Abstract

Tuberculosis (TB) is an infectious disease caused by Mycobacterium tuberculosis. Indonesia is one of the developing countries with high TB patients in the world. The most common detection used for TB diagnosis is sputum smear examination. Smear negative pulmonary TB still have a risk of infection and can develop into active. The GeneXpert molecular examination is a rapid diagnosis of TB by the real-time PCR method. The purpose of this study was to assess the validity of GeneXpert in smear-negative pulmonary TB compared to Lowenstein Jensen's culture. The design of this diagnostic test study is a cross-sectional study. The study was conducted on 40 people with smear-negative pulmonary TB suspect patients in Dr. Doris Sylvanus Regional Hospital. Sputum examination was performed with GeneXpert and compared with Lowenstein Jensen culture. The results of GeneXpert validity for diagnosing smear-negative pulmonary TB suspect are sensitivity 81.8%, specificity 96.5%, positive predictive value 90%, negative predictive value 93.3%, and accuracy 92.5%. It was concluded that GeneXpert has sensitivity, specificity, positive predictive value, negative predictive value, and high accuracy as a diagnostic tool in smear-negative pulmonary TB suspects.

Abstract

Androgen has an important role in regulation epididymal sperm maturation  Epididymal sperm maturation is occurs via interactions between sperm and proteins secreted by epididymal genes that are specifically expressed in a region-specific manner. Mice CD52 exhibited expression in all regions in epididymis. However, the regulation of its expression by androgen is needed to be known as a gene candidate that involved in sperm maturation. Objective: This study was aimed to analyze regulation of mice CD52 expression by androgen and a bioinformatics analysis was performed to predict functional domains of CD52. Methods: bioinformatics analysis using Motif Scan. For experiment, mice divided into 5 groups. The groups were control was injected ethanol 0,1 mL/day and others groups were  3, 5, 10, and 15 day used a androgen receptor antagonist, flutamide that dilute in ethanol at a dose of 50 mg/kg/day. And then quantitative real-time RT-PCR was used to analyse expression of CD52 and it’s normalized to Beta actin. Result: CD52 contained a two potential phosphorylation sites for protein kinase C and casein kinase II, N-myristoylation, and N-glycosylation that showed CD52 is secretory protein and predict it’s attached to sperm membrane. Expression of CD52 was decrease about 93% than control. Conclusion: CD52 expression depend on androgen.Keywords:  androgen; epididymis; sperm maturation; expression of CD52

Abstract

Gigitan nyamuk dapat membahayakan bagi kesehatan tubuh. Salah satu upaya untuk memberantas nyamuk dalam kehidupan sehari-hari yang umum digunakan adalah dengan menggunakan obat nyamuk. Obat nyamuk terbanyak yang dipakai di masyarakat Indonesia adalah jenis obat nyamuk bakar dan semprot. Kandungan dalam obat nyamuk yaitu berbahan aktif pyrethroid, yang memiliki efek bila dalam jangka waktu lama digunakan dapat menganggu kesehatan tubuh. Salah satunya gangguan terhadap reproduksi pada pria. Oleh sebab itu tujuan penelitian ini untuk mengetahui pengaruh obat nyamuk bakar dan semprot terhadap motilitas sperma tikus galur wistar. Penelitian eksperimental in vivo ini menggunakan desain post-test randomized control design dengan menggunakan 5 kelompok tikus. Kelompok ini terbagi menjadi kontrol, paparan obat nyamuk semprot 5 mL/hari selama 20 hari, paparan obat nyamuk semprot 5 mL/hari selama 35 hari, paparan obat nyamuk bakar 4,5 gr/hari selama 20 hari, paparan obat nyamuk bakar 4,5 gr/hari selama 20 hari. Hasil penelitian secara mikroskopis menunjukkan penurunan motilitas bermakna tidak signifikan pada kelompok tikus yang diberi obat nyamuk semprot dan bakar sejak dipaparkan 20 hari lamanya dan penurunan paling signifikan pada obat nyamuk bakar 35 hari. Obat nyamuk semprot dan bakar yang digunakan rutin dan dalam waktu lama dapat menurunkan motilitas sperma.

Abstract

Indonesia is one of the countries set as a corona disaster emergency with the increasing Covid-19 case. One of the factors that caused an increase in Covid-19 cases was the lack of public awareness in implementing 5-M prevention behavior using masks, hand washing using soap and running water, keeping distance, staying away from the crowd, and limiting mobilization and interaction) in his daily life can be a risk transmission in society. Therefore, an approach was carried out in increasing public awareness of the new normal adaptation in the face of the Covid-19 pandemic condition by promoting the "4G Movement" (Go Green, Go Health, Go Clean and Go Smart) at the UPTD Puskesmas Tangkililing carried out by the Faculty Team Medicine of Palangka Raya University. This activity aims to increase public awareness and understanding of the importance of good health behavior by applying a strict health protocol in the condition of the Covid-19 pandemic as a step in supporting government programs and maintaining personal, family, and community health in anticipation of the spread of Covid-19. It implements this activity in the form of a lecture/socialization, demonstration, free pocket book distribution, and educational video playback in animation. This activity runs smoothly and occurs interactive dialogue with respondents in the UPTD Puskemas environment. The 4G movement is one of the programs that contribute positively and can be applied by the community in increasing compliance in implementing a health protocol.

Abstract

WHO has declared COVID-19 a pandemic. Increasing body immunity is crucial for defense against infectious diseases. The immune system is influenced by several factors, including diet, environmental factors, lifestyle, age, stress, and hormones. Utilizing medicinal plants can make traditional drinks that are useful for increasing body immunity. The Ministry of Health Letter Statement No. HK.02.02/IV.2243/2020 concerning the Utilization of Traditional Medicines for Health Maintenance, Disease Prevention, and Health Care are attached with ingredients to increase body resistance. The community of RT 04 Bereng Village has a large area to grow family medicinal plants (TOGA) such as ginger, lemongrass, turmeric, Curcuma, and lime. These spices contain curcuminoid compounds which function as immunostimulators in boosting the immune system. In addition, herbs such as ginger, ginger, turmeric, and lemongrass contain high enough antioxidants so that they can ward off free radicals in our bodies. This service aims to provide information about processing TOGA into traditional drinks that can increase body immunity during the COVID-19 pandemic. This activity has stages of delivering education through pocketbooks, direct practice processing drinks from TOGA, and giving TOGA seeds. The results of this community service are in the form of herbal drinks, which are expected to increase the community's immunity; then can improve knowledge and skills in processing TOGA, especially in the neighborhood of RT 04 Kelurahan Bereng.

Abstract

WHO merekomendasikan pengambilan spesimen untuk diagnosis COVID-19 pada dua lokasi, yaitu dari saluran napas atas (swab nasofaring atau orofaring) atau saluran napas bawah [sputum, bronchoalveolar lavage, atau aspirat endotrakeal]. Tenaga medis harus memperhatikan keselamatan kerja dengan menggunakan APD (alat pelindung diri) yang tepat karena adanya kontak langsung dengan pasien terduga COVID-19 ketika mengambil sampel yang berisiko tinggi terinfeksi. Kasus COVID-19 di Indonesia meningkat setiap hari dan harga APD melambung tinggi dan terjadi kelangkaan. Mencermati kejadian tersebut maka sebagai wujud implementasi Tri Dharma Perguruan Tinggi, dosen UPR mengambil peran aktif dalam pengabdian masyarakat melalui program dosen pendukung SDM unggul untuk memberikan bilik swab dalam melindungi tenaga medis dari penyebaran infeksi secara langsung. Target dalam pelaksanaan pengabdian masyarakat ini adalah RS TNI-AD Palangka Raya. Metode yang digunakan adalah metode serah-terima alat dan demonstrasi pemakaian sebagai solusi kepada mitra yaitu RS TNI-AD Palangka Raya. Kesimpulan dari program pengabdian ini dengan adanya pemanfaatan bilik swab di RS dapat meminimalisir paparan penyebaran infeksi secara langsung ke tenaga medis serta sebagai alternatif saat krisis kelangkaan APD level 3

Abstract

Background: The most commonly found disease-causing microorganisms are the Salmonella typhi and Staphylococcus epidermidis bacteria. S.typhi is a gram-negative bacterium that causes typhoid fever, and S.epidermidis is a gram-positive bacterium that attacks mucous membranes and human skin. Sangkareho (Callicarpa longifolia Lam.) is one of the indigenous herbal plants of Central Kalimantan, which is empirically used as a wound medicine, diarrhea, diabetic, anti-inflammatory, and can be an antibacterial agent.Objective: This study used six groups are concentrations of 10%, 25%, 50%, and 100%, Chloramphenicol as a positive control, and dimethyl sulfoxide (DMSO) as a negative control. Methods: The method used for this test is the well diffusion method. In Nutrient Agar (NA) media, holes were made to insert 40 μl of each extract concentration, positive and negative control with four repetitions, then incubated for 18-24 hours at 37ºC. Data analysis using the One Way Anova test.Results: The research findings showed that ethanol extract of sangkareho leaves with all concentrations had an inhibitory effect against both bacteria tested.Conclusion: The ethanol extract of sangkareho leaves at the concentration of 10% was found to be the optimum and at the concentration of 100% was the maximum effectiveness for inhibiting S. typhi and S. epidermidis.

Abstract

The higher the production produced in agriculture, the higher the waste generated. For example, such as rice, the waste produced is very large, namely rice husks. Rice husk that can be utilized and developed for fuel needs. The method used was preparation, socialization and training which was carried out in Pangkoh Hulu Village, Pandih Batu District, Pulang Pisau Regency, Central Kalimantan. Charcoal briquettes are made from rice husks that are burned, mashed, and filtered until clean then mixed with tapioca flour and water, after which the briquettes are molded and dried to harden for 3-5 days. Digital marketing training in the form of packaging and branding is carried out to obtain added value so that it can be marketed by local residents. The participants who attended were the surrounding community, PKK women, and the X 2022 National Community Service Program students. There were approximately 30 participants who attended the activity and the Pangkoh Hulu Village community received and welcomed the training that we carried out. Making rice husk charcoal briquettes is one way to process rice husk waste so that it can become a selling point that can be marketed.

Abstract

AbstrakHand sanitizer merupakan salah satu bahan antiseptik yang yang praktis agar tubuh terhindar dari penyakit yang disebabkan oleh mikroorganisme. Hand sanitizer berbasis alkohol digunakan berlebihan dan terus menerus dapat mengakibatkan iritasi pada kulit. Kandungan yang terdapat dalam bahan alami sebagai alternatif pengganti alkohol antara lain flavonoid dan terpenoid yang terdapat di daun sembung (Blumea balsamifera). Penelitian ini bertujuan untuk mengetahui efektivitas gel hand sanitizer dari ekstrak daun sembung pada berbagai konsentrasi. Serbuk daun sembung diekstraksi secara maserasi menggunakan etanol 70%, kemudian dibuat gel dengan konsentrasi 10%, 15%, dan 20% ditambah carbopol 940, TEA, serta metil paraben. Sediaan gel hand sanitizer dievaluasi melalui uji organoleptis, pH, dan homogenitas selama 4 minggu. Setelah itu, dilakukan pengujian daya antiseptik dan aktivitas antimikroba terhadap Staphylococcus aureus, Escherichia coli, dan Candida albicans. Sediaan gel hand sanitizer ekstrak daun sembung memiliki konsistensi kental, warna hijau kehitaman, aroma khas daun sembung, homogenitas baik, dan nilai pH 6. Sediaan ini juga memiliki daya antiseptik yang baik mampu mengurangi jumlah koloni hingga 95% sama dengan sediaan gel berbasis etanol. Sediaan gel konsentrasi 20% memiliki zona hambat yang sangat kuat terhadap S. aureus dan C. albicans, serta kuat melawan E. coli. Gel hand sanitizer ekstrak Blumea balsamifera menunjukkan aktivitas antimikroba dan dapat digunakan menjadi kandidat agen antimikroba organik alami.Kata kunci: Antimikroba; Antiseptik; Ekstrak Blumea balsamifera; Hand sanitizer  AbstractHand sanitizer is one of antiseptic so that our body avoids diseases caused by microorganism. Hand sanitizer based on ethanol using continuously can cause skin irritation. The secunder metabolit contained in herbal as an alternative to hand sanitizer based ethanol contained flavonoid and terpenoid, are found in sembung leaves (Blumea balsamifera). The aim of this study is to determine the effectiveness of hand sanitizer gel from sembung leaf extract at various concentrations. Sembung leaf was extracted by maceration using 70% ethanol, and then gel was made with concentrations of 10%, 15%, and 20% plus carbopol 940, TEA, methyl paraben. The hand sanitizer gel preparation was evaluated through organoleptic, pH, and homogeneity tests for 4 weeks. After that, antiseptic activity and antimicrobial activity were tested against Staphylococcus aureus, Escherichia coli, and Candida albicans. The hand sanitizer gel preparation of sembung leaf extract has a thick consistency, blackish green color, a distinctive aroma of sembung leaves, good homogeneity, and a pH value of 6. The gel with a concentration of 20% had a very strong zone of inhibition against S. aureus and C. albicans, also strong against E. coli. Blumea balsamifera extract hand sanitizer gel exhibits antimicrobial activity and can be used as a candidate natural organic antimicrobial agent.

Abstract

Background: Tuberculosis (TB) is a chronic infectious disease caused by the bacterium Mycobacterium tuberculosis. The sputum Acid-Fast Bacillus (AFB) examination is a widely used diagnostic test, but it is difficult to control a minimum of 5,000 germs / ml of sputum to get a positive result. The GeneXpert MTB / RIF examination is an automatic machine with easy and fast use that uses the principle of real-time nested PCR and molecular technology for the examination of Mycobacterium tuberculosis (MTB) and rifampicin resistance (RR). Purpose: To determine the relationship between the results of sputum AFB examination and the results of rifampicin resistance GeneXpert MTB / RIF in pulmonary TB patients at Doris Sylvanus Hospital, Palangka Raya for the period 2018-2019. Methods: This study used an analytic observational method with a cohort study design. The subjects of this study were all pulmonary TB patients who had been examined for sputum AFB and GeneXpert MTB / RIF at Doris Sylvanus Regional Hospital for the period 2018-2019. Data analysis used SPSS program with Chi Square test (p <0.05). Results: The results of statistical tests showed that the Sputum BTA result variable had a significant relationship with the GeneXpert MTB / RIF outcome variable. So  the result is p = 0.009. Conclusion: There is a significant relationship between the results of sputum AFB examination and the results of rifampicin resistance of GeneXpert MTB / RIF in pulmonary TB patients at Doris Sylvanus Hospital, Palangka Raya for the period 2018-2019.
