Abstract

Infeksi kulit oleh Staphylococcus aureus memerlukan alternatif bahan antibakteri yang efektif dan dapat diformulasikan untuk penggunaan topikal. Penelitian ini bertujuan memformulasikan ekstrak etanol daun glodokan tiang (Polyalthia longifolia) menjadi gel, mengevaluasi mutu fisiknya, dan menguji aktivitas antibakteri terhadap S. aureus. Serbuk simplisia sebanyak 500 g dimaserasi menggunakan etanol pro analysis dan menghasilkan 35 g ekstrak kental dengan rendemen 7%. Gel dibuat dalam empat formula, yaitu F0 tanpa ekstrak, F1 1%, F2 1,5%, dan F3 2%. Evaluasi meliputi organoleptik, homogenitas, pH, viskositas, daya sebar, daya lekat, stabilitas enam siklus selama 12 hari, iritasi, dan aktivitas antibakteri dengan metode difusi sumur. Hasil skrining fitokimia menunjukkan ekstrak positif mengandung flavonoid. Seluruh formula homogen, mempunyai pH 4,80–6,06, daya sebar 5,5–6,8 cm, daya lekat 11,15–21,72 detik, dan viskositas 19.720–19.980 mPa·s. Tidak ditemukan perubahan organoleptik bermakna setelah cycling test dan tidak tampak iritasi pada 12 sukarelawan. Diameter zona hambat F1, F2, dan F3 berturut-turut sebesar 15,80 ± 0,56; 16,63 ± 0,51; dan 17,83 ± 0,68 mm, sedangkan F0 tidak menghasilkan zona hambat. Formula F3 menunjukkan aktivitas antibakteri terbesar dan memenuhi parameter mutu fisik yang dievaluasi.

Abstract

Introduction: Peperomia pellucida (L.) Kunth, commonly known as Chinese betel leaf, is a medicinal plant widely used in traditional medicine for various ailments. The plant is reported to contain diverse secondary metabolites with potential pharmacological activities, including antioxidant properties. However, comprehensive information on its phytochemical constituents and antioxidant potential remains limited, particularly for extracts of Indonesian origin. Objective: This study aimed to determine the phytochemical profile, chemical constituents, and antioxidant activity of the ethanol extract of P. pellucida leaves. Methods: Dried leaf powder (100 g) was extracted by maceration using 96% ethanol (1:10 w/v) for 3 × 24 hours at room temperature. Phytochemical screening was conducted using standard qualitative tests for alkaloids, flavonoids, glycosides, saponins, steroids/terpenoids, and tannins. Chemical composition was analyzed using Gas Chromatography-Mass Spectrometry (GC-MS). Antioxidant activity was evaluated using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical-scavenging assay, with IC₅₀ values determined by linear regression analysis. Vitamin C served as the positive control. Results: The extraction yielded a concentrated ethanol extract of 11.8% (w/w). Phytochemical screening revealed the presence of flavonoids, glycosides, saponins, and terpenoids/steroids, while alkaloids and tannins were not detected. GC-MS analysis identified 14 chromatographic peaks, with major compounds including methyl hexadecanoate (14.12%), methyl 9-octadecenoate (13.61%), and triacontanoic acid. The ethanol extract exhibited very strong antioxidant activity with an IC₅₀ value of 41.47 µg/mL, compared to vitamin C (IC₅₀ = 4.083 µg/mL). The linear regression equation for the extract was y = 0.4291x + 32.2016 (R² = 0.9985). Conclusion: The ethanol extract of P. pellucida leaves contains various secondary metabolites and demonstrates very strong antioxidant activity. Although GC-MS analysis was predominantly characterized by fatty acid esters, the observed bioactivity likely results from synergistic contributions of multiple bioactive constituents. Further studies employing LC-MS/MS are recommended to identify specific compounds responsible for the antioxidant activity and to quantitatively determine total phenolic and flavonoid contents to establish their correlation with the observed bioactivity.

Abstract

The misalignment between vocational education curricula and dynamic industrial requirements contributes significantly to graduate unemployment in the chemical manufacturing sector. This study evaluates a community engagement program designed to strengthen industrial chemistry instruction through the collaborative development and handover of industry integrated teaching modules at SMK Negeri 1 Air Putih in partnership with PT Bakrie Renewable Chemicals. Utilizing a non-empirical, participatory implementation framework, the intervention focused on upstream instructional enhancement without experimental sampling. The program generated a standardized annual teaching module encompassing effective lesson planning, industrial case studies, and safety protocols, thereby shifting instructional approaches toward case based and project based learning. Strategic institutional synergy established clear roles, placing pedagogical design under teacher ownership while the corporate partner provided technical context and document production support. A sustainable roadmap incorporating expert validation, periodic content updates, and digital media integration was established to maintain curriculum relevance. The findings demonstrate that upstream instructional support effectively bridges the school to work transition and reinforces vocational learning quality.
