Abstract

Berdasarkan hasil penelitian riset kesehatan dasar, konsumsi pangan masyarakat Indonesia saat ini sebagian besar belum sesuai nutrisi sehat dan seimbang khususnya konsumsi sayuran dan buah-buahan. Hal ini menimbulkan kurangnya implementasi konsumsi pangan sehat masyarakat desa. Penyuluhan dan edukasi menu nutrisi sehat berbasis produk lokal menjadi penting dilakukan untuk mendukung Gerakan Masyarakat Hidup Sehat (GERMAS) serta pemanfaatan pangan lokal yang bernutrisi sehat. Salah satu desa dengan potensi sumber daya lokal yang melimpah sebagai produk lokal pangan nutrisi sehat adalah Desa Karangwidoro, Kecamatan Dau, Kabupaten Malang. Kegiatan penyuluhan ini merupakan upaya pemenuhan program pengabdian kepada masyarakat DPP/SPP Departemen Biologi, Fakultas Matematika dan Ilmu Pengetahuan Alam Universitas Brawijaya Tahun 2022 untuk memberikan edukasi bagi masyarakat khususnya ibu-ibu PKK sebagai penggerak pertama dalam keluarga. Metode pelaksanaan dilakukan dengan 5 tahap yaitu kerangka pemecahan masalah; diskusi; sosialisasi penyuluhan; khalayak sasaran yakni perangkat desa, ketua pokja sandang dan kesehatan, ibu-ibu PKK; serta pelaksanaan kegiatan. Bentuk penyuluhan berupa edukasi pangan sehat, praktik menu sehat dan pengolahannya, serta pemaparan materi. Kegiatan penyuluhan dan edukasi menu sehat berjalan sukses dengan ibu-ibu kader PKK desa yang sangat antusias. Hasil kegiatan pre-test menunjukkan nilai 65–70 sebanyak 33%. Hasil post-test terdapat peningkatan menjadi 91–100 sebanyak 45%. Pendampingan dan pemantauan dilakukan oleh kader PKK kesehatan dan tim pengabdi di masyarakat secara door to door untuk mendukung keberhasilan program.

Abstract

Keterbatasan fasilitas dan keterampilan pembelajaran di laboratorium sains di lingkungan sekolah menengah (SMA) menjadi permasalahan umum yang dapat menurunkan minat dan pemahaman siswa di bidang sains. Permasalahan ini juga ditemukan di wilayah Bali Barat, di mana keterbatasan alat dan bahan serta keterampilan tenaga pendidik menjadi kendala dalam pembelajaran. Salah satu keterampilan pembelajaran di laboratorium sains yang penting serta membutuhkan alat dan bahan relatif sederhana adalah pengujian aktivitas antioksidan senyawa yang dapat diterapkan dalam pembelajaran di sekolah. Kegiatan pengabdian ini bertujuan untuk menguatkan pemahaman dan keterampilan pengujian aktivitas antioksidan pada guru dan siswa SMA di wilayah Bali Barat. Kegiatan pengabdian dihadiri oleh 40 guru dan 26 siswa dari 6 SMA negeri di wilayah Bali Barat yang dilaksanakan di aula Balai Taman Nasional Bali Barat (TNBB). Metode community-based interaction approach dilakukan melalui penyampaian materi dan diskusi interaktif serta praktikum pengujian aktivitas antioksidan menggunakan metode Ferric Reducing Antioxidant Power (FRAP) menggunakan alat dan bahan yang sederhana. Kegiatan pengabdian ini menguatkan pemahaman dan meningkatkan keterampilan guru dan siswa terkait sumber senyawa antioksidan, kapasitas antioksidan, prinsip dasar, dan praktik pengujian aktivitas antioksidan menggunakan metode FRAP.

Abstract

This research is a development research that uses the ADDIE Design Research and Development (R&D) model which consists of 5 stages, namely analysis, design, development, implementation, and evaluation. The purpose of this study is to describe the validity and practicality. Augmented reality-assisted flashcards on linear motion material to increase students' learning interest. Product validity was assessed by 3 experts consisting of aspects of material and construct assessment as well as aspects of media and design assessment with an overall average score of 3.64 with a very valid category. The practicality of the product is assessed from the readability aspect and the teacher's perception aspect by obtaining an overall average percentage of 95% in the very practical category. Based on the analysis of the results of the data, it was concluded that the product resulting from the development of augmented reality-assisted flashcards on linear motion material was valid and practical to increase students' interest in learning.Keywords: flashcards, augmented reality, learning interest, linear motion.DOI: http://dx.doi.org/10.23960/jpmipa/v24i1.pp291-297

Abstract

Lipase memiliki manfaat penting di bidang industri. Tujuan penelitian ini adalah untuk mendapatkan kapang lipolitik yang mampu tumbuh dan menghasilkan aktivitas lipase tinggi pada limbah ampas kelapa menggunakan metode solid state fermentation. Isolat kapang uji dipurifikasi kemudian dilakukan skrining dan seleksi kapang lipolitik dan dilanjutkan dengan produksi lipase menggunakan substrat ampas kelapa yang sebelumnya diukur kandungan biokimia. Hasil menunjukkan bahwa 8 isolat kapang lipolitik mampu tumbuh baik pada substrat ampas kelapa yang ditunjukkan dengan adanya sporulasi dan perubahan pH medium selama reaksi. Diantara kapang lipolitik tersebut, isolat kapang KLC-333 diketahui menghasilkan aktivitas hidrolisis lipase terbesar yaitu 13,33 U/ml dan volume produksi 46 ml. Biosintesis dan peningkatan produksi lipase dipengaruhi oleh kandungan nutrien di dalam substrat ampas kelapa.

Abstract

Biogrouting is a process that transforms soil or sand into calcarenite or sandstone by bacterial calcium carbonate precipitation . The mechanisms of this process are that the bacteria hydrolyze urea catalyzed by urease, and with the existence of dissolved Ca2+, a solid crystalline calcite or calcium carbonate (CaCO3) will be produced. The main advantage of biogrout is that soil or sand can be strengthened without interfering with the hydraulics of the treated soil or sand and without excavation or replacement. In this study, the isolation and identification of bacterial calcium carbonate precipitation , and characterization of urease produced by bacteria were conducted. In the isolation method, the enrichment method using urea as abacterial carbon source was carried out. The formation of crystalline calcite was observed by a light microscope. The urease enzyme activity was determined by the 3, 5-Dinitrosalicylic acid (DNS) method. The molecular identification of isolates was analyzed by the determination of 16S rRNA gene sequencing. As a result, 19 calcium carbonate precipitation have been isolated from soils, sands, water, and rocks collected from several areas in Indonesia. They showed calcite formation in a medium with urea. Molecular identification of isolates with high urease enzyme activity revealed that the isolates belong to the group Bacilli. The highest enzyme activity produced by ID10-U004 is 374.94 U/mL. The preliminary biogrouting experiment was also conducted using isolated strain, and the reaction was able to make the sand solid and stiff.

Abstract

Brown rice (BrR) is a nutrient-rich staple food and a potential source of γ-oryzanol, which has the potential to promote health effects in hypercholesterolemia. However, the availability and biological mechanism of γ-oryzanol in Indonesian BrR remain unexplored. This study aimed to characterize γ-oryzanol content in three Indonesian BrR varieties, Black Madras, Lawang, and UB BrR, and evaluate its biological functions for hypercholesterolemia. γ-Oryzanol was identified using HPLC, antioxidant activity by the ferric reducing antioxidant power (FRAP) method, and anticholesterol activity by the Lieberman-Burchard method. In silico analysis was performed to assess γ-oryzanol derivatives' interaction with microsomal triglyceride transfer protein (MTP). Black Madras BrR showed the highest γ-oryzanol content (0.3007 ± 0.0011 g). γ-Oryzanol was comprised of four main derivatives: cycloartenyl ferulate, 24-methylenecycloartanyl ferulate, campesteryl ferulate, and β-sitosteryl ferulate. Black Madras BrR exhibited strong antioxidant (IC50 18.89 ± 0.37 μg/mL) and anticholesterol (IC50 14.02 ± 0.23 µg/mL) activities. Docking simulations revealed that γ-oryzanol derivatives interact with MTP lipid-binding residues (Leu643, Ile666, Phe813, Val817) with the same binding energy as lomitapide. Molecular dynamics indicated γ-oryzanol stable interaction, closer to lomitapide. These findings proposed the potential bioactivity of γ-oryzanol from Black Madras BrR as antioxidant, anticholesterol, and a natural MTP inhibitor for hypercholesterolemia treatment.

Abstract

Rheumatoid arthritis (RA) is an autoimmune disease that can cause progressive damage to the joints of patients. The number of patients is expected to increase, along with the exact cause of this disease remains unknown. However, there are several risk factors associated with RA, including dysbiosis. The purpose of this study was to characterize the composition of intestinal microbiota in the RA and control groups through fecal analysis and reveal the association of microbiota composition with RA disease in Indonesia, especially Malang. Fecal samples were obtained from RA patients and controls. Fecal analysis was carried out through several stages, namely the calculation of total bacterial colonies, isolation and characterization of anaerobic bacteria, calculation of the Simpson diversity index, and DNA isolation. Analysis of bacterial composition profiles in fecal was carried out using 6 specific primer sets through PCR analysis. The results of the 16S rRNA PCR analysis showed different microbiota compositions between RA patients and controls. The number of Enterococcus bacterial group was lower in the control patients than the RA group, whereas the Lactobacillus bacteria decreased in RA patients. In addition, our study found that the existence of bacterial isolate 11 changed the composition of microbiota in RA patients, and the DNA band only appeared in Universal primers. The diversity of bacterial species can provide symbiotic and pathogenetic effects in RA patients.Keywords: Dysbiosis, intestinal microbiota, PCR, rheumatoid arthritis.
