Abstract

Background: In Central Sulawesi, Indonesia, Syzygium polyanthum leaves (SPL) have been traditionally used by society to reduce high blood pressure. However, the molecular mechanism behind this effect still remains unclear. Objective: This study aims to explore the antihypertensive molecular mechanisms of SPL using a network pharmacology approach.Method: Previous studies have investigated some bioactive compounds contained in SPL. Those studies identified 57 bioactive compounds, of which 31 were filtered for further analysis. Target proteins from SPL and disease were identified using GeneCards. Then, 62 selected target proteins which overlapped through Venn diagrams would be analyzed using Cytoscape software to produce compounds-target proteins’ network. Protein-protein interactions were assessed using STRING and visualized by Cytoscape. GO Function and KEGG Pathways were obtained using ShinyGo.Results: The results of this study revealed pathways associated with hypertension through the cellular senescence pathway with target proteins mTOR, ERK (also known as MAPK), p53, CycD, and IL6. The chemical constituents associated with these target proteins are hydroxychavicol, farnesol, naphthalene, phloretin, pyrogallol, and decanal, which were identified as compounds closely related to the target protein of hypertension. Conclusion: The bioactive compounds in SPL play a significant role in regulating hypertension by influencing various target genes, particularly those associated with cellular senescence. Keywords: Antihypertensive, bay leaf, cellular senescence, network pharmacology, Syzygium polyanthum

Abstract

There has been an increase in cases of sexual violence against children in the Ponorogo Regency, some of which have been made public through social media. Due to the courage of both those who commit sexual violence and the victims, this issue makes for an exciting area of research using the interaction theory, the social factor theory, and the coping theory. This study aimed to analyze the interactions of social factors and parents' coping models of the victims and perpetrators of sexual violence. It employed a qualitative approach through interviews, observations, and documentation. The findings revealed that all victims and perpetrators of sexual violence have an honest relationship with their parents. As a result, this act of sexual violence occurs due to factors in the victim's personality being close to their parents. The social factors of victims, parents, and perpetrators related to education are diverse. This crime occurs at all levels of education because, as Muzaham argues, family education is an essential element of character development that has not been implemented. The fact that all victims and perpetrators are from Java indicates a cultural shift in Javanese society due to social changes. While the concept of family is good, it falls short in building character. Focused coping is the primary mode of coping in the model. Some parents use emotional coping in the beginning, but they eventually switch to a more focused approach

Abstract

This study aims to find a negotiation model in the tradition of Javanese Muslim communities in resolving disputes regarding marriage and inheritance restrictions. When the reality of resolving marriage and inheritance disputes impacts family and community strife, this becomes crucial. The qualitative paradigm was used to construct this research, which included a series of observations and in-depth interviews with the disputed families and focus group discussions with informants in the East Java community. This research discovers three models for resolving the tradition of avoiding marriage and inheritance disputes in Javanese Muslim society with social components in a heterogeneous society. 1) negotiating the traditional cum theology model, 2) negotiating the tradition and law model, and 3) negotiating the traditional wisdom model. Furthermore, when many cultural traditions fade, the growing negotiation model can serve as the foundation for policy for local governments in generating variants of the nation's cultural traditions.

Abstract

Pluchea indica L. (Less), known as Indian camphorweed (common name) or Beluntas (Indonesia), is an antioxidant polyphenol-rich plant. In our previous studies, choline chloride-urea (CCU) at a molar ratio of 1:2 was one type of natural deep eutectic solvents (NADES) in the extraction of P. indica leaves (PIL) polyphenolics. The present study aims to evaluate the comparison of total phenolics (TPC) and flavonoids content (TFC) and antioxidant activity of CCU (1:2) extract of PIL obtained from ultrasonic-assisted extraction (UAE) and magnetic stirrer-assisted extraction (MSAE). Ethanol was used as a comparison to conventional extraction solvents. Determination of TPC and TFC was carried out using colorimetric with a microplate reader, while the antioxidant activity test was carried out using the ferric thiocyanate (FTC) method. The UAE is more effective in terms of PIL flavonoid extraction using CCU compared to MSAE, while PIL phenolic extraction with CCU is more effective with MSAE. CCU extract of PIL from extraction with MSAE at a concentration of 1.56% has an antioxidant capacity equivalent to 500 ppm ascorbic acid in inhibiting lipid peroxidation by about 70%. Thus, CCU combined with the MSAE can be recommended in further development of phenolic antioxidants extraction from PIL.

Abstract

Capsaicin, a potent analgesic, suffers from poor aqueous solubility and low bioavailability. Therapeutic deep eutectic solvents (THEDES) have emerged as a promising platform to enhance the solubility of poorly soluble drugs. This study employed the conductor-like screening model for real solvents (COSMO-RS) for the in-silico screening of capsaicin solubility in 34 lidocaine-based THEDES, comprising 10 hydrogen bond acceptors (HBAs) and 26 hydrogen bond donors (HBDs) at 1:1 and 1:2 molar ratios. The σ-profiles and σ-potentials of the components were analysed to understand the intermolecular interactions governing solubility. Our predictions revealed a remarkable solubility range from below 1 g/L to over 437 g/L. The betaine-lidocaine (1:1) system was identified as the optimal solvent, achieving a capsaicin solubility of 437.47 g/L at 333.15 K, attributed to betaine's zwitterionic nature facilitating multifaceted hydrogen-bonding. Small polyols like ethylene glycol also performed excellently. A consistent enhancement in solubility was observed in HBD-rich (1:2) compositions and with increasing temperature. Molecular interaction analysis confirmed a robust network of conventional and non-conventional hydrogen bonds within the optimal betaine-lidocaine-capsaicin system. This work demonstrates the power of COSMO-RS as a rational design tool for formulating high-loading THEDES-based drug delivery systems, with betaine-lidocaine emerging as a top candidate for advanced capsaicin topical formulations.

Abstract

Garcinia cowa Roxb. ex DC. is a plant belonging to the Clusiaceae family, and commonly dicovered in Southeast and South Asia. This study aimed to assess the efficiency of eight natural deep eutectic solvents (NaDES) in extracting total xanthones and phenolic compounds from the stem bark using ultrasound-assisted extraction as there is limited research on the use of NaDES in this plant extraction process. The study also examined their antioxidant properties. The NaDES were synthesized with choline chloride and betaine as hydrogen-bond acceptors, accompanied by various acids, alcohols, and glucose as hydrogen-bond donors. In comparison, ethanol was used as standard solvent. The NaDES exhibited higher densities than water, ranging from 1.059 to 1.244 g/cm³, with density increasing according to the number of hydroxyl groups present in the constituents. The total phenolic content (TPC) varied from 22.82 to 28.73 mg GAE/g extract, with NaDES1 (a combination of choline chloride, 1,2-propanediol, and water in a ratio of 1:3:1) showing the highest TPC at 28.73±0.18 mg GAE/g extract. It also exhibited significant antioxidant activities, as demonstrated by DPPH (28.98±0.03 µg/mL) and FRAP assays (43.66±1.51 mmol trolox/g dw). A significant negative correlation was observed between total xanthone, total phenolic content, and IC50 values. These findings suggest that NaDES, particularly NaDES1, have considerable advantages in extracting phenolic compounds and xanthones from G. cowa stem bark, resulting in enhanced antioxidant properties. This highlights the potential of NaDES as eco-friendly and effective solvents for the extraction of bioactive compounds from plant materials.
