Abstract

ABSTRACTCapability of general primer CK4/CK2 infectious bronchitis virus (IBV) to amplify of genomic IBV field isolates in Indonesia. Avian infectious bronchitis virus (IBV), the prototype of coronaviridae, is highly contangius, economically important pathogen of the chickens. Comparative sequence analysis of IBV S1 gene revealed regions that were conserved among serotypes. Two of conservesd region were used to develop degenarate general primer CK4/CK2 for amplifying IBV genomic RNA by RT-PCR. The aims of this study to demonstrated ability of general primer CK4/CK2 for amplified S1 gene IBV field isolates in Indonesia. The result of this is study suggest that only three IBV field isolates that can be amplified by primer CK4/CK2 that are I-14, I-37 and I-269.Key words: Capability, general primer CK4/CK2, infectious bronchitis virus, field isolates

Abstract

ABSTRACTPotential virus A/Ck/West Java/PWT-Wij/06 for Vaccin. Vaccination program for controllingavian influenza (AI) virus infection in poultry have been emerged during the past 5 years inIndonesia. However, due to the mutation character of this virus the available vaccines were nolonger effective. Therefore a new local isolate of avian influenza virus A/Ck/West Java/Pwt-Wij/2006 was studied. The virus, formulated as inactive vaccine, was injected in to 3 weeks oldof layer chickens intramuscularly. At 3 weeks after vaccination, vaccinated chickens werechallenged against seven local isolates of HPAI H5N1 intranasaly. Unvaccinated chickenswere included in the challenge test as control. Results showed that the vaccine produced100% protection against A/Ck/West Java/Pwt-Wij/2006 (homologous), A/Ck/West Java/1067/2003, A/Ck/West Java/Hj-18/2007, and A/Ck/Payakumbuh/BPPVRII/2007; produced 90%protection against A/Ck/BB149/5/2007, and 80% protection against A/Ck/West Java/Hamd/2006 isolates. The vaccine also stoppped viral shedding by day 5 to 7 after challange. Thisstudy indicate that the new local isolate of avian influenza A/Ck/West Java/Pwt-Wij/2006 hasgood potency as vaccine with broad spectrum against a range of AI viruses available inIndonesia.Key words: avian influenza, HPAI, H5N1,vaccine, poultry.

Abstract

Penelitian untuk mendapatkan bahan baru dengan kinerja tinggi telahbanyak dilakukan, khususnya di negara-negara industri. Salah satu programyang cukup pesat perkembangannya dan hingga sekarang sedang giatdilakukan penelitian, baik penelitian dasar maupun terapan adalah bidangnanosains dan nanoteknologi. Perkembangan nanosains dan nanoteknologidewasa ini telah mulai mendominasi dunia industri, dan bahkah telah mengeserteknologi mikro-elektronik (10-6 m) yang telah berperan dalam beberapadasawarsa terakhir (Edelstein, 1996). Nanomaterial, sebagai bagian darinanosains dan nanoteknologi, merupakan rekayasa material dalam ordenanometer (10-9 m) (Kebamoto, 2003). Hampir semua material dapat dibuatdalam bentuk nanokristal dan nanopartikel, untuk menghasilkan sifat lebihunggul, yang bergantung pada pemrosesan, manipulasi dan komposisinya(Gleiter, 1989, Kimura, 1995). Produk-produk industri diperkirakan akanmenggunakan nanomaterial secara komersial (Inoue, 2003) mulai tahun 2010(NSTC, 1993). Eksploitasi sejumlah mineral, seperti logam dan batuan, telahdilakukan di daerah penambangan intan rakyat di daerah Cempaka, KalimantanSelatan, dan difokuskan pada fragmen-fragmen yang dihasilkan daripenambangan intan. Ukuran fragmen-fragmen yang diperoleh ini berkisarantara 1,5 – 15 cm. Tumpukan fragmen-fragmen ini tidak berada jauh darisungai yang melintas di daerah penambangan, yaitu paling jauh 100 m danmasih merupakan daerah pelamparan sedimentasi sungai(sikumbang, 1994).Dari penelitian pendahuluan didapatkan kadar silika (SiO2) didaerah cempakaantara 94,4% - 99% (Wianto, 2008). Ketersediaan sumber alam berupabeberapa material di Banjarbaru, Kalimantan Selatan, seperti pasir silika, dapatdiolah lebih lanjut sebagai nanomaterial dengan karakteristik yang khas.Nanomaterial ini dapat dibuat dalam dalam ukuran nano (< 100 nm) melaluisintesis metode gaya berat kemudian melalui proses penggilinganmenggunakan planetary ball milling. Penggunaan peralatan X-ray diffractometer(XRD) dan scanning electron microscope (SEM) serta transmission electronmicroscope (TEM) akan menghasilkan karakteristik rinci nanosilika tersebut,menyangkut struktur, bentuk dan ukuran partikelnya serta agregasi yangmungkin terjadi. Hasil penelitian ini didapatkan material nano silika denganukuran antara < 100 nm setelah proses kopresipitasi sedangkan sebelumproses tersebut yaitu setelah proses ball mill didapatkan ukuran butir > 300 nmini berdasarkan hasil metode scherer XRD dan SEM. Sedangkan hasil analisiskualitatif dan kuantitatif didapatkan mineral SiO2 yang dominan yaitu 98 - 99%,ini setelah melalui proses pencucian untuk menghilangkan clay sertamenggunakan magnetik separator untuk menghilangkan material magnetik.

Abstract

ABSTRACTNipah Virus Seroepidemiology in Flying Fox and Pig Husbandry in Several Areasof Indonesia. Nipah is a dangerous zoonotic disease which was carried by flying fox.The disease had been occurred in Malaysia in 1999 and infect pigs and caused humandeath. Indonesia is adjacent country to Malaysia, hence, a serological study had beenconducted on 156 flying fox (P. vampyrus) sera from North Sumatera, West Java, CentralJava and East Java. Besides that, 2740 pig sera was randomly collected in differentprovinces to detect Nipah infection. Both flying fox and pig sera were tested usingELISA test to detect the presence of Nipah antibody. The results indicated that 37 from156 flying fox sera (23.7%) has antibodies against Nipah virus. Infections were occuredin all sampling sites with the prevalence varied from 18% to 33 %. Meanwhile, no pigsera tested (2740) had antibody against Nipah virus. Based on these results it can beconcluded that Nipah virus infections were occurred in flying fox in some parts inIndonesia, but not in pigs. It was suggested that the presence of Nipah virus in Indonesiashould be anticipated. Hence the distribution of its infection in pigs and human must beanticipated. Monitoring of Nipah infection in areas adjacent to Malaysia must be increasedto detect the entering of the disease in Indonesia.Keywords: Nipah, pigs, flying fox, serology

Abstract

ABSTRACTGenetic Mapping of Indonesian Avian Influenza Viruses 2007.Department of agricultureimplements vaccination as one a tool to control of avian influenza disease. The vaccinationprogram use virus seed such as H5N1, H5N2 and H5N9. Such as vaccination program for otherdiseases, avian influenza vaccine program have lack implementation in the field. In 2007,department of agriculture evaluated the AI vaccination program such as the master seedvaccine that can be used. Result of the evaluation showed that 11 of AI vaccines that weredistributed in Indonesia did not give protection more than 60% to Indonesian isolates in 2006(A/Ck/Pwt-Wij/06). From this point and many AI cases in the field in 2007, the aim of thisstudy was to conduct genetic diversity of avian influenza viruses which have circulated inIndonesia region. We used virus isolation for propagate the viruses, RT-PCR for identificationand DNA sequencing on HA1 region to analysis genetic diversity for genetic mapping anduseful for master seed candidate. The result of the study showed that there were 6 group ofgenetic diversity in 2007; Isolates from group 1, 5 and 6 can be used for AI vaccine candidate.Key words: genetic mapping, genetic diversity and avian influenza viruses

Abstract

Lapisan tipis karbon amorf terhidrogenasi (a-C:H) telah berhasil ditumbuhkan di atas substrat kaca ITO dengan teknik Radio Frequency – Plasma Enhanced Chemical Vapor Deposition (RF–PECVD) pada frekuensi standar 13,56 MHz. Efek variasi parameter laju aliran gas metana (CH4) murni dari 40 sampai 100 sccm terhadap sifat optik dan sifat listrik lapisan a-C:H diselidiki dalam penelitian ini. Parameter temperatur substrat, tekanan deposisi, daya RF, dan waktu deposisi dibuat konstan pada nilai 100 0C, 450 mTorr, 15 W dan 120 menit. Jenis fase, ketebalan, celah pita energi dan konduktivitas listrik lapisan a-C:H masing-masing dikarakterisasi dengan Difraksi Sinar – X (XRD), Scanning Electron Microscopy (SEM), spektroskopi UV-Vis dan probe 4-titik (Four-Point Probe). Hasil karakterisasi dengan XRD menunjukkan bahwa struktur lapisan a-C:H yang diperoleh berfase amorf. Peningkatan laju aliran metana dari 40 sampai 100 sccm dengan selang 20 sccm telah meningkatkan ketebalan lapisan tipis a-C:H pada nilai 641,5 - 1256 nm dengan laju deposisi 5,43 - 10,47 nm/menit serta peningkatan konduktivitas listrik lapisan pada rentang nilai 6,90x10-2 - 7,43x10-2 S/cm yang berakibat pada penurunan celah pita energi lapisan dari 2,55 eV menjadi 2,00 eV dengan meningkatnya laju aliran metana.

Abstract

Dalam studi ini, pengaruh fungsi kerja kontak depan terhadap kinerja sel surya struktur ITO/(p)a-Si:H/(i)a-Si:H/(n)a-Si:H/ZnO/Ag yang difabrikasi dengan metode RF-PECVD disimulasi menggunakan perangkat AFORS-HET. Berdasarkan pada hasil simulasi menunjukkan bahwa nilai WFITO divariasikan antara 4.95 to 5.55 eV dengan tujuan untuk menentukan nilai optimum WFITO dalam mendapatkan sel surya dengan efisiensi tinggi. Hasil simulasi menunjukkan bahwa nilai optimum fungsi kerja kontak depan pada antar-muka ITO/(p)a-Si:H adalah sekitar 5.45 eV yang menghasilkan efisiensi sebesar 7.22% (VOC = 1063 mV, JSC = 8.60 mA/cm2, FF = 78.93%) untuk sel surya Strukture-2. Hasil studi simulasi ini menghasilkan parameter-parameter optimum yang dapat dilanjutkan dengan eksperimen untuk mendapatkan sel surya yang lebih baik. Kata kunci: WFITO; sel surya; AFORS-HET; efisiensi.

Abstract

In this work, the imbalances in band gap energy between p-window layer and intrinsic layer (p/i interface) in p-i-n type solar cells to suppress charge recombination adopting with the addition of buffer layer, at p/i interface, namely solar cell structures without buffer (Cell A) and with buffer (Cell B). Using well-practiced AFORS-HET software, performances of Cell A and Cell B structures are evaluated and compared to experimental data. A good agreement between AFORS-HET modelling and experimental data was obtained for Cell A (error = 1.02%) and Cell B (error = 0.07%), respectively. The effects of dopant concentrations of the p-type and n-type were examined with respect to cell B for better performance by analysing the energy band diagram, the electric field distribution, the trapped hole density, the light J-V characteristics, and the external quantum efficiency. The simulated results of an optimised Cell B showed that the highest efficiency of 8.81% (VOC = 1042 mV, JSC = 10.08 mA/cm2, FF = 83.85%) has been obtained for the optimum dopant values of NA = 1.0 x 1019 cm-3 and ND = 1.0 x 1019 cm-3, respectively. A comparison between experimental data and simulation results for Cell B showed that the conversion efficiency can be enhanced from 5.61% to 8.81%, using the optimized values
