Abstract

Diabetes mellitus (DM) is a serious chronic disease that occurs when the pancreas does not produce enough insulin (a hormone that regulates blood sugar or glucose), or when the body cannot effectively use the insulin it produces. The basic health research report (RISKESDAS), shows that the prevalence of diabetes mellitus in Indonesia is 6.9%. North Sumatra data on the prevalence of diabetes mellitus is 1.8% or around 160 thousand people. Problems that arise in patients with DM include the emergence of DM ulcers. These diabetic ulcers or wounds usually often occur on the hands or feet of the patient. The prevalence of diabetic wounds in Indonesia is around 15%, the amputation rate is 30%, the mortality rate is 32% and diabetic wounds are the most common cause of hospitalisation at 80%. A natural material that can be used as an alternative biological therapy for wound management is putat air [Barringtonia racemosa (L). Spreng]. This plant contains many saponins that are useful as anti-inflammatory, antinociceptive and help the proliferation process by increasing collagen production. The effectiveness and convenience of using putat air (B. racemosa) seed extract on diabetic wounds is improved by formulating it into a gel dosage form with carbopol 940 as the base material. This study aims to identify the effectiveness of putat air (B.racemosa) seed gel on diabetic wound healing. The research design was quasy experiment pre post design. The number of samples was 32 people. The sampling technique was total sampling. The research was conducted at Asri Wound Clinic Medan. The instrument used for diabetic wound monitoring was Bates Jonson Assessment Tool (BWAT). Data analysis using paired t test. The results of the wound assessment of respondents for 4 times with an interval of 4 days. The 4th observation showed a decrease in score. Based on the BWAT wound healing continuum, the respondents' wound conditions were still in the wound degeneration phase. The majority of the wound area < 80cm; stage 3 depth; clear wound edges, not fused with the wound bed, GOA 2-4 cm > 50%; the edge of necrosis tissue is sticky, soft and there is black false scarring (black eschar); the amount of necrosis tissue > 50% of the wound bed; the skin colour around the wound is dark red; oedematous tissue < 4mm around the wound; granulation tissue 25 and epitalisation < 25%.  The statistical test results of the difference in mean 3.782 and P value = 0.001 (<0.05), it can be concluded that there is a significant difference in the wound healing scores of respondents before and after the intervention. This study proves that the gel of putat air (B. racemosa) seed extract can be used as an alternative to diabetic wound healing, especially in the inflammatory phase as auotolysis.

Abstract

Obat anti tuberkulosis primer sebagai standar terapi tuberculosis sering digunakan dalam bentuk kombinasi untuk memperoleh efektivitas pengobatan dan mencegah timbulnya resistensi. Salah satu bentuk kombinasi tersebut adalah kombinasi INH dan vitamin B6. Kombinasi ini bertujuan untuk untuk mencegah efek samping dari INH yaitu neuritis perifer Namun, kombinasi bahan aktif tersebut dapat menimbulkan masalah dalam analisis kuantitatif untuk kontrol kualitas sediaan. Penelitian ini bertujuan untuk mengembangkan metode spektrofotometri UV-Vis secara mean centering of ratio spectra pada analisis isoniazid dan vitamin B6 secara simultan pada sediaan tablet tanpa adanya pemisahan. Penelitian dilakukan dengan mengembangkan metode spektrofotometri teknik mean centering of ratio spectra pada analisis simultan INH dan vitamin B6 pada sediaan tablet, kemudian dilakukan uji validitas metodenya berdasarkan parameter validasi yaitu liniearitas, akurasi, presisi, selektivitas dan sensitivitas. Hasil penelitian menunjukkan bahwa aplikasi secara mean centering of ratio spectra pada penetapan kadar INH dan vitamin B6 berhasil diterapkan untuk analisis formulasi farmasi secara simultan, tanpa gangguan dari eksipien serta hasil parameter validasi berada dalam kisaran yang dapat diterima. Abstract

Buah bit telah lama dimanfaatkan masyarakat Afrika sebagai obat tradisional untuk meningkatkan stamina dan mengurangi masalah pencernaan karena kandungannya yang kaya akan karbohidrat, beta karoten, protein, serat, kalsium dan vitamin C. Di Indonesia sendiri khususnya di Desa Semangat, Kecamatan Merdeka, Kabupaten Karo, pengolahan buah bit hasil tani masyarakat kurang dapat dioptimalkan karena rasanya yang langu dan baunya yang kurang menyenangkan membuat masyarakat kurang menyenangi buah ini. Pengabdian masyarakat ini bertujuan untuk memberikan alternatif kepada masyarakat Desa Semangat dalam mengolah buah bit menjadi hidangan minuman dan makanan sehat. Tim telah melakukan sosialisasi, edukasi dan pendampingan dalam mengolah buah bit menjadi minuman dan makanan sehat berupa sirup dan dodol. Sebelum kegiatan pengabdian masyarakat dilakukan, tingkat pemahaman dan minat masyarakat berada dalam rentang 16-68%, sedangkan setelah kegiatan pengabdian masyarakat dilakukan, tingkat pemahaman dan minat masyarakat berada dalam rentang 80-100%. Pengabdian masyarakat yang telah dilaksanakan ini telah terbukti meningkatkan minat dan pemahaman masyarakat mengenai pemanfaatan buah bit menjadi olahan produk pangan berupa dodol dan sirup konsentrat. Selain bermanfaat bagi kesehatan, olahan produk ini dapat menjadi produk usaha yang memiliki peluang daya saing yang tinggi ke depannya.

Abstract

DAGUSIBU adalah singkatan dari DA (Dapatkan obat dengan benar), GU (Gunakan obat dengan benar), SI (Simpan Obat dengan benar) dan BU (Buang obat dengan benar) adalah program tentang cara mengelola obat mulai dari cara mendapatkannya sampai membuangnya. Di Indonesia khususnya Desa Biru-Biru, Kecamatan Biru-Biru, Kabupaten Deli Serdang, pemahaman Masyarakat dalam menggunakan dan mengelola obat dengan cara yang baik dan tepat masih kurang. Pengabdian Masyarakat ini bertujuan untuk memberi edukasi kepada masyarakat terkait cara penggunaan obat yang baik dan benar, menjamin kehidupan yang sehat dan meningkatkan kesejahteraan sehingga diharapkan masyarakat khususnya masyarakat Desa Biru-Biru Kecamatan Biru-Biru Kabupaten Deli Serdang lebih edukatif dan informatif dalam menggunakan obat-obatan.

Abstract

Metamizole is a nonsteroidal antiinflammatory drug (NSAID) that functions as an analgesic, antipyretic, and antiinflammatory. Examination of active substance contents is a requirement that must be met to ensure the quality of drug preparations. The aims of this study were to develop and validate the Fourier Transform Infrared spectrophotometric method for the quantitation of metamizole content in marketed tablet preparation. Identification and determination of metamizole contents by Fourier Transform Infrared spectrophotometric method used methanol solvent in the wavenumber range 4000 cm–1 to 650 cm–1. The results showed that the specific wavenumbers of metamizole were 1649.3 cm–1; 1623.3 cm–1; and 1589.7 cm–1; and the contents metamizole in marketed tablet preparation ranged from (97.954 ± 0.121)% to (104.541 ± 0.257)%. From the validation method, the recovery result is 100.129%; the relative standard deviation is 0.057%; the limit of detection is 2.09526 mg/mL; the limit of quantitation is 6.34928 mg/mL; and the range 40 mg/mL to 60 mg/mL. The quantitation of metamizole contents can be carried out by Fourier Transform Infrared spectrophotometric method with accurate and precise quantitation results.

Abstract

Syzygium belongs to the guava tribe or Myrtaceae, originating from Southeast Asia. Syzygium is divided into two: small water guava known as Syzygium aqueum and large water guava known as Syzygium samarangense. This study aims to determine the mineral levels of iron, potassium, calcium, and sodium contained in guava and guava Semarang and to compare mineral levels between a guava and guava Semarang. Each sample was dry digested, and then quantitative analysis of iron, potassium, calcium, and sodium was carried out using atomic absorption spectrophotometry (AAS) with a wavelength of 248.3 nm; potassium 766.5 nm; calcium 422.7; and sodium 589.0 nm. The advantage of this method is that it can determine the content of minerals in small quantities without being influenced by other minerals. The results showed the levels of iron, potassium, calcium, and sodium in guava and guava Semarang, respectively: (0.2553 ± 0.0077) mg/100g and (0.1548 ± 0.0152) mg/100g; (70.2326 ± 1.5738) mg/100g and (45.7714 ± 0.9827) mg/100g; (3.8588 ± 0.1619) mg/100g and (0.4663 ± 0.0507) mg/100g; (8.1187 ± 0.3375) mg/100g and (6.1648 ± 0.1689) mg/100g. A comparison of mineral content between guava and guava Semarang shows that iron, potassium, calcium, and sodium minerals in guava are more significant than in guava Semarang. Statistically, the different tests of the average content of iron, potassium, calcium, and sodium between guava and guava Semarang using the F distribution found that the content of iron, potassium, calcium, and calcium sodium in guava was significantly higher than in guava Semarang.

Abstract

One of the medicinal Zingiberaceae plants which is regularly used by the society as a traditional remedy is turmeric rhizome. The pharmacological properties of turmeric rhizome include anti-inflammatory, anti-immunodeficiency, anti-viral. Anti-bacterial, anti-fungal, anti-oxidant, anti-carcinogenic, and anti-infectious. This study aimed to identify the immunomodulator activity of curcuminoid extract of turmeric rhizome against the increase of leucocyte and its components, lymphocyte and neutrophil in white Galur Wistar rats that had been injected with Staphylococcus aureus. These test animals were given curcuminoid extract in VCO 100 mg/ml on a dosage of 1 ml, 0,5 ml, 0,25 ml curcuminoid powder 200 mg/kg body weight. Curcuminoid was suspended in Na CMC0,5%, Levamisole 25 mg/kg of body weight suspended in Na CMC 0,5%, VCO 1 ml. For the negative control, merely Na CMC 0,5% was given. The extract was orally given daily for 14 days. On the 4th day, Staphylococcus aureus was injected intra-peritoneally. The rats’ blood was then drawn from the heart and was observed to calculate the volume of the leucocytes, lymphocyte, and neutrophil which was conducted using flow cytometry.The findings suggested that there was a significant difference between the immunomodulatory activity of the curcuminoid extract in VCO at the doses of 1 ml, 0,5 ml, 0,25 ml on the increase in leucocyte, lymphocyte, and neutrophil compared to that of the negative control.Therefore, the findings of this study indicated that the extracts of curcuminoid in VCO could increase the number of leucocytes, lymphocytes, and neutrophils. Thus, the curcuminoid extract of turmeric rhizome in VCO potentially possessed immunomodulatory activity.

Abstract

Nanoparticles are particles measuring 1-100 nm that were synthesized using metals, and silver was a commonly used metal because of its potential as an antibacterial agent. Physical and chemical methods were often used in synthesis but are less environmentally friendly. Alternatively, biological processes that utilize phytochemical compounds as reducing agents are preferred. One of the plants rich in phytochemical compounds was kluwih leaves (Artocarpus camansi Blanco). This study aimed to synthesize silver nanoparticles (AgNPs) mediated by kluwih leaves using microwave irradiation. Characterization of nanoparticles was carried out by visual observation and using UV-Vis spectrophotometry. Antibacterial activity was evaluated by determining MIC (Minimum Inhibitory Concentration) and MBC (Minimum Bactericidal Concentration), followed by examining DNA and protein leakage and biofilm formation using a microdilution method against S. epidermidis and P. aeruginosa. The results showed that AgNPs from kluwih leaves had been successfully formed and could be used as reducing agents in AgNPs biosynthesis. Visual color changed, and analysis of UV-Vis spectrophotometry showed peaks at 441 nm. The results of the antibacterial evaluation showed MIC 1.90 µg/ml (S. epidermidis) and 7.80 µg/ml (P. aeruginosa) also caused cell leakage and reduced biofilm formation significantly at a concentration of 1/2 MIC. These results were promising for further development in future research.

Abstract

Sweeteners, preservatives, and caffeine in soft drinks can pose health risks if consumed excessively. This study aimed to develop a method for the simultaneous determination of acesulfame, saccharin, cyclamate, aspartame, caffeine, benzoate, and sorbate in carbonated soft drinks. The analysis was conducted using reversed-phase high-performance liquid chromatography (RP-HPLC) with a dual-wavelength PDA detector (Shimadzu LC-20AD), a C18 column (Shimadzu), and a UV-Vis spectrophotometer (Shimadzu). Standard materials included acesulfame-K, saccharin-Na, cyclamate-Na, aspartame, caffeine, benzoate-Na, and sorbic acid. Samples were obtained from a supermarket in Medan, Indonesia. Optimization parameters included detection wavelength, mobile phase pH, column oven temperature, and mobile phase composition. Validation parameters assessed were linearity, limit of detection (LOD), limit of quantification (LOQ), accuracy, precision, and selectivity. The optimized method employed a mobile phase of phosphate buffer (pH 3.8) and methanol (80:20, v/v) at a flow rate of 0.55 mL/min and column oven temperature of 40 °C. Detection was carried out at 200 nm for cyclamate, caffeine, aspartame, and benzoate, and at 220 nm for acesulfame, saccharin, and sorbate. Validation results showed the method met all requirements, with recovery rates ranging from 95.21% to 99.82%, system suitability values of 1.0%–1.9%, and precision values between 1.65% and 2.48%. The method also demonstrated good selectivity. The concentrations of acesulfame, saccharin, cyclamate, caffeine, benzoate, and sorbate in the analyzed samples did not exceed the maximum permissible limits.

Abstract

ABSTRACT. Marketed pharmaceutical dosage forms must be checked to ensure that the drug in distribution meets the quality requirements. The Fourier transform infrared spectrophotometric method will provide new innovations which are low cost, environmental friendly, simple, and rapid for analysis. The aim of this research was to develop a single analytical method that can be used to determine levels of three different statin derivatives (simvastatin, atorvastatin, and rosuvastatin) in tablet preparations. The method used in this study was Fourier transform infrared spectrophotometric, a very simple sample preparation stage by solvent extraction with minimal solvent extraction without potassium bromide and without pellet press. The specific wave numbers of simvastatin, atorvastatin, and rosuvastatin were determined between 4000 cm–1 to 650 cm–1. The Fourier transform infrared spectrophotometric method had been successfully developed and successfully applied to the determination of levels of simvastatin, atorvastatin, and rosuvastatin in marketed tablet preparations containing single active pharmaceutical ingredient. The specific wave numbers of simvastatin, atorvastatin, and rosuvastatin obtained was 1712.7 cm–1; 1660.5 cm–1; and 965.4 cm–1, respectively. The results obtained in the content determination based on the peak height and peak area fulfill the requirements for active pharmaceutical ingredient assay ranging from 90.00% to 110.00%. The developed method has also been validated for accuracy, precision, specificity, linearity, range, limit of detection, and limit of quantitation. Thus, this method can be a valid alternative method, cheap, green, easy, and fast in determination of the levels of simvastatin, atorvastatin, and rosuvastatin in tablet preparations.
