuses on the structural characterization and heterologous expression of the ZmES4 peptide in Escherichia coli (E. coli). The gene encoding ZmES4 was obtained from the maize female gametophyte (NCBI Reference Sequence: NM_001112150.3) and cloned into the pET24d(+) expression vector using NcoI and XhoI restriction sites. Transformation into E. coli BL21 (DE3) cells enabled recombinant expression upon induction with isopropyl β-D-1-thiogalactopyranoside (IPTG). Sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and Bradford assays confirmed the expression of ORF-ZmES4, with protein concentrations ranging from 14.647 to 63.606 mg/mL. The successful expression of ZmES4 in E. coli highlights its potential application as a recombinant AMP for future plant disease management strategies.

Abstract

Abstrak Pada saat ini minat generasi muda semakin berkurang pada sektor pertanian, karena menganggap bertani adalah pekerjaan tradisional yang kurang bergengsi dan hasilnya tidak dapat untuk memenuhi kebutuhan hidup. Sektor pertanian banyak diisi oleh generasi tua yang masih bertahan ditengah kesulitan lahan yang semakin sempit dan harga pupuk yang semakin meninggi, oleh karena itu untuk membangun citra pertanian diperlukan sosialisasi pertanian dengan harapan mampu membuat generasi muda sadar akan pentingnya pertanian dengan segala potensi wilayah yang ada dan dimiliki. Desa Cinanjung terletak di Kecamatan Tanjungsari Kabupaten Sumedang, dahulu daerah ini merupakan daerah pertanian tetapi anak muda lebih memilih untuk bekerja di kota, walaupun hanya menjadi buruh atau kuli bangunan daripada menjadi petani di desanya. Tujuan dari pengabdian ini adalah masyarakat bisa meningkatkan potensi pemudanya dalam pemanfaatan pertanian organik serta mengetahui cara memaksimalkan potensi tersebut. Metode yang digunakan adalah Partisipatory Rural Appraisal (PRA), yaitu metode pendekatan perencanaan wilayah pedesaan melalui pelibatan masyarakat secara aktif dan efektif. Hasil dari pengabdian ini masyarakat di Desa Cinanjung memeiliki ketertarikan dalam bertani organik dan mengali potensi desa dalam kegiatan yang ada dalam program pengabdian ini.

Abstract

Komoditas Padi merupakan salah satu komoditas andalan di Desa Jagayabaya, Kecamatan Cimaung Kabupaten Bandung. Kesadaran masyarakat akan pentingnya bahan pangan organik terus meningkat. Keterbatasan pengetahuan pertanian organik dan penerapannya di Desa Jagabaya telah mendorong dilaksanakannya pengabdian masyarakat untuk meningkatkan pemahaman dan pengetahuan pertanian organik termasuk pembuatan pupuk cair organik untuk meningkatkan produksi pangan organik khususnya padi. Metode partisipatif digunakan untuk melibatkan mitra secara aktif dalam pelaksanaan pemahaman dan penerapan pertanian organik. Tahap-tahap pelaksanaan terdiri dari sosialiasi kegiatan, wawancara, penyuluhan, diskusi, dan praktek. Hasil kegiatan terjadi peningkatan ilmu, pengetahuan dan teknologi pertanian organik, cara pembuatan pupuk cair organik serta penerapannya.

Abstract

Yam bean Pachyrhizus erosus is widely cultivated in Central America, Africa, Asia and Pacific.  The objective of the research was to analyse genetic relationships of yam bean originated from Indonesia compared to its ancestor species from Mexico and Guatemala. Fourty three yam beans consisted of 35 genotypes from Indonesia and 8 introduced genotypes from Mexico and Guatemala were planted under complete block design with two replications. Fourteen significant different traits from 25 morphological leaf and flower properties traits have been used to analyse the genetic relationships. Results showed that yam beans from Sumatra differed from its relatives from the rest regions of Indonesia. Yam bean populations originated from Indonesia are closely related to its ancestor species from Mexico and Guatemala.   Key words :  Yam bean, Pachyrhizus erosus, genetic relationships
