Abstract

Penelitian ini bertujuan mengetahui gambaran klinis sapi pyometra sebelum dan setelah diterapi dengan antibiotik dan prostaglandin. Dalampenelitian ini digunakan enam ekor sapi betina yang didiagnosis menderita piometra berdasarkan pemeriksaan secara klinis dan ultrasonografi pada organ reproduksi. Sapi tersebut dibagi ke dalam dua kelompok perlakuan, masing-masing 3 ekor sapi untuk tiap kelompok. Kelompok I diterapi dengan 5 ml antibiotik (gentamicine, flumequine) ditambah 15 ml NaCl fisiologis dan PGF2 (Luprostiol) 12,5 mg secara intra uteri,sedangkan kelompok II diterapi hanya dengan menggunakan antibiotik. Hasil penelitian menunjukkan pada sapi yang didiagnosis piometra ditemukan adanya cairan yang penuh mengisi uterus (100%), korpus luteum persisten pada salah satu ovarium (100%), discharge di sekitar ekor, perineum, dan vulva yang berwarna kuning (50%), krem (33,3%), dan hijau keabu-abuan (16,6%). Sapi yang diterapi dengan antibiotik dan PGF2 menyebabkan pengeluaran leleran yang lebih cepat dan lebih banyak dibandingkan sapi yang diterapi hanya dengan antibiotik.

Abstract

The aim of this study was to determine the survival of embrionated eggs of Ascaridia galli. Adult female worms were obtained from lumen of intestine of native chickens in a slaughter house. Eggs obtained from the uteri of adult female worms were incubated in distilled water at room temperature for 20-31 days in order to develop A. galli infective eggs. The eggs were counted using stereomicroscope. The result showed that the amount of A. galli eggs were 1,045,478 and the amount of embrionated eggs were 935,300 (89.46%).Keywords: Ascaridia galli, embrionated eggs

Abstract

Penelitian ini bertujuan mengetahui efektivitas daun Datura metel Linn. dalam mengurangi cekaman transportasi pada domba. Penelitian dilakukan 3 tahap. Tahap pertama adalah menentukan dosis yang tepat dengan perlakuan 0, 5, 10, 15, 20, 25%, dan pekat pada hamster melalui kemampuan berputar pada roda berputar selama 1 menit, yang diobservasi pada waktu 10, 20, 30, 60, 120, dan 180 menit pascapemberian ekstrak daun Datura metel Linn. Tahap kedua menguji efek sediaan ekstrak daun Datura metel Linn. yang diberikan pada domba pratransportasi. Tahap 3, mempelajari bentuk penyajian dengan teknik celup. Lima ukuran partikel tepung daun Datura metel Linn. yaitu 60, 60-40, 40-30, 30-20, dan 20 mesh diuji kelarutannya dalam air selama 10 menit. Data diolah dengan deskriptif analisis. Hasil penelitian menunjukkan bahwa pemberian daun Datura metel Linn. untuk semua perlakuan memberikan sensasi tenang dan bertahan sampai 180 menit. Domba yang diberi ekstrak daun Datura metel Linn. memiliki derajat stres transportasi yang lebih rendah dibandingkan dengan tanpa pemberian ekstrak Datura metel Linn. Ukuran mesh 20 memberikan kelarutan yang paling tinggi. Dapat disimpulkan bahwa ekstrak daun Datura metel Linn. dapat mengurangi stres transportasi pada domba.

Abstract

Mesenchymal stem cells (MSC) derived from white adipose tissue are considered to be an appropriate therapeutic candidate for various human diseases due to their natural ability to maintain homeostasis. They have also been considered for use as a therapeutic alternative due to their sufficient availability and ease of harvesting. MSC play an important role in the process of adipogenesis and energy metabolism that is directly related to the pathogenesis of various degenerative diseases and metabolic syndromes. The purpose of this study was to obtain MSC cultures from white adipose tissue biopsied from three adult males Macaca fascicularis and to evaluate these cultures qualitatively and quantitatively regarding their characteristics and ability to differentiate. Polymerase chain reaction (PCR) was used to confirm the expression of mesenchymal stem cell-specific gene markers, and a specific growth medium was used to differentiate these cells into adipocytes, chondrocytes, and osteocytes. The results obtained showed that the MSC culture from M. fascicularis WAT had a fibroblast-like morphology with a spindle shape. The MSC markers of CD73, CD90 and CD105, were also expressed. The MSC culture was also able to differentiate into adipocytes, chondrocytes, and osteocytes. Based on these results, white adipose tissue from Macaca fascicularis was found to have potential as a source of MSC.

Abstract

The objective of this research was to analyze the composition of saturated fatty acid, unsaturated fatty acids, and cholesterol in meat of Bali bull fattened using forage crops in small holder farming. The experiment was conducted for 3 months using nine Bali bull aged 2.5-3.5 years old with the initial body weight of 227-290 kg in the feedlots of Bero Sembada Farmers Group, Laen Manen Sub District, Malaka Regency, East Nusa Tenggara. The experiment was conducted based on the fattening system of local farmers in terms of feeding, housing, and health management. Feed given during the experiment was Centrosema pubences, Clitoria ternatea, fresh corn straw, king grass, Leucaena leucocepala, natural grass, Pennisteum purpureum, and Sesbania grandiflora. At the end of this study, cattle were slaughtered and the composition of saturated fatty acid, unsaturated fatty acids and cholesterol were measured. Data were analyzed using a descriptive statistic. Results revealed that the total of saturated fatty acid and unsaturated fatty acids in meat were 36.628% and 26.739%, respectively, while the cholesterol reached 79.18 mg/100 g of meat. In conclusion, meat of Bali bull fattened using forage crops in small holder farming has higher saturated fatty acid than unsaturated fatty acids.

Abstract

Tujuan penelitian adalah mengetahui ultrastruktur oosit pasca vitrifikasi menggunakan transmisi elektron mikroskop. Ovarium dikumpulkan dari rumah potong hewan dan oosit dikoleksi dengan cara aspirasi dari folikel menggunakan jarum ukuran 18 G. Setelah dilakukan klasifikasi kualitas oosit, maka oosit yang berkualitas A dimaturasi secara in vitro. Oosit hasil maturasi dipaparkan ke dalam larutan vitrifikasi yang mengandung 30% EG + 0,5 M sukrosa dengan lama waktu paparan 3 menit, oosit dimasukkan ke dalam ministraw 0,25 cc (French straw), kemudian dipaparkan pada uap nitrogen selama 10 detik dan dimasukkan dalam kontainer nitrogen cair. Setelah 3 minggu ministraw diambil dan dilakukan penghangatan (warming) di udara selama 10 detik, dimasukkan dalam waterbath suhu 35 C selama 30 detik. Isi ministraw dituangkan ke dalam cawan petri dan oosit dibilas dua kali dengan sukrosa 0,5 M. Selanjutnya dilakukan pengamatan ultrastruktur oosit dengan transmisi elektron mikroskop. Hasil penelitian menunjukkan oosit hasil vitrifikasi mempunyai zona pelusida yang abnormal (fraktur), mem bran plasma mengalami lisis, beberapa butir korteks mengalami degenerasi dan perpindahan butir korteks di ruang perivitelin. Kesimpulan dari penelitian ini adalah terdapat perubahan ultrastruktur pada oosit setelah vitrifikasi.

Abstract

The purpose of this research was to analyse the quality and storability of native chicken semen at room temperature after diluted with physiological saline solution (NaCl) supplemented with noni fruit extract. Research methodology used was nested randomized design which variables observed covering the quality of chicken spermatozoa macroscopically and microscopically with the doses of noni fruit extract of 0 % (P0), 10 % (P1), 20 % (P2), and 30 % (P3). Rate of spermatozoa motility at 0, 1, 2, and 3 hours on P0 were 83.704.74, 63.309.63, 32.908.52, and 15.006.20; P1 were 85.305.44, 72.7010.06, 51.9011.75, and 28.505.68; P2 were 84.606.40, 78.508.59, 57.9010.73, and 33.709.06; and P3 were 81.807.30, 64.207.93, 40.3011.66, and 19.807.47, respectively. Rate of spermatozoa viability at 0, 1, 2, and 3 hours on P0 were 85.309.09, 65.106.15, 32.1011.86, and 10.309.09; P1 wer 87.406.22, 72.705.33, 50.8013.59, and 26.5010.99; P2 were 68.585.30, 77.704.79, 56.3013.76, and 32.7013.79; P3 were 81.208.04, 68.7010.40, 36.2016.61, and 16.9011.93, respectively. Rate of spermatozoa abnormality at 0, 1, 2, and 3 hours on P0 were 8.703.40, 12.204.42, 17.805.67, and 20.306.38; P1 were 7.802.04, 9.802.69, 13.604.45, and 16.505.19; P2 wer 8.404.33, 10.002.45, 12.505.21, and 15.803.71; and P3 were 9.603.41, 10.902.64, 17.105.61, and 21.208.16, respectively. It can be concluded that the addition of noni fruit extract at the dose of 10 % and 20 % in physiological saline solutin maintain the native chicken spermatozoa quality up to 2 hours post storage at room temperature.

Abstract

Penelitian ini bertujuan mengetahui efek minyak jintan hitam (Nigella sativa L.) terhadap spermatogenesis tikus Wistar yang terpapar asap rokok. Jenis penelitian adalah eksperimental murni dengan subyek 24 ekor tikus yang terpapar asap rokok selama 21 hari dan dirandomisasi dalam 4 kelompok yaitu kelompok kontrol dan kelompok perlakuan dengan pemberian minyak jintan masing-masing 0,1; 0,2; dan 0,3 ml. Penegakan hipotesis dilakukan dengan menggunakan Krusskal-Wallis dan Mann Whitney dengan batas derajat kemaknaan P0,05, interval kepercayaan 95%. Pemberian minyak jintan hitam dapat memperbaiki derajat spermatogenesis.

Abstract

This study aims to determine the growth inhibition and apoptosis induction of MCF-7 and T47D cancer cells by ethanol extract of Chromolaena odorata leaves. A post-test with control group design was used in this study. The extract was made by maceration with 80%ethanol and the tested concentrations used were 7.80 g/mL-500 g/mL with standard drug doxorubicin ranged from 1.56 g/mL-100 g/mL.The growth inhibition was determined by the MTT colorimetry method, apoptosis induction by double staining using acridine orange-ethidiumbromide, and the existence of apoptosis was proven immunocytochemically through the expression of Bcl-2 proteins. The results showed that the growth inhibition of MCF-7 was 100.29%-28.19% and T47D was 100.37%-16.01%. The IC50 values of MCF-7 was 327.34 g/mL and T47D was 135.16 g/mL. The presence of apoptosis was marked by finding the morphological change of cells such as dead, necrosis, and chromatin condensation cells. This suggests that interventions with ethanol extract of Chromolaena odorata leaves can induce apoptosis that has been proven by reducing the expression of Bcl-2 proteins.

Abstract

This research was aimed to study curative effect of sour soup seed powder on cattle invested with ticks and goat infected with scabies. This study was using 12 cattle invested with ticks and 12 goats with scabies. The cattle divided into 4 groups (S1, S2, S3 and S4) while goats were divided into 3 groups (K1, K2, and K3) equally. For cattle with ticks group S1 received water (control group), while group S2, S3, and S4 received 1%, 5%, and 10% sour soup powder respectively. Ticks that fell to the ground and not engorged were collected and identified. Statistical analysis showed that all concentrations of sour soup were effective in paralyzing and or killing ticks of the genera Boophilus sp. and Dermacentor sp. but were not effective against Rhipicepalus sp. For goats with scabies, groups K1, K2, and K3 received 1, 5 and 10% sour soup powder respectively mixed with water applied to whole area of infected and uninfected skin surrounding infected area. Number of mites per cm2 before and after treatment was counted. Statistical analysis showed that 1, 5, and 10% sour soup powder effective in reducing the number of scabies mites on day 1 and 7 after treatment and were significantly different from those number of mites before treatment (P0.01). Statistical analysis also showed that no significant difference among concentration of sour soup seed powder in decreasing the number of mites (P0.05).
