Abstract

ABSTRACT Diabetes Mellitus (DM) was a disease or disorder of chronic metabolism which be characterized by high blood sugar levels along with carbohydrate, lipid, and protein metabolism disorders as a result of insulin function insufficiency. Treatment of diabetes could be overcome by utilizing nutritious plants, one of which was thegaling leaves (Cayratia trifolia L. Domin.). The galing plant contained secondary metabolites such as alkaloids, phytosterol saponins, phenols, flavonoids and tannins. Flavonoids were suspected as an antidiabetic agent. This study aimed to determine the effectiveness of some fractions of galing leaves extract in lowering blood glucose levels at mice which be induced streptozotocin (STZ). The research design used was a completely randomized design consisting of five treatment groups consisting of positive control (glibenclamide 5 mg) and negative control (Na. CMC 0,5%) with three replications. The results showed that there were significant differences in each treatment group (Fhit> Ftabel 0.05) of the antidiabetic effect, where ethyl acetate fraction had the best effectiveness in reducing blood glucose levels in Balb / C male mice that were STZ induced when compared with positive control of glibeklamid. Abstract

Inflammation is a response process to tissue injury and infection. The inflammatory response causes the release of pro-inflammatory mediators such as IL-1, IL-6 and TNF-α which are the main cytokines in the acute inflammatory response. Plants that have the potential to be anti-inflammatory are notika leaves (Archboldiodendron calosericeum KOBUSKI). This research was conducted to determine the anti-inflammatory activity of the ethanol extract of notika leaves against the levels of Tumor Necrosis Factor Alpha (TNF-a) in wistar mice. This study used a post test only control group design. Using 5 treatment groups, namely positive control (diclofenac sodium), negative control (NaCMC 0.5%), 100 mg/kgBW extract test group, 200 mg/kgBW extract group and 300 mg/kgBW extract group. In vivo tests were carried out by induction of inflammatory substances, administering preparations and measuring TNF-α levels using the ELISA method. Data were analyzed using the One Way ANOVA test. The results showed that the mean levels of TNF-α in the positive control group (0.475 ng/L), negative control (0.816 ng/L), extract dose 100 mg/kgBW (0.67 ng/L), extract dose 200 mg/kgBW (0.7285 ng/L) and extract dosage of 300 mg/kgBW (0.7785 ng/L). The conclusion obtained is that the ethanol extract of notika leaves has anti-inflammatory activity against the levels of Tumor Necrosis Factor Alpha (TNF-a) in Wistar rats based on the reduction in levels with negative control and the best dose was 100 mg/kgBW.

Abstract

Notika leaves are traditional medicinal plants used as antimalarial medications by the Papuans. Notika leaf is an endemic plant with many benefits but has not been scientifically proven. Plasmodium falciparum is a species of malaria that damages up to 50% of red blood cells, and its schizogony process mainly occurs in the liver. This study aims to confirm the activity of the ethanolic extract of notika leaf in improving liver function by using an experimental pre and post-test-only control group design with five treatment groups of Wistar strain mice. The normal group was treated with 0.5% NaCMC, the negative group was treated with 0.5% NaCMC and Carbon Tetrachloride (CCl4), and the third, fourth, and fifth groups received extract with doses of 125 mg/kgBW, 250 mg/kgBW, and 500 mg/kgBW, respectively. The extract was administered for seven days and, on the eighth day, was induced with CCl4 intraperitoneally. The SGOT and SGPT levels in mice were measured using a 5010v5+ photometer, and liver histopathology was examined using HE (Hematoxylin-Eosin) dye. The SGPT levels in the normal, negative, third, fourth, and fifth groups were 20.6 U/L, 52.08 U/L, 32.8 U/L, 19.8 U/L, and 7.8 U/L, respectively. Meanwhile, the SGOT levels were 18 U/L, 54.2 U/L, 28.2 U/L, 17.8 U/L, and 7.4 U/L in the normal, negative, third, fourth, and fifth groups, respectively. Based on ANOVA analysis, the SGPT and SGOT levels of the mice liver-treated group with notika leaf extract show significant values of < 0.05. The extract exhibits hepatoprotective activity and liver histological characteristics that do not induce necrosis at a dose of 500 mg/kgBW.
