Abstract

Background: Environmental DNA (eDNA) metabarcoding is a new method for assessing biodiversity in which samples are taken from the environment via water, sediment or air from which DNA is extracted, and then amplified using common or universal primers in a polymerase chain reaction and sequenced using next-generation DNA. sequencing to produce thousands to millions of sequences. From this data, the presence of species can be determined, and overall biodiversity can be assessed.. Methods: The method used is a literature review using reputable national and international journals Results: Based on the results of the literature review, the use of eDNA metabarcoding can be used to analyze fish biodiversity. eDNA metabarcoding can be analyzed with primary data, databases, and bioinformatic pipelines. Conclusion: eDNA metabarcoding has been widely used to study fish communities in lakes and rivers, as well as coastal fish communities. Despite its considerable ability to detect fish species in large aquatic systems, many methods still need to be understood to improve the quality of primers, databases, and pipelines for analyzing fish biodiversity

Abstract

Background: The biodiversity of citrus plants is very abundant and many are cultivated, creating various types. However, the high demand makes orange farmers feel overwhelmed. This is what causes some orange cultivars to become rare. Characterization of the genetic diversity of citrus germplasm at the molecular level is an important and crucial step that can assist in the breeding and conservation process. The aim of this literature review is to provide information regarding molecular markers regarding the application of SSR molecular markers to analyze the genetic diversity of citrus. Method: The method used is a systematic literature review with the literature sources used coming from articles taken from national journals and international journals indexed by Scopus. Results: The application of molecular markers using the SSR method is useful for identifying and characterizing the genetic diversity of citrus germplasm so that it can assist in breeding and conservation actions. SSR has many advantages both in terms of its use and the results obtained. Conclusion: One application of molecular markers is to analyze plant genetic diversity. Simple Sequence Repeat (SSR) molecular markers can help to analyze the characteristics and genetic diversity of oranges so that they can assist in plant breeding and conservation purposes.

Abstract

This study aims to analyze the feasibility of fruit fly e-booklets (Bactrocera spp.) for grade X SMA students on biodiversity material. This study analyzes the validity, readability test, and response of e-booklets. The method used is Research & Development (R&D) with the development model used is ADDIE with three stages, namely analysis, design, and development. The feasibility of the e-booklet media used is determined based on validation from material experts and media experts. The response to the e-booklet is determined based on teacher and student responses. The readability test using the gap test was carried out in grade X. The results of the material validation got a percentage of 91.2% with a very valid category, media validation got a percentage of 91.6% with a very valid category, the readability test got a percentage of 98.7% with an independent level category, teacher responses got a percentage of 90%, and student responses got a percentage of 82% with a very good category. Based on the results of the study, it was concluded that the e-booklet media was very valid to use, positive responses were very good from teachers and students, and readability included an independent level or free which means easy to read.

Abstract

Background: The development of edible fruit quality is a vital process. Banana fruit ripening is a complex biochemical and physiological process that causes physical changes such as softening. Fruit aging and ripening are greatly influenced by transcriptional regulation. Transcription factors also play an essential role in regulating various biological processes. Therefore, this study aimed to explore information about the role of transcription factors in banana fruit ripening. Methods: A systematic review was conducted by searching the Scopus database indexed Q1-Q4 through the Watase UAKE application from 2018 to 2023 to find relevant studies on the role of transcription factors in banana fruit ripening. Results: From the search results, 157 articles were found, which were then narrowed down to 11 articles by considering the established inclusion criteria. This study identified 11 transcription factors that have the potential for fruit ripening. Conclusions: The findings of this article review indicate that there are transcription factors, namely MYB, MaWRKY49, MabHLH7, MaNAC42, MaMADS36, bZIP21, MaMYB3, MabHLH, MaERF012, NAC, and WRKY. The role of transcription factors can control fruit quality in agricultural commodities that play genetics in molecular biology. These findings indicate that knowledge of the role of transcription factors can provide insight into the development of innovations in agriculture in the future.

Abstract

Usaha keripik pisang “Noli” di Desa Kebondalem, Kec. Jambu, Kab. Semarang, Jawa Tengah memproduksi keripik pisang asin dan manis, belum memiliki varian baru. Kemasan yang digunakan sering bocor dan keripik melempem. Daya simpan keripik pisang terbatas dan mengalami ketengikan. Manajemen usaha masih dilakukan secara konvensional dan lantai tempat pencucian masih berlantai semen, terkesan kurang higienis. Tujuan kegiatan ini adalah menambah varian produk keripik pisang, memperbaiki manajemen keuangan, teknik pengemasan, dan tempat pencucian agar lebih bersih. Sekitar 75% kegiatan menggunakan metode praktik, sedangkan 25% berupa kegiatan informatif, diskusi, dan tanya jawab. Hasil kegiatan ini adalah pelaku UMKM berhasil membuat keripik pisang coklat bubuk, coklat lumer, matcha, red velvet, dan tiramisu, serta mengemasnya menjadi layak jual di toko oleh-oleh. Pelaku UMKM telah mampu membuat manajemen keuangan digital menggunakan aplikasi “Akutansiku” dan tempat usaha yang lebih bersih. Pelaku UMKM juga sudah mampu mengoperasikan mesin peniris minyak. Kesimpulan dari kegiatan ini adalah pelaku UMKM “Noli” berhasil memdiversifikasi produk keripik pisang, lebih melek manajemen keuangan digital, terampil mengemas produk, dan meningkatkan kebersihan tempat kerja, yang diharapkan dapat menguatkan ekonomi lokal, sehingga mendukung pencapaian Tujuan Pembangunan Berkelanjutan ke-8, 9, dan 12. Abstract

Anggrek merupakan tanaman dengan nilai ekonomis tinggi. Griya anggrek Mekar Sari merupakan salah satu rumah anggrek di wilayah Kalisegoro yang mempunyai potensi besar untuk berkembang. Kegiatan saat ini yang dilakukan di Griya Mekar Sari hanya pemeliharaan dan penjualan anggrek saja. Rutinitas ini memberikan dampak menurunnya antusiasme pengelola. Selain itu tidak beragamnnya produk anggrek yang ditawarkan menyebabkan turunnya omzet penjualan. Sejak berdiri hingga sekarang rerata penjualan anggrek hanya berkisar 4 tanaman perbulan. Jumlah penjualan ini masih rendah dan belum memberikan dampak ekonomi yang berarti bagi anggota Griya Anggrek Mekar Sari. Tingkat penjualan yang rendah antara lain disebabkan karena koleksi anggrek yang masih terbatas dan tidak adanya produk lain yangberkaitan dengan anggrek yang dijual. Perlu adanya suatu terobosan berupa diversifikasi produk yang dapat menarik pembeli dan juga meningkatkan antusime pengelola. Solusi dari berbagai permasalahan yang dialami oleh mitra akan dilakukan bersama-sama antara tim pengusul dengan mitra. Jabaran dari solusi tersebut adalah sebagai berikut: (1) Peningkatan koleksi anggrek. (2) Peningkatan diversifikasi produk anggrek. Diversifikasi produk yang ditawarkan adalah kit pemeliharaan anggrek (berisi media tanam, pot kecil dan bibit anggrek dalam botol kecil) dilengkapi dengan manual penggunaannya, gantungan kunci dan hiasan meja berisi tanaman anggrek, peralatan standar pemeliharaan anggrek, pot cantik homemade, dan juga produk berupa kemasan anggrek yang menarik. Diharapkan diversivikasi produk ini akan meningkatkan omzet penjualan dan juga meningkatkan motivasi bagi pengelola Griya Anggrek Mekar Sari.

Abstract

The conservation of  Vanda floresensis Motes has been conducted using in vitro techniques and has produced a large number of plantlets. These plantlets require acclimatization, but the optimal method has not yet been found. The aims of the research were to develop an acclimatization protocol by verifying the effectiveness of an acclimatization chamber (AC), chitosan spraying, and mixed medium composition on plantlet growth.  The research was conducted in a screen house of Universitas Negeri Semarang, using a factorial randomized block design with three factors. The AC factor consisted of two levels: inside and outside the AC. The chitosan concentration factor consisted of five levels, namely 0.0, 1.5, 3.0, 4.5, and 6.0 ppm. The composition of the mixed medium consisted of three levels. The observed variables included the increase in number, length, and width of the leaf, total chlorophyll content, and wilted leaf. Data were analyzed by ANOVA and Duncan test. It can be concluded that the use of AC, chitosan spraying, and mixed medium composition was effective in increasing leaf growth and total chlorophyll content and reducing wilted leaf.  Spraying chitosan of 1.5 - 3.0 ppm with the use of AC and spraying chitosan of 1.5 ppm with a mixed medium of brick pieces, coconut fibre, and tree bark chips with a ratio of 1:2:1 resulted in the highest leaf growth and total chlorophyll content, and the lowest wilted leaf. The results of this study can be used as an acclimatization protocol for orchid propagation for conservation purposes.

Abstract

Guru Biologi Madrasah Aliyah Kota Semarang yang mengajar Prakarya dan Kewirausahaan (PKWU) mengeluhkan semakin menurunnya kreativitas peserta didik. Mereka cenderung mereplikasi atau menduplikasi karya kakak kelas, sehingga nilai kreativitas dan inovasinya rendah. Kecenderungan ini harus segera diminimalkan. Teknik eco-print merupakan salah satu jawaban terhadap permasalahan tersebut karena hasilnya bersifat unik atau ekslusif. Oleh karena itu perlu dikenalkan dan dilatihkan keterampilan teknik eco-print kepada guru-guru Biologi MA Kota Semarang. Tujuan pengabdian pada masyarakat adalah mengenalkan, menambah pengetahuan dan keterampilan teknik eco-print pada guru-guru Biologi MA Kota Semarang. Mitra kegiatan ini adalah MGMP Biologi MA Kota Semarang dan diikuti oleh 10 guru terpilih. Kegiatan pengabdian pada masyarakat diawali dengan pretest, pengenalan tentang teknik eco-print, praktik teknik pounding, praktik teknik steaming, fiksasi warna, diskusi, posttest, dan tugas mandiri. Dari segi pengetahuan diperoleh N-gain= 0,502 (kategori sedang). Semua peserta aktif dalam kegiatan dan mampu menghasilkan eco-print dengan teknik pounding dan steaming menggunakan kain Primisima, dan berhasil membuat totebag eco-print dengan teknik pounding secara mandiri. Hasil tersebut menunjukkan bahwa dari segi pengetahuan teknik eco-print peserta mengalami peningkatan. Dari segi keterampilan semua peserta mampu membuat kain eco-print dengan teknik pounding maupun steaming. Secara mandiri, peserta sudah mampu membuat totebag eco-print dengan teknik pounding. Keterampilan peserta masih harus ditingkatkan dengan secara mandiri atau berkelompok dengan mencoba berbagai variasi untuk melatih kreativitas serta inovasi, sehingga mampu menularkan pengetahuan dan keterampilannya kepada para siswa.

Abstract

This study aims to determine the types of ferns in the Curug Winong and the results of the E-booklet product feasibility test from the research results. This research uses the Research and Development method with a size trap of 4 x 4 m. The results of the identification of ferns recorded 39 species of ferns which are divided into 17 families, 6 ordo, and 3 classes. The dominant percentage can be seen from the highest INP calculation, namely P. Biaurita and S. Ornate, while the exposed INP values are A. Evecta and G. Verrucosum. The index of plant diversity in Curug Winong is of moderate criteria. The results of the validation of material experts (77.78%), media (96.13%), teacher responses (91.67%), overlap test (83.66%), and student responses (83.90%). These results indicate that the E-booklet that was developed is suitable to be used as a learning resource.

Abstract

Livestock farming is an important sector in meeting the food needs of the community, especially for animal protein from meat and dairy products. Although livestock farming has great potential, livestock productivity still faces significant challenges, including inadequate husbandry systems, technological limitations, and a lack of understanding of reproductive management. In an effort to improve productivity, two approaches that can be applied are Good Farming Practices (GFP) and Artificial Insemination (AI) based on molecular biology. GFP includes principles of feed management, sanitation, livestock health, and a good environment, which are expected to improve livestock welfare and the quality of production results. On the other hand, IB uses advanced technology to improve the genetic quality of livestock and enhance reproductive efficiency, particularly with the integration of molecular biology techniques. This study aims to compare the effectiveness of both approaches in improving livestock productivity. By applying GFP and IB synergistically, it is hoped that the increasing market demand can be met without neglecting animal welfare aspects.
