Abstract

Antidesma bunius L. is a tree species that has ecological and economic benefits, and has many bioactivities. Propagation of this tree can be done in several ways, one of which is callus induction. Callus is a collection of undifferentiated cells that can develop into new plants with the help of ZPT. The purpose of this study was to determine the effect and optimum combination of ZPT 2,4-D and BAP on the induction of leaf callus of A. bunius L. The research design was a non-factorial Completely Randomised Design (CRD) with 4 treatments namely P0: MS without the addition of zpt; P1: MS + 1 ppm BAP + 3 ppm 2,4-D; P2: MS + 2 ppm BAP + 2 ppm 2,4-D; P3: MS + 3 ppm BAP + 1 ppm 2,4-D. Parameters observed included callus emergence time, percentage of callus explants, and callus morphology. The results showed that P2 is the best concentration for callus induction, by looking at the observed effect on callus emergence time of 6 HST, percentage of callus explants 56% and callus morphology which is white with crumb texture, which can develop into embryogenic callus

Abstract

This quantitative descriptive research aimed to describe the antimicrobial activity and phylogenetic relationships of Actinomycetes from the rhizosphere of pandan (Pandanus spp.). Soil samples were taken from three different villages in Gorontalo Regency, including Bontula Village, Asparaga Sub-district, Lombongo Village, Suwawa Tengah Sub-district, and Moutong Village, Tilongkabila Sub-district, Bone Bolango Regency. Isolation was carried out using SCA media, resulting in six Actinomycetes isolates with different colony morphologies. Antimicrobial activity tests were carried out using the cross-streak method against Escherichia coli, Staphylococcus aureus, Candida albicans, Fusarium oxysporum, and Neocosmospora solani. The results showed that only one isolate, IRzP-at.k, showed significant antimicrobial activity, against Escherichia coli and Staphylococcus aureus with inhibition zones of 13,31 mm and 13,67 mm, respectively. The IRzP-at.k isolate also showed inhibition against Candida albicans and Fusarium oxysporum with inhibition zones of 34,12 mm and 11,25 mm, but did not show inhibition against Neocosmospora solani. Phylogenetic analysis based on 16S rRNA gene sequences identified the isolate as closely related to Streptomyces vinaceusdrappus (97.42% similarity). This research indicated the potential of the IRzP-at.k isolate as a source of new antimicrobial compounds and strengthens the role of the pandan rhizosphere as a habitat rich in Actinomycetes.

ABSTRAK Elawati.2015. Efisiensi Penyerapan Logam Berat Tembaga (Cu) Oleh Tumbuhan Kangkung Air (Ipomoae aquatica Forks) Dengan Waktu Kontak Yang Berbeda. Skripsi. Jurusan Pendidikan Biologi, Fakultas Matematika dan IPA, Universitas Negeri Gorontalo. Dr. Novri Y. Kandowangko M.P sebagai Pembimbing I dan Dr. Djuna Lamondo, M.Si sebagai Pembimbing II. Penelitian ini bertujuan untuk mengetahui efisiensi penyerapan logam berat tembaga (Cu) oleh tumbuhan kangkung air (Ipomoea aquatica Forks) pada waktu kontak yang berbeda. Penelitian ini adalah penelitian eksperimen dengan menggunakan 4 perlakuan, yakni waktu kontak selama 7, 14, 21, dan 28 hari. Kemudian setiap wadah diberikan Cu(NO3) dengan konsentrasi 40 ppm. Pengukuran akumulasi logam Cu pada tumbuhan kangkung air dengan menggunakan Atomic Absorpsion Spektofotometri (AAS). Hasil penelitian menunjukan bahwa penyerapan tumbuhan kangkung air yaitu pada hari ke-7, kangkung air menyerap logam berat Cu sebanyak 26,27 ppm atau sekitar 65,67 %, pada hari ke-14 menyerap sebesar 28,25 ppm atau sekitar 70,62 %, hari ke-21 menyerap sebesar 26,38 ppm atau sekitar 65,95 % dan hari ke-28 menyerap sebesar 24,5 ppm atau sekitar 61,25 % yang terserap. Abstract

Yellow bamboo has great potential in various fields, but its conventional propagation has several obstacles. Tissue culture offers a solution to overcome these problems. The focus of this research is the use of appropriate sterilization for in vitro culture. Effective sterilization is a crucial first step in the success of tissue culture. In this study using sterilization in the form of using Nordox 56 WP, without using Nordox 56WP, using Antracol 5 gr solution, and using HgCl2. The parameters observed were the appearance of contaminants in the form of fungi and bacteria and the time of death of the explants. The results showed that the use of HgCl2 can inhibit contaminants and the time of explant death in yellow bamboo plants in vitro.

Abstract

Penelitian ini bertujuan untuk mendeskripsikan karakteristik morfoogi tanaman Lemon Suanggi (Citrus limon) yang tumbuh di wilayah pesisir Teluk Tomini. Penelitian ini menggunakan metode deskriptif eksploratif, dengan aksesi yang dikumpulkan dari lima kabupaten di dua provinsi, yaitu di Gorontalo dan Sulawesi Tengah. Data Morfologi diperoleh melalui pengamatan langsung dari batang, daun, buah, dan biji, mengacu pada Deskriptor untuk Jeruk oleh Plant Genetic Resoutces Institue (1999). Temuan menunjukkan bahwa ada delapan aksesi Lemon Suanggi (Citrus limon) dengan variasi karakteristik morfologi yang nyata. Perbedaan ini diamati pada ukuran dan bentuk daun, ketebalan kulit buah, jumlah biji, dan tekstur permukaan buah. Variasi morfologi ini dipengaruhi oleh kombinasi faktor genetik dan kondisi lingkungan seperti suhu, kelembaban, dan curah hujan di wilayah pesisir. Temuan ini menunjukkan keragaman yang signifikan yang memiliki potensi besar utuk menjadi landasan penting bagi konservasi, pemuliaan, dan pengembangan sumber data genetik tanaman lokal di masa depan.

Abstract

This study was conducted in the coastal area of Tomini Bay, Sulawesi Island, and it focused on Citrus maxima, a plant known for its unique fruit flesh colors, which range from yellow to pink, and its varying leaf stalk wings. The study aimed to analyze the variations in maturase K (matK) sequences, molecular characteristics, and phylogenetic relationships of two C. maxima samples from Tomini Bay compared to other C. maxima and Citrus species using data available in GenBank. The study utilized DNA barcoding with matK molecular markers, followed by phylogenetic tree construction using the maximum likelihood method and the Tamura 3-parameter model. The matK DNA sequences from both samples were 1,530 base pairs in length, with an AT content of 64.64%, a GC content of 35.36%, and no variable sites. Phylogenetic tree reconstruction revealed that C. maxima formed a monophyletic group. These samples were also grouped in the same phylogenetic branch as C. maxima Mato Buntan, C. maxima Suisho Buntan, and C. maxima Banpeiyu, with a similarity index score of 100%, which indicates a very close relationship. Despite morphological differences, genetic analysis showed significant similarities among the samples, confirming a close relationship with minimal genetic variation

Abstract

The learning process in high schools in West Papua tends to focus on students’ cognitive enhancement and ignore the aspect skill. School is the frontline to develop students’ skills. Metacognitive and process skills are the two skills needed by the students, and both should be trained and developed. These two skills help students to implement an investigation process in which the investigation process can be well utilized using inquiry-based learning. This study aims to find out the differences in metacognitive and process skills between students treated with inquiry-based learning and those who are taught using conventional learning. This experimental study was carried out at Grade X of senior high school students of SMA Negeri 1 Manokwari. The result reveals a significant metacognitive skill of 0.000 < α 0.05, and science process skill of 0.001 < α 0.05 of students who were taught using the inquiry-based learning more than those who were taught using the conventional learning. It can be concluded that there is a significant improvement in students’ metacognitive and process skills for those who are taught using inquiry-based learning compared to those who are taught using conventional learning. Therefore, inquiry-based learning has the potential to appropriately train students’ metacognitive and science process skill.

Abstract

Penelitian ini bertujuan untuk menganalisis kadar antosianin pada jagung lokal Gorontalo varietas Binthe Kiki, Binthe Momala, dan Binthe Pulo. Metode yang digunakan adalah penelitian deskriptif kuantitatif dengan teknik ekstraksi dan pengukuran antosianin menggunakan metode pH diferensial pada panjang gelombang 530 nm dan 700 nm. Hasil penelitian menunjukkan bahwa ketiga varietas mengandung antosianin dengan kadar yang bervariasi, yaitu Binthe Kiki sebesar 9,00 ppm, Binthe Momala sebesar 3,77 ppm, dan Binthe Pulo sebesar 3,13 ppm. Kandungan antosianin tertinggi ditemukan pada varietas Binthe Kiki, yang ditandai dengan warna biji orange. Perbedaan kadar antosianin antar varietas dipengaruhi oleh faktor genetik, warna biji, dan kondisi lingkungan. Temuan ini menunjukkan bahwa jagung lokal Gorontalo, khususnya varietas Binthe Kiki, berpotensi sebagai sumber pewarna alami sekaligus antioksidan bagi kesehatan manusia.

Abstract

One of the primary agricultural commodities that contributes significantly to global food security is maize (Zea mays L.), a monoecious cereal plant of the Poaceae family with distinct but identical male and female blooms. Local cultivars like Pulut and Momala in Gorontalo have exceptional promise that requires further study, particularly with regard to their anatomical makeup. This study Comparing the two types' stem and leaf anatomical features, including vascular tissue (xylem and phloem), epidermis, and stomata, is the goal of this study. A qualitative descriptive approach was used to make the observations, which were subsequently evaluated and shown as a comparison table. The results showed that at the age of 30 and 40 HST, the network of vessels (xylem and phloem) and epidermis on the stems and leaves of Pulut and Momala corn varieties reflected a development pattern that supported the transportation of water, minerals, and nutrients. The number of stomata in Pulut varieties decreased due to less than optimal growth, while Momala varieties showed an increase in the number of stomata which was influenced by the growth period with environmental conditions suitable for corn plants.
