Abstract

Simvastatin is a drug used to lower cholesterol levels in the blood. The mechanism of action of simvastatin is by inhibiting the enzyme 3-hydroxy-3-methylglutaryl coenzyme A (HMG-CoA) reductase, which competitively inhibits the cholesterol biosynthesis process in the body. Simvastatin is included in the class II BCS (Biopharmaceutics Classification System) drug, which has low water solubility and high permeability. The low solubility of simvastatin in water will affect its bioavailability in the body, which result in a therapeutic effect that is not achieved. Drug complexation is a method used to determine the solubility of a compound by adding a complexing agent. Complexing materials that are often used in other research are materials that do not have therapeutic activity such as cyclodextrin, but ini this study used complexing material from anti-microbial drug class by utilizing the reaction of glucoronide formation in the glucoronic acid structure of sulfanilamide. This research is a quantitative research with a laboratory experimental trial method which aims to see the effect of adding sulfanilamide complexing agents at various concentrations (25 ppm, 50 ppm, 75 ppm, and 100 ppm) on the solubility level of simvastatin using the acid-base titration method. The dissolved simvastatin concentration values ​​for F1, F2, F3, F4, and F5 were 0.261 ± 0.087, respectively; 0.348 ± 0.151; 0.416 ± 0.195; 0.444 ± 0.163; 0.539 ± 0.118. The results obtained showed that the higher concentration of the sulfanilamide complexing agent added, the greater solubility of simvastatin.

Abstract

Internal Quality Assurance of malaria is a mandatory activity carried out by the malaria examination laboratory which includes a whole series of malaria examinations from pre-analytical, analytical, and post-analytical carried out with the aim of obtaining good malaria microscopic examination results in the form of good quality preparations. So far, there has not been much internal quality assurance associated with quality assessment. The purpose of this study was to determine the relationship between internal quality assurance and the assessment of the quality of malaria preparations in the laboratory of RSUD Umbu Rara Meha Waingapu. This type of research is an analytical quantitative approach with a cross sectional design The sample in this study is the total sample. Seven laboratory officers who carried out microscopic examination of malaria were interviewed and observed regarding the implementation of internal quality assurance from the pre-analytical to post-analytical stages, then 23 positive preparations were taken in 2021 and macroscopic and microscopic observations were carried out and an assessment of the quality of malaria preparations was carried out. Statistical analysis using the Pearson test processed using SPSS 25. The results of the study found that the percentage for the implementation of Internal Quality Assurance was 86%, while the percentage of preparate quality was 82%. Based on the Pearson correlation test between the two variables of internal quality assurance and also the quality of malaria preparate obtained a p-value of 0.010 where p < 0.05 which means there is a relationship between internal quality assurance and the quality of malaria preparate.

Abstract

The sugar levels and protein levels in a urine sample may change over time after sampling. Such delays can affect the stability or degradation of components in the urine, including sugar and protein. Several studies have been conducted concerning that delay such as the difference in the length of time of the delay. But the information that the delay affects the sugar content and protein content of the sample, has not been studied much. The purpose of the study was to analyze the sugar levels and protein levels of urine samples that experienced delayed examination. This study was a laboratory study, with the sample being urine. Because of this, a clearance ethics test was carried out. Chemical Analysis of urine is the measurement of chemical components in urine, namely sugar (glucose), protein, uric acid, ketones, bilirubin, nitrite, and urine pH. Chemical analysis can help in the diagnosis of certain diseases or conditions, such as diabetes, kidney disease, urinary tract infections, or liver disorders. The result was that urine levels and protein levels dropped after a delay in examination for 1 hour and 2 hours. Postponement of the examination lowers the sugar level and protein content of the urine sample. By understanding the changes in sugar and protein levels that can occur during delayed urine examinations, healthcare practitioners can make more informed decisions in the diagnosis and management of patients.

Abstract

Daun sirih cina secara empiris dapat digunakan sebagai obat sakit kepala, obat jerawat, dan obat nyeri perut pada beberapa penelitian juga di laporkan ekstrak etanol tanaman daun  sirih cina (Peperomia pellucida) diduga memiliki kandungan antioksidan yang baik bagi tubuh. Namun belum ada penelitian yang melaporkan bagaimana aktivitas antioksidan dari ekstrak 2 propanol daun sirih cina dimana dari tingkat kepolaran pelarut propanol lebih polar dibanding etanol dan methanol. Kebaharuan dalam penelitian ini adalah menggunakan 2 propanol sebagai pelarut untuk mendapatkan ekstrak daun sirih cina. Tujuan penelitian untuk mengetahui bagaimana aktivitas ekstrak 2 propanol daun sirih cina. Metode yang digunakan untuk mendapatkan nilai aktivitas antioksidan. Penelitian ini merupakan penelitian eksperimental laboratorium. Daun sirih cina dikeringkan dan dihaluskan kemudian dimaserasi dengan larutan 2 propanol. Selanjutnya dilakukan pengujian aktivitas antioksidan dengan metode DPPH (1,1-Dyphenyl-2-Pycrylhydrazyl) menggunakan alat microplate reader pada panjang gelombang 520 nm. Adapun hasil uji aktivitas antioksidan ekstrak 2 propanol daun sirih cina dengan nilai IC50 603,03 ppm yang menyatakan tingkatan aktivitas antioksidannya termasuk dalam kategori lemah. Dapat disimpulkan bahwa antioksidan ekstrak daun sirih cina menggunakan pelarut 2 propanol tidak lebih baik dalam menarik senyawa aktif

Abstract

Yogurt is a functional food product that contains probiotic bacteria. The functional value of yogurt can be enhanced by the addition of inulin from dahlia flower tubers, which has prebiotic content. Inulin is a water insoluble fiber. The purpose of this study was to analyze the organoleptic quality of yogurt added with dahlia flower tuber inulin and its physico-chemical properties. Experimental research type. There were three treatments in this study, namely the addition of dahlia flower inulin as much as 3, 5, and 7% and one control. Krusskall Wallis test was conducted to see the treatment differences in color, aroma, texture and taste of yoghurt. Objective observations included pH measurement, lactic acid content, LAB count and glucose test. Organoleptic test results showed that the most preferred yogurt in terms of color and texture was yogurt without the addition of inulin. The most preferred yogurt flavor and aroma was yogurt with 7% inulin added from dahlia flower tubers. The Krusskall Wallis test results showed no difference in all treatments on the color, aroma, texture, and taste of yogurt. There was no effect of adding inulin from dahlia flower tubers on organoleptic quality. Yoghurt added with 7% inulin at pH level, glucose test, lactic acid content, and lactic acid content increased. Total LAB decreased. Yogurt with 7% inulin added met SNI standards.

Abstract

Kedondong (Spondias dulcis Parkinson) is a tropical fruit known for its rich vitamin C content and associated antioxidant activity. The challenges will maximize the benefits of vitamin C in kedondong fruit soluble in water, and the light will degrade and reduce its effectiveness as an antioxidant. Previous research has highlighted that the heating process affected the antioxidant activity and the ethanol extract kedondong is used to make hand body lotion. This research aims to exploit the potential of using kedondong fruit in the formulation of hand body lotion by utilizing its antioxidant activity. The study used the FRAP method using Trolox as a synthetic antioxidant so that the antioxidant capacity of kedondong juice hand body lotion is obtained which was equivalent to TEAC (Trolox Equivalent Antioxidant Capacity). The results obtained from the hand body lotion preparation of kedondong fruit juice were 4.5 mM TE⁄gram, while kedondong fruit juice was 23.04 mM TE⁄gram. The results of the quality evaluation test of hand body lotion preparations have good homogeneity, do not cause irritation, have stable pH, and have a moisturizing effect on the skin.

Abstract

Clean water plays a central role in human survival. The majority of clean water comes from groundwater. Shallow groundwater from dug wells does not necessarily have suitable quality for use because it is vulnerable to contamination. Bantan Sari is one of the villages located on Bengkalis Island. Besides having problem related to water availability, this area also has poor shallow groundwater quality. This study aims were to determine the quality of shallow groundwater in the study area and map its feasibility as a source of drinking water. This research is expected to be the basis for further research to improve groundwater quality on Bengkalis Island. The research methods included field data collection and laboratory analysis. The parameters measured consisted of color, TDS, DHL, pH, Fe and Mn levels. Based on the results of study, 14 dug wells were obtained. Groundwater color in study area were colorless (36%), yellow (7%), brown (21%), and reddish brown (36%). Groundwater TDS ranged from 59 mg/l – 560 mg/l. Groundwater DHL ranged from 107 mg/l – 1080 mg/l. Groundwater pH ranged from 6,49 – 6,74. Groundwater Fe levels ranged from 0,3 mg/l – 5,43 mg/l and groundwater Mn levels < 0,4 mg/l. Based on Minister of Health Regulation No.492/Menkes/Per/IV/2010 about drinking water quality requirements, it is known that only 1 sample (7%) is included in the drinkable category while the other 13 samples (93%) are included in non-drinkable category.

Abstract

Determination of ketorolac tromethamine levels based on United States Pharmacopeia (USP) using L7 column 4.6 mm x 25 cm; 5 μm with mobile phase tetrahydrofuran and buffer (30:70). Tetrahydrofuran has oxidative properties and has a high UV limit and the presence of peroxide impurities in it can affect the stability of the analyte so that the use of tetrahydrofuran is often avoided. Therefore, a method for analyzing ketorolac tromethamine was developed by applying of High Performance Liquid Chromatography (HPLC) using C18 column with acetate buffer pH 4.2 and methanol (20:80) as mobile phase, flow rate 1.0 mL/minute, wavelength 318 nm, and 10 μL injection volume. The validation results obtained a linearity value (r) 0.9982, value of LOD is 9.61 μg/mL and LOQ is 29.11 μg/mL. The results of precision test obtained %RSD equal to 0.776% and the accuracy test of the sample concentrations 80%, 100% and 120% show the %recovery are 94,79%, 99,42% and 92,83%, respectively. New degradants were formed with acid exposure of 80.66%, base exposure of 59.30%, temperature exposure (70-90°C for 3 hours) are 1.28%, 15.06%, and 18.40 %, respectively. Oxidation exposure (H2O2 1%, 3% and 5%) are 11.36%, 13.98% and 14.79%, respectively. All the research results can be concluded that the method development and validation results can be used for the analysis of ketorolac tromethamine and testing its stability.

Abstract

Donuts are a snack made from wheat flour and are popular among various groups. However, wheat flour contains gluten, which can cause the immune system of a person with celiac disease to react to gluten and attack the small intestine lining. Therefore, wheat flour needs to be substituted with alternative ingredients, one of which is sweet potato flour. This research aims to determine the effect of different types of sweet potato flour in making donuts on their chemical and organoleptic characteristics. The research design used was a Completely Randomized Design (CRD) with five treatments and three replications. Observation data were analyzed using ANOVA and Duncan's New Multiple Range Test (DNMRT) at α = 1%. The treatment in this study was the addition of variations of sweet potato flour (A = 50% white sweet potato, B = 50% purple sweet potato, C = 50% yellow sweet potato, D = 50% orange sweet potato, E = 50% cilembu sweet potato). Further test results showed that the addition of various variations of sweet potato flour had very significantly different effects on moisture, ash, fat, protein, carbohydrate content, and antioxidant activity. Based on organoleptic tests, the best treatment was treatment E (cilembu sweet potato flour) with an average value of 5.93 (very like), where the resulting donuts had the following chemical characteristics: moisture content (24.18%), ash content (2.10%), fat content (10.87%), protein content (14.96%), carbohydrate content (56.74%) and antioxidant activity (64.75%). Donuts with cilembu sweet potatoes have the most fragrant aroma, golden brown color, slightly sweet taste, and soft texture.

Abstract

Noodles are one of the carbohydrate alternatives to rice. To get healthy noodles, you need additional vegetables or fruit. Dragon fruit skin is high in antioxidants that function to ward off free radicals. The addition of tuna bone meal as an additional nutritional value in the form of potassium and mineral. The purpose of this study was to determine the best characteristics of red dragon fruit skin dry noodles with the addition of tuna fish bone meal which were the best in making dry noodles that the panelists liked. This study used a Completely Randomized Design (CRD) with 5 treatment levels and 3 replications. Observational data were analyzed using Duncan’s New Multiple Range Test (DNMRT) at 1% level. The treatment in this study was the addition of tuna fish bone meal to dried red dragon fruit skin noodles (A=2%, B=4%, C=6%, D=8%, E=10%). The results showed that the addition of tuna bone meal had highly significant different, ash content, protein content, antioxidant test and calcium content. Based on the organoleptic test, dry noodles with treatment B (Addition of 4% tuna fish bone meal) were most preferred by the panelists and met the quality requirements of SNI No 8217-2015 with a composition of water content (11,47%), ash content (0,45%), protein content (16,24%), antioxidant test (34,38%) and calcium content (30,79%).
