Abstract

Purpose – This study aims to describe the validity and practicality of augmented reality card media on biodiversity material for class X students of SMA Negeri 1 Telaga Biru, based on morphological studies of bananas (Musa spp).Method – This research used the research and development (R&D) method with the PLOM model. The media was developed using Assembler and Canva applications. Findings – The learning media developed in this research is in the form of augmented reality (AR) cards designed to improve students' understanding through interactive visualization of three-dimensional objects. This media combines print elements with digital technology, thus providing an interesting, contextual, and easy-to-understand learning experience. The results of the validity test showed that the media expert's assessment obtained an average score of 80% in the “Valid” category, while the material expert gave an average score of 85%, which was included in the “Very Valid” category. Overall, the average validity of the media reached 82% and was categorized as “very valid.” The practicality of the media was tested through teacher and learner response questionnaires, each of which obtained an average score of 95% and 89%, so that the total practicality reached 92% in the “Very Practical” category. Research Implications – Based on the results of the validity and practicality tests, the augmented reality card media is considered very valid and very practical, so it is feasible to use as a biology learning media, especially in high school biodiversity material.

Abstract

Bihe Village is one of the villages in Asparaga District, Gorontalo Regency, which is a center for coconut production. The abundant potential of coconut plants has yet to be utilized optimally by the people of Bihe village. Coconuts are only produced as copra, or coconuts are sold whole, without processing. Coconut plants have many derivative products, such as virgin coconut oil (VCO) and coconut peat. This problem prompted the implementation of socialization, training, and assistance in making VCO and Cocopeat for the Bihe village community. The aim is to increase the community's insight and skills regarding the diversification of coconut products so that they can provide additional income, which can improve the economy in Bihe Village. The training method is the Participatory Action Research (PRA) method, which involves the active role of the Bihe village community. The stages carried out are analysis of the actual conditions of society, planning, action, observation, and reflection. This method is carried out to provide an understanding of a) the weaknesses they have, b) the community's desire to overcome their shortcomings and weaknesses, c) together with the community to develop strategies and methods to solve the problem, and d) help the community overcome, solve, and find a way out. Forty participants attended this training and mentoring. After implementing the empowerment program, the post-test results showed that 62.5% of the community gave an excellent assessment, and 37.5% gave a good assessment of the usefulness of this activity program. Furthermore, 72.5% of participants were motivated to open a new coconut commodity business.

Abstract

Momordica cochinchinensis, locally known as Dumbaya in Gorontalo Province, is a traditional medicinal plant with underutilized potential due to the extremely hard morphology of its seed coat. This physical barrier limits the penetration of water, air, and nutrients, thereby reducing the success rate of seed germination and plant propagation. The challenge of propagating plants that are difficult to reproduce sexually, such as dumbaya, can be addressed through various approaches, one of which is asexual reproduction using tissue culture techniques. This study aims to address propagation constraints by applying tissue culture techniques supplemented with Plant Growth Regulators (PGRs), specifically BAP (Benzyl Amino Purine) and NAA (Naphthalene Acetic Acid), which are critical factors in successful in vitro plant regeneration. The combination of Benzyl Amino Purine (BAP) and Naphthalene Acetic Acid (NAA) is a commonly used Plant Growth Regulator (PGR) for inducing callus formation and organogenesis, where BAP is effective in stimulating shoot formation, while NAA plays a role in root induction. The research employed an experimental method using a Completely Randomized Design (CRD) with five treatment levels: H0: MS + 0 ppm NAA + 0 ppm BAP; H1: MS + 1.5 ppm NAA + 0.5 ppm BAP; H2: MS + 3.0 ppm NAA + 0.5 ppm BAP; H3: MS + 1.5 ppm NAA + 1.0 ppm BAP; and H4: MS + 3.0 ppm NAA + 1.0 ppm BAP. Each treatment was replicated three times. The results showed that treatment H1 produced the earliest callus formation, with an average callus initiation time of 7 days after planting.

Abstract

Banana (Musa spp) is a tropical fruit plant that has many benefits and important roles for the community. This study aims to provide information on the morphological characteristics of bananas in the Gorontalo Province and their phenetic relationships. This study uses a descriptive method to describe the diversity of morphological characters of bananas (Musa spp) with data collection using a survey study method (exploration) which is carried out by tracing or exploring the location of the Gorontalo Province, purposive sampling (Purposive Stratified Sampling) by seeing and observing directly the morphological characters of bananas (Musa spp). Data analysis using the Unweighted Pair Group with Mean of Arithmetic (UPGMA) method with the Gower Genelar Similarity Coeffiicient formula in the MVSP (Multi-Variate Statistical Package) v.3.1 computer program. The results of the study showed that there were 4 types of bananas found in the Gorontalo Province which showed unique morphological variations, namely Kepok Bananas have the characteristic of quite large leaf morphology, Abati Bananas have a unique yellowish color, Green Butota Bananas have morphological characteristics with very thick fruit and Wild Bananas have characteristics with a large number of stones. Based on the results of the clustering, 2 main groups were formed which separated at a cosiness of 0.643 (64.3%) found between Abati Bananas, Green Butota, and Wild Bananas, while Kepok Bananas showed a very distant relationship with a cosiness value of 0.482 (48.2%). The results of this study have the potential to support the preservation of local varieties and support the sustainable genetics management through the identification of varieties that are tolerant to salinity and drought.

Abstract

The lack of contextual teaching materials that highlight local potential is a challenge in biology learning in schools. This study aims to determine the validity and practicality of the learning module on genetic biodiversity in local Gorontalo corn plants. This study uses the Research and Development method. The learning module was developed using the ADDIE model, up to the limited trial stage. The research subjects were 25 grade X high school students. The learning module was validated by material experts and media experts, and the practicality of using the module in learning biodiversity material was tested. The validation results showed that the module was very valid with an average percentage of 98% from material experts and 93.77% from media experts. The practicality of the module was also very high based on teacher responses of 97.77% and student responses of 98.06%. From the results of the validity and practicality tests, it can be concluded that the learning module on genetic biodiversity in local corn plants can be used in learning Biology for grade X high school, biodiversity material in schools. Abstrak. Kurangnya bahan ajar kontekstual yang mengangkat potensi lokal menjadi tantangan dalam pembelajaran biologi di sekolah. Penelitian ini bertujuan untuk mengetahui validitas dan kepraktisan modul pembelajaran keanekaragaman hayati tingkat genetik pada tanaman jagung lokal Gorontalo. Penelitian ini menggunakan metode Research and Development.  Modul pembelajaran dikembangkan dengan menggunakan model ADDIE, hingga tahapan uji coba terbatas. Subjek penelitian adalah 25 peserta didik kelas X SMA. Modul pembelajaran divalidasi oleh ahli materi dan ahli media, serta diuji kepraktisan penggunaan modul dalam pembelajaran materi keanekaragaman hayati.  Hasil validasi menunjukkan bahwa modul sangat valid dengan rata-rata persentase 98% dari ahli materi dan 93,77% dari ahli media. Kepraktisan modul juga sangat tinggi berdasarkan respons guru sebesar 97,77% dan respons peserta didik sebesar 98,06%. Dari hasil uji validitas dan kepraktisan, maka dapat disimpulkan bahwa modul pembelajaran keanekaragaman hayati tingkat genetik pada tanaman jagung lokal dapat digunakan dalam pembelajaran Biologi kelas X SMA, materi  keanekaragaman hayati  di sekolah.

Abstract

Antidesma bunius L. is a tree species that has ecological and economic benefits, and has many bioactivities. Propagation of this tree can be done in several ways, one of which is callus induction. Callus is a collection of undifferentiated cells that can develop into new plants with the help of ZPT. The purpose of this study was to determine the effect and optimum combination of ZPT 2,4-D and BAP on the induction of leaf callus of A. bunius L. The research design was a non-factorial Completely Randomised Design (CRD) with 4 treatments namely P0: MS without the addition of zpt; P1: MS + 1 ppm BAP + 3 ppm 2,4-D; P2: MS + 2 ppm BAP + 2 ppm 2,4-D; P3: MS + 3 ppm BAP + 1 ppm 2,4-D. Parameters observed included callus emergence time, percentage of callus explants, and callus morphology. The results showed that P2 is the best concentration for callus induction, by looking at the observed effect on callus emergence time of 6 HST, percentage of callus explants 56% and callus morphology which is white with crumb texture, which can develop into embryogenic callus

Abstract

This quantitative descriptive research aimed to describe the antimicrobial activity and phylogenetic relationships of Actinomycetes from the rhizosphere of pandan (Pandanus spp.). Soil samples were taken from three different villages in Gorontalo Regency, including Bontula Village, Asparaga Sub-district, Lombongo Village, Suwawa Tengah Sub-district, and Moutong Village, Tilongkabila Sub-district, Bone Bolango Regency. Isolation was carried out using SCA media, resulting in six Actinomycetes isolates with different colony morphologies. Antimicrobial activity tests were carried out using the cross-streak method against Escherichia coli, Staphylococcus aureus, Candida albicans, Fusarium oxysporum, and Neocosmospora solani. The results showed that only one isolate, IRzP-at.k, showed significant antimicrobial activity, against Escherichia coli and Staphylococcus aureus with inhibition zones of 13,31 mm and 13,67 mm, respectively. The IRzP-at.k isolate also showed inhibition against Candida albicans and Fusarium oxysporum with inhibition zones of 34,12 mm and 11,25 mm, but did not show inhibition against Neocosmospora solani. Phylogenetic analysis based on 16S rRNA gene sequences identified the isolate as closely related to Streptomyces vinaceusdrappus (97.42% similarity). This research indicated the potential of the IRzP-at.k isolate as a source of new antimicrobial compounds and strengthens the role of the pandan rhizosphere as a habitat rich in Actinomycetes.

ABSTRAK Elawati.2015. Efisiensi Penyerapan Logam Berat Tembaga (Cu) Oleh Tumbuhan Kangkung Air (Ipomoae aquatica Forks) Dengan Waktu Kontak Yang Berbeda. Skripsi. Jurusan Pendidikan Biologi, Fakultas Matematika dan IPA, Universitas Negeri Gorontalo. Dr. Novri Y. Kandowangko M.P sebagai Pembimbing I dan Dr. Djuna Lamondo, M.Si sebagai Pembimbing II. Penelitian ini bertujuan untuk mengetahui efisiensi penyerapan logam berat tembaga (Cu) oleh tumbuhan kangkung air (Ipomoea aquatica Forks) pada waktu kontak yang berbeda. Penelitian ini adalah penelitian eksperimen dengan menggunakan 4 perlakuan, yakni waktu kontak selama 7, 14, 21, dan 28 hari. Kemudian setiap wadah diberikan Cu(NO3) dengan konsentrasi 40 ppm. Pengukuran akumulasi logam Cu pada tumbuhan kangkung air dengan menggunakan Atomic Absorpsion Spektofotometri (AAS). Hasil penelitian menunjukan bahwa penyerapan tumbuhan kangkung air yaitu pada hari ke-7, kangkung air menyerap logam berat Cu sebanyak 26,27 ppm atau sekitar 65,67 %, pada hari ke-14 menyerap sebesar 28,25 ppm atau sekitar 70,62 %, hari ke-21 menyerap sebesar 26,38 ppm atau sekitar 65,95 % dan hari ke-28 menyerap sebesar 24,5 ppm atau sekitar 61,25 % yang terserap. Abstract

Yellow bamboo has great potential in various fields, but its conventional propagation has several obstacles. Tissue culture offers a solution to overcome these problems. The focus of this research is the use of appropriate sterilization for in vitro culture. Effective sterilization is a crucial first step in the success of tissue culture. In this study using sterilization in the form of using Nordox 56 WP, without using Nordox 56WP, using Antracol 5 gr solution, and using HgCl2. The parameters observed were the appearance of contaminants in the form of fungi and bacteria and the time of death of the explants. The results showed that the use of HgCl2 can inhibit contaminants and the time of explant death in yellow bamboo plants in vitro.

Abstract

Penelitian ini bertujuan untuk mendeskripsikan karakteristik morfoogi tanaman Lemon Suanggi (Citrus limon) yang tumbuh di wilayah pesisir Teluk Tomini. Penelitian ini menggunakan metode deskriptif eksploratif, dengan aksesi yang dikumpulkan dari lima kabupaten di dua provinsi, yaitu di Gorontalo dan Sulawesi Tengah. Data Morfologi diperoleh melalui pengamatan langsung dari batang, daun, buah, dan biji, mengacu pada Deskriptor untuk Jeruk oleh Plant Genetic Resoutces Institue (1999). Temuan menunjukkan bahwa ada delapan aksesi Lemon Suanggi (Citrus limon) dengan variasi karakteristik morfologi yang nyata. Perbedaan ini diamati pada ukuran dan bentuk daun, ketebalan kulit buah, jumlah biji, dan tekstur permukaan buah. Variasi morfologi ini dipengaruhi oleh kombinasi faktor genetik dan kondisi lingkungan seperti suhu, kelembaban, dan curah hujan di wilayah pesisir. Temuan ini menunjukkan keragaman yang signifikan yang memiliki potensi besar utuk menjadi landasan penting bagi konservasi, pemuliaan, dan pengembangan sumber data genetik tanaman lokal di masa depan.
