Abstract

Abstrak. Pretrombin-2 merupakan prekursor trombin dan dapat berperan sebagai komponen lem fibrin (LF). LF merupakan biomaterial perekat yang dapat diaplikasikan sebagai pengganti teknik jahitan pasca operasi. Biomaterial ini terdiri dari trombin, fibrinogen dan faktor XIII sebagai komponen utamanya. Dalam LF, trombin rekombinan berperan sebagai enzim yang mengubah fibrinogen menjadi fibrin untuk membentuk benang-benang fibrin sehingga luka tertutup. Kajian ini bertujuan untuk memprediksi ekspresi gen pretrombin-2 (PT2) manusia sintetik pada Escherichia coli secara in silico. Urutan asam amino pretrombin-2 manusia yang diperoleh dari data GeneBank dirubah ke dalam bentuk wild type urutan nukleotida menggunakan software OPTIMIZER sesuai dengan kodon preferensi E. coli K12, Selanjutnya dianalisis dengan Graphical Codon Usage Analyzer dan dioptimasi secara menual berdasarkan preferensi kodon E. coli dari data Codon Usage Database. Untuk memprediksi tingkat ekspresinya pada inang E. coli, dipelajari kajian ekspresi PT2 secara in silico menggunakan perangkat lunak dalam jaringan OptimumGeneTM. Hasil analisis secara in silico urutan nukleotida PT2 hasil optimasi dan wild type terhadap kodon preferensi E. coli menunjukkan bahwa, persentase rata-rata kandungan GC hasil optimasi sebesar 54,71% sedangkan wild type 52,56%, PT2 hasil optimasi memiliki 100% kodon dengan frekuensi tinggi, sedangkan wild type hanya 67%, dan PT2 hasil optimasi memiliki nilai Codon Adaptasi Index  satu, sedangkan wild type hanya 0,89. Dengan demikian dapat diprediksi bahwa gen pretrombin-2 manusia sintetik kemungkinan akan diekspresikan tinggi di inang E. coli. Abstract

Abstract: Pretrombin-2 (PT2) is a thrombin precursor that plays a role in converting fibrinogen to fibrin for the process of wound recovery. This material can be applied instead of eye surgery suture technique. The intein-mediated refining system to purify the protein is attractive to be developed, since the protein is obtained by one purification step, capable of self-splicing, the protein can be diffused in to the N-terminal (PT2 cutting of the intein induced marker changes in pH and temperature) and on the C-terminal (cutting PT2 from the intein-induced marker of the thiol reagent). In this study, we purified the PT2 fusion of the expression of E. coli BL21 (DE3) Arctic Express in the intein-mediated chitin matrix column. PT2 was fused with a tag at its N-terminal position, containing the sequence of intein codes SspDnaB followed by chitin binder domain (CBD). Furthermore, the PT 2 fusion was expressed on the E. coli host, then purified in the chitin matrix column. The PT2 cutting process of the intein marker induced changes in the pH and temperature in the column. PT2 fusion was successfully purified in the intein-mediated chitin matrix. The PT2 fusion cut from the induced intein buffer marker at pH 6.5 and incubation at the temperature of 25 oC for 48 hours.Keywords: E. coli, expression, intein mediated purification, pH and temperature changes, PT2

Abstract

Program Vaksinasi Covid-19, diharapkan dapat mencegah penularan Covid -19 di masa mendatang serta menurunkan angka kesakitan dan kematian yang diakibatkan infeksi virus Sars cov-2. Berdasarkan hal tersebut, maka dilakukan kegiatan pengabdian kepada masyarakat, berupa penyuluhan kepada masyarakat di Wilayah Kota Cimahi. Penyuluhan diikuti oleh 40 peserta yang terdiri dari: 13 orang kader kesehatan, 9 orang anggota karang taruna dan 18 orang warga masyarakat Kelurahan Baros, Cigugur Tengah. Untuk mengukur efektivitas penyuluhan, dilakukan pengukuran tingkat pengetahuan dengan metode pre-test dan post-test kepada peserta. Nilai pre-test dan post-test kader dan karang taruna lebih tinggi dari pada warga masyarakat. Namun demikian semua katagori peserta menunjukkan peningkatan pengetahuan dengan nilai rata-rata pre-test adalah 7,28 dan post-test 8,13. Dengan demikian dapat disimpulkan bahwa terdapat peningkatan pengetahuan peserta setelah diberikan penyuluhan.

Abstract

Fruitghurt jambu biji merah merupakan produk fermentasi jambu biji merah menggunakan bakteri asam laktat (BAL),yaitu Lactobacillus acidophilus yang merupakan bakteri probiotik handal yang sangat bermanfaat bagi tubuh. Pada penelitian sebelumnya didapatkan bahwa penambahan tapioka 0,5% dan sukrosa 10% menunjukkan pertumbuhan Lactobacillus acidophilus yang paling baik. Tujuan penelitian ini adalah untuk mengetahui fruitghurt jambu biji merah yang paling disukai oleh panelis ditinjau dari warna, aroma, rasa, dan tekstur serta uji karakteristik fisikokimia biji jambu biji merah yang difermentasi oleh Lactobacillus acidophilus dengan penambahan sukrosa dan tapioka sebagai emulsifier. Metode penelitian dengan menggunakan uji organoleptik dengan kuesioner pada 30 panelis tidak terlatih yang merupakan pegawai Fakultas Kedokteran Universitas Jenderal Achmad Yani yang memenuhi kriteria inklusi dan eksklusi. Sedangkan uji karakteristik fisikokimia dilakukan dengan metode eksperimental di labolatorium yang meliputi uji fisik, yaitu pH,kadar air dan uji kimia, yaitu kadar protein, kadar lemak, dan kadar asam. Objek penelitian ini adalah fruitghurt sari jambu biji merah yang difermentasi oleh bakteri Lactobacillus acidophilus ATCC 4356 dengan penambahan tapioka 0,5%. Hasil uji organoleptik menunjukkan bahwa panelis lebih menyukai buah jambu biji merah hasil fermentasi Lactobacillus acidophilus dengan penambahan tapioka 0,5% ditinjau dari warna, aroma, rasa dan tekstur. Hasil uji fisikokimia fruitghurt dengan penambahan tapioka 0,5% yang meliputi pH, kadar air, kadar protein, kadar lemak, dan kadar asam semuanya memenuhi persyaratan SNI yoghurt. Abstract

Dyslipidemia is a lipid metabolism disorder characterized by an increase in total cholesterol, triglycerides, and Low-Density Lipoprotein (LDL), as well as a decrease in High-Density Lipoprotein (HDL). According to the 2018 Basic Health Research (Riset Kesehatan Dasar/RISKESDAS), 12.6% of the Indonesian population aged 55–64 years had high cholesterol levels along with very high LDL and triglyceride levels. The Mediterranean diet is a form of non-pharmacological therapy involving nutritional intervention for patients with dyslipidemia. This study aimed to examine the effect of adherence to the Mediterranean diet on lipid profiles and the Plasma Atherogenic Index (PAI) in dyslipidemic patients at a private clinic in Bandung. This research employed an observational analytic method with a prospective cohort design involving 30 patients with dyslipidemia. Subjects were selected using a systematic random sampling technique and categorized into three groups based on their adherence to the Mediterranean diet: low, medium, and high. At the end of the month, lipid profile parameters and PAI were measured. Data on total cholesterol, LDL, and HDL levels were analyzed using ANOVA and Tukey’s Post Hoc test, while triglycerides and PAI were analyzed using the Kruskal-Wallis and Mann-Whitney tests. The results showed significant differences in total cholesterol (p = 0.007), LDL (p = 0.019), HDL (p = 0.006), triglycerides (p = 0.005), and PAI (p = 0.036) across the adherence groups. These findings suggest that higher adherence to the Mediterranean diet is associated with improvements in lipid profile and PAI. The beneficial effects are likely due to the diet’s rich content of monounsaturated fatty acids (MUFA), polyunsaturated fatty acids (PUFA), phytosterols, and polyphenols, which positively influence lipid metabolism and reduce atherogenic risk. Abstract

Prethrombin-2 (PT2) is a thrombin precursor, which plays a role in the conversion of fibrinogen into fibrin during blood clotting process. Previous study reported that the expression of human prothrombin-2 (rhPT2) in Escherichia coli formed inclusion bodies. The aim of this study was to establish a strategy to express a soluble rhPT2 in E. coli. This study was animed to design and codon optimize human prethrombin-2 gene as well as to optimize the expression condition using four strains of E. coli. The codon adaptation index (CAI) of the unoptimized hpt2 gene was 0.336, with 56.8% GC content. After optimization, the CAI of optimized hpt2 became 1.000 with 53.1% GC content. The optimized gene was successfully cloned into pTWIN1 expression vector. Expression analysis indicated that only E. coli ArcticExpress strain could successfully express a soluble recombinant rhPT2 protein, with only part of rhPT2 being expressed in insoluble form. However, the rest of the E. coli strains used in the experiments failed to express the rhPT2 in soluble form. We are deducing that the success in achieving soluble expression was not only due to the availability of chaperonins Cpn60/Cpn10, which played a crucial role in the protein folding in E. coli ArcticExpress strain, but also due to the codon optimization of hpt2 gene.

Abstract

α-Amylase from Saccharomycopsis fibuligera R64 is a non-adsorbing raw-starch degrading enzyme, a unique characteristic. This character is difficult to explain in the absence of its three-dimensional structure. Here we discuss the expression of a-amylase from Saccharomycopsis fibuligera in Pichia pastoris and the effect of site directed mutagenesis on its activity. A model based on the structure of its homologs suggested mutation of codon of Tyr401 into that of a Trp residue. An activity study using whole cells P. pastoris showed similar substrate degradation rates by cells carrying either the native or mutant amylase encoding gene. However, the purified enzyme of the mutant strain showed faster starch hydrolysis.

Abstract

Prethrombin-2 is a thrombin precursor that has important role in blood coagulation. It is the smallest precursor which is activated into thrombin by FXa prior to coagulation process. However, as a commercial theurapetic protein in fibrin sealant component, prethrombin-2 must be activated by ecarin before used. Thus, the production process of this protein needs further purification. In order to eliminate ecarin activation step and to increase production efficiency, we designed, cloned and expressed the recombinant autoactivated human prethrombin-2 in Pichia pastoris SMD1168. The variant was designed with 4 mutations, E40A, D47A, G48P, and E52A, following the result of a previous study. The synthetic variant gene was first optimized to conform with P. pastoris codon preference. The optimized synthetic gene was cloned in pD912 plasmid using XhoI and SacII restriction enzymes. The transformed P. pastoris was selected on agar plate supplemented with 1,000 µg.mL-1 Zeocin as a selection marker. This study showed that autoactivated prethrombin-2 was succesfully expressed extracellularly by P. pastoris SMD1168. The activity of recombinant autoactivated prethrombin-2 using a chromogenic substrate S-2238 was 0.540 unit/mg. Taken together, these results demonstrated that autoactivated human prethrombin-2 was successfully produced extracellularly in P. pastoris.

Abstract

The use of alternative medicine, especially medicinal plants to treat various diseases including hyperlipidemia has increased over the last few decades in most countries around the world. Bitter melon (Momordica charantia) contains active ingredients such as flavonoids, tannins, saponins and polyphenols which have antihyperlipidemic effects. The study aims to determine the antihyperlipidemic effect of bitter melon aqueous extract and compare it with simvastatin in reducing total cholesterol and low- density lipoprotein (LDL), and increasing high-density lipoprotein (HDL) in male Wistar strain rats. The research is an experimental study with pre and post-test design, carried out in October-December 2021 at the Faculty of Medicine, Universitas Jenderal Achmad Yani, Cimahi Indonesia. Subjects consisted of 25 Wistar rats which were divided into five groups. The negative control group (standard diet), the positive control group (induced by a high-fat diet and propylthiouracil), and the three groups that were induced by a high-fat diet and propylthiouracil then given bitter melon extracts of 300 and 600 mg/ kg BW and simvastatin 10 mg/kg BW. The method of measuring cholesterol using the Cholesterol Oxidase-Perioxidase Aminoantipyrine (CHOD-PAP) enzymatic method principle with a Semi-automatic Chemistry Analyzer. Analysis using paired t-test. The results showed a significant difference (P<0.005) in HDL and total cholesterol levels in the 600 mg/kg BW dose group, and a significant difference in LDL level in the 300 mg/kg BW dose group. The Simvastatin group showed significant differences in HDL, LDL, and total cholesterol. This is presumably because the active substances in bitter melon can inhibit the HMG-CoA reductase enzyme so that it can reduce LDL and total cholesterol levels. It can be concluded that bitter melon aqueous extract at doses 300 and 600 mg/Kg BW can reduce total and LDL cholesterol levels, and increase HDL. Keywords: antihyperlipidemic, HDL, LDL, Momordica charantia, total cholesterol Efek Antihiperlipidemik Ekstrak Buah Pare (Momordica Charantia L.) pada tikus WistarAbstract Penggunaan pengobatan alternatif, terutama tanaman obat untuk mengobati berbagai penyakit termasuk hiperlipidemia telah meningkat selama beberapa dekade terakhir di sebagian besar negara di seluruh dunia. Pare (Momordica charantia L.) mengandung bahan aktif flavonoid, tanin, saponin dan polifenol yang memiliki efek antihiperlipidemia. Tujuan penelitian untuk mengetahui efek antihiperlipidemia ekstrak air buah pare dan membandingkannya dengan simvastatin dalam menurunkan kolesterol total dan low density lipoprotein (LDL), serta meningkatkan high density lipoprotein (HDL) pada tikus jantan galur Wistar. Penelitian ini merupakan penelitian eksperimental dengan desain pre and posttest, dilaksanakan bulan Oktober-Desember 2021 di Fakultas Kedokteran Universitas Jenderal Achmad Yani Cimahi Indonesia. Subjek penelitian adalah 25 ekor tikus Wistar yang dibagi menjadi lima kelompok. Kelompok kontrol negatif (diet standar), kelompok kontrol positif (diinduksi diet tinggi lemak dan propiltiourasil), dan tiga kelompok yang diinduksi diet tinggi lemak dan propiltiourasil kemudian diberikan ekstrak pare 300 dan 600 mg/kgbb dan simvastatin 10 mg/kgbb. Analisis menggunakan uji t berpasangan. Hasil penelitian menunjukkan perbedaan bermakna (P<0,005) kadar HDL dan kadar kolesterol total pada kelompok dosis 600 mg/kgbb, dan perbedaan kadar LDL bermakna pada kelompok dosis 300 mg/kgbb. Kelompok simvastatin menunjukkan perbedaan bermakna untuk HDL, LDL, dan kolesterol total. Hal ini diduga karena buah pare mengandung zat aktif yang dapat menghambat enzim HMG-CoA reduktase sehingga dapat menurunkan kadar LDL dan kolesterol total. Dapat disimpulkan bahwa ekstrak air buah pare dengan dosis 300 dan 600 mg/kgbb dapat menurunkan kadar kolesterol total dan LDL, serta meningkatkan HDL.Kata kunci: antihiperlipidemi, HDL, LDL, kolesterol total, Momordica charantia
