Abstract

Isolation and Identification of Cellulolytic Bacteria in Religious Ceremony Waste at Goa Lawah Temple Klungkung Cellulolytic bacteria are bacteria that can hydrolyze cellulose complexes into smaller oligosaccharides and finally glucose. Cellulolytic bacteria synthesize a set of enzymes that can hydrolyze cellulose called cellulase enzymes. This study aims to isolate and identify cellulolytic bacteria that have the potential to degrade cellulose by taking organic waste samples from TPS Pura Goa Lawah Klungkung. Characterization was carried out by growing selected pure isolates on CMC media and then dripping with congored 0.1% to test their cellulolytic potential. The results of bacterial isolation at 10dilution-8 obtained 28 bacterial isolates that were able to grow and utilize cellulose as a carbon source. From the isolation results obtained The 4 bacterial isolates each had different morphological characters capable of producing clear zones with a diameter of 10 mm: 5 mm respectively; 3 mm and 4 mm. The results of the isolation of bacterial colonies appeared to be round and irregular in shape, had clear and cream colors, had a convex and flat surface and all isolates had smooth edges. Type C bacterial isolates included gram-negative groups, bacterial isolates A, B and D included gram-positive groups and all bacterial isolates A, B, C and D were rod-shaped. The degradation ability based on the cellulolytic index value of bacterial isolate A was in the highest category compared to bacterial isolates B, C and D.

Abstract

ABSTRACT Isolation and Identification of Agrobacterium tumefaciens At Plant roses (Rosa sp.). Agrobacterium tumefaciens caused crown gall tumor in many of dicotyledonous plants. The purpose of this study tried to isolate and identify the A. tumefaciens from rose plant. Various techniques were used such as selection media, colonies shape and color, Koch's postulate test, DNA isolation and agarose gel elektroforesis. The results of this study showed that A. tumefaciens can be isolated through culturing in LB medium and selection AB medium that was specific for A. tumefaciens. The isolate caused crown gall tumor on carrot slice 3 weeks after inoculation by using a modified Koch’s postulate test. The characteristics of colony formed in this research are round shaped, cream coloured with pink tint, smooth edge, and convex elevation. DNA isolation and its running in agarose gel electroforesis showed positive result.   Key words : Agrobacterium tumefaciens, AB medium and Agarose gel electroforesis.

Abstract

Histopatology Siamese Citrus Leaves (Citrus nobilis) Infected by Citrus Vein Phloem Degeneration (CVPD) at mild, moderate and severe attacks. Aims of this study is to identify the presence of the bacteria Liberobacter asiaticum, cause of CPVD (Citrus Vein Phloem Degeneration) disease and to knowthe differences in the histology of Siamese citrus plant tissue that is attacked by mild, moderate and severe attacks. This research was in the Laboratory of Plant Structure and Development of the Faculty of Biology, University of Gajah Mada and conducted at the UPT of the Genetic and Molecular Biology Laboratory of Udayana University. Field observations and sampling were carried out in the Wanasari Banjar, Kintamani Village, Kintamani Sub-District, Bangli District, Bali Province. There are several symptomatic citrus plants and attacked by CVPD with various levels of severity by visual appearance. On the observed land it were found that the attack percentage was 70% and the intensity of the attack was 46.4%. PCR (Poly polymeration chain reaction) identification method using OI1 and OI2 specific primers. The DNA band 1160 bp was found in mild, moderate, and severe chlorosis symptomatic samples. Leaves without symptoms do not change shape in the epidermis, cortex, xylem and phloem, while form changes occur in phloem, xylem and cortex in symptomatic citrus plants.

Abstract

ABSTRACT Isolation and Characterization of Mycorrhizae Arbuskular Fungi in the Soil and Roots of Corn (Zea Mays L.) in the Sanur Kaja Village Mycorrhiza is an organism that is derived from the fungus that describes a form of symbiotic relationship between mutualisme functions with a high degree of plant roots. Benefits of mycorrhiza for plant growth and development as its host is to increase the absorption of nutrient elements of soils, as biological barrier against infection of root pathogen, enhancing the resilience of crops to drought and increasing hormone boosters grows.This research aims to identify a Arbuskular Mycorrhiza fungi isolated from corn plants rhizosphere in the village of Sanur Kaja. The results of this research show that the isolation and characterization of spores that develop on the roots of corn in the village of Sanur Kaja indicates a growing spores there is the genus Glomus sp. type of Spore that successfully identified a species of Glomus sp-1, Glomus sp-2, Glomus sp-3. Abstract

ABSTRACT Identification of Arbuscular Mycorrhiza through Microscopis Methode in Rhizosfer of Several Grass and Cacao ( Theobroma cacao L.) Arbuscular mycorrhiza fungi that live in a symbiotic mutualism with plant roots and helps the absorption of plant nutrients, and also able to live in various places. The study aimed to identifiy arbuscular mycorrhiza through microscopic methode in rhizosfer of several grass and cacao was conducted from December 2013 to March 2014. The method used in this study were spore isolation by wet sieving and root staining techniques. The results showed that arbuscular mycorrhiza spores and structures found in Imperata cylindrica L. are 2 types spores of genus Glomus (Glomales: Glomeaceae) with vesicular and hyphae structures. The presence of spores and structures in Paspalum notatum are Acaulospora (Glomales: Acaulosporaceae) and genus Gigaspora (Glomales: Gigasporineae) with arbuscular, vesicular and hyphae structure. Spores and structures of mycorrhiza found in the Pennisetum purpureum are 3 types of the genus Glomus with hyphae and arbuscular structure. Spores and structures mycorrhiza in the Cyperus rotundus are 1 type spore of the genus Gigaspora and 2 types of spores Glomus with internal hyphae structure. While spores and mycorrhiza structures in the cacao are 2 types spore of the genus Glomus with hyphae and vesicular structures.   Keywords: Glomus, Gigaspora, Acaulospora, Arbuscular Mycorrhiza Structure

Abstract

ABSTRACT Vesicular Arbuscular Mycorrhizae (VAM) Identification of Red Onion (Allium cepa L.) and Taro (Colocasean esculenta (L.)Schott) Rhizosphere and Its Spore Multiplication in Zeolite Media Efforts to increase agricultural production can be done with a variety of technologies. Technology is widely applied to farmers in the cultivation process is the use of inorganic fertilizers with high doses exceeding the dose of balanced fertilizer. In the production of onion crops in the field, also using inorganic fertilizer application on an ongoing basis which may result in damage to the ground both in terms of texture and structure. Red onions and taro roots are shallow causing both of these plants are unable to absorb nutrients to the maximum if exercised enough chemical fertilizer. This study was aimed to determine genus and species of VAM in red onion and taro rhizosphere, its colonization in root tissue, and to examine zeolite media compatibility with corn as a symbiont. Based on the results of the study, it was found three species of VAM spores of red red onion plant rhizosphere, namely: Acaulospora denticulata, A. laevis, Glomus ambisporum, and five species of VAM spores recovered from the rhizosphere of taro plants namely: Acaulospora foveata, A. koskei, A. capsicula, Scutellospora calospora and Glomus ambisporum. Infections were found in plants onion, taro and corn plants form symbiotic arbuscular, vesicles and inner spore. VAM from the rhizosphere of plants onion and taro can be reproduced using zeolite media and symbionts plant corn. Keywords: MVA, rhizosphere, Acaulospora, zeolite, inner spore

Abstract

Citrus (Citrus nobilis L.) Propagation Through Culture In Vitro Using Seed From Infected Plants Of Citrus Vein Phloem Degeneration (CVPD) Disease. The sample was taken in Belancan Village, Kintamani District, Bangli Regency, and invitro culture was conducted in UPT. Genetics Resource and Molecular Biology Laboratory in February to June 2018. This research purposes were to get the new citrus plant from citrus seed infected of CVPD disease explants with culture in vitro and found the presence of the bacterium Liberobacter asiaticus in results of citrus plants culture in vitro. The sampling method is using plants appearance. Obtained sample 1 is a plant that shows severe symptoms of CVPD disease and sample 2 plants that show no symptoms of CVPD disease. Seed Sample is cultured with MS0 medium for 8 WAP then proceed to acclimatization. The results of this research showed that the seed explants from both samples were able to grow well and did not have the characteristic of CVPD disease, but explant seed sample 2 in vitro culture result showed that the bacterial DNA bacterium Liberobacter asiaticus pathogen CVPD disease does exist in length 1160bp, so it cannot be expected to produce CVPD-free crops.

Abstract

Agrobacterium tumefaciens Mediated Genetic Transformation of acvB Gene in Potato (Solanum tuberosum L.). Genetic transformations are now routinely applied to plant mediated by Agrobacterium tumefaciens as the most convenient technique. This study aimed to prove the success of A. tumefaciens mediated genetic transformation in potato. A. tumefaciens LBA (pBI 121) and explant of potato shoot were used in this study. Explants were grown in vitro on Murashige and Skoog media. Transformation was implemented using smear technique by smearing A. tumefaciens to injured explant. Experimental groups consisted of two groups: control group which did not receive transformation treatment and treatment group receiving transformation treatment. Explant growth was observed through the presence of shoots, branches and the shoot height. Explants in the treatment group resulted in a higher number of shoots, branches, and shoot heights compared to control. Phenol compounds appear in explant epidermal tissue, indicating the wounds produced by A. tumefaciens infection, thus the gene predicted to be transformed. Identification by PCR is needed to prove the existence of the acvB gene in potato plants genome, using acvB specific PCR primer as the marker, such as (5?-CCCT CTAG AGAC CCGC GCCA AGGCG-3?) and (5?CGCG TCGA CCTT GTCG GAAAG -3?) with 540-bp in base pair size produced.

Abstract

Effectiveness Test of Several Concentrations of Kirinyuh Leaf Extract (Chromolaena odorata L.) Toward Development of Cabbage Crop Caterpillar (Crocidolomia pavonana F.). This research was conducted at the Laboratory of Plant Pests and Diseases of the Faculty of Agriculture, Udayana University from April 2018 to August 2018 to test the extract of kirinyuh leaves (Chromolaena odorata) on the development of biology of cabbage crop caterpillar (Crocidolomia pavonana). The completely randomized design (CRD) was used in this study, with four types of treatment of leaf extract concentration of 10%, 20%, 30%, 40%, and control. Each treatment was repeated 10 times. Observations were completed every day of the larval stage until the larvae do not carry out any activity anymore (dead), or until they become imago. The results showed that (1) the leaf extract of C. odorata plant which was used as research material had the potential to suppress the biological development of C. pavonanadi larvae in the laboratory, (2) the application of leaf extract concentration of 10% and 20% had an influence on the development of larvae which had developed slow compared to concentrations of 30% and 40%, where larvae experience rapid development of only 9 observations and (3) the most effective test leaf extract treatment was the treatment of leaf extracts of C. odorata concentrations of 40% and 30% followed by leaf extract concentrations of 20% and 10%.

Abstract

Online travel agent (OTA) Cina offers variety of tourism products. However, some of tourism products that offered by OTA through website seem not interesting for Chinese tourists. This research aimed to analyze factors that influence for the Chinese tourists consumer on the decision of buying tourismproduct in Bali through OTA. This research was conducted in Bali island, the number of samples taken was 100 Chinese tourists who has bought tourism product in Bali through OTA by using quota sampling technique. Data was analyzed with exploratory factor analysis. Results show there are five factorsthat influence for the Chinese tourists consumer on the decision of buying tourism product through OTA namely Product, OTA, Reference Group, Tourist Characteristic and Income. Finally,this study identify that factor product is the most influence for the Chinese tourists on the decision of buying OTA product.
