Abstract

Dengan perubahan gaya hidup manusia yang tidak sehat, terpapar radiasi sinar ultraviolet, agen fisika dan kimia secara berlebihan, luka yang lama tidak sembuh khususnya luka bakar dan infeksi virus, yang apabila dibiarkan dan tidak diobati bisa berkembang menjadi kanker. Pengobatan untuk tumor lebih banyak menggunakan obat-obatan sintetik yang juga dapat menimbulkan komplikasi dan kebanyakan belum menunjukkan hasil yang memuaskan, sehingga banyak penelitian ditujukan pada tanaman tradisional yang banyak tumbuh di Indonesia. Tujuan penelitian ini adalah untuk mengetahui potensi penghambat pembelahan sel dari ekstrak buah legundi (Vitex trifolia). Penelitian ini adalah studi eksperimental dengan menggunakan tikus Wistar yang dibagi dalam 5 kelompok yaitu kelompok kontrol normal (K1), kontrol positif (K2), P1, P2, P3. Ekstrak etanol buah V. trifolia dibagi dalam 2 dosis yaitu 0,5g dan 1g/kgBB/hari/oral selama 3 minggu. Hasil penelitian berupa 3 slide (60%) adalah tumor ganas (nonkeratinizing squamous cellcarcinoma), 1 slide (20%) adalah lesi atipik dan 1 slide (20%) adalah lesi jinak. Setelah pemberian terapi menggunakan buah V. trifolia, kelompok 2, 3, dan 4 yang memiliki lesi tumor mengalami penurunan ukuran (mengecil). Disimpulkan bahwa ekstrak etanol buah V. trifolia memiliki kemampuan untuk menginhibisi aktivitas proliferasi dan pertumbuhan dengan dosis pemberian 0,5 g dan 1 gr/hari.

Abstract

Tuberculosis is an infectious disease caused by Mycobacterium tuberculosis. Indonesia ranks 2nd largest in the world after India. Resistance to anti-tuberculosis drugs is one of the difficulties in treating tuberculosis. This research aim to determine the effectiveness of legundi (Vitex trifolia) leaf extract nanoherbal in killing Mycobacterium tuberculosis. This study was conducted with an experimental design using M. tuberculosis colonies taken from 25 samples of aspirate of tuberculosis patients with lymph nodes and then given the ethanol extract of legundi leaves (70% and 90%) with the process of making nanoparticle extracts. The evaluation of the observations was in the form of measuring the inhibition zone for bacterial growth which were categorized as weak ( 5mm), moderate (6-10mm), strong (11-20 mm) and very strong ( 21mm). The results of the study from 6 repetitions found that the inhibition zone category was 0% weak, 39% moderate, strong 52.3%, and very strong 8.7%. Comparison of the concentration of legundi leaf extract to the diameter of the bacterial growth inhibitory zone was analyzed using the Kruskal Wallis test, which found a significant relationship with p value = 0.000 ( 0.05). Legundi's leaf extract can be used as an antibacterial agent for M. tuberculosis.

Abstract

Tuberculous lymphadenitis (TBLN) is the most common form of extrapulmonary tuberculosis. However, the optimal diagnosis using Fine-Needle Aspiration Cytology (FNAC) or excisional biopsy is uncertain. This research aims to improve the diagnostic of TBLN with FNAC and immunocytochemistry (ICC) compared to the response to antituberculosis therapy. The cross-sectional study involved 43 patients with the criteria for TBLN diagnosis based on the appropriate clinical history of tuberculosis and indicative cytological results. Immunocytochemical examination employed rabbit-polyclonal to Mycobacterium tuberculosis (MTB) antibody (AB905). The MTB expression was found in 35 out of 43 cases (81%) that appropriate cytological features of the tuberculosis process. Meanwhile, eight out of 43 cases (19%) did not express MTB. Diagnostic tests for lesions with a positive cytologic appearance of TBLN and ICC were compared to the response to anti-tuberculosis therapy, revealing the sensitivity, specificity, positive predictive value, and negative predictive value of 95.2%, 75%, 95.2%, and 17%, respectively. Besides, Fisher's exact tests utilized to identify the relationship between two variables; p 0.05 was considered significant. This research found immunocytochemical study was a sensitive and specific tool for improving the diagnostic of TBLN.

Abstract

Woodworkers are particularly susceptible to inhalation of sawdust exposure. Exposure to sawdust for a long time causes various health problems and causes the damage of the epithelium to a barrier. This study aims to identify whether inhaled sawdust can cause changes in the nasal mucosa and the occurrence of an inflammatory process. This study was a cross-sectional analytical study. Data were collected from July 2021 to January 2022 from the nasal cavity swab of 35 woodworkers who had worked for 1 to 5 years. The assessment was carried out by cytological examination to identify epithelial changes in the form of mild and moderate dysplasia, while the inflammation was found in the form of acute and chronic inflammation. Twenty-six (74.3%) samples were normal, 7 (20%) samples had mild dysplasia, and 2 (5.7%) samples had moderate dysplasia. Whereas 20 (57.1%) were found to have no inflammation, 8 (22.9%) were with acute inflammation, and 7 (20.0%) were found to have chronic inflammation). Fisher’s exact test showed a relationship between the inflammatory process and epithelial changes (p 0.020). In conclusion, sawdust inhaled showed changes in the epithelial mucosa and inflammatory processes of the nasal cavity.
