Abstract

Sinularia gibberosa is one of the soft coral species reported to produce bioactive compounds for pharmaceutical’s potential. Soft corals produce compounds with antibacterial and antioxidant properties. This study aimed to obtain a bioactive profile of S. gibberosa, which could scavenge DPPH radicals and inhibit the growth of Listeria monocytogenes, Pseudomonas aeruginosa, and Salmonella typhimurium. The research included a sampling of soft corals, extraction by maceration, identification of chemical profiles using LC-HRMS, the assay for antibacterial activity (well-diffusion method), antioxidant assay (DPPH method), and total phenolic content (Folin–Ciocalteu method). Sampling was conducted in Palu Bay, Central Sulawesi, Indonesia. Based on monomorphic colonies and sclerites, soft coral samples were identified as Sinularia gibberosa. The chemical profiles of S. gibberosa extracts was dominated by trimethylethanolamine (22.03%), arachidonic acid (14.13%), (3R)-3-hydroxy-4-(trimethylazaniumyl)-butanoate (10.90%), (5Z,8Z,11Z,14Z,16R)-16-Hydroxy-5,8,11,14-icosatetraenoic acid (5.64%), Octyl decyl phthalate (4.18%), and N-Methylnicotinic acid (3.63%). The antibacterial assaying at 300 mg/mL of S. gibberosa extracts showed moderate to strong antibacterial activity, with the inhibition zone diameter of L. monocytogenes (15.11 ± 0.70 mm), P. aeruginosa (13.44 ± 0.84 mm), and S. typhimurium (19.78 ± 0.39 mm). S. gibberosa extracts showed strong antioxidant activity, with an IC50 of 54.69 ± 0.34 µg/mL and total phenolic content was 5423.76 ± 14.00 mg GAE/25 mg dry extract. Based on the results, S. gibberosa shows the potential for antibacterial and antioxidant activity. This study indicates that soft corals origin Palu Bay, Central Sulawesi, Indonesia, are potential organisms in the discovery and development of antibacterial and antioxidant agents.

Abstract

Antiradicals are substances with important functions for human health. Mangrove leaves are one of potential sources of natural antiradical. The objective of this research was to identify the type and fraction of mangrove leaves extract with the highest antiradical activity. The research procedure included sampling and extraction of mangrove leaves, assay of antiradical activity (DPPH), phytochemical assay and determination of IC50 for the fraction with the highest inhibition percentage. Mangrove leave samples (Rhizophora sp., Avicennia sp. and Sonneratia sp.) used in this study were collected from the Palu Bay coastal. Results showed the highest yield of extracts was from Rhizophora sp., followed by Avicennia sp. and Sonneratia sp. Inhibition percentage was higher from dried compared to fresh mangrove leaves. Additionally, the inhibition percentages of the ethanol fraction was higher than that of the ethyl acetate and n-hexane fractions, while Sonneratia sp. gave a higher inhibition percentage than Avicennia sp. and Rhizophora sp. The ethanol fraction IC50 was determined for Sonneratia sp. (46.05 ± 0.18 µg/mL), Avicennia sp. (88.41 ± 0.29 µg/mL) and Rhizophora sp. (103.95 ± 0.38 µg/mL). Phytochemical assays showed that the three ethanol fractions contained flavonoids, phenols (tannins) and steroids compounds. From the research, it can be concluded that the three leaves of mangrove potential as antiradicals.

Abstract

Bahari Jaya 2 and Sehati are partner fishermen groups located in Lere urban village, West Palu District, Palu, Central Sulawesi. In general, the catches from the two partner fishing groups are small shrimp (Penaeus sp.), with local names lamale. The purpose of IbM activities is to improve the skills and knowledge of members of the Bahari Jaya 2 and Sehati fishing group, to be able to run a processed business made from raw shrimp lamale. The projects carried out from April to November 2015. The method consists of 3 stages: 1) The stage of problem determination; 2) Stage presentation, negotiation and programming; and 3) Facilitation and mentoring of partners. The success indicator of this project; namely the two partner groups able to sustainably run a processing business. The results achieved by the two partner groups, i.e., 1) Partners can process lamale into noodles and nuggets, 2) Partners know business prospects for lamale noodle and nuggets, 3) Partners know quality control, quality management and sanitation in processing business, 4) Partners know the nutritional composition of the lamale noodle and nuggets, 5) Both groups of partners can manage profit-oriented sales businesses.

Abstract

Abstrak: Teknologi yang dipakai oleh pembudidaya udang di Sulawesi Tengah masih secara tradisional. Pelaku budidaya enggan melakukan peningkatan teknik budidaya dikarenakan kurangnya akses informasi ataupun percontohan yang tepat. Sasaran dan target kegiatan ini, yaitu para pembudidaya udang tradisional di Desa Lalombi, Kabupaten Donggala, Sulawesi Tengah, dengan harapan para pembudidaya mampu mengaplikasikan teknologi budidaya semi intensif. Tujuan pelaksanaan kegiatan ini, yaitu dapat meningkatkan produksi tambak udang tradisional dengan menggunakan teknologi semi intensif melalui mekanisasi dan budidaya yang modern, terukur dan berkelanjutan. Metode pelaksanaan kegiatan ini, yaitu survei lapangan, transfer teknologi ke mitra budidaya baik secara teoritis maupun praktek langsung. Pengumpulan data sebagai acuan monitoring. Dari kegiatan penerapan teknologi budidaya udang vannamei dengan metode semi intensif dengan luasan lahan 1600 m2, pembudidaya mendapatkan keuntungan sebesar Rp.3.288.750/bulan. Kegiatan penerapan teknologi masih diperlukan pendampingan bagi para pembudidaya. Oleh karena itu, tahap monitoring, evaluasi serta pendampingan terus dilakukan, agar para pembudidaya dapat mandiri menjalankan usaha budidaya udang vannamei dengan metode semi intensif.Abstract: The technology used by shrimp farmers in Central Sulawesi is still traditional. The farmers were reluctant to improve their shrimp culture techniques due to the lack of access to information or the right pilot project. The targets are local farmers in Lalombi Village, Donggala Regency, Central Sulawesi, are hoping that the farmers will be able to apply semi-intensive shrimp culture technology. This activity aimed to increase traditional shrimp ponds using semi-intensive technology by mechanizing modern, scalable, and sustainable aquaculture. This pilot project's method includes field survey and technology transfer of shrimp culture to partners, both theoretically and indirectly. The Data collection as a reference for monitoring. From the application of vannamei shrimp culture technology with the semi-intensive method on 1600 m2 pond, farmers get a profit of Rp.3,288,750/month. This shrimp farming pilot project still needs assistance for farmers. Therefore, the monitoring, evaluation, and assistance stages continue to be carried out so that the farmers can independently run the vannamei shrimp farming business with the semi-intensive method.

Abstract

Bioaktif antioksidan merupakan substansi yang penting bagi kesehatan manusia. Karang lunak telah diketahui memproduksi bioaktif dengan keragaman struktur dan aktivitas biologi, termasuk memproduksi bioaktif antioksidan. Tujuan penelitian ini adalah untuk mendapatkan bioprospeksi karang lunak dari perairan Teluk Palu sebagai penghasil antioksidan. Penelitian meliputi pengambilan sampel, identifikasi, ekstraksi (maserasi) karang lunak, skrining konstituen kimia, pengujian aktivitas antioksidan (penangkapan radikal DPPH), dan penentuan IC50. Pengambilan sampel dilakukan di Teluk Palu, pesisir desa Kabonga Besar, Kabupaten Donggala, Sulawesi Tengah. Berdasarkan bentuk koloni monomorfik, tujuh sampel karang lunak yang digunakan pada penelitian ini termasuk dalam genus Sinularia, Dampia dan Sarcophyton. Analisis konstituen kimia mengindikasikan adanya senyawa saponin, fenolik, triterpenoid, dan alkaloid. Hasil pengujian aktivitas antioksidan dengan metode DPPH, menunjukkan ekstrak kasar karang lunak menunjukkan persentase inhibisi radikal DPPH yang lemah. Hasil pengujian aktivitas antioksidan (metode DPPH) dari fraksi hasil partisi tujuh ekstrak kasar karang lunak berpotensi sebagai antioksidan dengan kategori kuat sampai lemah. Dari hasil penelitian dapat disimpulkan bahwa ekstrak karang lunak asal Teluk Palu berpotensi sebagai sumber antioksidan. Karang lunak di pesisir Desa Kabonga Besar didominasi oleh genus Sinularia. Antioxidant Activity of Soft Corals Extracts from Palu Bay, Central Sulawesi, IndonesiaAbstractThe antioxidants substances are important for human health. The research included sampling, identification, extraction (maceration) of soft corals, screening of chemical constituents, antioxidant activity assay (DPPH’s radical scavenger), and determination of IC50. Soft coral samples were collected from Palu Bay, coastal village of Kabonga Besar, Donggala District, Central Sulawesi. Based on the shape of the monomorphic colony, seven soft coral samples that were used in this study belong to the genus Sinularia, Dampia and Sarcophyton. Analysis of chemical constituents indicated the presence of saponin compounds, phenolics, triterpenoids, and alkaloids. The results of the antioxidant activity by DPPH method showed that soft coral crude extracts have a weak percentage of DPPH’s radical inhibition, while the results of the fraction had potential to be antioxidants with strong to weak categories. From the results of this study, it concluded that the soft coral extracts from Palu Bay have the potential as a source of antioxidants. The genus Sinularia dominates soft coral on the coast of Kabonga Besar Village.

Abstract

Weight problems are often a concern for teenagers. Be it overweight or underweight. The GADIS (Balanced Diet Action) program counseling was carried out to increase knowledge about a healthy and balanced diet based on these problems. Including adolescents in the Sangihe Islands Regency, North Sulawesi Province, based on the results of dietary observations carried out, namely, eating one time a day to lose weight. The activity was conducted on February 11, 2022, at SMKN 3 Tahuna, Sangihe Islands Regency. Activities are carried out in counseling with lecture methods, discussions, and demonstrations. Sixty-nine students attended the training. The action results in the form of an increase in knowledge of 8.9 through the consequences of pre-test and post-test assessments. This activity aims to increase understanding about regulating the balance of nutrients through the type and portion of each person's needs.

Abstract

Calcium is one of the important minerals in the physiological regulation of the immune system. Until now, tuna bone waste has not been widely used to fulfill human health needs. This study aims to obtained a high calcium sago cookies formulation by utilizing local resources, like seaweed (Caulerpa sp.) and tuna bone. Research methods include 1). Production of tuna bone porridge; 2). Production of lahe (Caulerpa sp.) porridge; 3). Production of sago flour; 4). Production of sago cookies; and 5). We were testing the chemical and physical quality of sago cookies. The univariate analysis showed that formula 1 sago cookies was preferred by the panelists and was close to the control formula. The chemicals analysis of the formula 1 sago cookies showed carbohydrates (57.95 ± 0.06%); water (5.09 ± 0.04%); fat (25.93 ± 0.08%); ash (5.90 ± 0.04%); protein (5.14 ± 0.10%); total energy (485.67 ± 0.05 kcal/100 g); energy from fat (233.33 ± 0.70 kcal/100 g); insoluble dietary fiber (20.29 ± 0.01 %); calcium (998.64 ± 8.60 mg/100 g); and phosphorus (5765.35 ± 40.36 mg/kg). The present findings confirm that panelists prefer the formula 1 sago cookies with a chemical composition according to quality requirements Indonesia National Standard (SNI 2973:2011) and nutritional adequacy recommended for Indonesian people (AKG 2019). In addition, the chemical composition contained in prototype sago cookies of formula 1 has the potential to become a high-calcium snack.

Abstract

Karang lunak merupakan bagian dari ekosistem terumbu karang, yang mengandung senyawa bioaktif dan dapat berperan sebagai alat pertahanan diri serta memiliki kemampuan sebagai senyawa antibakteri. Penelitian ini bertujuan untuk menginvestigasi potensi ekstrak karang lunak Sinularia sp. sebagai antibakteri dengan metode broth-dilution. Karang Lunak Sinularia sp. dikoleksi dari perairan Desa Kabonga Besar, Sulawesi Tengah, dan dimaserasi dengan diklorometana : methanol. Ekstrak Sinularia sp. dilakukan pengujian skrining metabolit kimia dan pengujian antibakteri dengan metode broth-diluttion. Hasil skrining ekstrak Sinularia sp. terdeteksi senyawa metabolit flavonoid, saponin, alkaloid dan steroid. Ekstrak Sinularia sp. memberikan pengaruh terhadap total koloni Escherichia coli dan Pseudomonas aeruginosa. Ekstrak Sinularia sp. dapat menghambat pertumbuhan E. coli dengan konsentrasi minimum 1.76 ± 0.10 mg/mL, sedangkan pada P. aeruginosa konsentrasi minimum yang dibutuhkan 1.85 ± 0.14 mg/mL. Konsentrasi bunuh minimum ekstrak Sinularia sp. yang dibutuhkan dalam membunuh E. coli, yaitu 7.06 ± 0.42 mg/mL, sedangkan pada P. aeruginosa, yaitu 7.38 ± 0.54 mg/mL. Penelitian ini disimpulkan bahwa senyawa bioaktif dari metabolit ekstrak karang lunak Sinularia sp. berpotensi sebagai senyawa antibakterial terhadap E. coli dan P. aeruginosa. Oleh karena itu, perlu diinvestigasi lebih lanjut dari identifikasi profil bioaktif karang lunak Sinularia sp.

Abstract

ABSTRAKSemi Refined Carrageenan merupakan salah satu produk carrageenan dengan tingkat kemurnian lebih rendah dibandingkan dengan refined carrageenan. Semi Refined Carrageenan merupakan tepung hasil ekstraksi rumput laut jenis Kappaphycus alvarezii berwarna putih kekuningan, dapat membentuk gel sehingga sangat berperan dalam industri makanan dan obat-obatan. Tujuan penelitian untuk mendapatkan konsentrasi pelarut KOH dan NaOH yang tepat dalam proses ekstraksi tepung Semi Refined Carrageenan rumput laut Kappaphycus alvarezii. Metode yang digunakan dalam penelitian ini adalah metode eksperimental, yaitu mengekstraksi Kappaphycus alvarezii menggunakan larutan alkali KOH dan NaOH yang direndam selama 4 menit. Hasil yang diperoleh menunjukkan bahwa rendemen tertinggi terdapat pada larutan KOH 5% (56,10%) dan larutan NaOH 3% (42,29%), kadar air pada larutan KOH 5% (15,45%) dan larutan NaOH 1% (16,50%), kadar abu larutan KOH  4% (4,85%) dan larutan NaOH 2% (3,50%), nilai pH terbaik terdapat larutan KOH 3% (8,14) dan larutan NaOH 3% (8,01) dan viskositas laju alir pada larutan KOH 3% (36,50 mL/detik) dan larutan NaOH 3% (38,10 mL/detik). Dari hasil penelitian dapat disimpulkan bahwa rendemen tertinggi terdapat pada larutan KOH 5% dan larutan NaOH 3%.Kata Kunci: alkali, carrageenan, KOH, Kappaphycus alvarezii, NaOHABSTRACTSemi Refined Carrageenan is a carrageenan product with a lower level of purity compared to refined carrageenan. Semi Refined Carrageenan is flour extracted from Kappaphycus alvarezii type of seaweed, yellowish white in color, can form a gel so that it plays a very important role in the food and medicine industry. The aim of the study was to obtain the correct concentration of KOH and NaOH solvents in the extraction process of Kappaphycus alvarezii seaweed Semi Refined Carrageenan flour. The method used in this study is an experimental method, namely extracting Kappaphycus alvarezii using an alkaline solution of KOH and NaOH soaked for 4 minutes. The results obtained showed that the highest yield was found in 5% KOH solution (56.10%) and 3% NaOH solution (42.29%), water content in 5% KOH solution (15.45%) and 1% NaOH solution ( 16.50%), ash content of 4% KOH solution (4.85%) and 2% NaOH solution (3.50%), the best pH values were 3% KOH solution (8.14) and 3% NaOH solution (8 0.01) and the viscosity of the flow rate in 3% KOH solution (36.50 mL/second) and 3% NaOH solution (38.10 mL/second). Abstract

Dagho Village is the center of fisheries industrialization in the Sangihe Islands, North Sulawesi. The only village that has a Coastal Fishery Port and a fishing industry is PT. Perindo Unit Dagho. The people of Kampung Dagho who live around PT. Perindo Unit Dagho especially fishermen's wives who do not have the ability to process fishery products and cannot generate money to help improve the welfare of their families. In general, they do not have more expertise in terms of skills in utilizing abundant fishery products. In Dagho Village, there are no Micro, Small and Medium Enterprises or similar businesses, both those engaged in agriculture, animal husbandry and fisheries. Small businesses in the village can be done by providing guidance and training to the village community and helping distribute products in the village. The method used in this research is a qualitative descriptive method with a series of stages arranged systematically. The stages are Research and Development, preliminary survey and Focus Group Discussion, problem identification, Kampung Dagho is a central industrialization, needs analysis, Target Audience Determination, Implementation Phase, and Program Evaluation. This activity was carried out at the Dagho Village Hall, Tamako District. The activity begins with a field survey for the stage of preparing a work plan, activity time and targets. In this activity, partners are given motivation to be able to carry out entrepreneurial activities so that they can increase group and individual income. The activity team from the State Polytechnic of North Nusa Tenggara provided material related to marketing strategies in selling products to the local market. flying fish. Kampung Dagho merupakan sentral industrialisasi perikanan yang ada di Kepulauan Sangihe Sulawesi Utara. Satu-satunya Kampung yang memiliki Pelabuhan Perikanan Pantai dan Industri perikanan yaitu PT. Perindo Unit Dagho. Masyarakan Kampung Dagho yang bermukim di seputaran PT. Perindo Unit Dagho khususnya istri-istri nelayan yang tidak memiliki kemampuan dalam melakukan pengolahan hasil perikanan dan tidak dapat menghasilkan uang guna membatu mensejahterakan keluarganya. Umumya mereka tidak memiliki keahliah yang lebih dari segi kertanpilan dalam memanfaatkan produk hasil perikanan yang melimpah. Di Kampung Dagho belum adanya Usaha Mikro Kecil Menengah ataupun usaha sejenisnya, baik yang bergerak dalam bidang pertanian, peternakan maupun perikanan. Usaha kecil diperkampungan bisa dilakukan dengan memberikan bimbingan dan pelatihan kepada masyarakat kampung dan membantu mendistribusikan produk di perkampungan. Metode yang digunakan pada penelitian ini adalah metode deskriptif kualitatif dengan sebuah rangkaian tahapan yang disusun secara sistematis. Tahapnya adalah Research and Development, survey pendahuluan dan Focus Group Discussion, identifikasi masalah, Kampung Dagho merupakan merupakan central industrialisasi, analisis kebutuhan, Penetapan Khalayak Sasaran, Tahap Pelaksanaan, dan Evaluasi Program. Kegiatan ini dilaksanakan di Balai Kampung Dagho Kecamatan Tamako. Kegiatan diawali dengan survei lapangan untuk tahap penyusunan rencana kerja, waktu kegiatan dan sasaran. Pada kegiatan ini mitra diberikan motivasi untuk dapat melakukan kegiatan berwirausaha sehingga dapat meningkatkan pendapatan kelompok hingga perorangan. Tim kegiatan dari Politeknik Negeri Nusa Utara memberikan materi terkait strategi pemasaran dalam menjual produk ke pasar lokal. Berdasarkan hasil yang telah dicapai dalam kegiatan Program Kemitraan Masyarakat Stimulus dapat disimpulkan bahwa di Kampong Dagho terbentuk satu UMKM baru yang bergerak dalam bidang pengolahan hasil perikanan khususnya produk bakso ikan layang.
