Abstract

This study aimed to evaluate the genetic potential of local meat chickens (LMC) to get the candidate of parent stock through the development of the selection method applied, as well as economical and practical approach and adjusment of nutritional requirement, especially protein and energy balance. Research was divided into 2 groups, where the female and male parent was reared for 2 months and the offspring (F1) was reared for 4 months and selected. The parent was mated by the method of artificial insemination (AI). Method of selection was used on the orientation of the meat chicken production parameters with a positive correlation with the gene expression Diet treatments were formulated by optimal nutrition standards in order to drive maximum primary production of local meat chicken, contained 17, 19, and 21% of protein. The result showed that feeding diet with 19% protein can stimulate optimum growth and food conversion of ALPU male, whereas female growth LMC in line with the increasing availability of protein in the diet, wherein feeding 21% of protein result in optimal growth until age 4 months.

Abstract

Penelitian ini bertujuan mengetahui persentase peningkatan sel beta Langerhans pankreas yang mengalami nekrosis setelah diinduksistreptozotosin berulang.Tiga puluh ekor mencit jantan galur Balb-c, umur 12-14 minggu dengan bobot badan 30-40 g dikelompokkan menjadi 2kelompok perlakuan.Kelompok I (KI)diberikan pelarut streptozotosin danKelompok II (KII) diberikan streptozotosin dengan dosis 40 mg/kg bbdalam 50 mM natrium sitrat buffer pH 4,5.Semua perlakuan diberikan secara intraperitoneum selama 5 hari berturut-turut. Pemeriksaan kadarglukosa darah dan berat badan dilakukan pada hari ke-0, 7, 14, 21, dan 28 sesudah pemberian perlakuan. Hewan percobaan dari masing-masingkelompok dieutanasiasebanyak 2 ekor pada hari ke-7, 14, 21, dan 28 setelah pemberian perlakuan untuk pewarnaan Gomori. Data pengamatanberat badan adalah42,862,3 vs 32,787,4 gdan kadar glukosa darah puasa 95,892,5 vs 255,7787,60 mg/dl menunjukkan perbedaan yangsignifikan (P0,05) antara kedua kelompok perlakuan, sedangkan persentase sel beta yang mengalami nekrosis antara kedua kelompok perlakuan4,570,47 vs 64,114,10juga menunjukkan perbedaan yang signifikan (P0,05). Induksi dosis rendah streptozotosin secara berulang dapatmenyebabkan peningkatan persentase nekrosis sel beta Langerhans pankreas.

Abstract

The aims of the research were to evaluate resistance of Staphylococcus aureus against several antibiotics and to detect the gene encoding ofmethicillin resistant Staphylococcus aureus (MRSA) in broiler. Twenty three Staphylococcus aureus isolated from infected broiler in Yogyakartawere used. Genotypic identifications were done based on the occurrence of gene nuc and 23SrRNA. The resistance assay to several antibioticsrevealed that Staphylococcus aureus were resistant to penicillin 78% (18/23), doxycyclin 56% (15/23), gentamicin 26% (6/23), tetracyclin 22% (5/23), erythromycin 13% (3/23), and methicillin 9% (2/23). Detection of gene encoding MRSA on 23 isolats of S. aureus showed that 8 (34.8%) isolates contain gene mecA.Key words: Staphylococcus aureus, broiler, resistant, methicillin, mecA

Abstract

Tujuan penelitian ini adalah mengetahui, mengkaji, dan menganalisis potensi seledri sebagai herba antistres dan imunomodulator dengan cara menghambat sekresi kortisol plasma pada tikus Wistar (Rattus norvegicus) yang diinduksi kejutan listrik. Penelitian ini merupakan penelitian eksperimental laboratoris, pre and postest with control group design. Sampel yang digunakan adalah 13 ekor tikus Wistar yang dibagi dalam 3 kelompok: P0, kelompok kontrol (kejutan listrik); P1, kejutan listrik + ekstrak seledri 0,14 mg; dan P2, kejutan + ekstrak seledri 0,72 mg. Kejutan listrik dan ekstrak seledri masing-masing diberikan selama 7 hari. Kadar kortisol diukur menggunakan metode enzyme-linked immunosorbent assay (ELISA). Analisis data menggunakan uji analisis varian satu arah. Hasil penelitian menunjukkan bahwa nilai rata-rata kadar kortisol serum kelompok P0 sebelum dan setelah diberikan perlakuan mengalami peningkatan, sedangkan kelompok P1 dan P2 mengalami penurunan (P0= 1,88 vs 3,65; P1= 4,59 vs 2,63 dan P2= 3,00 vs 2,99). Pemberian ekstrak seledri cenderung menurunkan kadar kortisol serum meskipun secara statistik tidak berbeda secara signifikan (P>0,05). Disimpulkan bahwa ekstrak seledri belum sepenuhnya berperan sebagai herbal antistres pada tikus Wistar yang mengalami stres.

Abstract

The purpose of this study was to analyze how the effect of each increase in Body Mass Index (BMI) on the increase in blood pressure (BP). The research design was a laboratory experiment with 52 non-obese and obese rats as subjects (non-obese, n=14 and obese, n=38). Rats were induced obesity using a high-fat diet for four weeks. BMI used is based on the Lee Index with obesity indicators if 300 g/cm. Data analysis using independent sample t-test, simple linear regression test, and Pearson correlation test; (p0.05). The results: BP in non-obese rats was lower than obese rats (89.007.38 vs 190.114.42; p=0.001*), BMI was positively associated with BP (p=0.001*); an increase in BMI will increase BP (p=0.001*), there is a strong correlation between BMI and BP (r=0.977; p=0.001*). The regression equation (Y = 707.42 + 2.75 X) indicates that every 1 g/cm increase in BMI will be followed by an increase in BP of 2.75 mmHg. The analysis of R-square = 0.942 shows that the accuracy of the linear equation model is 94.2%. In conclusion, BMI has a strong positive correlation with BP, an increase in BMI will increase BP; obesity induces high blood pressure in rats.

Abstract

The purpose of this study was to determine the type of insecticides which Aedes aegypti mosquitoes are resistant to, so that other susceptibleinsecticides still can be used. The study was a cross sectional epidemiological study with cluster sampling in Sub-district Wongsorejo,Banyuwangi, Muncar, Tegaldlimo, Kalibaru which is considered sufficient to represent Banyuwangi Regency. Mosquito samples appropriate tothe characteristics were isolated and tested for insecticide-resistance primer namely voltage gated sodium channel (VGSC) to determine thespecific resistance expressed in mosquitoes. The result of resistance test using WHO standard method showed that Aedes aegypti mosquito fromBanyuwangi Regency were resistant to cypermethrin 0.25% and malathion 0.8%. There was a VGSC coding gene with 250 bp band detectedusing polymerase chain reaction (PCR) technique which was associated with the resistance of Aedes aegypti mosquitoes in Banyuwangi Regencyto organophosphate insecticides (malathione) and pyrethroid insecticides (cypermethrin).

Abstract

Penelitian ini bertujuan mengetahui pengaruh pemberian hormon PMSG sebelum kawin terhadap gambaran darah merah induk domba bunting. Sebanyak 14 ekor induk domba bunting dengan kisaran bobot badan antara 20-25 kg dikelompokkan ke dalam dua kelompok, masing-masing terdiri atas 7 ekor domba. Perlakuan pertama terdiri atas induk domba bunting hasil perkawinan alami tanpa diberikan hormon PMSG (P0) dan perlakuan kedua terdiri atas induk domba bunting yang diberikan 125 IU PMSG (PG 600, Intervet, Holland) sebelum dikawinkan (P1). Sebelum diberikan hormon PMSG, domba penelitian diserentakkan berahinya dengan menyuntikkan 10 mg/ekor prostaglandin (PGF2, LutalyseTM, Pharmacia Upjohn Company, Pfizer Inc.) sebanyak dua kali dengan interval sebelas hari. Penyuntikan hormon PMSG dilakukan bersamaan dengan penyuntikan hormon PGF2 yang kedua. Domba betina yang estrus dikawinkan secara alami dengan pejantan yang telah diseleksi. Pada hari ke-30 setelah dikawinkan, dilakukan pemeriksaan kebuntingan dan jumlah fetus dengan menggunakan ultrasonografi (USG). Sampel darah untuk pemeriksaan jumlah sel darah merah, kadar hemoglobin, dan nilai hematokrit diambil pada periode kebuntingan bulan ke-1, 2, 3, 4, dan 5. Perlakuan hormon PMSG sebelum perkawinan meningkatkan jumlah sel darah merah, nilai hematokrit, dan kadar hemoglobin induk domba bunting. Peningkatan jumlah sel darah merah, nilai hematokrit, dan kadar hemoglobin terjadi pada periode awal kebuntingan dan menurun pada akhir masa kebuntingan.

Abstract

The morphological characteristics of the stomach and esophagus of five adults bandicoot (Echymipera kalubu) with average 1.160.29 kg in body weight and 38.24.76 cm in body length were studied macroscopically, microscopically and histochemically. The bandicoot esophagus were found relatively long about one third of body length. The esophageal glands were identified along the esophagus and the number decreased gradually from cranial to caudal region. In addition of smooth muscle, skeletal muscles were also examined in the external muscle layer of esophagus up to the esophageal junction. The stomach of the E. Kalubu had short lesser curvature and three glandular regions of cardiac, fundic and pyloric glands were observed, respectively. The cardiac glands area was small and concentrated in the cranial part of the stomach. The fundic glands area occupied about half parts of the stomach. Mucous surface epithelial cells, mucous neck cells, parietal cells and chief cells were found in the entire fundic gland. The parietal cells were the most abundant cells in the major curvature distributed from the basal to neck area of the glands, but few in the minor curvature. Chief cells were mostly distributed in the basal gland. The pyloric glands region was observed in caudal part of the stomach. Staining with alcian blue-periodic acid Schift (AB-PAS) showed various distribution and intensity of neutral and acid mucopolysaccharides in the esophageal and stomach mucosa. High concentration of neutral and acid mucopolysaccharides were detected in esophageal and stomach glands with various concentration. Morphological characteristics of the esophagus and stomach were assumed to be related to the digestive processes in the gastrointestinal tract of bandicoot.

Abstract

Penelitian ini bertujuan melakukan identifikasi dan mengetahui persentase orangutan yang terinfeksi endoparasit (cacing) pada orangutan excaptive yang berada di Suaka Margasatwa Sungai Lamandau. Sampel feses orangutan yang diperiksa sebanyak 30 individu yang didapatkan dicamp Gemini dan Siswoyo. Pemeriksaan feses dilakukan secara makroskopis dan mikroskopis. Pemeriksaan mikroskopis dilakukan dengan menggunakan dua metode yaitu metode Ridley dengan larutan sodium acetate, acetic acid, dan formaldehyde (SAF) fiksatif dan metode apung. Hasil pengamatan menunjukkan adanya infeksi cacing gastrointestinal sebesar 56,7%. Hasil penelitian menunjukkan bahwa orangutan positif terinfestasi telur cacing tipe ascarid; trichurid; dan strongiloid masing-masing sebesar 43,3; 16,7; dan 6,7%. Orangutan dapat mengalami infeksi lebih dari satu jenis tipe telur cacing.

Abstract

Penelitian ini bertujuan membandingkan efek pemberian fraksi n-heksan, etilasetat, dan metanol dari ekstrak daun kesum dalam memperbaiki kerusakan organ paru akibat paparan benzopirena. Penelitian dilakukan secara in vivo dengan menggunakan hewan model terpapar benzopirena. Pembuatan hewan model kanker terpapar benzopirena dilakukan secara intraperitoneal dengan dosis 200 mg/kg yang dilanjutkan dengan pengobatan menggunakan fraksi n-heksana, etil-asetat, dan metanol dari ekstrak daun kesum. Uji keberhasilan penelitian dilakukan dengan pengukuran kadar malondialdehid (MDA) dan pemeriksaan histologis organ paru dengan pewarnaan hematoksilin-eosin (HE). Kadar MDA tikus kontrol; tikus terpapar benzopirena; tikus terapi fraksi n-heksana; tikus terapi fraksi etil-asetat; dan tikus terapi fraksi metanol masing-masing adalah 8,44; 6,24; 7,21; 8,47; dan 5,27 ppm. Tingkat kerusakan organ paru tikus kontrol, terpapar benzopirena, terapi fraksi n-heksan, terapi fraksi etil-asetat, dan fraksi metanol masing-masing adalah normal (0,728), strong (3,002), light (1,687), moderate (2,600), dan strong (3,060). Fraksi n-heksana merupakan fraksi yang paling bagus dalam memperbaiki kerusakan organ paru hewan model akibat paparan benzopirena.
