Abstract

Hydrolase enzymes are a class of enzymes used to break down the organic substrates into simpler molecules. Hydrolase enzymes play a role in biocontrol, industry, and public health. One of the hydrolase enzyme producers is bacteria. Serratia marcescens strain MBC1 is a collection in the Microbiology Laboratory of the Department of Biology, Faculty of Mathematics and Natural Sciences, University of Lampung which enzymatic activities haven’t been known yet. This research aimed to determine the enzymatic activity of S. marcesescens strain MBC1 that can be used as a candidate for biocontrol agents, biosurfactant producers, industry, or in the health sector. The screening was carried out using a qualitative method plate assay using selective solid media for each test, with three replications for each test. The data obtained were analyzed statistically using analysis non-parametric kruskall-wallis test. The lipolytic activity of the S. marcescens strain MBC1 showed high efficacy with an enzymatic index of 5.52 and amylase, cellulases, protease, mannanase, and chitinase with enzymatic indexes, respectively, 4.17, 1.50, 0.69, 0.35, and 0.27. This enzyme activity of this isolate can be used as a candidate for biological control agents,biosurfactant producers, industry, or the health sector.

Abstract

More than 70% of the earth's oceans are known to cover the surface of the earth. High biodiversity makes the ocean a habitat for microorganisms such as tunicates. Tunicates can associate with other microorganisms such as Pseudonocardia carboxydivorans so that they can produce bioactive compounds including antibiotics, pesticides and antitumors. The initial stage to identify a bioactive compound is by isolating DNA. This study aims to determine the optimization of bacterial DNA extraction using promega and qiagen extraction kits. This study consists of 3 treatments, namely rejuvenation, bacterial identification and concentration measurement. The results showed that the optimization of using qiagen extraction kit was relatively superior at 1,989 ng/µl - 2,000 ng/µl with 1 hour. While in the optimization of promega extraction, the resulting purity is 1,500 ng/µl - 1,943 ng/µl with a processing time of 2 hours.

Abstract

Hutan Kemasyarakatan (HKm) termasuk hutan negara. Pemanfaatannya adalah untuk pemberdayaan masyarakat sekitar hutan. Untuk itu dalam pengelolaannya masyarakat harus dilibatkan agar tujuan dari pemberdayaan itu dapat tercapai. Peningkatan pendapatan menjadi tolak ukur untuk melihat kesejahteraan masyarakat, diantaranya adalah dengan pemberian stup (kotak) lebah Trigona (Apis trigona). Pemberian stup lebah ini bertujuan untuk membantu petani lebah di Hutan Kemasyarakatan Kecamatan Pagar Dewa Lampung Barat guna menambah pendapatannya. Hal itu disebabkan karena kurangnya stup lebah yang tersedia serta kurangnya sumber pakan menjadi hambatan dalam meningkatkan hasil panen madu oleh petani. Disini, artikel ini memberikan gambaran dan penjelasan tentang pelaksanaan program kerja Hutan Kemasyarakatan melalui pemberian stup lebah Trigona kepada Kelompok Tani Hutan (KTH) Wonorejo di Pekon Basungan, Lampung Barat. Pemberian stup lebah dilaksanakan menggunakan pendekatan kualitatif dimana modelnya adalah model naratif. Hasilnya, Kelompok Tani Hutan Wonorejo mendapatkan tambahan stup lebah dan tambahan sumber pakan untuk meningkatkan hasil panen madu. Artikel ini lebih lanjut diharapkan dapat menambah wawasan dalam bidang pertanian dan kehutanan khususnya petani untuk mengembangkan hasil hutan bukan kayu berupa pembudidayaan lebah madu. Kata Kunci: Hutan Kemasyarakatan, Peningkatan Pendapatan, Lebah Trigona, Hasil Hutan Bukan Kayu  Community Forests (Hutan Kemasyarakatan/HKm) are state forests. The use is for empowering communities around the forest. For this reason, in its management, the community must be involved so that the goals of empowerment can be achieved. Increasing income is a benchmark for assessing community welfare, including providing trigona bee stup. The aim of providing bee stup is to help bee farmers in the Pagar Dewa District Community Forest, West Lampung, to increase their income. This is because farmers are unable to increase their honey output due to a shortage of feed sources and bee stup. Here, this article provides an overview and explanation of the implementation of the Community Forest work program by providing Trigona bee stup to the Wonorejo Forest Farmers Group (Kelompok Tani Hutan/KTH) in Pekon Basungan, West Lampung. The provision of bee stup is carried out using a qualitative approach where the model is a narrative model. As a result, the Wonorejo Forest Farmers Group received additional bee stup and additional food sources to increase honey yields. It is intended that this essay will deepen readers' understanding of the domains of forestry and agriculture, particularly helping farmers create forest-based products that are not timber by raising honey bees.

Abstract

Eggplant (Solanum melongena L.) is an authentic plant native to tropical climates, widely favored by the community, and available at an affordable price. The demand for eggplant in Indonesia reached 69,963 tons in 2015 and has continued to increase to 74,081 tons. Based on FAO data in 2016, Indonesia is one of the eggplant-producing countries, ranking sixth in the world after China, India, Egypt, and Turkey. The Laguna F1 eggplant variety is known for its quality and resistance to bacteria-causing wilting. To minimize the cultivation time of eggplants, it is necessary to use readily available natural fertilizers, one of which is coconut water. The utilization of coconut water as fertilizer is due to its content of various hormones that play a crucial role in plant growth, namely auxin, cytokinin, and gibberellin. The purpose of this research is to investigate the effects of coconut water and atonic, determine the optimum concentrations, and understand the interaction between coconut water and atonic in the growth of eggplant plants. The research was conducted in the Botany Laboratory of the Department of Biology, Faculty of Mathematics and Natural Sciences, Universitas Lampung, in October-November 2019, using a 2x3 factorial design. The first factor involved coconut water at concentration levels of 25%, and 50%, and compared with a control group (0%). The second factor was the atonic solution with a concentration of 10 ml/l, compared with a control. The treatments were repeated in five replications, resulting in 30 experimental units. Observed variables included plant height, fresh weight, dry weight, relative water content, and chlorophyll content. The results of this research can be summarized as follows: the application of coconut water and atonic has a positive effect on the growth of eggplant plants. Additionally, the combined induction of coconut water at a concentration level of 25% and atonic at 0% demonstrated superior outcomes in terms of plant height, fresh weight, dry weight, relative water content, and chlorophyll content in eggplant plants.

Abstract

Desa Suka Mulya terletak di Kecamatan Pagar Dewa Kabupaten Lampung Barat.Sebagian besar orang di desa tidak tahu bagaimana menggunakan limbh rumah tangga, terutama nasi basi. Mengolah sampah dari sumbernya sendiri, yang dapat mengurangi volume sampah, adalah solusi untuk masalah sampah rumah tangga yang meningkat. Menggunakan beras basi sebagai pupuk akan mengurangi jumlah sampah organik yang ada di lingkungan. Masyarakat hanya memanfaatkan sisa nasi untuk pakan ternak seperti unggas, tetapi informasi ini dapat membantu mereka mengubah sisa nasi menjadi pupuk cair. Dengan demikian, sampah di masyarakat dapat dikurangi dan pemanfaatannya akan lebih menguntungkan. Untuk mendapatkan hasil pertumbuhan terbaik, pupuk harus digunakan secara teratur selama minimal dua minggu

Abstract

The global market demand for vaname shrimp in Indonesia is increasing every year. Vannamei shrimp is a mainstay of fisheries commodities. Cultivators often encounter vibriosis disease caused by Vibrio sp. Therefore, it is necessary to pay attention to the quality of the feed by adding supplements from synbiotics and empon-empon. This study aims to determine the effect of synbiotics and empon-empon on the growth of white shrimp (Litopenaeus vannamei) larvae in controlling Vibrio sp. Parameters measured included daily length growth, survival, water quality, total bacteria, and total vibrio. Water quality and total bacteria are growth-supporting factors. The treatments consisted of positive control (C+), negative control (C-), Synbiotic 1 (S1), Synbiotic 2 (S2), Synbiotic 3 (S3), Synbiotic 4 (S4), and empon-empon (E). The synbiotics used were Bacillus sp 1x 1010. as a probiotic, 2 ppm yam paste as a prebiotic and 1 ppm  ginger, 1 ppm white turmeric, and 1 ppm black cumin. And the combination of the three types of empon-empon with a concentration of 0.5 ppm (treatment S4 and E). The results showed that the S2 treatment had a significant effect (P>0.05) on the daily length of the fry with an average of 0.05±0.06 mm. The S4 treatment had a significant effect on survival with a percentage of 83.6%. The lowest total Vibrio was shown in the S4 treatment with a value of 0.45±0.5 logs CFU/ml. The total density of bacteria was shown in the S2.treatment with a value of 2.28 ±log CFU/ml.

Abstract

Cooking oil contains fatty acids made from high-quality vegetable oil so it can be used for environmentally friendly food products. However, repeated heating processes can damage the molecular structure of the oil. So that it can cause several negative impacts such as on health if consumed too often and damage to the ecosystem in the surrounding environment if disposed of carelessly. To overcome these negative impacts, cooking oil waste can be processed into products such as laundry soap. This community service activity was carried out to educate the community, especially those in Gebang Village, Pesawaran Regency, about the dangers of repeated heating of cooking oil, as well as to provide alternatives to process it into more useful products. The results of this community service activity showed an increase in understanding for participants, this can be seen from the increase in the post-test scores worked on by participants and from the results of discussions during the activity.

Abstract

This activity was carried out in Banjar Agung Udik Village, Pugung District, Tanggamus Regency. The diversity of flora in Banjar Agung Udik Village creates great potential for the development of nature-based art and creativity. The use of natural materials for art and creativity will not only produce unique and aesthetic works of art, but also have high economic potential. One method that can be used is the ecoprint technique, so a deep understanding of the ecoprint technique, research on appropriate natural materials, and training for the local community are needed. The PKM activity was carried out in 2 stages, the first stage was the delivery of material through lectures and discussions, the second stage was a demonstration/practice of making patterned cloth with the ecoprint technique. The evaluation results showed an increase in the pre-test and post- test scores of 2.75 or 13.75%. The conclusion of this activity is that the knowledge, insight, and skills of the community in Banjar Agung Udik Village have increased, especially regarding the use of the ecoprint technique in making patterned cloth.

Abstract

Plankton are organisms that live or float in microscopic water and have an important role in aquatic ecosystems. The Way Umpu River is widely used by local residents as a development area in the agricultural, mining, fishery, transportation, industrial, and domestic needs of the community. Research on plankton diversity was carried out in the Way Umpu River, Way Kanan Regency, Lampung Province from August 2021 to February 2022. The purpose of this study was to determine the plankton diversity index and water quality supported by physical and chemical parameters. Sampling used a survey method based on land use at 7 research stations. The results found that there were 22 types of plankton grouped into 10 classes, namely 4 groups of zooplankton, namely Class Rhizopoda, Euglenophyceae, Ciliatea, and Oligotrichea, and 6 groups of phytoplankton namely Class Cyanophyceae, Bacillariophceae, Trebouxiophyceae, Zygnemophyceae, Chlorophyceae, and Fragilariophyceae. The diversity index (H') of all research stations ranged from 1.06 to 1.77. This shows that the diversity of plankton in the Way Umpu River, Way Kanan Regency is classified as moderate and the water conditions are categorized as moderately polluted with a value range of 1<H'<3.

Abstract

Diseases caused by bacteria can usually be combated by administering antibiotics and using antiseptics. However, there are some groups of pathogens, such Escherichia coli as well as Staphylococcus aureus, which are highly resistant to the antibiotics amoxicillin, penicillin, tetracycline and chloramphenicol. One of the efforts to control these bacteria is by using bioactive compounds derived from plants. Banana plants contain secondary metabolite compounds flavonoids, saponins, and tannins that can act as anti-infective agents, including muli banana (Musa acuminata Linn.). Banana plant roots as natural ingredients can be an alternative to antiseptics because they contain antibacterial compounds with milder side effects than long-term use of alcohol-based antiseptics that can reduce skin moisture. This study was conducted to determine the efficacy of banana muli root extract (M. acuminata Linn.) as an antibiotic and antiseptic. The hypothesis in this study is that the root extract at a certain concentration is thought to have effectiveness as an antibiotic (E. coli) and antiseptic (S. aureus). In this study, the Kirby-Bauer method was applied by agar diffusion and the results of the inhibition zone test were formed around the paper disc. The results showed that the optimum efficacy of muli banana root extract (M. acuminata Linn.) as an antibiotic (E. coli ATCC 25922) was at a concentration of 95% (2.96 mm) and as an antiseptic (S. aureus ATCC 25923) at a concentration of 75% (9.1 mm).
