Abstract

Populasi Eleusine indica dari perkebunan kelapa sawit di Kabupaten Batu Bara secara menyeluruh belum pernah dilaporkan resisten-glifosat. Penelitian ini bertujuan untuk melaporkan sebaran populasi dan mengklasifikasi resistensi E. indica terhadap herbisida glifosat pada perkebunan kelapa sawit di Kabupaten Batu Bara. Penelitian ini dilaksanakan di Lahan Pusat Penelitian Gulma Fakultas Pertanian Universitas Sumatera Utara pada Oktober 2016 - Agustus 2017. Penelitian ini menggunakan glifosat dengan dosis rekomendasi 720 g b.a.ha-1 dan tiga ulangan. Populasi ESU0 (dari Lapangan Bola Politeknik Negeri Medan) sebagai pembanding. Analisis data menggunakan software IBM SPSS Statistics 20. Hasil penelitian menunjukkan bahwa populasi E. indica yang paling resisten (63,33%) pada perkebunan kelapa sawit di Kabupaten Batu Bara terdapat pada ESU4.11 (afdeling 4 Kebun Tanah Gambus). Populasi E. indica yang tergolong resisten-glifosat 83,33% (10 populasi), tergolong moderat resisten-glifosat 16,67% (2 populasi) dan tidak ada populasi yang sensitif-glifosat pada dosis rekomendasi 720 g b.a.ha-1 pada perkebunan kelapa sawit di Kabupaten Batu Bara.

Abstract

To fulfill the demand of soybeans, the expansion of the planting area can be one of the choices through the conversion of marginal land by utilizing suboptimal land, such as saline soil. One of the plant breeding techniques to obtain tolerant plants to salinity is the use of chemical mutations, such as colchicine. The study aimed to reveal the effect of salinity on the characters of soybean plants (mutated with colchicine) and the effect of colchicine on the number of chromosomes and ploidy level. This study was conducted from January to March 2020 and carried out in the plastic house of Faculty of Agriculture, Universitas Sumatera Utara. The experiment was arranged in a Randomized Block Design with salinity levels (0 dS.m-1, 2 dS.m-1, 4 dS.m-1, 6 dS.m-1) as treatments and consisted of ten replications in each treatment. The result showed that the soybean plants were not successfully induced by colchicine at a concentration of 0.04 % with soaking duration of 10 hours, as indicated by the number of chromosomes and analysis of ploidy level. The soybeans that had been induced were planted in saline soil. There were significant effects observed on the productive branch, number of filled pods, and the weight of seeds per plant. Colchicine treatment did not successfully affect the number of chromosomes of soybeans but had an impact on the chromosome pattern. In addition, the salinity treatment of 4 dS.m-1 significantly affected the number of filled pods compared to the treatment of 6 dS.m-1.

Abstract

Eleusine indica populations on oil palm plantations in Deli Serdang Regency have not been reported to be resistant to glyphosate. This study was aimed to report distribution and resistance classification of E. indica population on oil palm plantations in Deli Serdang Regency. The research was conducted on Weed Research Center in Faculty of Agriculture, Universitas Sumatera Utara from October 2016 until August 2017. This research used Randomized Block Design with recommended dose at 720 g a.i.ha-1 of glyphosate and three replications. Population ESU0 (from Politeknik Negeri Medan Ball Field) as a comparison. Data on average survival, mortality, dry weight, resistance classification of E. indica were analyzeds using IBM SPSS Statistics 20 software. The results showed that there were 12 E. indica populations classified as glyphosate-resistant (56.52%), 7 glyphosate-resistant developing (30.43%), and 4 populations classified as glyphosate-susceptible (13.04%) with the recommended dose at 720 g a.i.ha-1 from oil palm plantations in Deli Serdang Regency.

Abstract

Gamma ray irradiation, especially applied at low doses, is one of methods to produce plant genetic diversity by means of micromutation. The objectives of this research was to determine the radiosensitivity of Argomulyo variety of soybeans (Glycine max (L) Merr) by gamma ray irradiation and to estimate genetic variablity of the crop after  irradiation. To determine the radiosensitivy, the seeds of Argomulyo variety were treated with gamma ray irradiation at  200 Gy, 400 Gy, 600 Gy, 800 Gy, and 1000 Gy. Curve-fit analysis was employed to find the Lethal Doses 50 (LD50) value . To evaluate the genetic variablity after irradiation with gamma ray, seeds of Argomulyo was irradiated under LD50  at doses of 0 Gy, 50 Gy, 100 Gy, 150 Gy, and 200 Gy.  The results showed that LD50 value was 457.17 Gy. The study found that for Argomulyo variety the LD20 was 490.93 Gy and the LD50 was 457.17 Gy.  Genetic variability was observed in various agronomic characters of M2 generation after low dose of gamma ray irradiation treatment of 50, 100, 150 and 200 Gy. The highest genetic variability was found at 200 Gy for plant height, number of productive nodes, and number of pods.  The variability of agronomic characters of soybean after irradiation was due to genetic factors. The highest heritability estimates was observed in plant height and number of productive pods. The succeding generation of putative mutants will be selected for productivity and drought tolerance.

Abstract

SSR marker has some merits such as quickness, simplicity, rich polymorphism and stability, thus being widely applied in genetic diversity analysis, molecular map construction and gene mapping. the purpose of this study was to determine polymorpic test and heterozygosity in F4 soybean (Glycine max (L.) Merril) progeni saline resistant characters using SSR (Simple Sequence Repeats) markers. This research was conducted in Biomolecular Laboratory, Socfindo Seed Production Laboratory (SSPL), Kebun Bangun Bandar Village Martebing District Dolok Masihul Regency Serdang Bedagai on December-May 2017. The number of samples were used 44. The five SSR primers (QS080465, QS1101, QS1112, QS100011, and Sat_091) used were specific primers, with a band pattern that was clearly visible around one or two bands. The percentage of polymorphic primers (PLP) of these three populations is high, all populations with a PLP of 100% of the saline resistant character. The effective allele number (Ne) of  7,160 for the progeny population is lower than the number of observed alleles (Na) of 10,000 which means that many progeny individuals are homozygous. The expected heterozygosity (He) value of 0.854 in the progeny population was higher than the observed heterozygosity (Ho) value of 0.027. The overall average observed heterozygosity (Ho) was 0.009 lower than the overall expected heterozygosity (He) of 0.61. Abstract

The objective of this research was to obtain a genotype of selected putative mutant withgood agronomic traits and high yielding on the optimum condition and its adaptability todrought stress in M3 generation. The study result showed that there were differences in thegrowth and production of each population as a result of irradiation in optimum conditions anddrought stress conditions. The population as a result of irradiation at 150 Gy and 200 Gy had a highermean value for the plant height trait, the number of productive branches, the number of productivenodes and the number of productive pods compared to irradiation population of 50 Gy, 100 Gy andthe control population of Argomulyo at optimum conditions. The decrease in the growth anddevelopment happened to all the characters observed in the irradiation population in droughtconditions compared to when grown in optimum conditions. The decrease can be seen in thecharacters of plant height, the number of pods and seed weight per plant. The growth and developmentof the population irradiation of 150 Gy and 200 Gy were more influenced by drought conditionscompared to other irradiation populations. The selection in the optimum conditions produced 291high yielding genotypes and the selection in drought conditions generated 202 genotypes andadapted to drought environments.

Abstract

The existence of educational institutions is very important and strategic to educate a quality generation. The target of this service program is junior high school in Medan, namely MTs P.P. Raudhatul Hasanah at the Ar-Raudlatul Hasanah Islamic Boarding School in Medan. Waste management and utilization and through the provision of soil microorganisms into compost and its role in land use through hybrid maize cultivation in the MTs P.P. school environment. Raudhatul Hasanah .. In addition, the need for the application of hybrid maize cultivation in the MTs P.P. school environment. Raudhatul Hasanah. Availability of organic waste treatment facilities as a means to process waste into compost. In addition, the USU PPM proposing team will provide multipurpose bins for the MTs P.P. school environment. Raudhatul Hasanah. The proposing team will practice compost processing and safeguarding hybrid corn in the school area. It is expected that later it will provide good direct benefits in the form of good food crops in the form of indirect benefits as knowledge and practice of MTs P.P. students. Raudhatul Hasanah. Partner involvement through community service programs by the community service team of MTs students. P.P. Raudhatul Hasanah is very high. This is indicated by the willingness of MTs school students. P.P. Raudhatul Hasanah was very enthusiastic and the active participation of students who attended the program was very satisfying. In addition this activity also helps MTs school students. P.P. Raudhatul Hasanah in studying and knowing waste management and its use through counseling and demonstrations. The success of the program in schools in MTs P.P. Raudhatul Hasanah Medan is largely determined by the participation of partners at each stage of the activities carried out. Partners namely schools in MTs P.P. Raudhatul Hasanah Medan really helped the implementation of community service programs in the USU community.

Abstract

The waste problem in Indonesia is still a problem that has not been resolved until now. The increase in population will also be accompanied by an increase in the volume of waste from human activities. The problem of plastic waste has become common, especially in big cities, suburbs and rural areas. Efforts to recycle plastic waste have been continuously carried out, especially for rural communities, but village communities often underestimate the recycling of plastic waste. This is due to the lack of counseling and guidance on independent and creative businesses and efforts to recycle plastic waste into useful products. One of the efforts to recycle plastic waste is to make bricks which raw material comes from plastic waste. In the process of making plastic bricks, there is a burning process, but this will not cause environmental pollution. Sidodadi Village located in Batang Kuis, Deli Serdang, is an area that has the potential to grow fast. Therefore, the development of Sidodadi Village in the social and economic aspects can be done by developing creative industries. To follow up on this effort, the USU community service team offers a teaching program on making bricks from plastic which can be used as a product of creative industry businesses with various advantages, including to reduce air pollution and environmental pollution.
