Abstract

The objective of this study was to determine the effect of resveratrol supplementation in KA-EN 3B-egg yolk dilution on sperm quality of Bangkok chickens. The semen was collected from 2 adult male Bangkok chickens with an age of 12 months and a body weight of 2.5 kg. The collected semen was divided into 5 treatments with 5 replicates each. The treatments used were KA-EN 3B-egg yolk (KEY), KEY + 0.02% resveratrol, KEY + 0.04% resveratrol, KEY + 0.06% resveratrol, and KEY + 0.08% resveratrol. The parameters observed were sperm motility, viability, and abnormalities during 24 hours, 48 hours, and 72 hours of storage in the refrigerator. The results showed that resveratrol supplementation in the diluent had no effect (P0.05) on sperm motility, viability, and abnormalities during storage. This study concluded that supplementation of resveratrol in KA-EN 3B-egg yolk diluent did not affect the quality of Bangkok chicken sperm during storage.

Abstract

The aim of this study was to determine the expression of the P53 gene and testicular weight of male mice (Mus musculus) given Bulo forest honey and exposed to cigarette smoke. The experimental animals used in this study were 24 male mice which were divided into 4 treatment groups, each group consisting of 6 mice. Mice in group P0 were considered as control group (without treatment), while mice in group 1 (P1) were exposed to 2 cigarettes smoke, while mice in groups 2 (P2) and 3 (P3) were exposed to 2 cigarettes smoke and given Bulo forest honey at a dose of 0.8 ml/day and 0.4 mL/day, respectively. The data obtained were analyzed using one way ANOVA followed by using the Least Significant Difference (LSD) test. The results showed that Bulo forest honey administration did not statistically affect (P0.05) the ratio of testicular weight and P53 gene expression in male mice (Mus musculus) exposed to cigarette smoke for 30 days. It is concluded that the administration of Bulo forest honey at dose of 0.8 mL and 0.4 mL has not been able to increase the testicular weight ratio and reduce the expression of the P53 gene in male mice.

Abstract

This study aims to understand the histopathological changes in the pseudo-pregnant rabbit uterus concerning the duration of ovaries inside the uterus during ovarian transplantation. The study used nine New Zealand White (NZW) pseudo-pregnant female rabbits aged 3-5 years, weighed 1.5-2.9 kg. After a 30-day adaptation period, all rabbits were divided into three treatment groups: K1 (n=3) underwent ovarian transplantation for 3 days, K2 (n=3) for 5 days, and K3 (n=3) for 7 days. Pseudo-pregnant was induced using PMSG and hCG. The rabbits were intramuscularly injected with 100 IU of PMSG, followed by intravenous injection of 75 IU of hCG three days later. Ovary transplantation was performed on the 8th day (day 0 being the day of hCG injection). Uterus was collected after the transplantation procedure according to the respective treatment groups. The results showed that in K1, K2 and K3, there was hyperemia in the endometrial epithelium and infiltration of inflammatory cells. Hyperemia and necrosis were more severe in K1 compared to K2 and K3. In K3, the histopathological appearance of the endometrium tended to be normal, with less damage. In conclusion, the histopathological findings of hyperemia and necrosis in the uterus undergoing ovarian transplantation lasted for 3 days compared to 5 and 7 days.

Abstract

The research was conducted to find out the effect of Aloe vera gel administrated orally onreproductive performance of female rats. Fifteen two months old female rats were alloted to fivetreatments of per oral Aloe vera gel administration: P0 (control), P1(1 mg /g body weight), P2 (2 mg/gbody weight, P3 (3 mg/g body weight), and P4 ( 4 mg/g body weight). The gel were administrated everyday for four weeks. The body weight were measured on the same day as last treatment. The animal thenmated to proven male on 1:1 rasio. Positively mated animals were sacrificed on D-10 after mating tocollect uterus and ovary so the number of fetus, corpus luteum, and ovary weight can be determined. Thetreatments significantly decreased (P0.05) the average body weight gain. The P0 (4.1 g/head/day) wasdifferent to P1 (2.69 g/head/day), P2 (2.87 g/head/day, and P4 (2.68 g/head/day), but was not different toP3 (3.39 g/head/day). The ovarium weight were not different (P0.05) among the treatments, so did thenumber of corpus luteum (CL), the number of fetuses and the CL: fetus rasio.Keywords: Aloe vera, rats, reproductive performance .

Abstract

Penelitian ini bertujuan mengetahui persentase kebuntingan kembar pada kuda di Pulau Jawa dan Madura dan menguji tingkat keberhasilan enukleasi vesikel embrio dengan panduan ultrasound. Sebanyak 354 induk kuda dikawinkan secara alami dengan kebuntingan mencapai 57,6% (204 ekor) dan ditemukan 11 kebuntingan kembar (5,4%). Kebuntingan kembar dengan 2 vesikel embrio pada 10 induk dan 3 vesikel embrio pada satu induk. Pengurangan embrio dengan memberikan tekanan probe terhadap vesikel embrio berpanduan gambaran ultrasound terhadap 6 ekor kuda bunting kembar berhasil pada satu ekor induk kuda (16,6%) dan melahirkan satu anak. Semua kontrol kebuntingan kembar pada 5 induk kuda mengalami abortus pada usia kebuntingan 7-9 bulan. _____________________________________________________________________________________________________________________

Abstract

The purpose of this study was to evaluate the effectiveness of cysteamine supplementation on the maturation medium and/or in vitro fertilization medium with regards to improving the normal fertilization rate of sheep oocytes, which are characterized by the formation of two pronuclei. Grade A and B oocytes were matured in medium-199 with 0.3% bovine serum albumin (BSA), 10 IU/mL follicle stimulating hormone (FSH), 10 IU/mL human chorionic gonadotrophin (hCG), and 50 g/mL gentamicin added for 24 hours in a 5% CO2 incubator at 39 C. The treatment group was divided into the following groups: a control group with no cysteamine (P1), a group with 100 M cysteamine in the maturation medium (P2), a group with 100 M cysteamine in the fertilization medium (P3), and a group with 100 M cysteamine both in the maturation and fertilization medium (P4). The fertilization was carried out by incubating sperm-oocytes for 12 hours and then staining them with 1% aceto-orcein to observe the formation of a pronucleus. Normal fertilization rates obtained by each treatment group were 56.5% (P1), 57.1% (P2), 57.8% (P3), and 59.9% (P4) with no significant difference (P0.05) between groups. It was concluded that 100 M cysteamine supplementation in both the maturation medium and fertilization medium was not able to increase the normal fertilization rate of sheep oocytes.

Abstract

This research was carried out to determine the morphology and morphometry of Haemonchus contortus in goats. Adult female parasites were collected from abomasum of naturally infected goats and stored in lactophenol. Images of the parasite were acquired using lucida camera and measurement was performed using Axiovision LE software. Basic characters of the morphology and morphometry of Haemonchus spp. were identified by determining the value of the discriminant function of three parameters on spicule. Vulvar morphology was identified by vulvar flap shape. Data were presented in terms of mean, standard deviation, and percentage. A hundred percent of the parasites were H. contortus with discriminant function value 0.63. The size of female parasites was longer and wider (25.5-32.6 mm; 0.38-0.63 mm) than that of male ones (17.3-20.0 mm; 0.24-0.33 mm). There were only 2 varieties of vulvar flap, linguiform (81%), and knobbed (19%). There were 5 subclasses identified in the linguiform type, including linguiform A (39%), linguiform B (22%), linguiform C (17%), linguiform I (1%), and a new subclass of linguiform D (2%). Based on the the type of vulvar flap, H. contortus found in goats in Yogyakarta were different to those found in previous researches. The findings could be used in identifying the parasite species in small ruminants.

Abstract

Penelitian ini bertujuan mengetahui ekspresi insulin pada pankreas mencit (Mus musculus) yang diinduksi streptozotocin berulang denganpewarnaan imunohistokimia yang berguna sebagai hewan model diabetes melitus. Tiga puluh ekor mencit jantan galur Balb-C, umur 12-14 minggu dengan bobot badan 30-40 g dikelompokkan menjadi 2 kelompok perlakuan, masing-masing kelompok terdiri atas 15 ekor. Kelompok 1 (K1) diberikan pelarut streptozotocin, sedangkan kelompok 2 (K2) diberikan streptozotocin dengan dosis 40 mg/kg bobot badan dalam 50 mM natrium sitrat bufer pH 4,5 secara intraperitoneal sebanyak 0,5 ml selama 5 hari berturut-turut. Hewan percobaan dari masing-masing kelompok dieutanasia sebanyak 2 ekor pada hari ke-0, 7, 14, 21, dan 28 setelah perlakuan, selanjutnya mencit diperfusi dan dinekropsi untuk mengambil jaringan pankreas sebagai sampel pemeriksaan imunohistokimia dengan metode streptavidin peroksidase menggunakan antibodi mouse anti-insulin (1:300). Berdasarkan uji statistik menggunakan analisis varian, ekspresi insulin pada sel beta Langerhans pankreas K1 lebih tinggi dibandingkan K2 (P0,05). Waktu pengamatan dan interaksi antara kelompok dan waktu pengamatan menunjukkan perbedaan yang signifikan (P0,05). Induksi dosis rendah streptozotocin secara berulang dapat menurunkan jumlah ekspresi sel beta Langerhans pankreas yang imunoreaktif terhadap insulin.

Abstract

The aim of this study was to examine the preservation of sentul crossbreed chicken semen in ringer lactate egg yolk diluent supplemented with various monosaccharide. Semen was collected from three roosters using massage method. Immediately after collection, the semen was evaluated macroscopically and microscopically. Semen with more than 70% motility was divided into four tubes. Each of them diluted with ringer lactate egg yolk glucose (RLEYG), ringer lactate egg yolk fructose (RLEYF), ringer lactate egg yolk xylose (RLEYX) and ringer lactate egg yolk mannose (RLEYM). Semen was stored in refrigerator (5o C) for sixty hours and evaluated every twelve hours for spermatozoa motility and viability. Results showed that no significant difference (P0.05) among diluents used on spermatozoa quality parameters after dilution and during preservation. Semen quality decrease during storage and at sixty hours of storage, the motility and viability of spermatozoa ranging from 48.332.56 to 55.422.26% and 58.592.87 to 64.832.42%, respectively. This research conclude that glucose, fructose, xylose and mannose can be used as energy source for roosters semen during preservation.

Abstract

Penelitian ini bertujuan mengetahui peran zink pada kesembuhan luka dalam kondisi berat badan berlebih. Dalam penelitian digunakan 20 ekor tikus Sprague Dawley jantan berumur tiga bulan (150-200 g). Tikus dibagi secara acak menjadi empat kelompok (A, B, C, dan D), masing-masing kelompok terdiri atas lima ekor. Kelompok A dan B diberi pakan normal, sedangkan kelompok C dan D diberi pakan lemak tinggi. Setelah dua bulan perlakuan pakan, seluruh tikus dioperasi untuk membuat luka irisan pada kulit, kemudian luka ditutup dengan jahitan. Luka jahitan pada kelompok A dan C diberi aplikasi topikal unguenta vaseline album (tanpa zink), sedangkan luka jahitan pada kelompok B dan D diberi aplikasi topikal unguenta zink 10% dengan bahan dasar vaselin album. Lima hari setelah operasi, seluruh tikus diambil darahnya untuk dianalisis terhadap jumlah leukosit total kemudian diikuti pengambilan jaringan kulit untuk pemeriksaan histopatologis dan penghitungan jumlah leukosit di jaringan. Hasil penelitian menunjukkan adanya interaksi yang signifikan antara pakan dan aplikasi topikal terhadap jumlah leukosittotal dalam darah (P0,05). Jumlah leukosit di jaringan mengalami penurunan setelah aplikasi topikal zink, meskipun hasilnya tidak signifikan (P0,05). Analisis histopatologis menunjukkan epidermis sudah tertutup pada seluruh tikus perlakuan, namun demikian sebagian tikus masih menunjukkan dermis yang terbuka, yaitu kelompok A sebanyak 60%, kelompok B sebanyak 60%, kelompok C sebanyak 60%, dan kelompok D sebanyak 40%. Dapat disimpulkan bahwa proses kesembuhan luka setelah pemberian topikal zink pada tikus yang diberi pakan lemak tinggi menunjukkan hasil yang baik, yaitu ditandai dengan penutupan lapisan epidermis dan dermis, serta menurunnya jumlah leukosit di jaringan.
