Abstract

Citrus Vein Phloem degeneration (CVPD) is an important citrus disese, which damaged citrus plantation and causing decrease of citrus production. In Indonesia, the CVPD disease caused by Liberobacter asiaticum bactery and the disease spread out by vectir insect Diaphorina citri and using infected bud in wood grafting. In infected citrus plant, two specific protein molecules with molecular weigt 16 kDa and 66 kDa are found. These protein molecules are not found in healthy citrus plant. The immunogenicity of PS16 accumulated on leaf of citrus plant infected by CVPD is known yet. The research material were leaves of citrus plant infected CVPD, leaves of healthy citrus plant and reagent used these research are for isolation of the total protein leaf of citrus plant, SDS-PAGE electroforesis, electroelution of PS16, ELISA Methods, Dot-Blot Method, anti-PS16 as aprimery antibody and secondary antibody is anti-Rabbit IgG Conjugated AP. The result of the research showed that of PS16 accumulated on leaf of citrus plant infected CVPD has immunogenic character. It is indicated by increase of the titer anti-PS16 after first immunization ang 2nd booster by indirect ELISA method and can be used to induce antibody (anti-PS16) and so showed that positive reaction between PS16 with anti-PS16. It is indicated by purples dark blue on cellulose membrane by Dot Blot method.

Abstract

Red cabbage is a plant that is rich in antioxidant, one of its secondary metabolites, antosianin that gave the purple color on red cabbage has multiple health effects for human such as preventing cancer, tumor, and many more. The use of this secondary metabolites in plants nowadays is very popular, not only for its pharmacological effects, but also as an antibacterial, coloring agent, flavoring agent, natural pesticide, etc. Therefore, this study is needed to look for another potentially benefical compound in the plant, such as red cabbage, a plant that is more than just a salad. The aim of this study was to determine the effect of the types of extraction solvents on phytochemical compound of red cabbage plant leaves. The dried off plant macerated with two kinds of solvents, aquades, and ethanol. After about +3 days, both of the extract got evaporated with rotary evaporator. The yields of evaporating will be tested with GCMS analysis. The result showed that kind of extraction solvents do affect the yield from evaporating, and GC-MS analysis. Aquades showed to have a better capability in both of extraction, or on the analysis with GC-MS.

Abstract

Kelor (Moringa oleifera) is a plant that grows in tropical climates including Indonesia. All parts of this plant can be used as medicine and other natural products. The purpose of this study was to determine the bioactive compounds contained in moringa stem bark and the level of toxicity expressed by an LC50 value of less than 1.000 ppm so that it can be used as traditional medicine and other natural products. Identification of bioactive compounds in moringa stem bark using the GC-MS analysis method and toxicity test on Artemia salina Leach shrimp larvae using the Brine Shrimp Lethality Test (BSLT) method. Based on the results of the analysis using GC-MS showed that moringa bark extract contains 14 bioactive compounds. The highest bioactive compounds in the 3 peaks were secondary metabolites in the form of phenols and monoterpenoids. The toxicity test of moringa bark extract against Artemia salina Leach larvae showed that the higher the concentration, the higher the percentage of mortality of Artemia salina Leach, so that the calculated LC50 value of moringa bark extract was toxic, which was 149,689 ppm.

Abstract

Coffee fruit contains high cellulose and lignin. Cellulose is the most abundant polysaccharide found in plants, cellulose in nature forms crystals so that cellulose is difficult to degrade. Cellulose can be broken down using cellulase enzymes produced by microorganisms. One of the microorganisms capable of producing cellulase enzymes which are used to hydrolyze cellulose into simpler products, namely glucose, is cellulolytic bacteria. The purpose of this study was to identify bacterial isolates that ferment in coffee cherries. The isolation results obtained 97 bacterial colonies that grew, based on observations of color, shape, edges, and surface, they could be grouped into four groups of bacterial isolates named M1, M2, M3, and M4 bacterial isolates, with each cellulolytic index (IS) of 3,00; 0,75; 2,00 and 3,00. The results of the gram stain test obtained one gram-negative bacterial isolate (M2 rod-shaped) and three gram-positive bacterial isolates (M1 in the form of a focus, M3 and M4 in the form of a rod). Bacterial isolates M1 and M3 degraded the earliest coffee cherries with an average of 2.66 days after inoculation and no later than 6 day after inoculation in M2M4 and M3M4 bacterial isolates. The ability to degrade bacterial isolates in coffee cherries was obtained 2 - 5.34 days faster than the control.

Abstract

The Abundance of Thrips parvispinus Karny and Its Natural Enemies in Chili Crops (Capsicum annuum L.). This aims of the research is to search the abundance of T. parvispinus Karny and its natural enemies on chili crops (Capsicum annuum.L). The research started from May 2021 to August 2021, located in Selat Village, Banjarangkan District, Klungkung Regency, and at the Plant Pest Laboratory, Faculty of Agriculture, Udayana University. The study was conducted using a descriptive method by conducting a diagonal survey of 20 samples of chili plants that were attacked by T. parvispinus. Observations were made by observing 10 leaves and 10 flowers of chili plants. The results showed that the abundance of T. parvispinus in the vegetative and generative phases fluctuated where the highest population of T. parvispinus occurred in plants aged 10 WAT with an average population of 17,57 individuals per plant then decreased in plants aged 11 WAT with an average population 11,12 individuals. There were two species of predators that preyed on T. parvisvinus, namely Coccinela transversalis with an average population of 23.3 and Chrysoperla carnea with an average of 18,2 individuals. There was one of nymph parasitoid to suppress the development of T. parvispinus namely Ceranisus spp with an average parasitization level 9,8 %.

Abstract

Bulung boni (Caulerpa sp.) is a type of algae that spreads in almost all Indonesian oceans. Generally, bulung boni grow in shallow water and attach to the sand substrate. This study aimed to determine the bioactive compounds of bulung boni and how they differ from other algae. The identification of bulung boni bioactive compounds was conducted using the GC-MS (Gas Chromatography-Mass Spectrometry) method. Based on the results of GC-MS analysis, bulung boni contains 10 compounds that mainly included into alkane group, i.e. 1-dodecene, Tetradecene, Decene, Pentadecene, Hexadecene, Dodecanol, Haxadecanoic acid, Heptadecene, Oktadecane, and Nanodecene.

Abstract

Phytochemical Content and Antioxidant Activity of the Leaves of the Senduduk Plant (Melastoma malabathricum L.) using the GC-MS (Gas Chromatography-Mass Spectrometry) Method. Senduduk (Melastoma malabathricum L) is one of the plants that can be used as traditional medicine. This study aims to determine the types of active compounds, levels of active compounds, and levels of antioxidant compounds in the leaves of the senduduk plant. This research lasted for 3 months. The method used in this study is a descriptive method with laboratory analysis techniques. Active compound analysis testing with GCMS analysis, testing and phytochemical levels, as well as determining the activity and levels of antioxidants. The results showed that the active compounds in senduduk leaf extract were mostly dominated by phenolic compounds and saponin compounds. The phenolic compound group contains active compounds 1,2,3-Benzenetriol and Vitamin e dl-.alpha.-Tocopherol with a percentage of 26.252% and the saponin group contains active compounds by fatty acids, namely hexanacanoic acid and linoleic acid with an area proportion of 14.25. %. The levels of active compounds from the phytochemical screening of the active compound group with the strongest levels were Alkaloids, Phenolics, and Saponins. The active compound group with moderate levels was only terpenoid active compounds, while the active compounds with sufficient levels contained steroids, flavonoids, and tannins which were tested by the Liebermann, Buchard, and Meyer reagent examination methods. The antioxidant activity in the extract of the leaves of this senresiden is strong enough, namely 70,607 ppm.

Abstract

Comparison of Total Protein Molecular Weight of Lettuce (Lactuca Sativa L.) Plant Factory and Planted in the Ground Using SDS-PAGE. Lettuce (Lactuca sativa L.) is a plant that contains nutrients and protein that are good for human needs. Lettuce is often found in Indonesian agriculture because lettuce has growing conditions that are very suitable for the Indonesian climate. The purpose of this study was to determine the comparison of protein profiles of lettuce plants grown on the ground with those grown using the hydroponic plant factory system. This research was conducted from May to June 2023 at the Laboratory of PT Genetika Science Indonesia, Tangerang City. This research stage consists of sampling, protein extraction, electrophoresis, and data analysis. Comparisons were made by comparing the two sample data that had been carried out by electrophoresis by looking at the molecular weights of each sample. Plants grown in the soil had higher molecular weights than those grown in the plant factory and each had 12 different proteins. Both samples produced two other same protein bands of 69.65 kDa and 11.97 kDa.

Abstract

Identification of Phytochemical Compounds and Antioxidant Activity Test of Ethanol Extract of Cocoa Bean Shell Waste (Theobroma cacao L.) Trinitario Varieties. Cocoa is one of the mainstay commodities of plantations in Indonesia, contributing to the country's foreign exchange earnings. Byproducts such as fruit skins and pulp are produced during the production of dry cocoa beans, and waste in the form of cocoa bean husks is produced during the processing of dry cocoa beans into chocolate products. So far, cocoa bean husk waste has not been used optimally, leading to a low economic value. The purpose of this study was to determine the bioactive compounds in the skin and the levels of antioxidant compounds in cocoa bean husk waste so that it could be used as a raw material for medicine. The GC-MS analysis method was used to identify bioactive compounds in cocoa bean husk waste, and the DPPH method was used to test antioxidant activity. According to the results of the GC-MS analysis, the cocoa bean husk waste extract contains four compounds with quality value higher than 90. These four compounds are Alpha-Copaene, Copaene, Caryophyllene, and Caffeine. The antioxidant test results of the cocoa bean husk waste extract revealed an IC50 value of 86,213 ppm, indicating strong antioxidant activity because the IC50 value is between 50-100 ppm.

Abstract

Phytochemical Content Identification and Antioxidant Activity Test of Arabica Coffee Leaves (Coffea arabica) in the Catur Village and Robusta Coffee Leaves (Coffea canephora) in the Petang Village. Coffee is considered one of valuable crops commodity, and its cultivation generates waste in the form of coffee leaves. Indeed, it is necessary to make use of this waste for the better Environment, in this case, researchers took the sample of arabica leaves in Catur village and the sample of robusta leaves in Petang village. Researchers conducted a study to identify compounds and determine the antioxidant activity of arabica coffee leaves (Coffea arabica) and robusta coffee leaves (Coffea canephora). The researchers employed GC-MS and for phytochemical analysis and the DPPH method to evaluate antioxidant activity. The results revealed that the viscous extract of both arabica and robusta coffee leaves contained Caffeine as the most prominent compound. Caffeine is known for its stimulating effect on the central nervous system and anti-inflammatory. In the viscous extract of arabica coffee leaves contained 48 substances and the viscous extract of robusta coffee leaves contained 9 substances were identified with a qual >90. The IC50 value for the viscous extract of arabica coffee leaves was measured at 91,837 ppm, indicating a strong-class antioxidant activity. In contrast, the IC50 value for the viscous extract of robusta coffee leaves was 120,972 ppm, representing a medium-class antioxidant activity.
